Screening of Resistance Genes Linked to Brown Planthopper Using STS Marker in Aromatic Rice Germplasm

  • Brown planthopper (BPH) is a serious insect pest of rice crop throughout rice growing countries, and yield loss due to its infection can be up to 60%. This study aimed to evaluate efficiency of molecular markers for screening BPH resistance accessions among 86 aromatic rice germplasm Eighty-six accessions of aromatic rice germplasm included two accessions of Tongil type (bred in Korea), 28 accessions of japonica type and 56 accessions of indica type. We applied eight STS markers (pBPH9, pBPH19, pBPH20, pBPH21, AJ09-b, RG457L, RG457B, and 7312.T4A) which were linked to four of BPH resistance genes, Bph1, Bph13(t), Bph10, and Bph18(t) respectively. One japonica type accession, 415XIr352, and six indica type accessions possessed one or four positive bands when tested with four STS markers linked to Bph1 gene. One indica type aromatic rice, Basmati9-93, showed the target bands linked to the Bph10 gene. The other accessions did not show same fragments as the respective resistant lines. Bph13(t) is the most widely introduced resistance gene and only one accession showed positive bands implying that this accession might harbor Bph10 and Bph18(t) genes. Three aromatic accessions, Domsiah, Khao Dawk Mali 105 and 415XIr352 showed gene pyramiding of Bph1 and Bph13(t). Two indica aromatic rice, Ds 20 and Basmati 9-93, possessed at least two BPH resistance genes, Bph1, Bph18(t) and Bph13(t), Bph18(t), respectively. These results indicates that aromatic rice germplasm have narrow diversities of BPR resistance genes.

A New Rice Variety 'Hwaweon 4' with Durable Resistance to Rice Blast (벼 도열병 저항성 신품종 '화원4호')

  • 'Hwaweon 4' was developed from a cross between the African upland cultivar, 'Moroberekan' and 'Ilpumbyeo' based on marker-aided backcross selection. The recurrent parent 'Ilpumbyeo' is a high grain quality cultivar with medium to late maturity. 'Hwaweon 4' is nearly isogenic to 'Ilpumbyeo' except a small Moroberekan introgressed segment on chromosome 4 harboring the resistance gene for blast. The preliminary and replicated yield trial was conducted at Chungnam National University in 2006 and 2007. The local adaptability test was carried out by the National Seed Management Office (NSMO) from 2008 to 2009. This cultivar was registered to NSMO with a cultivar designated as 'Hwaweon 4'. This cultivar averaged 76 cm in culm length and has medium growth duration. Milled rice of 'Hwaweon 4' is translucent and the grain quality traits are comparable to those of the recurrent parent. It has low protein content. The yield potential of 'Hwaweon 4' in grain was about 6.31 MT/ha at the ordinary fertilizer level for two years. This variety showed highly resistance reaction at the blast nursery test at four locations and also at the sequential planting method. This resistance is due to the resistance gene designated as Pi45(t) on chromosome 4 introgressed from the donor parent, 'Moroberekan'. The Pi45(t) gene would be useful inenhancing resistance to blast in rice breeding program.

Construction of New P4-Derived Vector Plasmid Containing Tetracyclin Resistance Marker for the Bacteriophage P2-P4 System (박테리오파아지 P2-P4 시스템을 위한 tetracyclin resistance marker 함유 P4 유도체 벡터 플라스미드 조성)

  • To develop vector plasmid for the bacteriophage P2-P4 system which is a useful experimental tool for the study of viral capsid assembly, we constructed a new P4-derived vector plasmid starting from P4 ash8 sid71 With recombinant DNA technology, a portion of P4 genome was deleted and tetracyclin resistance gene (terR) was introduced into P4 genome to give P4 selectivity. Resulting P4 ash8(sid71) terR was 12.09 kb long and could be converted to a viable bacteriophage with P2 infection. The burst size of induced bacteriophage form of P4 ash8(sid71) terR was determined. The CsCl buoyant equilibrium density gradient experiment of new P4 derivative suggested the upper limit of packaging capacity in P2-size head.

A Study on the Bow Hull Form Design of Full Ship Considering the Nonlinear Waves (비선형파를 고려한 비대선의 선수선형설계에 관한 연구)

  • This paper introduces a new hull form design method for the bow of a full ship, by actively applying the relation between the fore-body hull form and its wave resistance characteristics. For the hull form design, the Series 60($C_B=0.8$) hull is chosen as the parent ship, and Kracht's charts are used to determine the parameters of the bulbous bow in the early stages of hull form design. Several hull forms have been tested in order to obtain enough hull form variations with various bow shapes and design parameters in the search of the best design. In order to investigate the resistance characteristics of the designed hull forms, numerical simulations with corresponding model tests have been rigorously performed. For the numerical simulations, the Marker-density method is employed to track the nonlinear phenomena of the free surface(program IUBW). Model tests have also been performed to achieve an improved research performance using the designed hulls. Both numerical and experimental results show that the wave resistance of the hull forms can be effectively diminished if the bows are designed using the method introduced in this research. It is also expected that this research can facilitate better productivity in hull form design, especially at the preliminary design stage of a full ship type vessel.

Spheroplast fusion of pseudomonas spp. using plasmid as selective marker (선별표지로 plasmid를 이용한 pseudomonas spp.의 원형질체 융합)

  • Antibiotic resistance plasmids (RP4, pMG1, R679 and R91-5) were used as primary selective markers to detect the fusants in Pseudomonas spp. By using plasmid marker, clones containing both plasmids of parental strains were obtained as fusants by direct selection. The spheroplast fusion was occured not only between strains of interspecies, but also between strains of intergenus such as Pseudomonas and E. coli. The frequencies of fusant formation were variable from $2.8\times 10^{-1}$ to $6.0\times 10^{-4}$ -4/. In addition, chromosomal recombinants were formed among the clones with both parental plasmids in frequencies of 0.44-1.3%. The fusant formation frequency between interspecies of intraspecies was not different markedly, but stability of plasmids in fusants correlated with the phylogenic smilarity of the parental strains.

Molecular Markers for Sulfadoxine/Pyrimethamine and Chloroquine Resistance in Plasmodium falciparum in Thailand

  • Drug resistance is an important problem hindering malaria elimination in tropical areas. Point mutations in Plasmodium falciparum dihydrofolate reductase (Pfdhfr) and dihydropteroate synthase (Pfdhps) genes confer resistance to antifolate drug, sulfadoxine-pyrimethamine (SP) while P. falciparum chloroquine-resistant transporter (Pfcrt) genes caused resistance to chloroquine (CQ). Decline in Pfdhfr/Pfdhps and Pfcrt mutations after withdrawal of SP and CQ has been reported. The aim of present study was to investigate the prevalence of Pfdhfr, Pfdhps, and Pfcrt mutation from 2 endemic areas of Thailand. All of 200 blood samples collected from western area (Thai-Myanmar) and southern area (Thai-Malaysian) contained multiple mutations in Pfdhfr and Pfdhps genes. The most prevalent haplotypes for Pfdhfr and Pfdhps were quadruple and double mutations, respectively. The quadruple and triple mutations of Pfdhfr and Pfdhps were common in western samples, whereas low frequency of triple and double mutations was found in southern samples, respectively. The Pfcrt 76T mutation was present in all samples examined. Malaria isolated from 2 different endemic regions of Thailand had high mutation rates in the Pfdhfr, Pfdhps, and Pfcrt genes. These findings highlighted the fixation of mutant alleles causing resistance of SP and CQ in this area. It is necessary to monitor the re-emergence of SP and CQ sensitive parasites in this area.

Developmental Changes of Recessive Genes-mediated Cucumber mosaic virus (CMV) Resistance in Peppers (Capsicum annuum L.)

  • Cucumber mosaic virus (CMV) is one of the most important viral diseases in pepper (Capsicum annuum L.), and several genes for resistance were reported in Capsicum spp. In Korea, a single dominant gene that is resistant to $CMV_{Fny}$ and $CMV_{P0}$ has been used for breeding. Recently, a new strain ($CMV_{P1}$) was reported that could infect cultivars resistant to both $CMV_{Fny}$ and $CMV_{P0}$. Therefore, breeding of more robust CMV-resistant cultivars is required. In this study, we surveyed the inheritance of $CMV_{P1}$ resistance and analyzed the location of the resistance loci. After $CMV_{P1}$ inoculation of various germplasms and breeding lines, one accession (ICPN18-8) showed no visual symptoms at 15 dpi (days post inoculation) but was susceptible after 45 dpi, and one resistant line (I7339) showed resistance until at 45 dpi. The latter line was used for tests of resistance inheritance. A total of 189 $F_2$ plants were examined, with 42 individuals showing resistance at 15 dpi and a phenotype segregation ratio close to 1:3 (resistant:susceptible plants). In a lateral ELISA test at 45 dpi, 11 plants showed resistance, and the segregation ratio was changed to 1:15. These results indicate that resistance in C. annuum 'I7339' is controlled by two different recessive genes; we named these resistance genes 'cmr3E' and 'cmr3L,' respectively. To locate these two resistant loci in the pepper linkage map, various RAPD, SSR, and STS markers were screened; only nine markers were grouped into one linkage group (LG). Only one RAPD primer (OPAT16) was distantly linked with cmr3E (22.3 cM) and cmr3L (20.7 cM). To develop more accurate markers for marker-assisted breeding, enriching for molecular markers spanning two loci will be required.

Rice OsACDR1 (Oryza sativa Accelerated Cell Death and Resistance 1) Is a Potential Positive Regulator of Fungal Disease Resistance

  • Rice Oryza sativa accelerated cell death and resistance 1 (OsACDR1) encodes a putative Raf-like mitogen-activated protein kinase kinase kinase (MAPKKK). We had previously reported upregulation of the OsACDR1 transcript by a range of environmental stimuli involved in eliciting defense-related pathways. Here we apply biochemical, gain and loss-of-function approaches to characterize OsACDR1 function in rice. The OsACDR1 protein showed autophosphorylation and possessed kinase activity. Rice plants overexpressing OsACDR1 exhibited spontaneous hypersensitive response (HR)-like lesions on leaves, upregulation of defense-related marker genes and accumulation of phenolic compounds and secondary metabolites (phytoalexins). These transgenic plants also acquired enhanced resistance to a fungal pathogen (Magnaporthe grisea) and showed inhibition of appressorial penetration on the leaf surface. In contrast, loss-of-function and RNA silenced OsACDR1 rice mutant plants showed downregulation of defense-related marker genes expressions and susceptibility to M. grisea. Furthermore, transient expression of an OsACDR1:GFP fusion protein in rice protoplast and onion epidermal cells revealed its localization to the nucleus. These results indicate that OsACDR1 plays an important role in the positive regulation of disease resistance in rice.

Improvement of Selection Efficiency for Bacterial Blight Resistance Using SNP Marker in Rice (SNP 마커를 이용한 벼 흰잎마름병 저항성 선발 효율 증진)

  • Discovery of single nucleotide polymorphisms (SNPs), including small insertions and deletions, is one of the hot topics in genetic research. The most common type of sequence variant consists of single base differences or small insertions and deletions at specific nucleotide positions. Significance of SNPs in rice is increasing for genetic research, positional cloning and molecular breeding. $F_2$ 170 lines and $F_3$ 194 lines derived from Sangjuchalbyeo/HR13721-53-3-1-3-3-2-2 Were used for Searching SNP markers related to bacterial blight resistance. Sangjuchalbyeo is susceptible to bacterial blight, but HR13721-53-3-1-3-3-2-2 has Xa1 gene resistant to bacterial blight. Individual lines were inoculated with $K_1$ race of bacterial blight and resistant or susceptible was evaluated after 3 weeks from inoculation. The genotypes of population were analysed by PCR-RFLP for SNP marker developing. The segregation of $F_2\;and\;F_3$ population showed almost 3:1, 1:1 ratio, respectively. Analysis of genotype using SNP marker is capable of confirming resistance for $K_1$ race and genotype through amplifying the gene using 16PFXal primer and digested the PCR product with Eco RV. There were close relation between resistance test for $K_1$ race and SNP marker genotype. Especially, DNA analysis using SNP marker is capable of judging homozygote/heterozygote in $F_2$ population compared with resistant test for Kl race. So, it seems to improve the selection efficiency in disease resistant breeding.

The Use of Aureobasidin A Resistant Gene as the Dominant Selectable Marker for the Selection of Industrial Yeast Hybrid (산업용 효모 Hybrid의 선별을 위한 우성선별표지로서의 Aureobasidin A 내성유전자의 이용)

  • For the strain improvement of the industrial polyploid yeast strain through hybridization and protoplast fusion, a dominant selection marker other than a recessive marker such as the auxotrophic marker was required for the selection of the resulting hybrids. In the present investigation, the aureobasidin A resistant gene was tested in relation to whether it can be used as the dominant selectable marker for the isolation of hybrids of the yeast Saccharomyces. The plasmid pAUR112, carrying the gene responsible for resistance to aureobasidin A, was introduced into the haploid yeast strain K114/YIp. From the rare-mating between polyploid C6 and haploid K114/YIp carrying pAUR112, many hybrids were obtained from the agar medium containing 0.5 ${\mu}g$/ml of aureobasidin A. The hybrids exhibited characteristics derived from both of the parental strains; and the cell sizes of the hybrids were larger than those of the parental strains. These results showed that the aureobasidin A resistant gene could be successfully used as the dominant selectable marker for the isolation of yeast hybrids resulting from rare-mating.
