Effects of Lacquer (Rhus verniciflua) Meal on Carcass Traits, Fatty Acid Composition and Meat Quality of Finishing Pigs

  • This experiment was conducted to evaluate the effects of feeding different levels of lacquer (Rhus verniciflua Stokes) meal on the growth performance, carcass traits, fatty acid profile and meat quality of longissmuss dorsi (LD) muscle in finishing pigs. Pigs (n = 117; Landrace$\times$Yorkshire$\times$Duroc; initial body weight $80{\pm}0.4kg$) were allotted to three dietary treatments and fed lacquer at 0, 2 and 4% of the diet for five weeks. Inclusion of lacquer meal in the diets of pigs had no influence on their growth performance, carcass yield, loin eye area and fat free lean; however, pigs fed lacquer diets had lower backfat (linear, p = 0.006; quadratic, p = 0.004). Pigs fed increasing levels of lacquer meal had lower moisture (linear, p<0.001; quadratic, p = 0.008), crude fat (linear, p<0.001) and crude protein (linear, p<0.001; quadratic, p = 0.002) in LD muscle. The LD muscle of pigs fed lacquer meal had lower pH (linear and quadratic, p<0.05) at 6, 8 and 10 days, and linearly lower thio-barbituric acid reactive substances (TBARS, p<0.01) at 8 and 10 days and water holding capacity (WHC, p<0.05) at 3, 6, 8 and 10 days. The fatty acid composition of LD muscle revealed linearly lower stearic (p = 0.034) and total saturated fatty acid (p = 0.049) with increasing dietary lacquer meal levels. In general, higher lightness, redness and yellowness values were observed in LD muscle of pigs fed 2% lacquer meal on day 0 and subsequently on 3, 6, 8 and 10 days of refrigerated storage. The results of the current study suggest that lacquer meal can be incorporated up to 4% in the diet of finishing pigs without any adverse effects on performance; moreover, improvements in the meat quality during refrigerated storage can be obtained by inclusion of lacquer meal in the diet of finishing pigs.

  • This experiment was conducted to explore the efficacy of lacquer (Rhus verniciflua) supplementation on growth performance, nutrient digestibility, carcass traits and serum biochemical values in commercial broilers. Eight hundred and forty Hubbard broiler chicks (1d old) were randomly distributed and allotted to four dietary treatments for five weeks. Each treatment had 5 replicates with 42 birds each. The dietary treatments were (1) control (2) 1% lacquer, (3) 2% lacquer and 4) 4% lacquer supplied as meal in the diet. Supplementation with lacquer improved weight gain (p = 0.0960) showing a linear trend during the starter phase (0-3 wk), but weight gain, feed intake and feed conversion ratio remained unaffected at the finisher phase (4-5 wk) and overall (0-5 wk). The nutrient digestibility studies conducted after 18 and 35 days of experimental feeding showed a linear (p<0.05) increase in digestibility of CP and ether extract at both measurement timess. The carcass studies were conducted on two birds per replicate (10 per treatment) at the end of both 3 and 5 wk. The dressing percentage, liver weight, heart weight, abdominal fat and the breast meat expressed as a percentage of live weight, did not differ significantly due to treatments at both phases. The serum cholesterol and high-density lipoprotein (HDL) showed a linear decrease (p = 0.0683 and p = 0.0322, respectively) as the level of supplementation increased at 3 wk; at 5 wk serum cholesterol, HDL and triglyceride levels decreased significantly showing a positive linear effect of lacquer on fat metabolism. The meat color values such as lightness, redness and yellowness did not reveal any significant trend. Overall, it could be concluded that lacquer supplementation at higher levels did not affect growth performance, but had a positive impact on fat metabolism by influencing fat digestibility and reducing serum cholesterol and triglyceride.

한국산 옻칠의 우루시올 성분 분석 (Analysis of the Urushiol in Korean Lacquer)

  • 한국에서 옻나무(Rhus verniciflua)는 전통적으로 약용식물로 오랫동안 사용되어 왔다. 민간요법으로 위장병 치료를 목적으로 옻나무껍질을 닭과 함께 넣어 끊여서 백숙형태로 옻닭으로 식용하였다. 그러나 일부 사람에게서 피부발진이 발생하는 극심한 알러지 반응을 일으키는 나무로 인식되어 왔다. 알러지 발생을 유발시키는 우루시올은 옻나무의 수지상 옻칠 액의 주성분이다. 이 성분의 화학적 구조는 카테친의 기본구조에 알킬기인 불포화지방산(C$_{15}_{\sim}_{17}$)이 곁사슬로 붙어 있다. 곁사슬의 불포화지방산 성분은 칠기의 고분자화 및 경화과정에 중요한 역할을 하며, 옻칠공예품의 색상 및 품질에 커다란 영향을 미친다. 한국에서 생산되는 원주산 옻칠액의 우루시올성분은 역상 컬럼을 사용하여 고속 액체 크로마토그라피 (HPLC)법으로 정제하여 ESI(-), FAB(+) 질량분석기(MS)로 분석한 결과 분자량 340인 Hetadecatetetraenyl catechol 이었다.

생칠과 화칠의 우루시올 조성 및 생리활성 비교 (Comparison of Urushiol Composition and Biological Activity between Fresh Sap and Fire Distilled Sap of Lacquer Tree)

  • For the comparison of the urushiol composition and biological acitivity between the fresh sap and fire distilled sap of lacquer tree(Rhus vernicifera), we analysed the urushiol composition by HPLC and EI-MS, and investigated the antioxidative , antimicrobial and anticancer acitivities according to solvent fractionations. There was no difference in the urushiol composition between fresh and fire distilled saps of lacquer tree. The hexane frqctionsof two saps showed a strong DPPH radical scaverging activity (RC50 : 7.0-7.5$\mu\textrm{g}$). They also showed a strong antifungal activity onthe spore germination of Cladosporium herbarum(MIC : 8$\mu\textrm{g}$/ml), whereas they have no or low activity against the bacteria(BAcillus subtilis , Escherichia coli). In addition , hexane and butanol fractions of two saps showed a strong inhibitory activity against cultured tumour cell lines (GI50 : 0.35, 12.29$\mu\textrm{g}$/ml) in vitro. These results confirm that the fresh sap and fire distilled sap might have the similar urushiol compositions and biological activities.

옻나무 우죽의 수피생산량 분석 (Bark Production Analysis on Top Branch of Rhus verniciflua)

  • 본 연구는 옻을 채취한 옻나무에서 수피생산량을 파악하기 위하여 옻나무의 직경에 따른 중량과의 관계와 우죽의 중량에 따른 수피와 건피량을 조사한 결과 옻나무중량에 대한 우죽중량의 비율은 평균 $48\%$였는데, 중량이 큰 나무들이 우죽비율이 높게 나타났으며, 옻나무 직경이 최소한 8cm정도는 되어야 우죽에서 1kg이상의 건피를 생산할 수 있는 것으로 분석되었다. 옻나무 우죽의 수피를 측정한 결과 생피무게는 $1.1{\sim}l.5kg$에 해당하는 옻나무가 $43.3\%$로 가장 많았고, 다음은 $0.5{\sim}1.0kg$$37.5\%$로 조사되어 전체 우죽의 $80\%$이상이 1.5kg이하인 것으로 나타났다. 또한 생피를 말린 건피의 무게는 $0.6{\sim}1.0kg$에 해당하는 나무들이 $50.8\%$로 가장 많았으며, 다음이 $0.1{\sim}0.5kg$으로 $25\%$로 나타나 전체 옻나무의 $75\%$정도가 1.0kg이하인 것으로 조사되었다. 그리고 우죽중량에 대해 생피중량이 차지하는 평균비율은 $27\%$였으며, 우죽중량에 대한 건피중량의 평균비율은 $16\%$로 측정되었는데, 생피를 건조시켰을 때 평균건피중량은 생피의 $61\%$로 나타나 생피의 수분함유율이 평균 $39\%$로서 건피 1kg을 생산하기 위해서는 우죽 6.25kg이 소요되는 것으로 분석되었다.

산지별 옻피의 항산화 활성과 옻닭국 기호특성 비교 (Antioxidative Activities of Rhus verniciflua Bark from Different Area)

  • 본 연구에서는 지역별 옻나무 껍질의 총 폴리테놀 함량, 플라보노이드 함량, 항산화능과 우루시올 함량을 측정하고, 옻나무 피를 첨가한 옻닭 국물의 관능검사를 실시하였다. 원주산 옻나무 껍질의 total polyphenol, flavonoids, DPPH free radical 소거능, ABTS radical scavenging activity 등이 함양산 및 중국산에 비해 높은 수치를 나타내었다. Urushiol 함량은 함양산이 $4.59{\pm}0.04$ ppm으로 가장 높은 함량을 나타내었지만, 중국산 옻나무 껍질에서는 검출되지 않았다. 옻나무 껍질을 첨가한 옻닭 국물의 관능검사에서는 원주산을 첨가한 옻닭국물의 선호도가 가장 높은 것으로 나타났다.

Antioxidative Activity of Urushiol Derivatives from the Sap of Lacquer Tree (Rhus vernicifera Stokes)

  • The authors isolated four olefinic catechols, commonly referred to as urushiol, from the sap of Korean lacquer tree(Rhus vernicifera STOKES) with stronger antioxidative activities than $\alpha-tocopherol$. The hexane extract with a free radical scavenging activity was purified by silica and ODS gel column chromatography. The active compounds were identified by MS and $^1H-NMR$ as 3-[8'(Z),11'(Z),14'-pentadecatrienyl]catechol, 3-[8'(Z),11'(Z)-pentadecadienyl]catechol, 3-[8'(Z)-pentadecenyl] catechol, and 3-pentadecylcatechol. All of these compounds showed strong free radical scavenging activities on 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical, in which 3-pentadecylacatechol exhibited the highest activity ($IC_{50}$: $1.2{\mu}g/ml$). They also showed a significant inhibitory activity on lipid peroxidation ($IC_{50}$: 2.1 - 3.5 ${\mu}g/ml$). The antioxidative activity of 3-pentadecylcatechol on DPPH radical and lipid peroxidation is approximately two times greater than that of $\alpha$-tocopherol. The results suggest that the urushiol derivatices may contribute to the preservative characteristics effective against oxidative stress and could be a good source for industrial applications including a coating material.

Cytotoxicity of Urushiols Isolated from Sap of Korean Lacquer Tree (Rhus vernicifera Stokes)

  • Cytotoxicities of four urushiols, congeners isolated from the sap of Korean lacquer tree (Rhus vernicifera Stokes), to 29 human cancer cell lines originated from 9 organs were evaluated. Their values of 50% growth inhibition were below $4{\mu}g/ml$, and showed cell line specific cytotoxicity. The present results is the first report on the cytotoxicity of urushiols suggesting that they would have an anticancer activity to human cancer cells.

옻나무 추출물 첨가에 따른 영지버섯의 가나도마난디올 생합성 증대 및 멜라닌 생성 저해효과 (Enhancement of Ganodermanondiol and Anti-melanogenesis Effect of Ganoderma lucidum by Rhus verniciflua Extract Supplementation)

  • 본 연구는 옻나무 추출물의 첨가가 영지버섯 미백활성 물질인 가나도마난디올의 생합성에 미치는 영향을 조사하기 위하여 수행하였다. 옻나무 열수 추출물을 1, 5, 10, 15%의 비율로 영지버섯 균사발효 시 첨가하고 발효 균사 추출물의 HPLC분석을 수행한 결과, 가나도마난디올 생합성이 첨가하지 않은 대조군에 비해 유의적으로 증가함을 확인하였다. 또한, 옻나무 열수 추출물이 첨가되어 발효된 영지 균사 추출물의 B16F10 세포주에 대한 멜라닌 생합성 저해능을 평가한 결과 첨가하지 않은 대조군에 비해 유의적으로 저해능이 증가함을 확인하였고, MTT분석을 통해 B16F10 세포주에 대한 세포생존율에도 영향을 미치지 않은 것을 확인하였다. 결과적으로 옻나무 추출물이 영지버섯균사의 미백활성 물질인 가나도마난디올 생합성을 촉진하여 향후 옻나무 추출물 첨가 영지버섯균사 발효 추출물이 효과적인 화장품 성분으로 사용 가능할 것으로 사료된다.

The hexane extract with a inhibitory effects on reverse transcriptase was purified by silica and ODS gel column chromatography. The active compounds were identified by MS and $^1H-NMR$ as 3-[8' (Z), 11' (Z), 14' -pentadecatrienyl]catechol, 3-[8' (Z), 11' (Z)-pentadecadienyl]catechol, 3-[8' (Z)-penta- decenyl]catechol, and 3-pentadecylcatechol. All of these compounds showed strong inhibitory effects on reverse transcriptase of AIDS, in which 3-pentadecylcatechol exhibited the highest activity $(IC_{50}\;:\;10.87\;{\mu}g/ml)$.
