Isolation of Symbiotic Frankia EuIK1 Strain from Root Nodule of Elaeagnus umbellata (보리수나무 뿌리혹으로부터 Frankia EuIK1 공생균주의 분리)

  • The root nodules of Elaeagnus umbellata were coralloid-shape due to repeated dichotomous branching of nodule meristem. The filamentous endophyte with vesicle cluster ranging from 30 ${\mu}{\textrm}{m}$ to 60 ${\mu}{\textrm}{m}$ in diameter was present only in the cortical cells. The isolated endophytes in vitro culture showed typical Frankia morphology, consisting of highly branched hyphae ranging from 0.8 ${\mu}{\textrm}{m}$ to 1.0 ${\mu}{\textrm}{m}$ in diameter, terminal and intrahyphal sporangia varing in shape and size up to 60 ${\mu}{\textrm}{m}$ in length and laminated vesicles. Its infectivity and effectivity were confirmed by induction of nitrogen-fixing root nodules on the inoculated seedlings of two Elaeagnus species. Consequently, the isolate was confirmed as a true symbiont of Elaeagnus umbellata root nodule and named Frankia EuIK1.

EFFECT OF "STEP-DOWN" AND "BALANCED FORCE" PREPARATION METHODS ON THE SHAPE OF THE ROOT CANAL (Step-down과 Balanced force 근관성형술식에 의한 근관 형태의 변화)

  • This study was performed to investigate the effect of root canal shaping techniques on the change of the shape of prepared root canal. 40 mesiobuccal canals of recently extracted mandibular 1st and 2nd molars were divided into 4 groups and shaped by step-down/balanced force technique, step-down/step-back technique, step-back technique and conventional technique respectively. The change of the shape of root canal was traced by superimposing the radiographs obtained before and after shaping of each root canal. The results were as follows. 1. By the experimented techniques except conventional technique, the root canals were more shaped in convex side of apical area and in concave side of most curved and coronal area than in the other sides(P<0.05). By conventional technique, the root canals were more shaped in convex side than in convave side from apex to orifice(P<0.05). 2. By step-down/balanced force technique, the cancave sides at C and D points of proximal view and C point of clinical view were more shaped than the convex side(P<0.05). Through the entire canal, the concave side was more shaped than the convex side in proximal view(P<0.01). But there was no statistical difference between both sides in clinical view. 3. By step-down/step-back technique, the change of root canal shape was not statistically different in concave and convex sides at each point of both views(P>0.05). And through the entire canal in proximal view, there was no statistical difference in shaping percentage between both sides. But through the entire canal in clinical view, the concave side was more shaped than the convex side(P<0.1). 4. By step-back technique, the convex side at B point of clinical more shaped than the other sides(P<0.05). Through the entire canal in proximal and clinical views, there was no statistical difference in shaping percentage between both sides. 5. Comparing the total shaping percentage among techniques, that in conventional technique was the greatest numerically, and followed by the percentages in step-down/step-back, step-down/balanced force and step-back technique. But, in proximal view, shaping percentages were not statistically different among techniques(P>0.05, ANOVA test). In clinical view, shaping percentages in step-back and conventional techniques were statistically different(P<0.01, ANOVA test). * Proximal view: radiograph taken in mesiodistal direction. * Clincal view: radiograph taken in faciolingual direction.   • Stratified American ginseng(Panax quinquefoilium L.) seeds were planted in a shaded greenhouse at four depths and in four different soil types to observe effects on emergence rate and root size. Seeding depth affected seedling emergence rates and the number of days required to complete emergence. The shape of the roots was affected by the texture of soil, especially percentage of sand.

A Shape Control of Welded Joints to Improve Fatigue Strength (피로강도 향상을 위한 용접이음부의 형상제어에 관한 연구)

  • When U-ribs of steel deck plates are connected at the field, overhead welding should be done with backing strips. Misalignments may occur and lead to eccentric moments as well as high stress concentrations at welded joints. In this study, stress analyses and fatigue tests were carried out. Stress analyses for U-ribs' welded joints with backing strips were performed with different misalignments, root shapes, root gaps, and backing strip sizes. From the stress analyses, the stress concentration factors increased with increasing misalignments and root gaps. With the fixed misalignments and root gaps, the stress concentration factors obtained in the case of the semi-circle root shape were lower than those in the case of the right-angle root shape. It was verified that backing strip sizes have little influence on stress concentration factors. The fatigue tests for U-ribs' welded joints with backing strips indicated that increased misalignments shorten fatigue life drastically and cracks usually initiate at the root of the base metal and are propagated to the weld bead surface. Based on the results of the stress analyses, root-shape control methods were developed to mitigate stress concentration by changing welding condition control, radius curvature, and flank angle.

Saponin Contents, Histological and Cytological Characteristics of Ginseng Root with Physiological Disorder (생리장해 인삼의 Saponin 함량과 조직 및 세포학적 특성)

  • This study investigated ginsenosides and tissue characteristics of roots injured by physiological disorder, rusty and rough skin. After separation to cortex and stele parts of healthy, rusty (red) and rough skin roots, respectively, the contents of saponin and ginsenosides were analyzed. And also, the histological and cytological characteristics of cortex and stele parts were investigated. Crude saponin contents were little different among healthy, rusty (red) and rough skin root and ginsenesides as - Rgl, - Re and - Rbl were largely detected both in stele and cortex part. The ratio of PT/PD showed about 1:1 in three kinds of root. In histological study, destoryed cells in epidermis of rusty(red) root, and those in epidermis and exodermis of rough skin root were observed. The cells in cortex of rusty (red) and rough skin root have generally nucleus with unfixed shape, unequal cell wall, large number of vacuole and mitochondris, and unidentified dark substances compared to healthy root. But in cell of stele tissue, most of organellE seems to be normal except a small number of cells in rough skin root.

Effects of Treatment with the Extract from the Root Bark of Morus alba on the Cell Composition and the Shape Change of Microorganisms (상백피 추출물이 미생물의 균체성분 및 형태 변화에 미치는 영향)

  • The ethanol extract from the root bark of Morus alba showed the strongest antimicrobial activity on the growth of almost all the tested microorganisms which were food-borne pathogens and food-related microorganisms. Therefore, fatty acid composition, amino acid composition and shape change of microorganisms treated with the ethanol extract from the root bark of Morus alba were examined. In effects of treatment with the ethanol extract on the fatty acid compositions of B. subtilis, S. aureus and E. coli, fatty aicd compositions such as hexadecanoic acid (16:0) and octadecanoic acid (18:2) of the tested strains were increased but pentadecanoic acid (15:0) heptadecanoic acid (17:0) and acid (16:1) and octadecenoic acid (18:1) of E. coli were decreased. The ethanol extract did not significantly affect the aminn acid composition of the tested strains. Transmission electron micrographs of microorgani는 treated with the ethanol extract exhibited morphological changes that irregularly contracted cell surface in S. aureus and destructed cell walls in B. subtilis and E. coli.

Comparison of Plant Growth and Morphological Characteristics Among the Korean Ginseng, the American Ginseng and the Bamboo Ginseng (고려인삼, 미국삼 및 죽절삼의 생육 및 형태적 특성 비교)

  • An investigation was conducted to ascertain the basic information on characteristics of growth and morphological characters among the Korean (Panax. ginseng), the American (Panax. quinquefolium) and the Bamboo (Panax. japonicus) ginseng. In aerial parts growth of the ginseng species by age, The Korean ginseng and American ginseng's stem and leaf growth was alike in 2-4 years old, but growth cycle changed in 6 years old. The Korean ginseng was more vigorous than the American ginseng. The Korean ginseng roots were highly observed in ratio of red skin roots among three species, whereas The American ginseng roots were highly infected by root rot. It seems to be variable depending on growing stage and species. The Korean ginseng flowered about the middle of May, the American ginseng early June, and the Bamboo ginseng was late of May, The berry color of the ginseng species was observed, The Korean and American ginseng's mature berry color was red, The Bamboo ginseng's berry was three type of color and shape. In root characteristics of the seedling, Korean (p. ginseng), American (p. quinquefolium) ginseng's root shape was similarity in type, the bamboo ginseng showed different type, which root length and root weight was smaller than those of ginseng. In morphological characters of Leaf surface, pollen, and stoma, the Korean ginseng and American ginseng had crystal rosette on epidermis cell, but the Bamboo ginseng didn't has crystal rosette. Pollen shape observed tricolpate pollen and size was media type among the ginseng species, and also guard cell was anomocytic type, which were observed by scanning electronic microscope.

A STUDY OF ROOT CANAL SHAPE FOLLOWING CANAL PREPARATION OF VARIOUS CANAL ENLARGING INSTRUMENTS (수종 근관확대 기구를 이용한 근관 형성후의 근관형태 변화에 관한 연구)

  • The purpose of this study was to evaluate the resulting root canal angulation and shape change after using various enlarging instruments. The mesial canals (120) of extracted human mandibular 1st and 2nd molars were randomly divided into 6 groups; Control, K-type me, Heliapical me, Canal Master me, sonic and ultrasonic instrumentation group. Vertical angulation of each canal was determined by a straight line through the long axis of canal and another straight line through the apical foramen to intersect at the point where the canal began to leave the long axis of the tooth. By recording and comparing the measured angles of the each set of pre-and postinstrumentation. Then, the roots were sectioned horizontally in the apical, middle, coronal thirds and the canal shapes examined, as was the mesiodistal canal diameter as it relates to the external root surface. The results were as follows: 1. Instrumentation using K-type me group resulted in the highest mean change in angulation ($9.900^{\circ}$) (p < 0.005), while Sonic Air MM 3000 group resulted in the least degree of straightening canals ($8.250^{\circ}$) (p < 0.005). 2. Canal Master file group resulted in the best canal shape at the three levels (P < 0.005). 3. Measured minimal mesial root width produced Heliapical me group at the apical 1/3 level, Sonic Air MM 3000 group at the middle 1/3 level, Heliapical me group at the coronal 1/3 level (P < 0.005). 4. Measured minimal distal root width produced Sonic Air MM 3000 group at the apical 1/3 level (P < 0.05), Heliapical me group at the middle 1/3 level (P < 0.005), Canal Master me group at the coronal 1/3 level (P < 0.005). 5. HeIiapical me group produced more increased canal diameter than any other groups (P < 0.005).

  • Plasma arc welding(PAW) technology is a proven process that has already been adopted by other industrial fields and recently has been considered to join the tank structure of LNG carrier. The purpose of this study is to introduce PAW process for the root welding of stainless steel pipes instead of TIG welding. There are distinctive features of the PAW compared to TIG welding; higher energy density that can increase welding speed by more than twofold, and longer arc length that can be controlled to trace seam line easily because of allowable gap between workpiece and torch. However, PAW process is also very sensitive to the root gap and misalignment due to the characteristics of long and narrow arc shape. So, we have done various experiments to establish the allowable fit-up condition by changing welding parameters including arc length, with or without filler metal, groove shape, and obtained satisfactory result.

Assessment of Root and Root Canal Morphology of Human Primary Molars using CBCT (CBCT를 이용한 유구치의 치근 및 근관의 형태학적 평가)

  • The purpose of this study is to evaluate the morphological characteristics of the roots and roots canals of primary molar teeth using Cone-Beam Computed Tomography (CBCT). A total of 68 CBCT images of children aged 4 - 5 years was used for this study. A total of 160 molar teeth were analyzed. Various parameters such as the number of roots and canals, length of root and root canal, and the angulation and shape of the roots were analyzed. All maxillary primary molars had 3 roots. The presence of 2 root canals in 1 root was only observed in the mesiobuccal root of maxillary primary second molars. Most mandibular primary molars had 2 roots, and most mesial roots had 2 root canals. Concerning the length of the roots, the palatal root of the maxillary primary molar was found to be longest whereas the distobuccal root was shortest. In mandibular primary molars, the mesial root was longer than the distal root. In maxillary primary molars, the palatal root had the greatest angulation whereas the distal root has the greatest in mandibular molars. The root and root canals of maxillary primary molars were more curved in shape whereas mandibular primary molars were straight.
