Norcantharidin Anti-Angiogenesis Activity Possibly through an Endothelial Cell Pathway in Human Colorectal Cancer

  • The present study was based on the unexpected discovery that norcantharidin exerted anti-angiogenesis activity when effects on growth of human colon cancer were studied. The aim was to further verify this finding and explore possible mechanisms using a tumor xenograft model in nude mice. We confirmed that norcantharidin (5 or 15 mg/kg) could inhibit angiogenesis of human colon cancer in vivo. In vitro, crossing river assay, cell adhesion assay and tube formation assay indicated that NCTD could reduce the migration, adhesion and vascular network tube formation ability of HUVECs. At the same time, the expression levels of VEGF and VEGFR-2 proteins which play important roles in angiogenesis were reduced as examined by western blotting analysis. Taken together, the results firstly showed NCTD could inhibit angiogenesis of human colon cancer in vivo, probably associated with effects on migration, adhesion and vascular network tube formation of HUVECs and expression levels of VEGF and VEGFR-2 proteins.

Vascular Plants of Construct-Reserved Site of Ecological Stream, Sohyeoncheon in Gyeongju-si (경주시 생태하천 예정지인 소현천의 관속식물상)

  • The purpose of this study is to investigate the flora of Sohyeoncheon which is planned for ecological restoration and river maintenance from upstream to downstream, and also to collect the ecological data to cope with the change of the ecosystem of Sohyeoncheon. The numbers of vascular plants were summarized as 336 taxa including 82 families, 233 genera, 298 species, 1 subspecies, 33 varieties and 4 forms. The rare plants were 3 taxa including Aristolochia contorta Bunge, Prunus yedoensis Matsum.(planting) and Koelreuteria paniculata Laxmann. The Korean endemic plants were 4 taxa including Populus tomentiglandulosa T.B.Lee(planting), Lespedeza maritima Nakai, Forsythia koreana (Rehder) Nakai(planting) and Weigela subsessilis (Nakai) L.H.Bailey. The specific plants by floristic region were 15 taxa including 1 taxon of grade V, grade IV and grade II each, 4 taxa of grade III and 8 taxa of grade I. The naturalized plants were 60 taxa and the invasive alien plants were 3 taxa. The results of analysis by vertical structures showed that naturalized plants included 31 taxa of upstream, 53 taxa of midstream and 38 taxa of downstream. An analysis of crossing structures showed that 2 taxa of water channel, 3 taxa of low waterside, 41 taxa of high waterside and 48 taxa of bank.

Application of Geophysical Results to Designing Bridge over Large Fault (대규모 단층대를 통과하는 교량설계를 위한 물리탐사의 활용)

  • During the core drilling for the design of a railway bridge crossing over the inferred fault system along the river, fracture zone, extends vertically more than the bottom of borehole, filled with fault gouge was found. The safety of bridge could be threatened by the excessive subsidence or the reduced bearing capacity of bedrock, if a fault would be developed under or around the pier foundation. Thus, a close examination of the fault was required to rearrange pier locations away from the fault or to select a reinforcement method if necessary. Geophysical methods, seismic reflection method and electrical resistivity survey over the water covered area, were applied to delineate the weak zone associated with the fault system. The results of geophysical survey clearly showed a number of faults extending vertically more than 50m. Reinforcement was not desirable because of the high cost and the water contamination, etc. The pier locations were thus rearranged based on the results of geophysical surveys to avoid the undesirable situations, and additional core drillings on the rearranged pier locations were carried out. The bedrock conditions at the additional drilling sites turned out to be acceptable for the construction of piers.

  • 하천을 횡단하는 구조물은 흐름을 지체시켜 상 하류에 급격한 수위차를 발생시키며, 이에 동반되는 국부유속의 증가는 제방 및 하상을 세굴시킨다. 제방 및 하상 세굴은 하천횡단구조물의 구조적 안정성에 영향을 주어 불안정한 상태에 이르게 할 수 있다. 이와 같은 현상을 방지하기 위하여 하상유지시설 및 제방 보호공 등의 대책을 하천설계기준에 제시하고 있다. 하천설계기준에는 하천횡단구조물 보호시설의 종류, 설치길이 등이 명시되어 있으나, 보호공 제원 선정에 대한 구체적인 설계공식을 제시하지 않아 보완이 필요한 상황이다. 보호공 제원 선정 설계기준은 설계유속을 1차원 단면평균 유속으로 활용하는 국외의 설계공식을 활용할 수 있다. 그러나, 하천횡단구조물 주변의 흐름은 매우 복잡하므로 국외 설계공식을 그대로 적용하기에 앞서서 이에 대한 검토가 필요하다. 본 연구에서는 하천횡단구조물 주변의 흐름을 2차원 수치모형으로 재현하고 그 결과를 1차원 수치모의 결과 및 수리실험 관측결과와 비교하였다. 실험수로에 각기 다른 제원의 보호공을 설치하고 관측한 임계상태의 수리상황을 1, 2차원 수치모형으로 재현하였다. 각각의 모형에서 도출된 수리상황 결과를 비교 분석하여 보호공 제원 결정 공식을 검토하였다. 또한, 실제 하천구간에 임의의 보와 수제를 설치하고, 계획홍수량에서의 흐름특성을 비교하였다. 1, 2차원 모형에 의하여 도출된 관계를 분석함으로써, 하천횡단구조물 설계시 적용할 수 있는 보호공 제원 결정 설계공식을 제안하였다.

MR damping system for mitigating wind-rain induced vibration on Dongting Lake Cable-Stayed Bridge

  • The Dongting Lake Bridge is a cable-stayed bridge crossing the Dongting Lake where it meets the Yangtze River in southern central China. Several intensive wind-rain induced vibrations had been observed since its open to traffic in 1999. To investigate the possibility of using MR damping systems to reduce cable vibration, a series of field tests were conducted. Based on the promising research results, MR damping system was installed on the longest 156 stay cables of Dongting Lake Bridge in June 2002, making it the worlds first application of MR dampers on cable-stayed bridge to suppress the wind-rain induced cable vibration. As a visible and permanent aspect of the bridge, the MR damping system must be aesthetically pleasing, reliable, durable, easy to maintain, as well as effective in vibration mitigation. Substantial work was done to meet these requirements. This paper describes field tests and the implementation of MR damping systems for cable vibration reduction. Three-years reliable service of this system proves its durability.

Water environmental change by river-crossing structure (하천횡단구조물 설치에 따른 수환경 변화 분석)

  • 하천의 횡단구조물은 하천을 횡단하는 시설물로 하상유지공, 보 등이 있으며 현재 국내 하천에는 수많은 보가 존재한다. 하천에 설치된 보는 하천 유황을 변화시키기 때문에 보 상 하류로부터 상당히 멀리 떨어져 있는 하도 구간에 대해서도 수환경을 변화 시키게 된다. 이로 인하여 하천을 터전으로 생활하는 다양한 생물의 서식처를 손상시키며, 어류와 야생 생물 각 개체의 생활상을 흔들어버린다. 이에 본 연구에서는 금강을 대상으로 횡단구조물 중 보 설치 이전과 이후의 수질의 변화에 따라 생태환경의 변화특성을 분석하였다. 연구대상지인 금강의 수질자료는 보 설치 이전 6년간(2003년 ~ 2008년)과 보 설치 이후 7년간(2011년 ~ 2017년)의 자료를 조사하였고 수생태환경의 변화를 분석하기 위한 부착조류, 저서성 무척추동물, 어류를 9년간(2008년 ~ 2016년) 조사하였다. 금강에 보가 설치된 이후 본류 전체적으로 BOD와 SS, T-N 등의 농도가 낮아지는 등 개선된 것으로 나타났지만 COD는 개선효과가 전혀 없었고, 보지점에서는 수온증가는 미미하지만 Chl-a가 계속적으로 증가하여 최근까지 금강에 녹조발생을 일으키고 있다고 판단된다. 금강본류 전체적으로 저서성무척추동물과 어류의 개체수가 감소하는 추세이며 수생태 건강성분석 결과 다른생물종에 비해 어류의 건강성등급이 하향하는 추세로 악화되고 있음을 확인하였다. 이는 Chl-a와 COD의 증가로 판단되며 최근들어 BOD도 증가하고 있어 앞으로 어류건강성이 저하가 우려되기 때문에 하천의 유속증가나 수문개방을 통해 건강성을 회복할 필요가 있다고 판단된다.

  • 대구시민의 삶의 질 향상에 크게 기여하는 신천은 홍수, 친수 및 하천환경 관리에 대한 필요성이 증가되는 상황에서 신천 복원을 위하여 하천횡단구조물 및 생태조사가 필요한 실정이다. 신천 내에 위치하고 있는 14개의 하천횡단구조물은 신천의 흐름을 가로막고 있어 수질악화 및 생물이동을 저해하고 있으며, 집중호우 등으로 인하여 구조물 주변에 대한 하천관리가 어려운 것이 현실이다. 특히 신천은 인구가 밀집해 있는 도심을 관통하는 하천으로 친수공간, 홍수 및 하천환경 관리에 대한 필요성이 증가하고 있다. 본 연구에서는 신천의 수질과 어류의 생태성의 상관관계를 확인하고 횡단구조물에 대한 해결방안을 제시하고자 한다. 현지조사 결과 총 3목 6과 10종의 어류가 관찰되었으며, 법정보호종인 멸종위기 야생생물 및 천연기념물은 관찰되지 않았으나, 고유종으로 긴몰개와 참갈겨니, 꺽지, 동사리 4종이 출현하였으며, 환경부 지정 생태계교란 생물 블루길 1종이 관찰되었다. 이러한 결과를 바탕으로 어류생물지수(FAI)를 검토하였을 때, 하류지역으로 갈수록 E등급으로 매우 나쁨의 환경상태를 나타내었다. 이는 상류에서 하류로 갈수록 비정상개체수가 증가하고 하상의 단순화 및 유기물의 축적으로 출현 종수의 감소에 따른 결과로 판단된다. 이러한 결과를 바탕으로 신천의 평균 FAI 지수는 14.71점으로 E등급의 매우 나쁨의 환경상태를 나타내는 것으로 분석되었다.

Infiltration characteristic of modified slurry and support efficiency of filter cake in silty sand strata

  • To improve the understanding of infiltration characteristic of modified slurry and the support efficiency of filter cake in silty sand strata, the slurry infiltration (SI) and filter cake formation (FCF) were investigated in a laboratory apparatus. The water discharge and the excess pore pressure at different depths of silty sand strata were measured during SI. The relationship between permeability coefficient/thickness ratio of filter cake (kc/ΔL) and effective slurry pressure conversion rate of filter cake (η) were analyzed. Moreover, the SI and FCF process as well as the modification mechanism of CMC (carboxymethyl cellulose) were clarified. The experimental results indicate the formation of only external filter cake in the silty sand strata. The slurry particles obtain thicker water membrane after being modified by CMC, which blocks partial water path in filter cake and decreases the water discharge significantly. The silty sand excavated from tunnel face also contributes to the water discharge reduction. The kc of the external filter cake ranges from 3.83×10-8 cm/s to 7.44×10-8 cm/s. The η of the external filter cake is over 96%, which decreases with increasing kc/ΔL. A silty sand content within 10% is suggested during construction to ensure the uniformity of the filter cake.

The Walkable Green Street Design for "Dangjae-Gil" (당재길 걷고싶은 녹화거리 설계)

  • This paper presents a streetscape design for "Dangiae-Gil"which is located at 126-1 Yangpyung-2dong, Yeongdeungpo-gu, Seoul and is about 500m in length and about 24m in width. The design goals are to make a green street on which people want to walk and people can walk and rest safely and pleasantly. To achieve these goals, concepts of environmentally-friendliness, placeness, safety, amenity, vitality, connectivity, and democracy are developed. For pedestrian safety, shared street concepts, such as crank, slalom etc. are adopted. The site is divided into 5 thematic spaces, such as "Village Entrance Space", "Culture Street", "Dangsan Park", "Nature Street", and "Ferry Space". The Village Entrance Space, which is an entrance of the Dangjae-Gil and a welcoming space, is for communicating information about the area. "Dangnamu"(zelkova tree) and signs are introduced here. The Culture Street is for experiencing past and present culture of the area. Colored tiles and plant boxes attached to benches are introduced. The Dangsan park is a sacred space where modem people can feel the sacredness of nature arid of being in a refuge. Dangjib, Dangnamu, multi-purpose plaza, athletic facilities, and playground for infants are introduced. The Nature Street is a space for feeling and teaming nature which has disappeared from the area leading to the river and a space for community participation. The elementary school walls were demolished and nature education spaces, such as butterfly and dragonfly garden, ecological pond, wildflower garden, etc., which are related to school education, are introduced. The Ferry Space is a space symbolizing a old ferry crossing and an entrance plaza to a bridge for "Sunyu-do\" . A boat-shaped deck, an elevator for handicap people, and parking space are introduced. In conclusion, sustainable management schemes for the site are suggested.sted.

Estimating magnitude of suspended sediment transport in ungauged east coastal zone (미계측 동해안 유역의 토사유출 규모의 평가에 관한 연구)

  • Coastal sediment archives are used as indicators of changes on shore sediment production and fluvial sediment transport, but rivers crossing coastal plains may not be efficient conveyors of sediment to the coast. In some case there is a net loss of sediment in lower coastal plain reaches, so that sediment input from an upstream exceeds the sediment yield (SY) at the river mouth. The main source of sediment in coastal area is the load from land. In Korea, data on suspended SY are limited owing to a lack of logistic support for systematic sediment sampling activities. This paper presents an integrated approach to estimate SY for ungauged coastal basins, using a soil erosion model and a sediment delivery ratio (SDR) model. For applying the SDR model, a basin specific parameter was validated on the basis of field data. The proposed relationships may be considered useful for predicting suspended SY in ungauged basins that have geologic, climatic and hydrologic conditions similar to the study area.
