Restorer Genotype for Male Sterile Cytoplasm of Genetic Resources Moderately Resistant to Phytophthora capsici in Capsicum Pepper

  • KC00256, KC00406, KC00462, KC00463, KC00820, and KC00821, the genetic resources that have previously been reported as moderately resistant to Phytophthora capsici, as well as the line KC01322, a new source of moderate resistance introduced from Laos, were tested against two strains (Pc003 and Pc005) of P. capsici. We also determined the nuclear restorer genotypes of these lines, in regards to their interaction with cytoplasmic male sterility, through crossing the resources with cytoplasmic male sterile Punggok-A (Srfrf) and determining the fertility of the $F_1$ hybrids. The studied lines exhibited a low level of resistance to both the strains of P. capsici compared to highly resistant CM334, but their response was fairly consistent for both P. capsici strains. KC00406, KC00462, KC00463, and KC01322 produced stable, male fertile $F_1$ plants indicating that they are restorers with genotype N(S)RfRf. KC00821 produced male sterile $F_1$ plants and was identified as a maintainer with genotype Nrfrf. The $F_1$ plants of the KC00820 cross, however, set a few male fertile flowers in the greenhouse at seedling stage, then became male sterile after being transplanted to the plastic greenhouse soil in May and remained so to the end of the growing season. Therefore, KC00820 is an unstable maintainer with genotype Nrfrf. The moderate resistance exhibited by these genetic resources may be integrated into breeding programs aimed at promoting higher levels resistance via recurrent selection or hybridization.

Outcome of Single Agent Generic Gemcitabine in Platinum-Resistant Ovarian Cancer, Fallopian Tube Cancer and Primary Peritoneal Adenocarcinoma

  • Single original gemcitabine is commonly used as salvage treatment in platinum-resistant ovarian cancer, fallopian tube cancer and primary peritoneal adenocarcinoma (PPA) with a satisfactory outcome. However, efficacy data fro this regimen are limited. We therefore conducted a retrospective study to evaluate the outcome of patients who received single-agent generic gemcitabine (GEMITA) after development of clinical platinum resistance. The study period was between May 2008 and December 2010. Gemcitabine was administered intravenously in two different schedules: 1,000 $mg/m^2$ on day 1,8, and 15 every 28 days; and on days 1 and 8 every 21 days with the same dosage. Administration was until disease progression was noted. The response rate was evaluated using the Gynecologic Cancer Intergroup (GCIG) criteria while toxicity was evaluated according to WHO criteria. Sixty-six patients met the inclusion criteria in the study period. Two-thirds of them received gemcitabine as the second and third line regimen. The overall response rate was 12.1%. The median progression free survival and overall survival was 2 and 10 months, respectively. With the total 550 courses of chemotherapy,the patients developed grades 3 and 4 hematologic toxicity as follows: anemia, 1.5%; leukopenia, 13.7%; neutropenia, 27.3%; and thrombocytopenia, 3.0%. In conclusion, single agent generic gemcitabine revealed a modest efficacy in patients with platinum-resistant ovarian cancer, fallopian tube cancer and PPA without serious toxicity.

Characterization of Anthranilate Synthetase from a 5-methyltryptophan Resistant Mutant(MR1) in Maize (옥수수 5-methyltryptophan 저항성 돌연변이주(MR1)의 Anthranilate Synthetase 특성)

  • 5-methyltryptophan(5MT) resistant mutant plants (MRl) were analyzed for characterization of anthranilate synthetase (AS) and tryptophan synthetase (TS) enzymes. The enzyme was measured in crude extracts from MR1 and control seedlings of Danggin inbred line. There was no significant difference in the level of AS between MR1 and control seedlings when grown on MS medium without 5MT. However, MR1 seedlings grown on MS medium with 25mg/L 5MT showed the level of AS twice higher than that of control seedlings. The activity of AS was inhibited to 50% in untreated plants when 4mg /L L-tryptophan was added to their extracts. Extracts from MR1 plants required about four times higher concentration of amino acid to cause equal inhibition. In the TS assay, the activity observed in MR1 seedlings was four times higher than that of control seedlings. We have also isolated and sequenced the gene which encoding the tryptophan synthetase B subunit (TSB) from maize. The gene encodes polypeptides with high homology to TSB isolated from other plants, and is expressed in all the developmental stages examined. Northern hybridization analysis indicated that the gene expression in MR1 seedlings grown on MS medium showed a higher level than in control seedlings.

  • PI201232, a pepper line resistant to Phytophthora capsici Leonian, was crossed with PI2713222 and P1163192, resistant to Xanthomonas campestTis pv. vesicatoria (Doidge) Dye and inheritance of the resistance to each disease and genetic relationship between the two disease resistances was studied. Non-hypersensitive resistance to race 3 of X. c. pv. vesicatoria of PI271322 was inherited in a quantitative mode. Resitance to P. capsici of PI201232 was inherited in a mode close to two dominant alleles. Hypersensitive resistance to race 1 of X. c. pv. vesicatoria of PI271322 was inherited in a mode of single dominant allele. PI 163192 was consistently resistant to both race 1 and race 3 of X c. pv. vesicatoria and the resistance was inherited in a quantitative mode with high dominance effect. Resistance to X. c. pv. vesicatoria was inherited independently from resistance to P. capsici.

Enhanced Sensitivity to Gefitinib after Radiation in Non-Small Cell Lung Cancer Cells

  • Background: Epidermal growth factor receptor (EGFR) tyrosine kinase inhibitors, gefitinib and erlotinib, are effective therapies for non-small cell lung cancer (NSCLC) patients whose tumors harbor somatic mutations in EGFR. The mutations are, however, only found in about 30% of Asian NSCLC patients and all patients ultimately develop resistance to these agents. Ionizing radiation has been shown to induce autophosphorylation of EGFR and activate its downstream signaling pathways. In the present study, we have tested whether the effect of gefitinib treatment can be enhanced after ionizing radiation. Methods: We compared the PC-9 and A549 cell line with its radiation-resistant derivatives after gefitinib treatment with cell proliferation and apoptosis assay. We also analyzed the effect of gefitinib after ionizing radiation in PC-9, A549, and NCI-H460 cells. Cell proliferation was determined by MTT assay and induction of apoptosis was evaluated by flow cytometry. Caspase 3 activation and PARP cleavage were evaluated by western blot analysis. Results: PC-9 cells having mutated EGFR and their radiation-resistant cells showed no significant difference in cell viability. However, radiation-resistant A549 cells were more sensitive to gefitinib than were their parental cells. This was attributable to an increased induction of apoptosis. Gefitinib-induced apoptosis increased significantly after radiation in cells with wild type EGFR including A549 and NCI-H460, but not in PC-9 cells with mutated EGFR. Caspase 3 activation and PARP cleavage accompanied these findings. Conclusion: The data suggest that gefitinib-induced apoptosis could increase after radiation in cells with wild type EGFR, but not in cells with mutated EGFR.

Molecular biological characteristics and analysis using the specific markers of leaf folder-resistant GM rice (혹명나방저항성 GM 벼의 분자생물학적 특성 및 특이 마커를 이용한 검정)

  • In recent years, several genetically modified (GM) crops have been developed worldwide through the recombinant DNA technology and commercialized by various agricultural biotechnological companies. Commercialization of GM crops will be required the assesment of risks associated with the release of GM crops. In advance of the commercial release of GM crops, developer should submit the several information on GM crops for approval. In this study, we carried out to provide the molecular data for the risk assessment of GM rice containing insect-resistant gene, modified Cry1Ac (CryIAc1). Through the molecular analysis with CryIAc1 induced GM rice, we confirmed the steady integration and expression of transgene, the transgene copy number, the adjacent region sequences of inserted gene into rice genome, and the transgene stability in progenies. For the qualitative PCR detection methods, specific primer pairs were designed on the basis of integration sequences, and construct- and event-specific detection markers were developed for leaf folder-resistant rice, Cr7-1 line. From these results, we demonstrated that the molecular data and the PCR detection methods of leaf folderresistant GM rice could be acceptable to conduct the biosafety and environment risk assessment.

Identification of Cisplatin-Resistance Associated Genes through Proteomic Analysis of Human Ovarian Cancer Cells and a Cisplatin-resistant Subline

  • Chemoresistance to cancer therapy is a major obstacle to the effective treatment of human cancers with cisplatin (DDP), but the mechanisms of cisplatin-resistance are not clear. In this study, we established a cisplatin-resistant human ovarian cancer cell line (COC1/DDP) and identified differentially expressed proteins related to cisplatin resistance. The proteomic expression profiles in COC1 before and after DDP treatment were examined using 2-dimensional electrophoresis technology. Differentially expressed proteins were identified using matrix-assisted laser desorption/ ionization time-of-flight mass spectrometry (MALDI-TOF-MS) and high performance liquid chromatography-electrospray tandem MS (NanoUPLC-ESI-MS/MS). 5 protein spots, for cytokeratin 9, keratin 1, deoxyuridine triphosphatase (dUTPase), aarF domain containing kinase 4 (ADCK 4) and cofilin1, were identified to be significantly changed in COC1/DDP compared with its parental cells. The expression of these five proteins was further validated by quantitative PCR and Western blotting, confirming the results of proteomic analysis. Further research on these proteins may help to identify novel resistant biomarkers or reveal the mechanism of cisplatin-resistance in human ovarian cancers.

Characterization of a Novel Necrotic Response of Glycine max Line 'PI96188' to Xanthomonas axonopodis pv. glycines

  • Typical susceptible symptoms of the bacterial pustule disease caused by Xanthomonas axonopodis pv. glycines are pustule and chlorotic haloes that usually occur in leaves of Glycine max plants. The soybean genotype 'PI96188' showed an atypical response to all tested races X. axonopodis pv. glycines, accompanied with necrosis without chlorotic haloes on the underside of the necrotic symptoms. X. axonopodis pv. glycines 8ra grew to levels from 10 to 100 fold lower on PI96188 than on susceptible cultivar Jinjoo1, but 10-fold higher than on the resistant cultivar CNS. The chlorophyll content in PI96188 leaves remained unchanged until 12 days after bacterial infection. Ultrastructural observation showed that the infected leaf cells of PI96188 had intact normal chloroplasts compared to those of the susceptible cultivar Jinjoo1. Chloroplast degradation or the absence of chloroplasts in cells of the infected tissues was observed in Jinjoo1. Senescence-related ACS7 gene was significantly induced in PI96188 compared to those in Jinjoo1 at 2 days after inoculation. While photosynthesis-related rbcS gene showed the dramatic change in Jinjoo1, this gene was constitutively expressed in PI96188. However, expression of the defense-related genes, such as peroxidase and isoflavone synthase in the infected PI96188 leaves was similar to that in Jinjoo1. Together, these results suggest that the novel necrotic symptom in PI96188 is a kind of resistant response different from a typical hypersensitive response in the resistant genotypes.

Characteristics of Rice Mutants Resistant to 5- Methyltryptophan (벼 5-methyltryptophan 저항성 돌연변이체의 특성)

  • TR75, a rice (Oryza sativa L. var. Sasanishikj) mutant resistant to 5-methyltryptophan (5MT) was segregated from the progenies of its initial mutant line, TR1. The 5MT resistance of TR75 was inherited in the M$_{8}$ generations as a single dominant nuclear gene, and was also expressed in callus derived from seeds, roots, and anthers as well as in the seedlings. The callus induced from these organs could grow at 50 mg/1 of 5MT, whereas the growth of wild-type callus was completely inhibited even at 25 mg/1. The seedlings of TR75 did not show resistance to L-azetidine-2-carboxylic acid, S-2-aminoethyl-L-cysteine, p-fluoro-DL-phenylalanine. The content of free amino acids in the TR75 homozygous seeds increased approximately 1.5 to 2.0 fold compared to wild-type seeds. Especially, the contents of tryptophan, phenylalanine and aspartic acid were 5.0, 5.3 and 2.7 times higher than those of wild-type seeds, respectively.y.

Marker-Assisted Foreground and Background Selection of Near Isogenic Lines for Bacterial Leaf Pustule Resistant Gene in Soybean

  • Bacterial leaf pustule (BLP) caused by Xanthomonas axonopodis pv. glycines is a serious disease to make pustule and chlorotic haloes in soybean [Glycine max (L). Merr.]. While inheritance mode and map positions of the BLP resistance gene, rxp are known, no sequence information of the gene was reported. In this study, we made five near isogenic lines (NILs) from separate backcrosses (BCs) of BLP-susceptible Hwangkeumkong $\times$ BLP-resistant SS2-2 (HS) and BLP-susceptible Taekwangkong$\times$ SS2-2 (TS) through foreground and background selection based on the four-stage selection strategy. First, 15 BC individuals were selected through foreground selection using the simple sequence repeat (SSR) markers Satt486 and Satt372 flanking the rxp gene. Among them, 11 BC plants showed the BLP-resistant response. The HS and TS lines chosen in foreground selection were again screened by background selection using 118 and 90 SSR markers across all chromosomes, respectively. Eventually, five individuals showing greater than 90% recurrent parent genome content were selected in both HS and TS lines. These NILs will be a unique biological material to characterize the rxp gene.
