Skeletal System of the Hermaphroditic Rivulus Rivulus ocellatus marmoratus (Teleostomi: Aplocheilidae) (암수동체성 점박이송아리 Rivulus ocellatus marmoratus (진구아강: 점박이송사리과)의 골격계)

  • The complete osteology of the laboratory-reared hermaphroditic rivulus, Rivulus ocellatus marmoratus, was described. The skeletal system of this species consisted of more than 430 unit bones classified into 68 kinds. The osteological characters of this species were basically similar to those of the members of the Atherinomomha. The loss of circumorbitals, ectopterygoids, first pleural ribs and intermuscular bones was, however, noticed as the prominent features of this species. Anatomical and systematic implications of the observations were discussed.

  • The spontaneous bilateral asymmetry was analyzed in the hermaphroditic fish Rivulus marmoratus to obtain base line data on the developmental stability of this fish. The results obtained were as follows; 1. Eight kinds of countable anatomical characters except numbers of otolith and radii branchiostegi showed bilateral asymmetry at the frequency ranging from 9.8% to 64.7%. 2. Asymmetry index was not significantly different from that of gonochoristic fish species previously reported. Genetical implication of the results was discussed in relation to developmental stability and homozygosity of this species.

  • The full sequence analysis of 16 different $\beta-actin$ genes isolated from a single Rivulus marmoratus was performed. The numbers of isolated $\beta-actin$ genes varied from 1764 to 1769. They showed different amino acid residues at the exon 1 region. We named this new gene R. marmoratus $\beta-actin$ 2 gene. Intraspecific variation of R. marmoratus $\beta-actin$ 2 gene was also examined. Major differences of 16 isolated $\beta-actin$ genes, compared to already reported R. marmoratus $\beta-actin$ gene (GenBank AF168615), were observed in both deduced amino acid sequences (1~2%) and intron 2 sequences (4~5%). In this paper, we confirmed the intraspecific variation of $\beta-actin$ gene in this species.

Effects of Acute Acid Stress on Hatching and Mortality of Hermaphroditic Teleost, Rivulus marmoratus(Cyprinodontiformes; Rivulidae)

  • The effects of acute acid stress on hatching success and hatching period of laboratory-reared hermaphroditic fish Rivulus marmoratus were examined. The effects of acute acid toxicity on mortality was also determined in three life stages of this fish. There was a significant negative effect of acid stress on hatching performance in the R. marmoratus embryos. The hatching success was only 5% at pH 3.5 compared to over 78% at pH higher than 4.0. The hatching period was also delayed by low pH treatments. The larval and juvenile stages were more sensitive to acid toxicity on mortality than the adult stage, but larvae and juveniles showed similar sensitivity. The 96-h LC50 value was pH 3.8 in larval and juvenile stages and pH 3.3 in adult stage.

cDNA Cloning and Tissue Distribution of Two Parvalbumin Isoforms from the Hermaphrodite fish Rivulus marmoratus(Cyprinodontiformes, Rivulidae)

  • We isolated two parvalbumin cDNAS by expressed sequence tag analysis (1,577 ESTs in total) from the self-fertilizing fish Rivulus marmoratus (Cyprinodontiformes, Rivulidae). Two isoforms of parvalbumin genes showed high similarity to those of carp at 88% and 91% amino acid residues identity, respectively, and showed 79.8% similarity between two parvalbumin isoforms. Of 1,577 ESTs from R. marmroatus sequenced, parvalbumin 1 gene was most abundant. This gene was strongly expressed in the order of muscle, eye, and brain, while it was expressed slightly in other tissues. In this paper, we discussed on the R. marmoratus parvalbumin genes on its sequence and basic characteristics.

  • 점박이송사리에 관한 해부학적 자료축적의 일환으로 부속지골격의 형댈 발생 및 골화 진전양상을 조사하였다. 각 부속지의 구성골은 전피성골이 연골성골보다 먼저 골화되었다. 부속지의 기조는 가슴지느러미, 구리지느러미, 등 및 됫지느러미, 그리고 배지느러미의 순으로 골화되었다. 골화 진전에 따라 초기 자어시 유함되어 있던 수직지느러미의 기골은 방사기골과 중담기골로, 가슴지느러미의 오초-견갑골은 오훼골과 견갑골로 분리, 분화되었다. 그러나 골화진전에 따른 구성 골간의 유합은 나타나지 않았다. 5가지 부속지의 골화 완성은 가슴지느러미, 꼬리지느러미, 등지느러미, 됫지느러미 그리고 배지느러미의 순으로 이루어졌다.
