무지개송어의 바이러스성 출혈성 패혈증에 관한 기초적 연구 (A Basical Study on Viral Haemorrhagic Septicemia of Rainbow trout, Salmo gairdneri)

  • 무지개송어(Salmo gairdneri)의 바이러스성 출혈성 패혈증에 관하여 연구하였다. 병어의 hematocrit값은 정상어보다 매우 낮게 나타났으며, GOT와 GPT값은 정상어보다 병어에서 약간 높게 나타났다. 병어의 혈청과 장기의 마쇄액을 CHSE-214세포에 감염시켜 세포변성 효과를 조사한 결과 감염 24시간 후에 세포변성이 나타났다.

  • 광견병(rabies)은 국제수역사무국에서 지정한 동물전염병 분류 List B에 속하며, 전염병 코드 번호는 B 058이다. 본 병의 병명은 영어로는 , 불어로는 , 스페인어로는 로 표기하며, 특히 인체에서의 본병에 대하여는 로 표현한다. 우리말로는 동물에서의 질병을 <광견병>, 인체에서의 본병을 <공수병>으로 부르고 있다. 병인체는 Rhabdoviridae에 속하는 Lyssavirus이며, 최대 잠복 기간(maximum incubation period)은 6개월로 알려져 있다. 인수공통전염병(zoonosis)의 1종이다. 매개체(vector)로 작용하는 절지동물은 없는 것으로 알려져 있다. 광견병과 관련하여, 동물의 국제교역을 위한 동물검역 시행에 있어서 반드시 적용하여야 할 사항인 국제수역사무국(O.I.E) 제정 동물위생 규약(Zoo-sanitary Code)을 번역하여 소개한다.

Rabies immune status in the stray and companion dogs in Korea

  • Rabies virus (family Rhabdoviridae, genus Lyssavirus, RV) is the causative agent of rabies in mammals. We conducted a sero-epidemiological survey for RV using sera from South Korean stray and companion dogs in the present study. A total of 533 canine serum samples were collected between February 2006 and December 2007 and were screened for rabies immunity with a neutralizing peroxidase linked assay. Both companion (49.1%) and stray (60.1%) dogs demonstrated RV seropositivity. Regional RV antibody prevalence was measured in the Jeju (87.5%), Gyeonggi (62%), Gyeongsang (59.1%), Jeonra (42%), Chungcheong (37.9%), and Gangwon (30.4%) provinces. Prevalence increased with age but did not exceed 80% in any age group. Stray and companion dogs had RV antibody prevalence values of 26.7% and 23.7%, respectively. Seroprevalence was significantly associated with age $({\chi}2\;=\;9.46;\;p\;=\;0.024)$ for companion dogs, although this association was not evident in stray dogs. There were no significant differences in age between stray and companion dogs and no gender differences in RV seroprevalence. Our results suggested that a widespread and reinforced vaccination program must be applied to Korean dogs.

동아시아 지역에 있어 전염성조혈기괴사증 바이러스 (IHNV)의 분자역학 및 병독성의 변화 (Molecular epidemiology and virulence changes of infectious hematopoietic necrosis virus (IHNV) in East Asia)

  • Causative agent of infectious hematopoietic necrosis (IHN) belonging to genus Novirhabdovirus (Rhabdoviridae). Economic losses caused by IHNV are serious in mainly Oncorhynchus spp. including rainbow trout O. mykiss and Atrantic salmon Salmo salar. IHNV was initially found by endemic presence in U.S. West Coast for sockeye salmon fry O. nerka and chinook salmon fry O. tshawytscha in the 1950s, and it has spread to Japan, Korea and Taiwan in the 1970s, and also to Italy and France in the 1990s. Currently, IHNV is detectable in many parts of the world, including Russia and South America. Mortality due to IHNV infection in fish with ${\leq}0.5g$ of body weight reaches 60% to 100%, while the mortality reduces by fish growing. In recent years, onset of IHNV infection has increased also in fish with large sizes. Here, we introduce molecular epidemiology and virulence changes of IHNV in East Asia, furthermore, we discuss on future prospects in IHNV researches.

Molecular Characterization and Survey of the Infection Rate of Orchid fleck virus in Commercial Orchids

  • Orchid fleck virus (OFV) is an unassigned plant virus in the family Rhabdoviridae. OFV was isolated from Cymbidium sp. showing oval necrotic lesions on their leaves in Korea, and designated as OFV-NHHS1. The complete nucleotide sequence of the RNA1 (6,413 nt) (GenBank accession no. AB516442) and RNA2 (6,001 nt) (GenBank accession no. AB516441) was determined in this study. RNA1 and RNA2 contained five and one ORF respectively. RNA1 encodes nucleocapsid (N) of 49 kDa, ORF2 of 26 kDa, ORF3 of 38 kDa, ORF4 of 20 kDa and glycoprotein (G) of 61 kDa proteins, whereas RNA2 encodes a single polymerase of 212 kDa. OFV-NHHS1 shared extremely high similarity of 98.6-100% and 98.9-99.6% in nucleotidle and amino acid sequences with a Japanese isolate, OFV-so, respectively. However, the N, G and L of OFV-NHHS1 revealed 6.9-19.3%, 7.3-12.0%, and 13.4-26.6% identities to those of 29 Rhabdoviruses, respectively. To survey the infection rate of OFV in commercial orchids in Korea, 51 Cymbidium sp., 10 Phalaenopsis sp., 22 Oncidium sp. and 21 Dendrobium sp. plants that showed typical viral symptoms were collected. RT-PCR with specific primers for detection of Cymbidium mosaic virus (CymMV), ORSV and OFV showed high infection rate by ORSV alone and double infection by ORSV and CymMV. One of the orchids tested was infected with OFV. This is the first report of the complete nucleotide sequences of OFV isolated in Korea.

광견병바이러스에 대한 단크론항체 생산 및 특성 (Production and characterization of monoclonal antibodies against rabies virus)

  • Rabies virus which belongs to the genus Lyssavirus of the family Rhabdoviridae is known as a highly neurotropic virus and causes fatal encephalitis accompanied by severe neurological symptoms in almost all mammals, including humans. In this study, monoclonal antibodies (MAbs) against rabies virus were produced, characterized and applications of MAbs as diagnostic reagents were assessed Spleen and inguinal lymph node cells from Balb/c mouse immunized with purified rabies virus were fused with SP2/O myeloma cells using polyethylene glycol (PEG) and hybridoma cells producing rabies virus-specific MAbs were screened by an indirect fluorescent antibody test. A total of ten MAbs were produced against rabies virus. The protein specificity and neutralizing activity of MAbs were determined by Western blot analysis and fluorescent antibody virus neutralization test, respectively. As a result, two MAbs, 5G3 and 6H4 had specificity for nucleoprotein (N protein) and two other MAbs, 5B1 and 5C1 had neutralizing activity for rabies virus. Some MAbs recognized the rabies virus-infected bovine brain stem cells by immunohistochemistry (IHC) assay. In conclusion, it was confirmed that MAbs produced in this study were rabies virusspecific and could be used as reliable diagnostic reagents for the detection of rabies virus.

  • 바이러스성 출혈성 패혈증 바이러스(VHSV)는 넙치를 포함한 어류 양식의 막대한 피해를 일으키는 바이러스 병원체이며, VHSV가 생성하는 6개의 바이러스 단백질들 중에서 NV 단백질이 병원성에 관여하는 것으로 알려져 있다. VHSV-감염 넙치를 이용한 전사체 마이크로 어레이의 선행 분석 결과에 의하면 VHSV 감염이 해당과정 효소들의 mRNA 발현을 억제함으로써 넙치 세포에서 ATP 생성을 감소시켰음을 알 수 있었다. 이들 결과를 토대로, 본 연구에서는 VHSV NV 단백질이 해당과정 효소인 glucokinase의 발현에 미치는 영향을 검토하였다. 본 연구의 결과에 의하면, NV 단백질은 넙치 세포에서 glucokinase의 mRNA 발현을 감소시켰으며, 새롭게 동정한 glucokinase의 유전자 프로모터의 활성 실험결과, NV 단백질이 glucokinase의 프로모터 활성을 저해함을 알 수 있었다. 이와 같은 작용 결과들로 인하여 VHSV NV 단백질의 발현이 세포 내로의 포도당 흡수 또한 감소시켰다. 이러한 결과들은 VHSV NV 단백질이 유전자 발현의 전사 수준에서 음성적으로 해당과정의 효소 발현을 조절함을 의미하며, 결국 세포 내 에너지의 결핍으로 넙치의 폐사로 이어질 가능성을 보여주는 것이다.
