A STUDY ON METHODOLOGY FOR IDENTIFYING CORRELATIONS BETWEEN LERF AND EARLY FATALITY

  • The correlations between Large Early Release Frequency (LERF) and Early Fatality need to be investigated for risk-informed application and regulation. In Regulatory Guide (RG) -1.174, while there are decision-making criteria using the measures of Core Damage Frequency (CDF) and LERF, there are no specific criteria on LERF. Since there are both huge uncertainties and large costs needed in off-site consequence calculation, a LERF assessment methodology needs to be developed, and its correlation factor needs to be identified, for risk-informed decision-making. A new method for estimating off-site consequence has been presented and performed for assessing health effects caused by radioisotopes released from severe accidents of nuclear power plants in this study. The MACCS2 code is used for validating the source term quantitatively regarding health effects, depending on the release characteristics of radioisotopes during severe accidents. This study developed a method for identifying correlations between LERF and Early Fatality and validates the results of the model using the MACCS2 code. The results of this study may contribute to defining LERF and finding a measure for risk-informed regulations and risk-informed decision-making.

Quantitative Analysis of Hyangsayukgunja-Tang Using an Ultra-Performance Liquid Chromatography Coupled to Electrospray Ionization Tandem Mass Spectrometry (LC-MS/MS를 이용한 향사육군자탕의 주요성분의 함량분석)

  • The aim of this study was to quantitatively analyze for quality assessment of eighteen marker compounds, including homogentisic acid, 3,4-dihydroxybenzaldehyde, spinosin, liquiritin, hesperidin, ginsenoside Rg1, liquiritigenin, ginsenoside Rb1, glycyrrhizin, 6-gingerol, atractylenolide III, honokiol, costunolide, dehydrocostuslactone, atractylenolide II, nootkatone, magnolol, and atractylenolide I, in Hyangsayukgunja-tang using an ultra-performance liquid chromatography-electrospray ionization-mass spectrometer. The column for separation of eighteen marker components were used a UPLC BEH $C_{18}$ analytical column ($2.1{\times}100mm$, $1.7{\mu}$) and kept at $45^{\circ}C$ by gradient elution with 0.1% (v/v) formic acid in water and acetonitrile as mobile phase. The flow rate and injection volume were 0.3 mL/min and $2.0{\mu}l$, respectively. The correlation coefficient of all marker compounds was ${\geq}0.9914$, which means good linearity, within the test ranges. The limits of detection and quantification values of the all analytes were in the ranges 0.04-1.11 and 0.13-3.33 ng/mL, respectively. As a result, five compounds, homogentisic acid, 3,4-dihydroxybenzaldehyde, spinosin, liquiritigenin, and atractylenolide I, in this sample were not detected and the amounts of the 13 compounds except for the 5 compounds were $8.10-6736.37{\mu}g/g$ in Hyangsayukgunja-tang extract.

Extraction of Red Ginseng Extract by Impulse Vacuun System (Impulse-Vacuum System을 이용한 홍삼엑스의 추출)

  • This study was carried out to establish the extraction method of red ginseng extract without saponin decomposition. Red ginseng was extracted with impulse vacuum system and multi-stage extraction method. Crude saponin content of red ginseng extract (RGE) from impulse vacuum system was 5.4-5.9%, while that of RGE from multi-stage extraction method was 8.2-8.3%. However, HPLC Patterns indicated that saponins of RGE from impulse vacuum system were hardly decomposed, while those of RGE from multi-stage extraction method were decomposed, especially in ginsenoside -Rgl and -Re saponin. Also, the yields of red ginseng by impulse vacuum system were 15 to 20 times higher than that of multi-stage extraction method.

Verification of Effectiveness of Wearing Compression Pants in Wearable Robot Based on Bio-signals (생체신호에 기반한 웨어러블 로봇 내 부분 압박 바지 착용 시 효과 검증)

  • In this study, the effect of wearing functional compression pants is verified using a lower-limb wearable robot through a bio-signal analysis and subjective fit evaluation. First, the compression area to be applied to the functional compression pants is derived using the quad method for nine men in their 20s. Subsequently, functional compression pants are prepared, and changes in Electroencephalogram (EEG) and Electrocardiogram (ECG) signals when wearing the functional compression and normal regular pants inside a wearable robot are measured. The EEG and ECG signals are measured with eyes closed and open. Results indicate that the Relative alpha (RA) and Relative gamma wave (RG) of the EEG signal differ significantly, resulting in increased stability and reduced anxiety and stress when wearing the functional compression pants. Furthermore, the ECG analysis results indicate statistically significant differences in the Low frequency (LF)/High frequency (HF) index, which reflect the overall balance of the autonomic nervous system and can be interpreted as feeling comfortable and balanced when wearing the functional compression pants. Moreover, subjective sense is discovered to be effective in assessing wear fit, ease of movement, skin friction, and wear comfort when wearing the functional compression pants.

  • Ginseng, whichhas long been used for its medicinal properties, has recently been investigated by scientific research to identify its components and evaluate its efficacy. Recently, two components of ginseng, Rg3 and compound K, have been attracting attention and various functional materials containing these materials have been developed and investigated. In this study, compound K wasproduced using yeast (Saccharomyces servazzii, GB-07) and To be used for industrialization. The antioxidant and anti-inflammatory activity of compound K (containing 20 g/g) ginseng fermented extract was investigated. In the fermented ginseng extract, the free radical DPPH was scavenged in a concentration-dependent manner and the production of ROS was inhibited in macrophages(RAW 264.7 cell). Moreover, the LPS-induced inflammatory cytokines $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$ were suppressed. These results suggest the possibility of industrialization viathe development of products containing compound K through future process development and various efficacy tests.

Bio-antimutagenic effects of water extract from Rehmannia glutinosa Liboschitz in SOS Chromotest (SOS Chromotest에서 숙지황 물 추출물의 세포내 항돌연변이 효과)

  • The antimutagenic activity of the water extract of Rehmannia glutinosa Liboschitz (RG) on the mutagenicity induced by 4-nitroquinoline 1-oxide (4-NQO), N-methyl-N'-nitro-N-nitrosoguanidine (MNNG), mitomycin C (MMC), $aflatoxin\;B_1\;(AFB_1)$ and benzo(a)pyrene [B(a)P] were studied using the SOS Chromotest with Escherichia coli PQ37. The water extract of RG was separated into methanol soluble and methanol insoluble parts. The methanol soluble part exhibited higher inhibition effects than the methanol insoluble part against the mutagenic activities of five mutagens. Step-wise fractionation of methanol soluble part was done using methanol, ethyl acetate and water. Among these fractions, water fraction had the strongest inhibitory effects against the mutagenenicity of five model mutagens, showing $4.5{\sim}29.5%$ inhibition, but the $AFB_1$ mutagenic potency was increased slightly by ethyl acetate fraction. The water fraction was further partitioned by sephadex LH-20 column chromtography, and 9 subfractions were obtained. The fraction III showed the strongest inhibitory effects with dose response against the mutagenic activities induced by all the tested chemical mutagens. The inhibition rates of fraction III at concentration of $400\;{\mu}g/assay$ were 29%, 35%, 38%, 25% and 24% against 4-NQO, MNNG, MMC, AFBl and B(a)P, respectively. The fraction III also exhibited a strong bio-an-timutagenicity against 4-NQO and $AFB_1$ by showing more than 40% inhibition.

Steaming and Ultrasonic extraction conditions for enhancing the ginsenoside contents and anti-aging efficacy of the Ginseng sprout leaf/stem (새싹삼 잎 줄기의 진세노사이드 함량 및 항노화 효능 증진을 위한 증숙 및 추출조건)

  • 인삼은 우리나라에서 오랜 역사동안 많은 연구가 진행되어 왔으며, 현재는 다양한 방법으로 홍삼과 흑삼으로 만들어 식품, 화장품, 의약품 등 다양한 방면으로 사용하고 있다. 본 연구에서 시중에서 구매한 새싹삼(인삼새싹) 잎/줄기에 함유된 진세노사이드(Re, Rg1, Rb1, Rg3, Rh1) 함량을 높이기 위하여 증숙과 초음파 추출조건에 관한 연구를 수행하여 우수한 항노화 소재를 개발하기 위하여 실시하였다. 실험은 새싹삼 잎/줄기를 증숙 온도와 시간의 조건에서 진세노사이드 함량이 가장 높은 조건을 선정하였으며, 선정된 조건의 새싹삼 잎/줄기에 파장과 출력에 대한 조건으로 초음파 추출을 진행하여 진세노사이드가 가장 높은 함량을 보이는 조건을 선정하였다. 그 결과 새싹삼 잎/줄기추출물(GSE; Ginseng Sprout Extract)의 진세노사이드 함량은 4.8 mg/g으로 확인되었으나 증숙공정을 통해 8.82 mg/g으로 함량이 증가되었으며, 상기 증숙된 새싹삼 잎/줄기에 초음파공정을 적용하여 추출한 새싹삼 잎/줄기초음파추출물(SU-GSE; Steaming & dry Ultrasonication-Ginseng Sprout Extract)에서는 최대 10.65 mg/g으로 함량이 증가되었다. 반면, 새싹삼 뿌리의 진세노사이드는 2.30 mg/g으로 확인되었으나 증숙공정을 통해 4.95 mg/g으로 함량이 증가되었으며, 초음파추출공정을 통해 최대 5.82 mg/g으로 함량이 증가된 것을 확인할 수 있었으나, 새싹삼 잎/줄기에 비해 진세노사이드 함량이 낮은 것을 확인하였다. 항노화 소재로의 활용가능성을 평가하기 위하여 새싹삼 잎/줄기추출물 GSE와 SU-GSE에 대한 세포생존률, 항산화 및 항노화에 대한 효능평가를 진행하였으며 GSE의 경우 $100{\mu}g/ml$에서 세포생존률이 82.4%를 보인 반면 SU-GSE에서는 $1,000{\mu}g/ml$의 농도에서 101.8%의 세포 생존률을 보였다. 항산화 활성의 경우 GSE와 SU-GSE $100{\mu}g/ml$ 농도에서 각각 52%와 81%의 항산화 활성을 나타냄으로써 SU-GES의 조건에서 항산화 활성이 우수한 것으로 확인되었다. 또한, 항노화 활성에 대한 실험결과 MMP-1 유전자 발현에 대한 억제율을 비교한 결과 GSE와 SU-GES $100{\mu}g/ml$의 농도에서 각각 18%와 29%의 억제율을 보임에 항노화 소재로의 활용가능성을 확인하였다.

  • The revised U.S. Nuclear Regulatory Commission(NRC), Regulatory Guide(RG) 1.20, rev.3 requires the evaluation of the potential adverse effects from pressure fluctuations and vibrations on piping and components for the reactor coolant, steam, feedwater, and condensate systems. Detailed vibration analyses for the systems attached to the steam generator are very difficult, because these piping systems are very complicated. This paper suggests a screening method for the flow-induced vibration of acoustic resonances and pump-induced vibration of the piping systems attached to the steam generator in order to conduct the APR1400 comprehensive vibration assessment program. This paper seeks to address the areas such as potential vibration sources, and methods to prevent the occurrence of acoustic resonances and pump-induced vibration of piping systems attached to the steam generator, for conducting the APR1400 comprehensive vibration assessment program. The screening method in this paper will be used to estimate the flow-induced vibration of the piping systems attached to the steam generator for the APR1400.

Induction of Forkhead Class box O3a and apoptosis by a standardized ginsenoside formulation, KG-135, is potentiated by autophagy blockade in A549 human lung cancer cells

  • Background: KG-135, a standardized formulation enriched with Rk1, Rg3, and Rg5 ginsenosides, has been shown to inhibit various types of cancer cells; however, the underlying mechanisms are not fully understood. In this study, we explored its effects in A549 human lung cancer cells to investigate the induction of Forkhead Class box O3a (FOXO3a) and autophagy. Methods: Cell viability was determined by sulforhodamine B staining. Apoptosis and cell cycle distribution were analyzed using flow cytometry. The changes of protein levels were determined using Western blot analysis. Autophagy induction was monitored by the formation of acidic vesicular organelles stained with acridine orange. Results: KG-135 effectively arrested the cells in G1 phase with limited apoptosis. Accordingly, a decrease of cyclin-dependent kinase-4, cyclin-dependent kinase-6, cyclin D1, and phospho-retinoblastoma protein, and an increase of p27 and p18 proteins were observed. Intriguingly, KG-135 increased the tumor suppressor FOXO3a and induced the accumulation of autophagy hallmark LC3-II and acidic vesicular organelles without an increase of the upstream marker Beclin-1. Unconventionally, the autophagy adaptor protein p62 (sequestosome 1) was increased rather than decreased. Blockade of autophagy by hydroxychloroquine dramatically potentiated KG-135-induced FOXO3a and its downstream (FasL) ligand accompanied by the cleavage of caspase-8. Meanwhile, the decrease of Bcl-2 and survivin, as well as the cleavage of caspase-9, were also drastically enhanced, resulting in massive apoptosis. Conclusion: Besides arresting the cells in G1 phase, KG-135 increased FOXO3a and induced an unconventional autophagy in A549 cells. Both the KG-135-activated extrinsic FOXO3a/FasL/caspase-8 and intrinsic caspase-9 apoptotic pathways were potentiated by blockade of autophagy. Combination of KG-135 and autophagy inhibitor may be a novel strategy as an integrative treatment for cancers.

Screening of Antimicrobial Activity Compounds from Korea Ginseng Fine Root (고려인삼의 세근을 이용한 항균성 물질 탐색)

  • The study was performed to evaluate the antibacterial and antiviral activities of ginseng fine root in order to search for antibacterial substances. Among 8 kinds of fermentation strains, Lactobacillus plantarum was selected based on viable cell count and antibacterial activities during incubation. Optimum conditions of ginseng fine root fermentation for L. plantarum were incubation at $35^{\circ}C$ for 48 hr in 5% ginseng fine root broth. That methanolic extract of fermented ginseng fine root broth was observed to be antibacterial and have antiviral activities. The results of paper disc method of non-fermented extract and fermented extract measured against E. coli was 11 mm and 20 mm, S. aureus was 15 mm and 22 mm, respectively. Shaking flask method was observed to inhibit the growth E. coli and S. aureus in fermented extract by 99.9%. However, antiviral activity of Feline calicivirus (FCV) was mostly activated. Fermented extract was used to investigate the compositional changes of ginsenosides on HPLC analysis. By fermentation, ginsenoside Rg1, Re and Rd were increased, with Rd showing a significant increase of 50 ${\mu}g/g$. These results suggest that ginseng fine root extract is a useful resource.
