The Effects of Gyumsuyukgunjun on the injured Tracheal tissue induced $SO_2$ in Rats (김수육군전(金水六君煎)이 $SO_2$에 의한 흰쥐의 호흡기조직손상(呼吸器組織損傷)에 미치는 영향(影響))

  • This experiment was performed in order to study the effects of Gyumsuyukgunjun on the injured tracheal tissue induced $SO_2$ in rats. Healthy adult male rats weighting about 250g were divided into 4 groups - the Normal group, the Control group, the group of Gyumsuyukgunjun administration for 5 days after $SO_2$ gas exposure (Sample I), and the group of Gyurnsuyukgunjun administration for 10 days before and for 5 days after $SO_2$ gas exposure (Sample II). The results were obtained as follows ; 1. In the trachea Control group, the lesion of the ciliated epithelium was severe and the mucus secretion of the respiratory tract was increased significantly. 2. In the trachea of Sample I group, the lesion of the ciliated epithelium and the mucus secretion of the respiratory tract were decreased compared with Control group. 3. In the trachea of Sample II group, the lesion of the ciliated epithelium and the mucus secretion of the respiratory tract were decreased compared with Control and Sample I group. According to the above results, Gyumsuyukgnjun has significant effects on the injuried tracheal tissue caused by $SO_2$ in rats.

The Effects of Acute Osmotic Stress on Innate Immunity of Nile Tilapia (Oreochromis niloticus)

  • The effects of osmotic stress on the non-specific immune response of Nile tilapia, Oreochromis niloticus, were investigated. Osmoregulatory mechanism of tilapia has been studied, but less information is available about innate immune response of O. niloticus faced with hyperosmolality. Acute osmotic stress was elicited by transferring tilapia from freshwater (FW) to 24 psu seawater (SW). Non-specific immune parameters including lysozyme activities of plasma and head kidney (HK), alternative complement pathway (ACP) activity in plasma, phagocytic capacities of spleen and HK immune cells, and respiratory burst activity of immune cells in both HK and spleen were analyzed. Lysozyme activities were increased at 1 h and 30 h after transfer to SW, but decreased at 10 h after SW transfer. Conversely, ACP activity increased 10 h after SW transfer. Phagocytic capacity increased slightly at 1 h and 5 h after SW transfer, and respiratory burst activity showed an increase in superoxide release at 10 h after SW transfer. Taken together, these results indicate that the exposure of tilapia to hyperosmotic conditions has immunostimulatory effects on cellular and humoral immune reactions.

  • In order to investigate experimentally the clinical effects of Chungsimjeongchun-Tang that have been widely used for cardiac asthma, dyspnea and coughs secondary to the appoplexy and cardiac diseases, the water extract of Chungsimjeongchun-Tang was conducted to the effects on the anti-histamine action and respiratory system in mice, guinea-pigs, rabbits and cats. The following results of Chungsimieongchun-Tang were obtained. Spontaneous motility of the isolated ileum of mice and rabbits was strongly suppressed and contractions of the isolated ileum of mice and guinea-pigs induced by acetylcholine chloride, barium chloride and histamine were remarkably inhibited. Anti-histamine action was noted. Antitussive action on the mechanically irritative cough in cats was recognized. Expectorant action on the leakage of dye in the respiratory tract of rabbits was also shown.

Pathogenic effects of porcine reproductive and respiratory syndrome virus isolates in swine tracheal ring culture

  • Pathogenic effects of 29 different porcine reproductive and respiratory syndrome(PRRS) virus isolates were investigated in swine tracheal ring(STR) cultures by examining their effects on the ciliary activity of STR. Inhibition of ciliary movement and destruction of the tracheal epithelium were seen between 72 and 96 hours postinoculation(PI). Virus replication was demonstrated by examining viral infectivity of the supernatants from the STR cultures. PRRS virus antigen in macrophages was detected by a streptavidin-biotin complex(ABC) immunoperoxidase method. Of the 29 PRRS virus isolates, 8 isolates were classified into pathogenic, and the remaining 21 isolates were determined as mildly pathogenic or apathogenic viruses. These results suggest that STR examination may be used as a method for predicting pathogenic variability of PRRS virus isolates.

Effects of in vivo Synergism of Eunkyo-san, a Poly-Herbal Formula and Rufloxacin against Klebsiella pneumoniae Respiratory Infections (은교산(銀翹散)과 Rufloxacin의 병용투여(倂用投與)가 Klebsiella pneumoniae 호흡기감염(呼吸器感染)에 미치는 영향)

  • In order to evaluate the in vivo synergic effect of Eunkyo-san with quinolone antibiotics, rufloxacin (RUFX), the viable bacterial numbers and histopathological changes were monitored after experimental respiratory infection with Klebsiella peumoniae NCTC 9632. The obtained results were as follows : In RUFX group, the viable bacterial numbers were significantly decreased compared to those of control group and these were more dramatically decreased compared to those of single treatment with RUFX, respectively in concomitant treated groups with Eunkyo-san. In control group, severe infiltration of inflammatory cells, hemorrhage and hypertrophy of alveolar linings were demonstrated at microscopical levels. However, these abnormal histopathological changes were significantly decreased compared to those of control group in RUFX group, and these were more dramatically decreased compared to those of single treatment with RUFX, respectively in concomitant treated groups with Eunkyo-san. In RUFX group, the LSA% (luminal surface of alveolar%) were significantly increased compared to those of control group and these were more dramatically decreased compared to those of single treatment with RUFX, respectively in concomitant treated groups with Eunkyo-san. According to these results, it is considered that in vivo antibacterial activity of RUFX group was dramatically increased by concomitant use of Eunkyo-san against K. pneumoniae NCTC 9632 infection of respiratory tract.

Studying of the Effects of Gamichungsangboha-tang water-extract and ethanol-extract on $IFN{\gamma}$, IL-4, T-bet, GATA-3, c-maf in Asthma Model (가미청상보하탕(加味淸上補下湯)의 물과 에탄올 추출법에 따른 항(抗)알레르기 효과(效果)에 대(對)한 연구(硏究))

  • Objective: This study aimed to identify the different effects of GMCSBHT water-extract and ethanol-extract on Th1/Th2 differentiation by monitoring Th1/Th2 specific cytokine secretion patterns and the transcriptional activities of T-bet, GATA-3, c-maf, $INF{\gamma}$ and IL-4. Materials and Methods: Spleen cells from eight week-old BALB/c mice were cultured in GMCSBHT extracts containing medium without activation for 24 hours and with activation for 48 hours. CD4+ T cells were isolated and mRNA expression levels of $INF{\gamma}$, IL-4, T-bet, GATA-3, c-maf by RT-PCR and secretion cytokines levels of $IFN{\gamma}$, IL-4 by ELISA were analyzed. Results: GMCSBHT extracts didn't have mitogenic effects on the unstimulated CD4+ T cells. In Th1 skewed condition, GMCSBAHT water extract had no significant effects on mRNA expression levels of $IFN{\gamma}$, T-bet and c-maf, but inhibited mRNA expression levels of IL-4, GATA-3. It showed significantly increased secretion cytokine levels of $IFN{\gamma}$, but had no significant effect on secretion cytokine levels of IL-4. In Th2 skewed condition, GMCSBHT ethanol extract inhibited mRNA expression levels of $INF{\gamma}$, IL-4, GATA-3 and c-maf significantly, but had no significant effects on mRNA expression levels of T-bet. It had no significant effects on secretion cytokine levels of $INF{\gamma}$, but showed remarkable inhibitory effects on secretion cytokine levels of IL-4. Conclusion: Results suggest that on Th1/Th2 deviation, GMCSBHT water extract has both amplifying effects on Th1 differentiation and inhibitory effects on Th2, but GMCSBHT ethanol extract has stronger inhibitory effects on Th2 differentiation than on Th1.

Effects of Korean Medicine in Idiopathic Pulmonary Fibrosis Patients - A Retrospective Chart Review (한방병원에 내원한 특발성 폐섬유화증 환자 82명에 대한 임상적 특징 분석; 후향적 차트 리뷰)

  • Objective: This study was designed to analyze the clinical features of idiopathic pulmonary fibrosis patients who attended a Korean medicine hospital and the treatment effects through retrospective chart reviews. Methods: The medical records of 82 outpatients who had been diagnosed with idiopathic pulmonary fibrosis and visited the Allergy, Immune, and Respiratory System Department of Kyung Hee Korean Medicine Hospital from 8 January 2015 to 8 January 2021 were retrospectively reviewed. To assess the treatment outcomes, we used the FVC (Forced Vital Capacity), DLCO (Diffusing capacity of the Lung for CO), 6-minute walk test, and HRCT (High Resolution Computed Tomography). Results: The study group consisted of 28 females and 54 males. The median age of the patients was 67.98±11.44 years. The chief complaints were cough (n=51) and dyspnea (n=49). Other frequent symptoms were general weakness (n=8), weight loss (n=4), and a fever (n=2). A total of 77 (93.90%) patients were prescribed Korean herbal medicine, and 52 (63.41%) patients were treated with acupuncture, moxibustion, cupping therapy, ICT, or pharmacopuncture. After treatments, FVC, DLCO, the 6-minute walk test, and HRCT were maintained or worsened slightly. Conclusions: This study presented the characteristics of idiopathic pulmonary fibrosis patients treated by Korean medical therapies, and further studies of Korean medical treatments for idiopathic pulmonary fibrosis patients would be valuable.

The Effect of Chair Backrest on Respiratory Function in Prolonged Sitting Position

  • Purpose: The purpose of this study was to determine the effects of a chair backrest on respiratory function after prolonged sitting. Methods: Twenty-four young healthy subjects (12 males and 12 females) volunteered to participate in this study, and were equally allocated to a backrest (n=12) or a without backrest group (n=12). A spirometer was used to measure the respiratory functions of all subjects. Results: The chair with backrest group were significant difference in forced vital capacity (FVC), Forced expiratory volume in 1 second (FEV1), and peak expiratory flow (PEF) after sitting for 1 hour, compared with chair without backrest group (p<0.05). The chair with backrest group showed a significantly decreased in FVC, FEV1, and PEF. Conclusion: Using a chair without a backrest may help to reduce lung function deterioration as compared with a chair with a backrest.

  • In domestic rabbits, pasteurellosis is a primary respiratory disease entity caused by Pasteurella multocida. Respiratory pasteurellsos in rabbits is highly contagious due to dissemination of the organism in the nasal discharge (Benirschke, et al., 1978). The disease reflects a varied and complex host-parasite relationship. Acute infection with high mortality occurs sporadically especially in stressed rabbits less than 10 weeks old, in the form either an enzootic pneumonia, infection of the upper respiratory tract or septicemia. In addition to respiratory form, infection with P. multocida results in a variety of disease processes in rabbits including rhinitis, otitis media, otitis interna, abscesses, metritis, orchitis and meningoencephalaitis (Manning et al., 1989). The deleterious effects of pasteurellosis are common and pose serious problems for rabbit breeders and researchers (Flatt, 1974). (omitted)

Physiological effects of indole acetic acid (IAA) on chlorella ellipsoidea (Chlorella의 생리에 미치는 Indole acetic acid의 영향)

  • To study the effect of IAA on the growth of Chlorella, the alage wre cultured on the media for six days by bubbling $_{2}$ enriched air under 10K lux at 20-$25^{\circ}C$. The culture media were made by adding a concentration of $10^{-3}$M, $10^{-4}$M, and 0M(as a control) IAA to the standard media. During the period of culture, Chlorella was smapled for the given time of interval and photosynthetic and respiratory activities were measured by Warburg manometer and change of chemical components of Chlorella was determined by spectrophotometry after the Chlorella cell was fractionated by Schmidt-Thannhauser method. 1) Photosynthetic and respiratory activities were enhanced by IAA ; especially the enhancement of respiratory activity was so remarkable. 2) As to the chemical components of Chlorella, carbohydrates and amino acids were reduced a little but phosphate, RNA, DNA, and protein were increased by $10^{-3}$M IAA ; the increase of RNA, in particular, was noticable. 3) The above results suggest that the enhancement of growth of Chlorella, by IAA and ATP induced by respiratory activity accelerated with IAA enhanced RNA synthesis, resulting in an increase of protein synthesis.
