고추냉이 물재배시 묘크기가 생육 및 수량에 미치는 영향 (Effect of Seedling and Division Nursery Stock Size on Growth and Yield in Water Culture Condition of Wasabia japonica Matsum)

  • 1. 소양댐의 수질을 조사, 분석하여 일본 주산지인 정강(靜岡)과 비교한 결과 pH는 비슷하고, 전기 전도도는 $0.03{\sim}0.2mmho/cm$ 안에 들어 재배에 적당한 조건이었다. 2. 소양댐 용수의 용존성분중 질소, 인산은 일본보다 약간 높고, K는 낮았다. 황은 높은 수준이었고, Mg는 매우 낮았다. 3. 분주묘의 초장 변화는 대묘에서는 이중 sigmoid, 중묘, 소묘는 S형 생장을 하였다. 4. 분주묘의 근경중 분포에서 40g이상의 개체는 대묘는 87%, 중묘는 80%, 소묘는 54%의 분포를 보였고, 상품 근경중은 대묘 723kg/10a, 중묘 639kg/10a 이었다. 5. 실생묘의 묘 크기별 초장 변화 곡선은 묘 크기에 상관없이 S형 생장을 나타냈다. 6. 실생묘의 묘 크기별 상품율은 대묘 60%, 중묘 13%, 소묘 20%, 상품근경중은 대묘 395kg/10a, 중묘 102kg, 소묘 160kg 이었다. 7. 고추냉이 주요 형질간 상관관계에서 초장은 엽병장, 엽장, 엽폭, 엽병중과 엽폭은 엽중, 엽병중과 정의 상관관계를, 근경중은 주경엽수, 근경장, 근경폭, 엽중, 총중량과 고도의 정의 상관관계를 보였고, 엽병중, 총근경중과도 유의성 있는 상관을 보였다.

  • 국내에 분포하는 대사초절에 속하는 3분류군의 외부형태형질을 분석하고 분자계통학적 연구를 수행하여 이들 간의 유연관계를 밝혔다. 대사초절 내의 기본종인 대사초의 외부형태학 특징을 재검토하여 기존 문헌과의 차이를 파악하였으며, 식물상 문헌 및 표본을 통해 국내 분포도를 작성하였다. 문헌에 언급된 형질들 중에서 잎의 형태는 생육지 조건에 따라 변이가 심하였지만, 지하경 생장 양상, 잎과 과낭의 털의 존재 등은 종수준에서 구분하는데 좋은 형질이었다. 분자계통학적 연구에서 털대사초는 독립된 종으로 처리되는 것을 지지하였다.

창출이 제 2형 당뇨병 흰쥐에 미치는 영향 (The effects of Atractylodes japonica Koidz. on type 2 diabetic rats)

  • Objectives: Type 2 diabetes mellitus is a metabolic disease characterized by insulin resistance and high blood glucose level from progressive insulin secretory defect. The rhizome of Atractylodes japonica Koidz. (AJ) has been used for treatment of retention of water in oriental medicine. The aim of this study is to examine the effects of AJ on type 2 diabetes rats. Methods: Type 2 diabetes was induced by 60% high fat diet and low dose streptozotocin. Rats were divided into 4 groups (n = 6); Nor (normal control group), Con (diabetic group treated with vehicle), Met (diabetic group treated with 200 mg/kg metformin) and AJ (diabetic group treated with 100 mg/kg AJ). The body weights and food intakes were measured during the treatment period. After 4 weeks treatment, blood glucose level, HOMA-IR, and protein expressions of IRS-1, p-IRS-1, PPAR-${\gamma}$, and GLUT4 were measured, and histopathological examination of beta cell was performed. Results: Compared with the control group, blood glucose level and HOMA-IR were reduced in rats treated with AJ. Impaired beta cells in pancreas of rats were recovered and phosphorylation of IRS-1 was increased in rats treated with AJ. And also, protein expressions of PPAR-${\gamma}$ and GLUT4 were increased by treatment of AJ. Conclusions: The results suggest that Atractylodes japonica Koidz. may have anti-diabetic effect on type 2 diabetic rats through regulation of blood glucose level and insulin resistance. Therefore Atractylodes japonica Koidz. may have positive effects on patients with type 2 diabetes.

거대억새 단지 조성에 따른 토양 특성 변화 (Change in Soil Properties after Planting Giant Miscanthus)

  • Miscanthus has received wide attention as an option for biomass production in Korea, recently. New strain of giant miscanthus has been developed and was planted in two large trial sites (184 ha) in the lower reaches of the Geum River. To evaluate the susceptibility of the giant miscanthus as an bioenergy crop for the future, we investigated the influence of the giant miscanthus on the soil properties. The particle size, CEC, pH, EC, T-N, T-P, heavy metal total concentration, and heavy metal fractions of soil samples collected from abandoned field, 1 year old giant miscanthus field (1st Year GM), and 2 year old giant miscanthus field (2nd Year GM) at different depths of 0~15, 15~30, and 30~45 cm in April and August were analyzed. Results showed that the CEC and pH of the soil of the giant miscanthus field were lower than those of the soil of abandoned field. The EC of 2nd GM was lower than that of abandoned field, indicating that the giant miscanthus may facilitate soil desalination process. The organic and sulfide fraction and residual fraction of heavy metals in the giant miscanthus field was higher than in abandoned field, due to the low pH of the giant miscanthus field and the excretion of phytosiderophores by rhizome of the giant miscanthus. This study showed that the giant miscanthus can influence on the soil properties and further study for long term is needed to elucidate the interaction between the soil and the giant miscanthus.

저년생 인삼의 잠아 및 화서형성에 관한 연구 (Study on the Forulation of Dormancy Bud and Inflorescence in Young Ginseng Plant)

  • 저년생 인삼의 잠아 및 화기형성시기와 양상은 고년생 인삼의 이차적인 양상과 상이하기 때문에 1년생 묘삼의 잠아발달 및 다식된 2년생 인삼의 화기형성과정을 조사관찰하였던 바 그 결과를 요약하면 다음과 같다. 1. 2년생 이상의 인삼 잠아조직은 4월중순경 분제조직의 조기포분제로 시작하는데 1년생 묘삼에서는 유근발근시인 3월 하순경 시작하는 것으로 추측되었으며 잠아의 위치는 자엽간 줄기하단 문부위에 생성되었다. 2. 3년생 이상의 인삼 화기분화는 전년도 형성된 뇌두조직내에서 형성되지만 2년생 인삼의 화기는 줄기선단 엽병사이의 경단분제조직에서 분제세포의 분열과 함께 분화, 형성되었다.

인삼(Panax ginseng)에 존재하는 Calcium Oxalate 결정체의 분포, 유형 및 미세구조 (Occurrence, Type and Ultrastructure of Calcium Oxalate Crystals in Panax ginseng)

  • 본 연구는 인삼에 존재하는 calcium oxalate 결정체의 분포 위치, 형태 및 미세구조에 관하여 밝히고자 광학현미경, 조직화학적 방법 및 주사 전자현미경적 방법 등을 사용하여 연구 하였다. 인삼의 결정체는 calcium oxalate의 성분으로 구성되며, 결정체는 몸통뿌리, 지근, 뇌두, 줄기, 장엽병, 소엽병, 꽃대 등 대부분의 기관에서 관찰되었다 특히 지상부는 잎에서,지하부는 뇌두에서 가장 많은 결정체가 관찰되었다. 인삼에 존재하는 결정체는 silver nitrate-rubeanic acid를 사용한 조직화학적 반응을 실시한 결과 calcium oxalate치 성분으로 확인되었다. 또한 주사전자현미경으로 관찰한 결과 삼각뿔 형태의 결정체가 다수 모여서 형성된 유형, 프리즘 형태의 소결정이 모여서 형성된 유형 및 얇은 판상의 소결정이 모여서 형성된 유형 등 3가지 형태로 구분 할 수 있었다.

Antioxidant Effects of Scutellaria baicalensis Georgi Against Hydrogen Peroxide-induced DNA Damage and Apoptosis in HaCaT Human Skin Keratinocytes

  • In this study, we investigated whether S. baicalensis rhizome ethanol extract (SBRE) has antioxidant capacities against oxidative stress induced cellular damage in the HaCaT keratinocytes. Our results revealed that treatment with SBRE prior to hydrogen peroxide ($H_2O_2$) exposure significantly increased the HaCaT cell viability. SBRE also effectively attenuated $H_2O_2$ induced comet tail formation, and inhibited the $H_2O_2$ induced phosphorylation levels of the histone ${\gamma}H2AX$, as well as the number of apoptotic bodies and Annexin V positive cells. In addition, SBRE exhibited scavenging activity against intracellular ROS generation and restored the mitochondria membrane potential loss induced by $H_2O_2$. Moreover, $H_2O_2$ enhanced the cleavage of caspase-3 and degradation of poly (ADP-ribose)-polymerase as well as DNA fragmentation; however, these events were almost totally reversed by pretreatment with SBRE. Furthermore, SBRE increased the levels of HO-1 associated with the induction of Nrf2. Therefore, we believed that SBRE may potentially serve as an agent for the treatment and prevention of neurodegenerative diseases caused by oxidative stress.

천마(天麻)가 뇌실질출혈 흰쥐의 신경세포 자연사에 미치는 영향에 대한 면역조직화학적 연구 (An Immunohistochemical Study on Effect of Gastrodiae Rhizoma against Neuronal Apoptosis Following Intracerebral Hemorrhage in Rats)

  • Objectives: This study was carried out in order to examine the effects of Gastrodiae rhizoma(GR) ethanol extract on neuronal apoptosis in intracerebral hemorrhage(ICH)-induced rats. Methods: ICH was induced by the stereotaxic intrastriatal injection of bacterial collagenase type VII in Sprague-Dawley rats. GR was orally given once a day for 3 days after ICH. Histological changes of the peri-hematoma regions were observed by cresyl vioIet staining. Bcl-2-associated X protein(Bax), B-cell blastoma 2(BcI-2) and terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling(TUNEL) expressions in the affected regions were performed by immunohistochemistry. Results: 1. GR reduced apoptotic bodies and swelling neurons in the peri-hematoma regions of ICH-induced rats. 2. GR significantly reduced TUNEL positive cells in the peri-hematoma regions of ICH-induced rats. 3. GR significantly reduced Bax positive cells in the peri-hematoma regions of ICH-induced rats. 4. GR did not influence Bcl-2 expression in the peri-hematoma regions of ICH-induced rats. Conclusions: These results suggest that GR has neuroprotective effects against ICH-induced apoptosis.

Determination of ginsenosides in Asian and American ginsengs by liquid chromatography-quadrupole/time-of-flight MS: assessing variations based on morphological characteristics

  • Background: Asian ginseng and American ginseng are functional foods that share a close genetic relationship and are well-known worldwide. This article aims to investigate the correlation between morphological characteristics and the inherent quality of Asian and American ginsengs. Methods: In this study, an ultra-HPLC-quadrupole/time-of-flight MS (UHPLC-Q/TOF-MS) method was established for the quantitative analysis of 45 ginseng samples. The method developed for determination was precise and accurate. Results: The results showed that Asian ginseng samples with the same growing time (with the same or similar number of stem scars) that had a thinner main root, a longer rhizome and more branch roots contained greater amounts of ginsenosides. For American ginseng, two tendencies were observed in the relationship between the diameter of the main root and contents of ginsenosides. One tendency was that samples with thinner main roots tended to contain higher levels of ginsenosides, which was observed in the samples sold under the commercial name pao-shen. Another tendency was that samples with thicker main roots contained higher contents of ginsenosides, which was observed in the samples sold under the commercial name pao-mian, as well as in samples of American ginseng cultivated in Jilin, China. Conclusion: An approach using ultra-HPLC-quadrupole/time-of-flight MS was successfully established to link morphology and active components for evaluating the quality of Asian and American ginsengs. Clear correlation between visible morphological features and quality of Asian and American ginsengs was found. People can see the difference; this means consumers and vendors can evaluate ginseng by themselves.

Overexpression of ginseng UGT72AL1 causes organ fusion in the axillary leaf branch of Arabidopsis

  • Background: Glycosylation of natural compounds increases the diversity of secondary metabolites. Glycosylation steps are implicated not only in plant growth and development, but also in plant defense responses. Although the activities of uridine-dependent glycosyltransferases (UGTs) have long been recognized, and genes encoding them in several higher plants have been identified, the specific functions of UGTs in planta remain largely unknown. Methods: Spatial and temporal patterns of gene expression were analyzed by quantitative reverse transcription (qRT)-polymerase chain reaction (PCR) and GUS histochemical assay. In planta transformation in heterologous Arabidopsis was generated by floral dipping using Agrobacterium tumefaciens (C58C1). Protein localization was analyzed by confocal microscopy via fluorescent protein tagging. Results: PgUGT72AL1 was highly expressed in the rhizome, upper root, and youngest leaf compared with the other organs. GUS staining of the promoter: GUS fusion revealed high expression in different organs, including axillary leaf branch. Overexpression of PgUGT72AL1 resulted in a fused organ in the axillary leaf branch. Conclusion: PgUGT72AL1, which is phylogenetically close to PgUGT71A27, is involved in the production of ginsenoside compound K. Considering that compound K is not reported in raw ginseng material, further characterization of this gene may shed light on the biological function of ginsenosides in ginseng plant growth and development. The organ fusion phenotype could be caused by the defective growth of cells in the boundary region, commonly regulated by phytohormones such as auxins or brassinosteroids, and requires further analysis.
