Analysis of metabolites in wheat roots in response to salinity stress

  • Salinity stress is one of the most important abiotic stresses and severely impairs plant growth and production. Root is the first site for nutrient accumulation like as $Na^+$ in the plant. To investigate the response of wheat root under salinity stress, we executed the characterization of morphology and analysis of metabolites. Wheat seeds cv. Keumgang (Korean cultivar) were grown on the moist filter paper in Petri dish. After 5 days, seedlings were transferred to hydroponic apparatus at 1500 LUX light intensity, at $20^{\circ}C$ with 70% relative humidity in a growth chamber. Seedlings (5-day-old) were exposed to 50mM, 75mM, 100mM NaCl for 5 days. Ten-day-old seedlings were used for morphological characterization and metabolite analysis. Root and leaf length became shorter in high NaCl concentration compared to following NaCl treatment. For confirmation of salt accumulation, wheat roots were stained with $CoroNa^+$ Green AM, and fluoresce, and the image was taken by confocal microscopy. $Na^+$ ion accumulation rate was higher at 100mM compared to the untreated sample. Furthermore, to analyze metabolites in the wheat root, samples were extracted by $D_2O$ solvent, and extracted sample was analyzed by 1H NMR spectroscopy. Fourteen metabolites were identified in wheat roots using NMR spectroscopy. Methanol and ethanol were up-regulated, whereas formate, aspartate, aminobutyrate, acetate and valine were down-regulated under salinity stress on roots of wheat. Fumarate had no change, while glucose, betaine, choline, glutamate and lactate were unevenly affected during salinity stress.

Biological Efficacy of Streptomyces sp. Strain BN1 against the Cereal Head Blight Pathogen Fusarium graminearum

  • Fusarium head blight (FHB) caused by the filamentous fungus Fusarium graminearum is one of the most severe diseases threatening the production of small grains. Infected grains are often contaminated with mycotoxins such as zearalenone and trichothecences. During survey of contamination by FHB in rice grains, we found a bacterial isolate, designated as BN1, antagonistic to F. graminearum. The strain BN1 had branching vegetative hyphae and spores, and its aerial hyphae often had long, straight filaments bearing spores. The 16S rRNA gene of BN1 had 100% sequence identity with those found in several Streptomyces species. Phylogenetic analysis of ITS regions showed that BN1 grouped with S. sampsonii with 77% bootstrap value, suggesting that BN1 was not a known Streptomyces species. In addition, the efficacy of the BN1 strain against F. graminearum strains was tested both in vitro and in vivo. Wheat seedling length was significantly decreased by F. graminearum infection. However, this effect was mitigated when wheat seeds were treated with BN1 spore suspension prior to F. graminearum infection. BN1 also significantly decreased FHB severity when it was sprayed onto wheat heads, whereas BN1 was not effective when wheat heads were point inoculated. These results suggest that spraying of BN1 spores onto wheat heads during the wheat flowering season can be efficient for plant protection. Mechanistic studies on the antagonistic effect of BN1 against F. graminearum remain to be analyzed.

Feeding Value of Spent Mushroom (Pleurotus eryngii) Substrate (새송이버섯 수확후배지의 사료적 가치)

  • Spent mushroom substrates is composted organic material remaining after a crop of mushroom is harvested. The raw materials of mushroom substrates are same feed ingredient as corncobs, rice brown, wheat brown, cotton seeds and beet pulp. During the mushroom cultivation process, the mushroom substrates was used 15-25% by mushroom and 75-85% of mushroom substrates was remained in the SMS. Among of the spent mushroom substrates, spent mushroom substrates of pleurotus eryngii, Pleurotus ostreatus and Flammuliua velutipes is can be use the energy feedstock of animal feed. The cellulose content of spent mushroom(pleurotus eryngii) substrates containing the sawdust was high and total digestible nutrients (TDN) values was low. The spent mushroom(pleurotus eryngii) substrates fermented with cellulase and xylanase producing bacteria is may be used as an ingredient of feed in TMR for Hanwoo steer.

  • The parents and F$_1$ seeds from the all possible combinations of the diallel crosses among five upland varieties and a low-land variety were grown in green house in winter and also on paddy field in summer. The durations to heading of all F$_1$'s grown in green house at winter were shorter than those of their parents mean. The shorter duration to heading was due to dominace was partial. And the non-allelic gene interaction was not significant. But in case that the F$_1$'s and the parents were grown on paddy-field in summer. the durations to heading of F$_1$'s were various according to the cross combinations and the non-allelic gene interactions were significant.

Cereal Scab Causing Mycotoxicoses in Korea and Present Status of Mycotoxin Researches (한국(韓國)에서의 진균중독증(眞菌中毒症)을 일으키는 맥류(麥類) 붉은 곰팡이병 및 진균독소(眞菌毒素) 연구(硏究)의 현황(現況))

  • In 1963, a severe epidemic of cereal scab caused by Gibberella zeae occurred in southern Korea and to a less extent in central and northern Korea. In some areas losses were $80{\sim}100$ percent. The epiphytotic was due to heavy rainfall during the heading and flowering season which provided a favorable environment for this severe epidemic. Yield losses resulted a great social problem because of the resultant food and feed grain shortage, lose of seeds for planting the following crops and mycotoxicoses to man and animals. In the same year, a nationwide research committee was organized including plant pathologists, microbiologists, agronomists and biochemists under the juristiction of the Ministry of Agriculture and Forestry. The committee initiated research on etiology, epidemiology, and control of the disease and on the toxic effect of infected grains to man and animals. The present paper will review some research carried out in Korea on cereal scab with special reference to epidemiology and mycotoxicoses to animals and man. In addition, the present status of research in Korea on aflatoxins in foods and toxic moldy rice will be briefly reviewed.

Analysis of Physicochemical Properties of Nurungji Added with Various Materials (다양한 소재가 첨가된 누룽지의 이화학 특성 분석)

  • Increasing the rice consumption, seven kinds of Nurungji which served as a home meal replacer and snack were developed with some functional materials were added, and their physiochemical characteristics were analyzed. The water binding capacity was the highest in Nurungji with black sesame seed (264.13%), and the hardness was the lowest as 0.36 N in Nurungji with Tenebrio molitor. The reducing sugar content was the highest in Nurungji with sweet pumpkin (1.47%) when being soaked in water for 30 minutes. As a result of measuring 2,2-diphenyl-1-picryl-hydrazyl free radical scavenging activity of the tested Nurungji preparations, Nurungji with sweet pumpkin and beet showed the high scavenging activity against free radicals. As a result of sensory evaluation, overall acceptability was the highest in Nurungji with black sesame seeds (4.3). Physicochemical characterization and sensory evaluation of Nurongji added with various materials confirmed that they could be applied to various food products.

  • Water extracts from Polygonum aviculare and Salix koreansis markedly inhibited the germination of lettuce and rice seeds, indicating the presence of biologically active substances. The biochemical substances such as salicylic and+vanillic acid, tannic acid + gallic acid, p-coumaric acid, p-cressol, sinapic acid and catechol etc. belonging to phenolic compounds were detected in the cultured cells, suggesting that the secondary metabolites can be synthesized in plant cell and tissue culture. In addition, fatty acid like linolenic acid and organic acid such as oxalic acid were presented in the highest amount, 3.7 mg/g and 14.288 mg/g, respectively, which seem to be related to exhibiting phytotoxicity of P. aviculare. Petroleum ether extract exhibited another potential relating to inhibitory effect which needs further investigation. Calli from two plant sources were easily introduced by uses of 1.0 mg/l of 2.4-D and 0.1 to 0.2 mg/l of BAP in MS basal medium which can be implemented for a large scale production through cell culture.

  • Cell suspension cultures for Salix koreensis was well established at the supplements of 2, 4-D with cytokinin particulary the combination of 1.0 mg/l 2, 4-D with 0.1 mg/l of zeatin. These combined rates of phytohormones are also effective to callus induction from S, koreensis leaf and its multiplication. Cultured media exhibited the great inhibitory effect on the germination of rice, barnyard grass and lettuce seeds, indicating the presence of biologically active substances in media. Several phenolic compounds such as pyrogallol, sinapic acid, cinnamic acid, tannic + gallic and p-chlorobenzoic acid were detected in the cell suspension culture. The inhibitory effect exhibited by cultured media may be partly attributed to these phenalic compounds.

Characterization of a New High-lysine Mutant in Barley (Hordeum vulgare L.)

  • A chemical, MNU-induced hulless barley mutant line designated as 'Mutant 98 (M98)' was developed from a Korean hulless waxy barley cultivar, 'Chalssalbori'. The objective of the study was to determine the genetic basis of 'M98' and the possibility of using 'M98' as breeding parent to improve lysine level. Compared to 'Chalssalbori', 'M98' had large embryo and higher lysine content in both the embryo and endosperm. Significantly different lysine content in 'M98' and the other high-lysine barley mutant stocks was observed for two years. However, the genotype by year interaction was not significant. 'M98' was higher than the other high-lysine barley mutant stocks in the percentage of lysine of total amino acid composition (0.75%). The trait of shrunken endosperm of 'M98', which was typical in the high-lysine mutants, was inherited by a single recessive gene. Based on seed morphology and lysine content of $F_1$ seeds, 'M98' had a genetically different gene from the other high-lysine mutants for shrunken endosperm. Segregation of $F_2$ for plump/shrunken endosperm did not fit the expected ratio of Mendelian inheritance except for only one cross combination (GSHO1784 (lys1)/M98). The amino acid analysis of $F_5$ and $F_6$ progenies from the cross between 'M98' and 'Chalssalbori' revealed that the attempt to increase the range of lysine content of plump lines did not go beyond the limit of the average high-lysine barley germplasm.

Perceived Characteristics of Grains during the Choseon Dynasty - A Study Applying Text Frequency Analysis Using the Choseonwangjoshilrok Data - (조선왕조실록 텍스트 빈도 분석을 통한 조선시대 곡물에 관한 인식 특성 고찰)

  • This study applied the text frequency method to analyze the crops prevalent during the Chosunwangjoshilrok dynasty, and categorized the results by each king. Contemporary perception of grains was observed by examining the staple crop types. Staple species were examined using the word cloud and semantic network analysis. Totally, 101,842 types of crop consumption were recorded during the Chosunwangjoshilrok period. Of these, 51,337 (50.4%) were grains, 50,407 (49.5%) were beans, and 98 (0.1%) were seeds. Rice was the most frequently consumed grain (37.1%), followed by pii (11.9%), millet (11.3%), barley (4.5%), proso (0.8%), wheat (0.6%), buckwheat (0.1%), and adlay (0.05%). Grain chronological frequency in the Choseon dynasty was determined to be 15,520 cases in the 15th century (30.2%), 11,201 cases in the 18th century (21.8%), 9,421 cases in the 17th century (18.4%), 9,113 cases in the 16th century (17.8%), and 6,082 cases in the 19th century (11.8%). Interest in grain amongst the 27 kings of Choseon was evaluated based on the frequency of records. The 15th century King Sejong recorded the maximum interest with 13,363 cases (13.1%), followed by King Jungjo (8,501 cases in the 18th century; 8.4%), King Sungjong (7,776 cases in the 15th century; 7.6%).
