Effect of Temperature and Salinity on Production of Resting Egg in Korean Rotifer, Brachionus plicatilis (L and S-type)

  • Production of resting egg from the Korean rotifer, Brachiunus plicatilis (L and S-type) was investigated at different temperatures (L-type : 20, 24, $28^{\circ}C$, S-type : 28 32, $36^{\circ}C$) and salinities (10, 20,30 ppt). The rotifer was cultured in 25 ml test tube and fed on Nannochloris oculata. With regard to mixis rate, L-type rotifer showed higher rate at lower temperature, and the highest rate was observed at 20 ppt of salinity at each temperature of the experiment. However, for S-type rotifer, the optimum temperature and salinity were $28\~32^{\circ}C$ and 20 ppt, respectively. The highest number of resting egg was 173 eggs/ml in 16 days at $24^{\circ}C$, 10 ppt for L-type rotifer and 410 eggs/ml in 14 days at $28^{\circ}C$, 10 ppt for S-type rotifer. The maximum number of resting egg produced per 10,000 rotifers was 8,122 eggs at $20^{\circ}C$, 20 ppt for L-type rotifer and 8,700 eggs at $28^{\circ}C$, 20 ppt for S-type rotifer. The maximum number of resting egg produced $10^8$ cells of N. oculata was 50.7 eggs for L-type rotifer ($24^{\circ}C$, 20 ppt) and 79.6 eggs for S-type rotifer ($32^{\circ}C$, 10 ppt). The number of resting egg produced per day was $1\~11$ eggs/ml for L-type rotifers and $21\~35$ eggs/ml for S-type rotifer in 9 combination experiments. In this study, S-type rotifer is better than L-type rotifer in resting egg production, and the optimum temperature and salinity for resting egg production were $20^{\circ}C$, 20 ppt for L-type rotifer and $28^{\circ}C$, 20 ppt for S-type rotifer. This result shows the difference of Korean rotifer in the optimum condition for resting egg production from other rotifers reported earlier.

  • The effect of temperature ($24\~36^{\circ}C$ and salinities ($5\~30$ ppt) on hatching rate of the resting egg of the Korean rotifer, Brachionus plicatilis was investigated. The highest hatching rate of the resting egg was $85.7\%$ at $28^{\circ}C$ and 15 ppt. The hatching rates of the resting eggs with salinities were not significant at $326{\circ}C$ and $36^{\circ}C$. Hatching was faster at higher temperature. The resting eggs stored at $5^{\circ}C$ in dark hatched simultaneously within two days, however, those at $28^{\circ}C$ in dark hatched intermittently within 14 days. But, final hatching rate of the resting egg at $28^{\circ}C$ was higher at $28^{\circ}C$ than that at $5^{\circ}C$. With regard to drying temperature and time of resting egg, the resting egg dried at $30^{\circ}C$ for 1 hour showed the highest hatching rate ($46.4\%$).

The Resting Eggs of Marine Cladocerans in the Intertidal Sea-bottom Sediments of Gomso Bay, Korea: Distribution and Evidence of Egg Banks

  • We investigated the distribution of the marine cladoceran resting eggs in the intertidal sediments of Gomso Bay, Korea and compared the data with those in the subtidal sediments. The abundance of the eggs in the top 6cm of the sediments was higher than that in the other depths. The abundances of the eggs in seaward intertidal sediments below mean sea level (MSL) were higher than those in the sediments over MSL, but were not significantly different from those in the subtidal sediments. The distribution of the resting eggs of cladocerans in the intertidal sediments was affected by the grain-size and moisture content of sediments. The results of the present study suggest that the intertidal sediments as well as the subtidal sediments are a potential egg bank which plays important roles in population dynamics of zooplankton in coastal waters, in particular, recruitment of eggs into plankton.

Mass Production of Resting Egg of Korean Rotifer, Brachionus plicatilis (L and S-type) (한국산 Rotifer, Brachionus plicatilis (L과 S-type)의 내구란 대량생산)

  • Mass production of resting egg of the rotifer, Brachionus plicatilis was carried out in 1 $m^3$ tank for L-type rotifer and in 1 $m^3$ and 4 $m^3$ tank for S-type rotifer. L-type rotifer was fed on concentrated Chlorella + baker's yeast and only baker's yeast for 15 days. S-type rotifer was fed on concentrated Chlorella in 1 $m^3$ tank for 7 days and frozen Chlorella + baker's yeast in 4 $m^3$ for $6\~8$ days. Total number of resting egg and number of resting egg from $10^8$ rotifers and 1 g dry weight diet for L-type rotifer fed on Chlorella + baker's yeast were $54.5{\times}10^6$ eggs, $30.5{\times}10^6$ eggs and $100{\times}10^3$ eggs, respectively. These were higher than those of rotifer fed on baker's yeast only. Total number of resting egg and number of resting egg from $10^8$ rotifers and 1 g dry weight diet for S-type rotifer fed on concentrated Chlorella in 1 $m^3$ tank were $50\~104{\times}10^6$ eggs, $47\~82{\times}10^6$ eggs and $136\~260{\times}10^3$ eggs, respectively. In 4 $m^3$ tank with frozen Chlorella + baker's yeast, these were $149\~567{\times}10^6$ eggs, $36\~123{\times}10^6$ eggs and $131\~338{\times}10^3$ eggs, respectively. This result suggests that S-type rotifer are better than L-type rotifer for the mass production of resting egg of the Korean rotifer.

Survival Strategies of the Rotifer Brachionus rotundiformis for Coexisting with the Copepod Apocyclops borneoensis in Laboratory Culture

  • Interspecific relationship between a euryhaline rotifer Brachionus rotundiformis and a cyclopoid copepod Apocyclops borneoensis was investigated in the laboratory culture. In a mixed culture of B. rotundiformis and A. borneoensis, population growth of B. rotundiformis was suppressed from day 10, while growth in a monoculture population continuously increased throughout the experimental period. However, the population growth of A. borneoensis in the mixed culture did not markedly differ from that in a monoculture population. Suppression of B. rotundiformis growth coincided with a decrease in the numbers of both non-egg-bearing and egg-bearing females, and increasing resting egg formation. Growth of A. borneoensis was not affected by the presence of the rotifer. However, relative growth index of ovisac bearing females in the mixed culture was 1.62 times higher than that in the monoculture. Presence of the copepod did not greatly reduce the food available to the rotifer population. The rotifer B. rotundiformis responded in a unique way, to stresses such as physical damage (filtering by A. borneoensis) with the production of many resting eggs to increase its chances of survival.

Size and Resting Egg Formation of Korean Rotifer, Brachionus plicatilis and B. calyciflorus (한국산 Rotifer, Brachinus plicatilis와 B. calyciflorus의 크기 및 내구란 형성)

  • Sixteen strains of marine rotifer, Braohionus plicatilis were isolated from salt pond, estuary and lagoon. Among 16 strains, 2 strains were large (L)-type and the others were small (S) or ultra small (US)-type. Four strains of fresh water rotifer, B. calyciflorus were isolated from commercial fish ponds. The size of lorica and resting egg were measured. In B. plicatilis, the range of lorica length from S-type and S-type were $244.3{\~}255.3\;{\mu}m$ and $131.0{\~}165.8\;{\mu}m$, respectively. The major axis of resting egg in the marine rotifer were $93.7\~116.4\;{\mu}m$ for S-type and $142.4{\~}145.5\;{mu}m$ for L-type, respectively. In freshwater rotifer, B. calyciflorus, the size range of lorica and major axis of resting egg were $211.8\~229.9\;{\mu}m$ and $126.8\~140.2\;{mu}m$, respectively. The size of freshwater rotifer was larger than that of S-type marine rotifer, but smaller than that of L-type one. Growth and formation of resting egg of B. plicatilis were different among the strains. The maximum density of S-type and L-type rotifer was 753.3 inds./ml for H-S strain and 220 inds./ml for O-L strain, respectively. The largest production of resting egg of S-type and L-type rotifer were 86.7 inds./ml for YY-S strain and 45.8 inds./ml for O-L strain, respectively.

Production and Hatching Rate of Resing Egg of Korean Rotifer, Brachionus plicatilis (S-type) with Different Diets (먹이에 따른 한국산 S-tyoe Rotifer, Brachionus plicatilis의 내구란 생산과 부화율)

  • Dietary values of phytoplanktons, concentrated Chlorella and commercial yeasts were investigated for the resting egg production of the Korean rotifer, B. plioatilis S-type. The hatching rate of the resting egg of the rotifer fed on different diets was also studied. The highest production of the resting egg was 3,760 eggs/20 ml with N. oculata. However, the hatching rate was not significant among 4 phytoplankton species. The highest production of resting eggs among 9 diet groups ($\omega$-yeast, baker's yeast, 2 refrigerated Marine Chlorella, frozen Marine Chlorella, frozen Chlorella ($70\%$) + baker's yeast ($30\%$), frozen Chlorella ($30\%$) + baker's yeast ($70\%$), 2 refrigerated freshwater Chiorella) was 283 eggs/ml in the frozen Chlorella ($30\%$) +baker's yeast ($70\%$) and the refrigerated freshwater Chlorella. The highest number of resting egg from 10,000 rotifers and 1 mg dry wieght diet was 5,566 eggs and 2,131 eggs in the frozen Chlorella ($30\%$) + baker's yeast ($70\%$). However, the highest hatching rate of the resting eggs was $67.4\%$ in $\omega$-yeast. In this study, the results suggest that the baker's yeast with a small amount of frozen Chlorella seems to be good feeding regime for the economical production of resting eggs.

Growth and Production of Resting Eggs of Freshwater Rotifer, Brachionus calyciflorus Pallas at the Different Temperatures (수온에 따른 담수산 Rotifer, Brachionus calyciflorus Pallas의 성장과 내구란 생산)

  • The specific growth rate and production of resting ega of the freshwater rotifer, Brachionus calyciflorus Pallas was investigated at the different temperatures ($20\~30^{\circ}C$). The rotifer was cultured in 250 ml flask and fed on concentrated freshwater Chlorella. Specific growth rate of B. calyciflorus showed higher rate at higher temperature, but maximum density was observed higher at lower temperature, expect at $20^{\circ}C$. The production of resting egg with temperature was showed decrease on the basis on $26^{\circ}C$. The highest number of resting egg per ml and rotifer $10^4$ were 157 eggs and 810 eggs at $26^{\circ}C$, respectively. This result shows that the optimum temperature for mass culture and resting egg production of this freshwater rotifer were $30^{\circ}C$ and $26^{\circ}C$, respectively.

  • This study was performed to investigate production and hatching rate of the resting egg of freshwater rotifer, Brachionus calyciflorus fed the 5 different diets (ESP, freshwater Chlorella, baker's yeast, the mixture off$70{\%}\;ESP\;and\;30{\%}$freshwater Chlorelia, and the mixture of $30{\%}$ baker's yeast and $70{\%}$ freshwater Chlorella). The highest productions of resting eggs were 123.3 egg/ml and 126.7 egg/ml in the mixture of $70{\%}$ ESP and $30{\%}$ freshwater Chlorella, and the mixture of $30{\%}$ baker's yeast and $30{\%}$ and $70{\%}$ freshwater Chlorella, respectively, which were significantly better than ESP, freshwater Chlorella or baker's yeast alone (P<0.05). Hatching rate of the resting eggs from rotifer fed the mixture of $70{\%} ESP\;and\;30{\%}$ freshwater Chlorella was the highest, $71.9{\%}$ although this value was not significantly different compared with the mixture of $30{\%}$ baker's yeast and $70{\%}$ freshwater Chlorella, and freshwater Chlorella (P^lt;9.05). The results showed that the mixture of baker's yeast or ESP with the freshwater Chlorella seemed to be the best feeding regime for the production and hatching rate of resting egg of freshwater rotifer.

Resting Egg Production of Six Strains of Korean Rotifer, Brachionus plicatilis (S-type) (염분에 따른 한국산 Rotifer, Brachionus plicatilis (S-type) 6 strains의 내구란 생산)

  • Six strains (OKK, MOK, CHA, WON, 505 & PUA) of Korean rotifer, Brachionus plicatilis (S-type) were isolated from salt pond, and the resting egg production of these strains was investigated with the different salinities (10, 20, 30 put), Rotifer were cultured at $28^{\circ}C$ and 2,000 lux in 20 ml test tube with feeding Nannoohloris oculata. The maximum rotifer density was 2,050 inds./ml at 10 ppt for OKK strain, and MOK strain showed the highest specific growth rate (1.028) at 10 ppt. Mixis rate of CHA and WON strains increased with the lower salinity, while MOK strain increased the rate with salinity. PUA strain did not show the mixis rate even at the high rotifer density, and OKK strain showed the very low mixis rate ranging from 1.1 to $3.0\%$. Fertilization rate of CHA, MOK, SOS and WON strains was ranged from 10.4 to $68.8\%$, and OKK strain did not showed any fertilization rate. The highest production of resting egg in 20 ml test tube was 4,065 eggs at 10 ppt for WON strain. The results may suggest that the selection of rotifer strain and salinity are important factors for the mass production of resting egg.
