This study was conducted to determine the effect of soil physical characteristics on rhizome rot incidence of platycodon. Sampling sites were Keochang 4, Kimhae 7, Haman 6, Chinju 6 and Koseong 3 fields in Kyongnam province and Hongcheon 6 fields in Kangwon province. The root disease incidence rate was correlated with soil depth Y=-0.747X+88.19($R^2=0.394^{***}$), soil hardness Y=4.36X+8.93($R^2=0.201^*$), bulk density Y=104.7X-80.99($R^2=0.295^{**}$), clay content Y=1.24X+14.14($R^2=0.196^*$), porosity Y=-3.11X+215.9($R^2=0.220^*$) and silt content Y=-0.75X+67.85($R^2=0.178^*$). The yield was correlated with soil depth Y=0.263X+0.971($R^2=0.105^*$), clay content Y=-0.688X+32.74($R^2=0.158^*$), porosity Y=1.974X-93.19($R^2=0.231^{**}$) and silt content Y=53.05X-108.65($R^2=0.232^*$), The optimum cultivated land of perennial platycodon was soil depth over 1m, soil hardness under $5kg\;cm^{-2}$, bulk density $1.0Mg\;m^{-3}$, moisture content 13~17%. clay content 5~10%, porosity 58~63%, silt content 38~64% and soil texture of silt loam.
For the development of a multifunctional biocontrol agent, the siderophore-producing strain GL7 was isolated from a rhizosphere on chrome azurol S agar. The GL7 was identified as a strain of Pseudomonas fluorescents on the basis of their reactions to standard physicochemcial tests from Bergey's manual, API diagnostic test, and fatty acid analysis. P. fluorescents GL7 considerably inhibited spore germination and hyphal growth of phytopathogenic fungus Funsarium solani in a dual culture. In pot trials of bean with P. fluorescens GL7, the disease incidence was significantly reduced down to 5% from 70% of incidence in the untreated control. P. fluorescens GL7 also enhanced plant growth to nearly 1.5 times than that of the untreated control, promoting elongation and development of the roots. These results suggest that the plant growth-promoting P. fluorescens GL7 can play an important role in the biological control of soil-borne plant disease in a rhizosphere.
The activities of pectolytic and cellulolytic enzymes produced from slices of ginseng root infected with Cylindrocarpon destructains(Zins.) Scholtern were proportional to each concentration and reaction time. Activities of cellulase(Cx), endo-polygalacturonase(endo-PG), endo-polymethylg-alacturonase(endo-PMG), exo-polygalacturonase(exe-PG), and exe-polymethylgalacturonase(exo-PMG) were maximum on the 4th day after inoculation. No endo-PG and endo-PMG were detected at the first and second days, while exo-PG exo-PMG were active. On the 6th day, all pectic enzymes were completely lost, whereas Cx remained at a high concentration. pH optima of Cx, endo-PG, endo-PMG, exo-PG, and exo-PMG were 6.0, 5.5, 8.0, 7.0 to 7.5, and 8.5, respectively. Temperature optima of Cx, endo-PG, endo-PMG exo-PG, and exo-PMG were $66^{\circ}C\;53^{\circ}C\;41^{\circ}C\;37^{\circ}C\;and\;40^{\circ}C$, respectively. Cx was only inhibited by $0.05M\; Hg^{++}$ among 16 ions tested. Inhibitory effects of ions on pectolytic enzymes varied, however$M Fe^{+++}\;and\;0.05M\;Al^{+++}$ were the best in general. Among 8 fungicides, none of them inhibited all the enzymes studied at $0.1\%$, active ingredients. Exo-PG were highly inhibited by all of the fungicides, of which difolatan was the most inhibitory to all the pectic enzymes. $Ca^{++}\; at\; 0.02M\; and\;Fe^{+++}\;at\;0.02M$ completely inhibited all the pectolytic enzymes, and Cx was inhibited $30\%$ and $70\%$ at the same concentration, respectively Formalin almost inhibited exo-PG and exe-PMG at $0.8\%$ but not the other enzymes especially Cx. Difolatan at $0.8\%$ inhibited all the enzymes concerned above $80\%$. The cellulolytic and pectolytic enzymes of C. destructans must be closely associated with the ginseng root rot and should be inhibited to control the disease effectively.
This study was conducted to control the seed borne pathogens (Alternaria spp.) of carrot and to examine the seed germination rate by using 13 environment friendly agricultural materials instead of conventional chemicals for organic cultivation. The growth inhibiting effects on pathogens showed the different responses according to each agricultural material and effective 7 materials against seed borne pathogens were selected. Among 7 materials, the carrot seeds sterilized with plant extracts, Tanger Stop and Land Saver were not germinated at all. The germination rate of seeds sterilized with other materials showed the similar levels with reference chemical (Benlate-T) and non-treated seeds. Infection rates of seeds sterilized with seaweed extract, Bellopper for controlling A. radicina and plant extract, Ssial-100 for A. dauci were similar or low infection rate compared with reference chemical. The germination rate and root length of seeds sterilized with Bellopper and Ssial-100 was similar or superior to those of chemical sterilized seeds. The optimal condition seemed to be a little bit different depending on the concentration of materials. As a result, the sterilization of carrot seeds by using the environment friendly materials could be effectively utilized as a technology to inhibit the infection of seed-borne pathogens.

Background: Biocontrol agents are regarded as promising and environmental friendly approaches as agrochemicals for phytodiseases that cause serious environmental and health problems. Trichoderma species have been widely used in suppression of soil-borne pathogens. In this study, an endophytic fungus, Trichoderma gamsii YIM PH30019, from healthy Panax notoginseng root was investigated for its biocontrol potential. Methods: In vitro detached healthy roots, and pot and field experiments were used to investigate the pathogenicity and biocontrol efficacy of T. gamsii YIM PH30019 to the host plant. The antagonistic mechanisms against test phytopathogens were analyzed using dual culture, scanning electron microscopy, and volatile organic compounds (VOCs). Tolerance to chemical fertilizers was also tested in a series of concentrations. Results: The results indicated that T. gamsii YIM PH30019 was nonpathogenic to the host, presented appreciable biocontrol efficacy, and could tolerate chemical fertilizer concentrations of up to 20%. T. gamsii YIM PH30019 displayed antagonistic activities against the pathogenic fungi of P. notoginseng via production of VOCs. On the basis of gas chromatography-mass spectrometry, VOCs were identified as dimethyl disulfide, dibenzofuran, methanethiol, ketones, etc., which are effective ingredients for antagonistic activity. T. gamsii YIM PH30019 was able to improve the seedlings' emergence and protect P. notoginseng plants from soil-borne disease in the continuous cropping field tests. Conclusion: The results suggest that the endophytic fungus T. gamsii YIM PH30019 may have a good potential as a biological control agent against notoginseng phytodiseases and can provide a clue to further illuminate the interactions between Trichoderma and phytopathogens.
A severe ginseng root rot disease was occurred and became widespraed in 1973 affecting the ginseng cultivating plot around Kangwha and Gimpo-Gun of Kyunggi province. Soil samples were collected from the area of Wolgot-Myun, Gimpo-Gun, Kuunggi-Do, 1974. We examined general fungi and Fusarium sp. in soil layers and also in different kinds of soil of respective diseased, uncultivated and healthy areas, and found the following results. 1. In the diseased and uncultivated area, the content of moisture, organic matter and silt was greater than in the healthy area. 2. Contray to the above, the healthy area contained a greater amount of inorganic elements such as $P_2O_5$, K,Ca and of soil particle such as Cs and Fs. The degree of pH and content of Mg were even in the three types of soils. 3. General fungi were found in abundance in the diseased and uncultivated soils. It was observed that in all types of areas, general fungi reside in abundance in the rhizosphere, i.e., 10-15cm layers and that the closer the surface, the greater the numbers of fungi. 4. A great number of Fusarium sp. was found in the uncultivated area, and followed diseased and healthy areas. It was observed that in all types of areas, Fusarium sp. distributed in abundance in rhizosphere and that the closer the surface, the greater the numbers of Fusarium sp., with the numbers decreasing as the soil layers increase.

Bacillus subtilis strains isolated from different regions of Korea were screened for their plant growth promotion and induced systemic resistance (ISR) in tomato and red-pepper. The plant growth promotion on red-pepper and tomato revealed maximum plant height (22.73 cm) on red pepper treated with B. subtilis strain JE 21-1 and 30.18cm in case of tomato treated with B. subtilis strain JE 8-1. There was also significant improvement in root and shoot dry weight in both the plants. The strain JE 21-1 showed better promise for all growth parameters in red-pepper and tomato when compared to other strains and positive check BTH. Different strains screened in square plate method also revealed maximum plant height and leaf width, and suppressed anthracnose on red pepper in case of strain JE 21-1 at $10^6$ and $10^7$ cells/ml when compared to other strains. In all the bacterial inoculations the population was significantly high when compared to untreated check. In plant growth promotion with respect to fruit length and weight, fruit length was maximal in treating with JE 9-4 and ES 2-2, while fruit weight was maximal in treating with JE 3-6, ES4-2, ES2-2 and JE 21-2 on red pepper. In case of tomato, comparatively better fruit weight was in JE 21-1, ES 3-3 and JE 10-2 when compared to BTH and untreated control. The soft rot disease caused by Pectobacterium carotovorum SCCI was completely suppressed in case of transgenic tobacco harboring GUS gene related to PR1a and increased the level of salicylic acid significantly in combined application of JE 9-4 on par with BTH. Thus, this study clarified some potential Bacillus subtilis strains for plant growth promotion and ISR in red-pepper and tomato.
A leading sweet pepper cultivar, Keystone Resistant Giant #3, was crossed with a line with resistance to Phytophthora capsici, PI201232, for incorporation of the resistance and to study the inheritance of resistance to the disease. Seedlings of parents, $F_1$, $F_2$ and backcross populations of a cross between Keystone Resistant Giant #3 and PI201232 were inoculated with zoospore suspension of P. capsici at 36 days after seeding. Most of the $F_1$ seedlings survied the inoculation and this suggested that resistance is dominant over susceptibility. The number of survived plants in $F_2$ population was, however, much less than the killed. All the plants in a backcross to Keystone Resistant Giant #3 were killed. Therefore, the observed numbers did not fit the expected ratio for segregation of one or two dominant alleles as previously reported. The resistance to P. capsici appeared to be inherited in a quantitative mode in evaluation of root rot. Resistant individuals in $F_2$ population were selected and a breeding program for incorporation of the resistance to P. capsici by backcross method is continued.

Phytophthora capsici is one of major limiting factors in production of pepper and other important crops worldwide by causing foliage blight and rot on fruit and root. Increased demand for the replacement of fungicides has led to searching a promising strategy to control the fungal diseases. To meet eco-friendly agriculture practice, we isolated microorganisms and assessed their beneficial effects on plant health and disease control efficacy. A total of 360 bacterial strains were isolated from rhizosphere soil of healthy pepper plants, and categorized to 5 representative isolates based on colony morphology. Among the 5 bacterial strains (GJ-1, GJ-4, GJ-5, GJ-11, GJ-12), three bacterial strains (GJ-1, GJ-11, GJ-12) presented antifungal activity against P. capsici in an fungal inhibition assay. In phosphate solubilization and siderophore production, the strain GJ-1 was more effective than others. The strain GJ-1 was identified as Bacillus sp. using 16S rDNA analysis. Bacillus sp. GJ-1 was also found to be effective in inhibiting other plant pathogenic fungi, including Rhizoctonia solani, Pythium ultimum and Fusarium solani. Therefore, the Bacillus sp. GJ-1 can serve as a biological control agent against fungal plant pathogens.
