LC-MS/MS 와 GC-MS/MS 를 이용한 에센셜 오일 중 320 종 잔류농약 분석법 개발 (320 Pesticides Analysis of Essential Oils by LC-MS/MS and GC-MS/MS)

  • 에센셜 오일(essential oil)은 1 개의 단일 식물형 및 식물종이 만들어낸 향이 나는 식물 재료를 물리적인 방법으로 얻어낸 휘발성 물질로 방부, 살균, 항균 효과가 뛰어나 화장품, 향료, 아로마 테라피 치료 등의 목적으로 폭 넓게 사용되고 있다. 에센셜오일은 추출 및 농축과정을 거치게 되는데 이때 재배 과정 중 살포된 농약 또한 추출 및 농축이 됨으로써 인체에 유해할 수 있다. 본 연구에서는 에센셜 오일 중 320 종의 잔류농약을 분석하기 위하여 LC-MS/MS와 GC-MS/MS를 이용하였으며 기존의 정제과정인 hexane 대신 freezing 과정을 이용하여 전처리방법을 개선하였다. 에센셜 오일을 분석한 결과 기준치가 설정되어 있지 않은 chlorpyrifos, piperonyl butoxide, silafluofen 성분이 basil oil 및 clove leaf oil에서 검출되었다. 따라서 에센셜 오일에 대한 잔류농약 모니터링이 지속적으로 필요할 것으로 판단된다.

쌀 세척 및 취반 방법에 따른 isoprothiolane의 감소 효과 (Reduction effects of isoprothiolane during rice washing and cooking)

  • 본 연구는 쌀의 세척 및 취반 방법에 따른 isoprothiolane 잔류량을 GC-µECD를 이용하여 분석하였다. 세척 전 현미 잔류량은 17.03 mg/kg이었으며, 도정 후 백미는 1.67 mg/kg이었다. 세척 방법에서 물의 비율과 세척 횟수에 따라 현미와 백미의 isoprothiolane 잔류량은 19.3-59.3 및 43.1-66.5%이었으며, 세척물 온도가 5-40℃ 증가할수록 현미는 56.6-63.1%, 백미는 67.1-74.9% 감소하였다. 세척 방법을 손으로 살살 휘젓거나 비비는 방법으로(A, B 및 C)로 달리하였을 때 현미의 농약 감소율은 63.1-71.6%, 백미는 75.4-87.4%이었다. 현미를 압력밥솥과 전기밥솥을 이용하여 즉시 취반하였을 때 농약은 78.4 및 78.5%, 백미의 경우 94.0 및 94.0%로 감소하였으며, 30분간 불린 후 취반하였을 때 잔류농약은 현미에서 83.4%, 백미는 95.8% 감소하였다. 따라서 현미와 백미의 잔류농약 감소를 위한 가장 효과적인 세척 및 취반 방법은 40℃ 물, 7배 물로 6회 (2, 3회 차는 손으로 비벼서 세척) 세척 후 30분간 물에 불리기이다. 밥솥의 종류에 관계없이 취반할 경우 현미는 평균 83.4%, 백미는 95.8% isoprothiolane이 효과적으로 제거되는 것을 확인하였다.

논 토양 및 현미중 Imidacloprid의 잔류성 (Residue of Imidacloprid in Hulled Rice and Paddy Soil)

  • 논 토양과 현미 중 살충제 imidacloprid의 잔류성을 조사한 결과는 다음과 같다. 실내 조건의 토양 중 imidacloprid의 분해속도는 1차 반응식에 따랐으며 토양온도 및 토양 종류에 크게 영향을 받았다. $18-23^{\circ}C$의 토양중 imidacloprid의 반감기는 증식토에서 66.7-96.3일, 식양토에서 56.8-117.5일이었다. Arrhenius activation energy는 증식토에서 25.5KJ/mol, 식양토에서 50.3KJ/mol이었다. 포장조건하의 토양 중 imidacloprid의 분해는 처리 초기에는 매우 빨랐으나 점점 분해속도가 느려 1차반감기는 약 10일 정도이었으나 2차, 3차 반감기는 점점 길어져 처리량의 90%까지 분해되는데는 약 120일이 소요되었다. 현미 중 imidacloprid의 잔류량은 검출한계 0.01ppm이하이었다.

팽화 처리한 나락의 결정화도, 당화효율,농약잔류량 비교 (Comparison of Relative Crystallinities, Saccharification Effeciency and Diazinon Residue of Varietal Puffed Paddy)

  • 나락을 열풍팽화기로 팽화시켜 현미와 왕겨의 결정화도, 당화율, 농약잔류량의 변화를 비교하였다. 나락의 팽화처리로 쌀전분의 결정화도가 감소하였으나 왕겨의 결정화도는 큰 변화가 없었다. 찰벼의 팽화처리는 자숙처리에 비하여 엿기름 amylase에 의한 당화율이 높았으며 왕겨의 cellulase에 의한 가수분해율은 팽화처리시 $14{\sim}30%$ 상승되었다. 나락의 팽화처리로 왕겨와 현미 중의 농약(diazinon)의 잔류량 감소는 메벼에서 현저하였고, 팽화전에 비하여 현미와 왕겨에서 각각 42.7%, 47.6% 잔류되었다.

Comparison of 14C-radioactivity in rice-paddy soil exposed to atmospheric and elevated CO2 conditions after 14C-carbaryl treatment

  • This study was performed to investigate if elevated $CO_2$ affects the residue pattern of $^{14}C$ in the soil environment after $^{14}C$-carbaryl treatment $^{14}C$-carbaryl was applied on the rice plant-grown greenhouse soil exposed to atmospheric and elevated $CO_2$ conditions. $^{14}C$-radioactivity was measured in the rhizospheric soil and rice straw samples six months after $^{14}C$-carbaryl application. Significantly high radioactivity was observed in the soil exposed to atmospheric $CO_2$ as compared to that in the soil exposed to elevated C(h. Background level of radioactivity was observed in rice plant samples. These observations suggest the possibility that elevated $CO_2$ may affect residual radioactivity of $^{14}C$-carbaryl in the soil rather than that in the plant.

인삼 지상부를 첨가한 사료 급여가 닭에서 나타내는 효과 (Efficacy of orally administered ginseng stem and leaf in chickens)

  • Ginseng has been widely used in Korea as a natural medicine due to its saponin contents. Although the total amount of ginseng stem and leaf saponins (GSLS) is 4~5 times higher than that of saponin in the root, the root is mainly used. This is due to two reasons: nervous system-stimulant activity of GSLS and pesticide residues in GSLS. In this study, residual agricultural pesticides were removed from GSLS using two types of bacterial treatments. Two GSLS treatment groups of chickens (GSLS-1 and GSLS-2) were established. The chickens were fed 0.4% GSLS-1 or GSLS-2 mixed with crop. We then evaluated the effects of GSLS on bodyweight and several immune parameters. At the end of the experiments, chickens fed GSLS-1 and red ginseng saponin had significantly higher growth rates (16.6% and 8.0%, respectively) compared to the vaccine control group treated with Noblis Salenvac-T. The group fed GSLS-1 also had the highest IgG titer that was significantly different at the end of experiments compared to the other groups. These findings imply that GSLS-1 is a good candidate feed additive for the chicken industry.

계절에 따른 시판 신선 편이 샐러드 제품의 품질 평가 (Quality Evaluation of Fresh-cut Market Products by Season)

  • As a result of life-style changes, consumer's concerns of food have shifted from calories and nutrition to health and convenience. Fresh-cut products are one such new direction for fruit and vegetable consumption. In this study, the vitamin C, mineral, and pesticide contents of various fresh-cut products were analyzed. According to sensory evaluations, the key reason for a lower than expected overall acceptability of many fresh-cut products is that they are likely to have browning and can easily lose their freshness. Also, the sensory evaluation showed that shriveling, the degree of browning, softening around the cut edge, and off-flavors were the primary factors affecting the quality of fresh-cut products. As a nutritional factor of quality, vitamin C content was not practical with regard to fresh-cut lettuce because the level was very low. For product safety, residual pesticides were detected in the fresh-cut products, but the results showed that all items were under permitted levels and considered safe. In evaluation of the microbial levels of the fresh-cut market products, the levels of viable cells, mold, yeast, coliform bacteria, and enterobacteriaceae were not significantly different based on the summer and winter seasons. The levels of S. aureus and Listeria spp. in the products were higher during the summer season than the winter.

수생식물을 이용한 엔도설판(${\alpha},\;{\beta}$) 및 페니트로치온의 제거 (Phytoremediation of the pesticides, endosulfan (${\alpha}$ and ${\beta}$) and fenitrothion, using aquatic plants)

  • A phytoremediation study has been conducted to see if some known aquatic plants can remove the pesticides, endosulfan-${\alpha},\;{\beta}$ and fenitrothion which are frequently used in the crop protection and golf course management, and are likely to exist as residual pollutants in the aquatic ecosystems. Among the five aquatic plants tested in the microcosms, water lily Nymphaea tetragona Georgi showed the highest degradation efficacies (85~95%) for the three pesticides as opposed to the control(13~26%). The efficacies for the other plants were in the range of 46~80% in the order of Pistia stratiotes, Cyperus helferi, Eichhornia crassipes, and Iris pseudoacorus. Fenitrothion, an organo-phosphorus pesticide, was much more vulnerable to the phytoremediation than the organo-chlorine pesticides, endosulfan-${\alpha}$ and endosulfan-${\beta}$. The kinetic rate constants ($min^{-1}$) for removal of the three pesticides were more than 10 times higher than the control (non-planting) in case of Nymphaea tetragona Georgi. This aquatic plant showed kinetic rate constants about 2 times as much as the lower kinetic rate constants shown by Iris pseudoacorus. The reason for the highest degradation efficacy of water lily would be that the plant can live in the sediment and possess roots and broad leaves which could absorb or accumulate and degrade more pollutants in association with microbes. These results indicate that some of the selected aquatic plants planted near the agricultural lands and wetlands could contribute to remediation of pesticides present in these places, and could be applicable to protection of the aquatic ecosystems.

자외선과 광 촉매제를 이용한 감귤껍질 농약제거공정의 최적화 (Optimum Processing Conditions for Pesticides Removal in Mandarine Orange Peel by Ultraviolet Rays and Photocatalytic Materials)

  • UV-C와 -B type의 lamp를 이용한 혼합 자외선(UV)조사의 조사시간, 조사온도 및 광 촉매제인 $H_2O_2$의 분사농도변화를 이용하여 감귤껍질에 부착되어있는 농약을 제거하기 위한 공정의 최적화를 이루고자 하였다. Chloropyrifos에 대한 독립변수들의 영향력은 조사온도가 가장 컸으며 그 다음 조사시간, $H_2O_2$ 분사농도 순 이었다. Methidathion은 조사시간이 가장 컸으며 조사온도, $H_2O_2$ 분사농도 순으로 나타났다. EPN은 조사온도와 조사시간이 비슷하게 나타났고 $H_2O_2$ 분사농도가 가장 작았다. UV감귤껍질의 농약 잔존량이 가장 작게 나타난 처리조건으로 chloropyrifos와 EPN에 대한 최적조건으로는 조사시간 60분, 조사온도 $45^{\circ}C$, $H_2O_2$ 분사농도 1000 ppm으로 나타났다. 반면에 methidathion의 경우에는 최적조건으로는 조사시간 60분, 조사온도 $40^{\circ}C$, $H_2O_2$ 분사농도 1000 ppm으로 나타났다. 최적공정조건에서 chloropyrifos, methidathion 및 EPN의 잔존 량은 조사 전의 각각 46, 49 및 28% 수준이었다.

딸기 중 침투성 및 비침투성 농약에 따른 잔류특성 연구 (Comparison of Residue Patterns for Systemic and Non-systemic Pesticides in Strawberry)

  • BACKGROUND: The healthy food trend has encouraged the consumption of natural products, including berries. This trend is expected to increase the strawberry consumption. There has been a concern about the exposure of pesticides approved for use on strawberry. In this study, the dissipation patterns of systemic and non-systemic pesticides were evaluated in strawberry under plastic-covered greenhouse conditions. METHODS AND RESULTS: Cyflumetofen and dimethomorph were applied on strawberry in the critical GAP (Good Agricultural Practices). Strawberries were harvested at 0, 1, 2, 3, 5, 7 and 10 days after final application of the pesticides. The analyses of the residual pesticides were performed by HPLC-DAD with C18 column. The limits of quantitation (LOQ) of cyflumetofen and dimethomorph were 0.04 and 0.02 mg/kg, respectively. The recovery of cyflumetofen and dimethomorph were 88.1 ~ 103.3% and 79.0 ~ 110.2% for the spiked two levels (LOQ and 10LOQ), respectively. The biological half-lives of cyflumetofen and dimethomorph werer 7.5 and 8.9 days, respectively. The dissipation rates in strawberry were calculated by the statistics method at a 95% confidence level. The distribution showed that pesticides with low log Pow were indicated by the decreased dissipation rate and pesticides with similar log Pow and low solubility also showed the decreased dissipation rate. CONCLUSION: The residues of cyflumetofen and dimethomorph in strawberry at time 0 after the final application were below the established MRL in Korea. The dissipation behavior of systemic and non-systemic pesticides in strawberry is affected by their log Pow and water solubility values.
