PCR-based markers developed by comparison of complete chloroplast genome sequences discriminate Solanum chacoense from other Solanum species

  • One of wild diploid Solanum species, Solanum chacoense, is one of the excellent resources for potato breeding because it is resistant to several important pathogens, but the species is not sexually compatible with potato (S. tuberosum) causing the limitation of sexual hybridization between S. tuberosum and S. chacoense. Therefore, diverse traits regarding resistance from the species can be introgressed into potato via somatic hybridization. After cell fusion, the identification of fusion products is crucial with molecular markers. In this study, S. chacoense specific markers were developed by comparing the chloroplast genome (cpDNA) sequence of S. chacoense obtained by NGS (next-generation sequencing) technology with those of five other Solanum species. A full length of the cpDNA sequence is 155,532 bp and its structure is similar to other Solanum species. Phylogenetic analysis resulted that S. chacoense is most closely located with S. commersonii. Sequence alignment with cpDNA sequences of six other Solanum species identified two InDels and 37 SNPs specific sequences in S. chacoense. Based on these InDels and SNPs regions, four markers for distingushing S. chacoense from other Solanum species were developed. These results obtained in this research could help breeders select breeding lines and facilitate breeding using S. chacoense in potato breeding.

Purification and preliminary analysis of the ATP-dependent unfoldase HslU from the gram-positive bacterium Staphylococcus aureus

  • The gram-positive bacterium Staphylococcus aureus is a common cause of abscesses, sinusitis and food poisoning. The emergence of antibiotic-resistant strains has caused significant clinical issues worldwide. The HslU-HslV complex was first identified as a prokaryotic homolog of eukaryotic proteasomes. HslU is an unfoldase that mediates the unfolding of the substrate proteins, and it works with the protease HslV in the complex. To date, the protein complex has been mostly studied in gram-negative bacteria. In this study, we report the purification and crystallization of the full-length HslU from S. aureus. The crystal diffracted X-rays to a $3.5{\AA}$ resolution, revealing that the crystals belong to space group $P2_1$, with unit cell parameters of a = 166.5, b = 189.6, $c=226.6{\AA}$, and ${\beta}=108.1^{\circ}$. We solved the phage problem by molecular replacement using the structure of HslU from Haemophilus influenzae as a search model. The cell content analysis with this molecular replacement solution revealed that 24 molecules are contained in the asymmetric unit. This structure provides insight into the structural and mechanistic difference of the HslUV complex of gram-positive bacteria.

Nonlinear numerical analysis and proposed equation for axial loading capacity of concrete filled steel tube column with initial imperfection

  • The use of concrete filled steel tube (CFST) column is widely accepted due to its property of high axial load carrying capacity, more ductility and more resistant to earthquake specially using in bridges and high-rise buildings. The initial imperfection (δ) that produces during casting or fixing causes the reduction in load carrying capacity, this is the reason, experimental capacity is always less then theoretical one. In this research, the effect of δ on load carrying capacity and behavior of concrete filled steel tube (CFST) column have been investigated by numerically simulation of large number of models with different δ and other geometric parameters that include length (L), width (B), steel tube thickness (t), f'c and fy. Finite element analysis software ANSYS v18 is used to develop model of SCFST column to evaluate strength capacity, buckling and failure pattern of member which is applied during experimental study under cyclic axial loading. After validation of results, 42 models with different parameters are evaluated to develop empirical equation predicting axial load carrying capacity for different value of δ. Results indicate that empirical equation shows the 0 to 9% error for finite element analysis Forty-two models in comparison with ANSYS results, respectively. Empirical equation can be used for predicting the axial capacity of early estimating the axial capacity of SCFT column including 𝛿.

A Test Method of Drought Resistance of Rice Varieties Using Sloped-Ridges (경사휴에 의한 벼품종의 내한발검정법)

  • To investigate a possibility of screening for the drought resistance of rice varieties in the slope-ridges, this experiment was conducted in Youngnam Crop Experiment Station in 1982. Seeds were vertically seeded on the slope-ridges which has 30 degree slopes from water level at the front to 1 meter height at the back side. A significant positive correlation was observed between the height of slope-ridges and the PF values (Y=2.25 +0.007 X, r =0.990${\ast}\;{\ast}$).The heading date delayed, the culm length was shortened and the yield was decreased in accordance with the height of slope-ridges. Based upon the above results, it was possible to read the drought resistance of the rice varieties tested. Samnambyeo, Milyang 26, Jinjubyeo and Nongrimna 1 were classified as the resistant ones, while Knob 361-1-8-6-149 was susceptible one.

  • Construction of high-rise structures, formwork systems that can be installed quickly, resistant to external loads, can be used more than once, have become a necessity. Timber and composite timber materials are preferred in the formation of such formwork systems due to their durability, ease of assembly, light weight and easy to use more than one time. Formwork beams are the most commonly used structural component in the formation of such formwork systems, and these beams can be damaged for different reasons during their lifetime. In this study, H20 top P type timber formwork beams with 1800 and 2450 mm length which is among the products of DOKA(c) company is damaged under the effect of static loading up to a high load level of 85% of the maximum ultimate capacity and after being retrofitted using anchored CFRP strips, performance and behavior of the beams under the influence of various loading types such as static, fatigue and impact are investigated experimentally. Two different lengths of retrofitted timber formwork beams were tested by applying monotonic static, fatigue and impact loading and comments were made about the effects of the retrofit method on performance under different loading types.

A Medium Maturing Variety of Italian Ryegrass (Lolium multiflorum Lam.), 'IR605', with High Forage Productivity in Southern Region of Korea

  • This study was conducted at the National Institute of Animal Science from 2010 to 2017. As a variety that is sufficiently productive in the southern regions to replace imported varieties and sufficiently cold-resistant to be cultivated in the central-northern regions, "IR605" was developed and submitted to the Korea Seed & Variety Service in an application for protection. The novel Italian ryegrass variety "IR605" is a diploid with green leaves, a semi-erect growth habit before wintering, and an erect growth habit in the spring. "IR605" was a medium maturing variety with a heading date of around May 15th. "IR605" had a flag leaf width of 9.9 mm, flag leaf length of 26.7 cm, and plant length on the heading date of 100 cm, which was approximately 5 cm longer than "Kowinearly." The stem thickness and ear length of "IR605" are 0.08 mm thicker and 0.5 cm longer than those of "Kowinearly", respectively. The cold-resistance of "IR605" was weaker than that of "Kowinearly", but strong enough to be cultivated in Pyeongchang, Gangwon province. The dry matter yield of "IR605" (9,308 kg/hectare) was 20% higher than that of "Kowinearly", which was further pronounced in the southern region of Haenam, where there was a 52% increased (p < 0.05). The in vitro dry matter digestibility of "IR605" was 68.4% at which was slightly higher than that of "Kowinearly", The total digestible nutrients was 58.5%, which was slightly lower than "Kowinearly". Overall, the feed quality characteristics of "IR605" were similar to those of "Kowinearly".

A Black Waxy Hybrid Corn, "Heukjinjuchal" with Good Eating Quality (고품질 검정 찰옥수수 신품종 "흑진주찰")

  • A single cross hybrid, Heukjinjuchal, is a black color waxy corn (Zea mays L.) developed by the maize breeding team at NICS in 2008. Heukjinjuchal was produced by crossing two inbred lines, KBW24 as seed parent and KBW2 as pollen parent. Ear length and ear diameter of Heukjinjuchal was 16.0 cm and 4.4 cm, respectively. The ratio of kernel set length/ear length was 89%, similar with that of a check hybrid, Chalok1. It is resistant to Northern corn leaf blight disease and also lodging tolerant. Fresh ear yield of Heukjinjuchal comparable with those of a check hybrid, Chalok1 in regional yield trials for three years. Seed production of this hybrid is relatively easy because of well overlapping period of silking and pollen dispersal between the two parents. This cultivar would be well adaptable to the whole country.

Multivariate Characterization of Common and Durum Wheat Collections Grown in Korea using Agro-Morphological Traits

  • Developing improved wheat varieties is vital for global food security to meet the rising demand for food. Therefore, assessing the genetic diversity across wheat genotypes is crucial. This study examined the diversity of 168 durum wheat and 47 common wheat collections from 54 different countries using twelve agro-morphological parameters. Geumgang, a prominent Korean common wheat variety, was used as a control. Both qualitative and quantitative agronomical characteristics showed wide variations. Most durum wheats were shown to possess dense spikes (90%), while common wheats showed dense (40%) or loose (38%) spikes, with yellowish-white being the dominant spike color. The majority of the accessions were awned regardless of wheat type, yellowish-white being the main awn color. White or red kernels were produced, with white kernels dominating in both common (74%) and durum (79%) wheats. Days to heading (DH) and days to maturity (DM) were in the ranges of 166-215 and 208-250 days, respectively, while the culm length (CL), spike length (SL), and awn length (AL) were in the ranges of 53.67-163, 5.33-18.67, and 0.50-19.00 cm, respectively. Durum wheats possessed the shortest average DH, DM, and SL, while common wheat had the longest CL and AL (p < 0.05). Common wheats also exhibited the highest average one-thousand-kernel weight. Hierarchical cluster analysis, aided by principal component analysis, grouped the population into seven clusters with significant differences in their quantitative variables (p < 0.05). In conclusion, this research revealed diversity among common and durum wheat genotypes. Notably, 26 durum wheat and 17 common wheat accessions outperformed the control, offering the potential for developing early-maturing, high-yielding, and lodging-resistant wheat varieties.

Studies on the Wet-injury Resistance of Wheat and Barley Varieties III. Effect of Various Moisture Levels on the Top and Root Growth of Barley Crop (맥류 내습성에 관한 연구)

  • This experiment investigated the effect of various moisture levels on the top and root growth of barley and its yield. Resistant varieties to excess moisture injury shortened plant height, but tillered more, having creeping plant type compared with ordinary one, and greater portion of root of this varieties occured at the deeper soil depths with longer root length. However, susceptible one to excess moisture injury showed an inversed trend when compared with the untreated one. Oversaturated treatment compared with the untreated increased a little bit soil temperature and Eh of soil. Increased root activity and increased yield were obtained with resistant varieties compared with the untreated. Lower root activity and less yield than those of the untreated were observed in both extreme treatments. Yield reduction in both the extreme treatments was mainly due to reduction of tiller number, grain number and 1, 000-grain weight. Highest yield reduction was marked at the stage of internode elongation in the oversaturated and at the booting stage in the overdried treatment.

Comparative Assay Fungus Population and Resistant Genes about Magnaporthe grisea between Korea and China (한국·중국 벼 도열병균 생리형 분석 및 품종 저항성 특성 비교)

  • One hundred isolates of Magnaporthe grisea from Korea and China were characterized for pathogenicity using eight Korean differential varieties(KDV), six Chinese differential varieties(CDV) and six near isogenic lines(NILs) developed in China. The restriction length polymorphism of M. grisea isolates from each country also was analyzed using MGR586 as a probe. One hundred Korean isolates classified into 17 races on KDV were grouped into 29 pathotypes on Chinese near isogenic lines(NILs). Virulence of 46% of Korean isolates against all the six Chinese NILs indicated that the current six Chinese NILs alone was not enough to be used as differential varieties in Korea. Especially, susceptibility of the BL1 carrying resistance gene Pi-b to 70% of tested Korean isolates suggested that BL1(Pi-b) may not be a useful resistance source to Korean blast. Based on the virulence assays of M. grisea populations from each country were divided into two groups. About 50% of Chinese isolates showed similarity to the 30% of the Korean isolates. Especially, the isolates from northern part of China, where Japonica rice cultivars were grown, showed high similarity to the Korean isolates, while isolates from southern part of China, where Indica rice were mainly grown, showed low similarity to Korean isolates. The genome RFLPs of Korean isolates were quite different from those of southern part of China using MGR586 as a probe. These data indicated that the physiological and genetical characteristics of M. grisea population might be determined by strong interaction with cultivated rice.
