Phytochemical Screening of Korean Plants( I ) -On Urticaceae, Celastraceae, Rhamnaceae, Sterculiaceae and Rubiaceae- (국내(國內) 자생식물(自生植物)의 화학성분(化學成分) 검색(檢索)( I ) -쐐기풀과, 노박덩굴과, 갈매나무과, 벽오동과, 꼭두서니과에 대하여-)

  • The presence of saponin, terpenoid, steroid, anthraquinone and flavonoid were screened with the usual test methods in forty five(sixty four parts) Korean plants belonging to Urticaceae, Celastraceae, Rhamnaceae, Sterculiceae and Rubiaceae. Plants were extracted with methanol and fractionated with hexane, chloroform, ethyl acetate and butanol. Each fraction was tested for the components. The result showed that in saponin test, 21 plants were positive and 11 plants were weak positive; in terpenoid and steroid test, 26 plants were strong positive; in anthraquinone test, 20 plants strong positive; in flavonoid test, 31 plants were positive and 9 plants were weak positive.

Immune-enhancing Activity of Water Extracts for Each Part of 13 Species (Rhamnaceae) in Korea (국내 갈매나무과 13종에 대한 부위별 물 추출물의 면역 증진 활성)

  • This studied the immune-enhancing activity properties of water extracts from the leaves, branches, and fruit of 13 species (Rhamnaceae) collected during the bearing season (Berchemia berchemiifolia, B. floribunda, Hovenia dulcis, Paliurus ramosissimus, Rhamnella franguloides, Rhamnus crenata, R. davurica, R. koraiensis, R. parvifolia, R. ussuriensis, R. yoshinoi, Sageretia thea, and Ziziphus jujube). Immune-enhancing activity were studied using the nitric oxide (NO) production in RAW264.7 cells. Extracts of B. berchemiifolia, H. dulcis, R. franguloides, R. crenata, R. davurica, R. ussuriensis and S. thea showed strong immune-enhancing activity through NO production. In addition, the expression of immune enhancement-related cytokine genes (NOS, COX-2, IL-1β, IL-6 and TNF-α) were confirmed through PCR-electrophoresis. The results of this study suggest that Rhamnaceae extracts can be used as natural antioxidants and immune enhancer.

  • A hight performance liquid chromatographic methods was developed for the seperation and determination of seven alkaloids in "sanjoin" (the seeds of Zizyphus jujuba Rhamnaceae), a plant with potent sedative activity. A reverse phase system of Lichrosorb RP-Select B column and 0.05 M potassium phosphate buffer (pH = 3.5)-acetonitrile with gradient elution was employed. Two known alkaloids, juzirine and lysicamine, were newly isolated fom "sanjoin"."sanjoin".oin&".ot;.

  • The alkaloids of thirty five Korean plants which belong to Rhamnaceae, Sterculiaceae, Celastracea, Urticaceae and Rubiaceae were screened. Plant was extracted and solvent fractionated to yield ether soluble alkaloidal fraction and butanol soluble fraction. Each fraction was subjected to alkaloidal test by Dragendorff's reaction. The result showed eighteen plants strong positive, fourteen weak and six negative to alkaloidal test.

Antioxidant and immunoregulatory effects of Korean Rhamnaceae (국내 자생 갈매나무과 식물의 항산화 및 면역반응조절 효과 탐색)

  • We studied the antioxidative and immunoregulatory properties of 70% ethanol extracts from the leaves, branches, and fruit of 13 species of the Rhamnaceae collected during the bearing season (Berchemia berchemiifolia, Berchemia floribunda, Hovenia dulcis, Paliurus ramosissimus, Rhamnella franguloides, Rhamnus crenata, Rhamnus davurica, Rhamnus koraiensis, Rhamnus parvifolia, Rhamnus ussuriensis, Rhamnus yoshinoi, Sageretia thea, and Ziziphus ujube). Total phenolic and flavonoid content values were determined using calibration curves of gallic acid and rutin, respectively. ABTS and DPPH radical scavenging assays were performed to determine antioxidative potential. Anti-inflammatory effects were studied using the nitric oxide (NO) inhibition assay in LPS-induced RAW264.7 cells. Extracts of Berchemia berchemiifolia, Sageretia thea, and Ziziphus jujuba had the highest total phenol and flavonoid content values. Extracts of Berchemia berchemiifolia, Paliurus ramosissimus, Rhamnella franguloides, and Sageretia thea had the highest ABTS radical scavenging activities. Berchemia berchemiifolia and Sageretia thea had the highest DPPH radical scavenging activities. Furthermore, extracts of Berchemia berchemiifolia, Berchemia floribunda, Paliurus ramosissimus, Sageretia thea, and Ziziphus jujuba showed strong anti-inflammatory activity through inhibition of NO production. The results of this study suggest that Rhamnaceae extracts can be used as natural antioxidants and immunomodulators.

  • Sedative activity of Zizyphi fructus was evaluated by potentiation of hexobarbital-induced hypnosis test and its active principles have been characterized as nornuciferine and lysicamine. A new cyclopeptide alkaloid, daechucyclopeptide-1 was isolated together with zizyphusine.

  • As a series of the studies on the specific and indigenous plants of Mt. Halla, the constituents from stem of Rhamnus taquetii (Rhamnaceae) were investigated. From the water fraction of the MeOH extract, a new flavonol glycoside, kaempferide-3-O-${\alpha}$-L-rhamnopyranosyl(1->3)-${\alpha}$-L-rhamnopyranosyl(1-6)-${\beta}$-D-galactopyranoside, was isolated with three known compounds, quercetin, quercetin-3-methylether, kaempferol, by column chromatographic separation using Amberlite XAD-2, ODS-gel and Sephadex LH-20 and elucidated physico-chemical evidences($^1H-NMR,\;^{13}C-NMR$,IR, EI-Mass, FAB-Mass, and G.C.),respectively.

  • From the seeds of Zizyphus vulgaris var. spinosus, aporphine alkaloids : nuciferine, N-methylasimilobine, nornuciferine, norisocorydine, caaverine and tetrahydro benzylisoquinoline alkaloid : (+) coclaurine were isolated and identified. Zyzyphusine, a new quaternary aporphinium alkaloid form butanol soluble fraction was isolated and characterized by spectral data.

  • Sedative principles of the seeds of Zizyphus vulgaris var. sphinosus have been characterized as sanjoinine-A (frangufoline), nuciferine and their congeners. Also, heat treatment of sanjoinine-A-produced a more active artifact, sanjoinine-Ahl, which provides a scientific basis for heat-processing (roasting) of this Oriental medicine.
