Utilization of Sunflower Crop Residues as Feed in Small Ruminants

  • Sheep and goats in Pakistan have not been able to produce to the best of their potential. This may primarily be attributed to under feeding and malnutrition. Ranges have been depleted due to overgrazing and mismanagement and are not in position to feed the existing small ruminant population. To overcome the shortage of good quality fodder and balanced feed supply, the alternate means like cereal straws and other crop residues are being commonly used. Sunflower crop residues like stalks and heads provide a good quality forage for livestock. These crop byproducts are rich in crude protein and lower in crude fibre. Their inclusion in the diet of small ruminants at 20, 30 and 40 percent levels in ration has shown significantly (p < 0.05) increased feed intake and weight gain. Daily feed intake was 1,130, 1,180 and 1,750 g for sunflower crop residue, soybean crop residue and wheat straw, respectively, when added at the rate of 20 percent in the ration. The drymatter digestibility of sunflower, soybean crop residues and wheat straw was also comparable. The maximum performance of the animals was observed at the 20 percent level of inclusion of sunflower crop residue in the diet.

Cloning and Site-Directed Mutagenesis of Musca domestica Acetylcholinesterase for Enhancing Sensitivity to Organophosphorus and Carbamate Insecticides

  • Mature acetylcholinesterase (AChE) gene (gm, 1,836 bp) was cloned from the housefly and successfully expressed in the E. coli CodonPlus (DE3) RIL system (GM-E, 72 kDa) with a yield of 1,630 mU/g fresh cells. Using the gm, 10 kinds of mutants were constructed and expressed for enhancing sensitivity to insecticides. The sensitivity of these mutants to five kinds of organophosphate (OP) and three carbamate insecticides was investigated by measuring the apparent bimolecular inhibition constant ($k_i=k_2/K_d$). Surprisingly, the sensitivity of quadruple mutant IGFT was enhanced as much as 7-fold for acephate, 164-fold for demeton-S-methyl, 484-fold for dichlorvos, 523-fold for edifenphos, 30-fold for ethoprophos, 30-fold for benfuracarb, 404-fold for carbaryl, and 107-fold for furathiocarb, compared with that of GM-E, although the sensitivity of each single point mutant was slightly increased. These mutational studies indicated that (i) contradictory to Walsh et al. [39], the residue 327 is the important key residue for enhancing sensitivity as much as the residue 262, (ii) the residue 82 and additional residues of 234, 236, and 585 are also important, and (iii) sensitivity was cooperatively accelerated as the number of strategic mutations increased.

Enhancement of Water-solubilities of Protein-bound Polysaccharides Contained in the Basidiocarps of Ganoderma lucidum by Hydrolyzing with Chymotrypsin

  • Optimum conditions for hydrolysis were investigated to enhance water-solubilities of protein-bound polysaccharides in the basidiocarps of Ganoderma lucidum by treating chymotrypsin. We also attempted with Ganoderma lucidum residue remaining after extracting hot water-soluble compoents in Ganoderma lucidum. After hydrolyzing under optimum conditions (20 ppm chymotrypsin, 2% Gampderma lucidum or 6% Ganoderma lucidum residue, at pH 10 and at $ 40^{\circ}C$), the amounts of total protein and carbohydrate of hydrolysate were measured. Michaelis constant, $K_{m}$, and maximum rate, $V_{max}$, calculated by Lineweaver-Buck plot for the hydrolysis of Ganoderma lucidum were 1.73% and 0.073%/min respectively and those for hydrolysis of Ganoderma lucidum residue were 2.40% and 0.033%/min respectively. The amount of polysaccharide isolated from Ganoderma lucidum (100 g) treated with chymotrypsin was only 3.07 g, but significantly increased amount (14.34 g) of polysaccharides was isolated from Ganoderma lucidum residue (100 g) treated with chymotrypsin. The protein-bound polysaccharide was isolated from the non-hydrolyzed and hydrolyzed sample and molecular weights of the polysaccharide were measured by Sepharose CL-48 gel filtration.

Residue of Imidacloprid in Hulled Rice and Paddy Soil (논 토양 및 현미중 Imidacloprid의 잔류성)

  • The residue of imidacloprid in hulled rice and paddy soil was investigated. In laboratory conditions, the degradation of imidacloprid in the soils followed first-order reaction kinetic. The rate of degradation was influenced by soil temperature and soil type. The half-life of imidacloprid at $18-28^{\circ}C$ was 66.7-96.3 days in the heavy clay soil and 56.8-117.5 days in the clay loam soil. Arrhenius activation energy obtained from the temperature experiment was 25.5 KJ/mol in heavy clay soil and 50.3 KJ/mol in clay loam soil. In paddy field, the degradation of imidacloprid was fast during the initial period but the degradation rate was gradually slow. About 10 % of the initial amount remained in the soil 120 day after the application. The residual amount of imidacloprid in rice was below the detection limit, 0.01 ppm. The residue level in rice was lower than MRL 0.05ppm in Korea.

Gold Recovery from Geumsan Concentrate Using Microwave-nitric Acid Leaching and Lead-fire Assay (마이크로웨이브-질산용출과 납-시금법을 이용한 금산정광으로부터 금 회수)

  • This study aimed to effectively dissolve sulfide minerals through microwave-nitric acid leaching of invisible gold concentrate and then recover gold from the solid-residue with fire assay. For the purposes, this study conducted microwave-nitric acid leaching experiments to examine nitric acid concentration, time of microwave leaching, and sample addition effect. As results of the experiments, this study discovered that the weight loss rate of solid-residue increased as nitric acid concentration and microwave leaching time increased while weight loss rate decreased as sample addition increased. In an XRD analysis with solid-residue, it was discovered that pyrite completely disappeared when the nitric acid concentrate was 6 M and the microwave leaching time was 18 minutes. When a fire assay was carried out with solid-residue, gold particles with more content were recovered as nitric acid concentration and microwave leaching time increased whereas gold particles with more gold content were recovered as the sample addition decreased.

Establishment of Pre-Harvest Residue Limit (PHRL) of Emamectin benzoate during Cultivation of Amaranth (생산단계 비름 중 Emamectin benzoate의 잔류허용기준 설정)

  • This study was performed to investigate pre-harvest residue limit (PHRL) in amaranth, to estimate biological half-life of emamectin benzoate and identify the characteristics of the residue. Pesticides of standard and double appplication rate, were sprayed once on amaranth at 0, 1, 2, 3, 5, 7, 10, 14 days before harvest. Amaranth sample was extracted with acetonitrile and partitioned with dichloromethane, and pesticide residues were determined with LC/MS/MS. The limit of detection of emamectin benzoate was 0.01 mg/kg. Recoveries of emamectin benzoate ($B_{1a}$, $B_{1b}$) at two fortification levels of 0.1 and 0.5 mg/kg, $B_{1a}$ were $93.3{\pm}0.7%$ and $93.2{\pm}7.7%$, $B_{1b}$ were $106.6{\pm}1.9%$ and $80.5{\pm}6.6%$, respectively. The biological half-lives of emamectin benzoate were about 2.0 days at standard application rate and 1.7 days at double application rate, respectively. The PHRL of emamectin benzoate were recommended as 0.84 mg/kg for 10 days before harvest.

Biosorption of Methylene Blue from Aqueous Solution Using Xanthoceras sorbifolia Seed Coat Pretreated by Steam Explosion

  • Xanthoceras sorbifolia seed coat (XSSC) is a processing residue of the bioenergy crop. This work aimed to evaluate the applicability of using the steam explosion to modify the residue for dye biosorption from aqueous solutions by using methylene blue as a model cationic dye. Equilibrium, kinetic and thermodynamic parameters for the biosorption of methylene blue on the steam-exploded XSSC (SE-XSSC) were evaluated. The kinetic data followed the pseudo-second-order model, and the rate-limiting step was the chemical adsorption. Intraparticle diffusion was one of the rate-controlling factors. The equilibrium data agreed well with the Langmuir isotherm, and the biosorption was favorable. The steam-explosion pretreatment strongly affected the biosorption in some respects. It reduced the adsorption rate constant and the initial sorption rate of the pseudo-second-order model. It enhanced the adsorption capacity of methylene blue at higher temperatures while reduced the capacity at lower ones. It changed the biosorption from an exothermic process driven by both the enthalpy and the entropy to an endothermic one driven by entropy only. It increased the surface area and decreased the pH point of zero charge of the biomass. Compared with the native XSSC, SE-XSSC is preferable to MB biosorption from warmer dye effluents.

  • Approx. 200,000 bpd vacuum residue oil is produced from oil refineries in Korea. These are supplying to use asphalt, high sulfur fuel oil, and upgrading at the residue hydro-desulfurization unit. Vacuum residue oil has high energy content, however high sulfur content and high concentration of heavy metals represent improper low grade fuel. To meet growing demand for effective utilization of vacuum residue oil from refineries, recently some of the oil refinery industries in Korea, such as SK oil refinery and LG Caltex refinery, have already proceeded feasibility study to construct 435-500 MWe IGCC power plant and hydrogen production facilities. Recently, KIER(Korea Institute of Energy Research) are studing on the Vacuum Residue gasification process using an oxygen-blown entrained-flow gasifier. The experiment runs were evaluated under the reaction temperature : 1,100~1,25$0^{\circ}C$, reaction pressure : 1~6kg/$\textrm{cm}^2$G, oxygen/V.R ratio : 0.8~0.9 and steam/V.R ratio : 0.4-0.5. Experimental results show the syngas composition(CO+H$_2$) : 85~93%, syngas flow rate : 50~110Mm$^3$/hr, heating value : 2,300~3,000 ㎉/Nm$^3$, carbon conversion : 65~92, cold gas efficiency : 60~70%. Also equilibrium modeling was used to predict the vacuum residue gasification process and the predicted values were compared reasonably well with experimental data.
