Methylation Status of H19 Gene in Embryos Produced by Nuclear Transfer of Spermatogonial Stem Cells in Pig

  • The faulty regulation of imprinting gene lead to the abnormal development of reconstructed embryo after nuclear transfer. However, the correlation between the imprinting status of donor cell and preimplantation stage of embryo development is not yet clear. In this study, to determine this correlation, we used the porcine spermatogonial stem cell (pSSC) and fetal fibroblast (pFF) as donor cells. As the results, the isolated cells with laminin matrix selection strongly expressed the GFR ${\alpha}$-1 and PLZF genes of SSCs specific markers. The pSSCs were maintained to 12 passages and positive for the pluripotent marker including OCT4, SSEA1 and NANOG. The methylation analysis of H19 DMR of pSSCs revealed that the zinc finger protein binding sites CTCF3 of H19 DMRs displayed an androgenic imprinting pattern (92.7%). Also, to investigate the reprogramming potential of pSSCs as donor cell, we compared the development rate and methylation status of H19 gene between the reconstructed embryos from pFF and pSSC. This result showed no significant differences of the development rate between the pFFs ($11.2{\pm}0.8%$) and SSCs ($13.3{\pm}1.1%$). However, interestingly, while the CTCF3 methylation status of pFF-NT blastocyst was decreased (36.3%), and the CTCF3 methylation status of pSSC-NT blastocyst was maintained. Therefore, this result suggested that the genomic imprinting status of pSSCs is more effective than that of normal somatic cells for the normal development because the maintenance of imprinting pattern is very important in early embryo stage.

  • This study aimed to explain the factors that influenced an individual's decision to migrate. The method of analysis in this study was the estimation of the probit regression model with data from the Indonesian Family Life Survey (IFLS-5), which covered 30,000 individuals from 13 provinces in Indonesia. Data from IFLS-5 were longitudinal data, meaning that the study was looking for data consistently to get reliable data from respondents. The research variables to determine the individual's decision to migrate were education level, income level, employment status, marital status, land ownership status, health quality, gender, residence status, and poverty status. Individual decision to migrate as a dependent variable was placed as a dummy variable. The results showed that the level of education, income level, employment status, marital status, land ownership status, health quality, and poverty status significantly influenced an individual's decision to migrate. Meanwhile, gender and residence status did not significantly affect an individual's decision to migrate. This research recommends that it is necessary to pursue a policy of economic equality between regions because economic factors are the main trigger for an individual's decision to migrate. Policies to overcome economic disparities among regions will reduce the individual's decision to migrate.

Factors Indicating Culture Status During Cultivation of Spirulina (Arthrospira) platensis

  • Factors indicating culture status of two Spirulina platensis strains were monitored in a batch mode cultivation for 36 days. Changing mode in all factors showed a common turning point, indicating shift of cell or culture status. Mean biomass productivity was highly sustained until day 22, chlorophyll a concentration peaked on day 22, pH value was > 12 on day 22, coil number was abruptly shortened on day 22, and floating activity was sustained at greater than 79% after day 22, indicating that day 22 is a criterion reflecting phase-transfer in cell physiology in a batch culture system. Many of these changes may have been caused by increased pH, suggesting that pH control is essential for mass production of S. platensis. Fluctuations in floating activity were likely induced by the number of cellular gas vacuoles. Consequently, coil number per trichome and floating activity of S. platensis could readily act as simple indicators for determination of culture status or harvesting time of cells.

Nutritional Status, Nutrients Intakes, and Health Status of Young-old and Old-old Homebound Elderly in Korea (연소 · 고령 노인의 성별에 따른 건강 및 영양상태 비교 조사 연구)

  • Purpose: Number of the old-old elderly is rapidly increasing in Korea, but it is unclear whether there are differences between old-old and young-old elderly in nutritional status, nutrient intakes and health status. The gender differences in Korean elderly in these conditions also remains unknown. This study, therefore, investigated gender-associated differences in nutritional, health status and nutrient intakes and how they are related among young-old and old-old homebound elderly. Methods: Two hundred and eighty elderly who were attending a local elderly welfare center were recruited. Evaluation included demographic, nutritional and health status related data, nutritional intakes, and life style related factors including physical activity. Results: Of the 280 subjects, old-old were 147 (52.5%) and young-old were 133 (47.5%). Male old-old elderly had more often abdominal obesity than female old-old, but male old-old more often had malnutrition than female old-old. There were few differences in nutrient intakes between old-old male elderly and female elderly after energy intakes were controlled. Male old-old more often had less intake of beta carotene and Vitamin A than female old-old. On the other hand, male old-old elderly more often had hypercholesterolemia and hypertension than male young-old. Conclusion: Male old-old may be at a greater health and nutritional risk than female old-old. Targeted nutritional intervention for male old-old emphasizing antioxidant nutritional intakes may be warranted.

  • This paper reports the development work carried out in respect of a proposed application of Neural Network approach for the Korean Next generation Reactor (KNGR) now referred as APR-1400. The emphasis is on establishing the methodology and the approach to be adopted towards realizing this application in the next generation reactors. Keeping in view the advantages and limitation of Artificial Neural Network Approach, the role of ANN has been limited to plant status or to be more precise plant transient monitoring. The simulation work carried out so far and the results obtained shows that artificial neural network approach caters to the requirements of plant status monitoring and qualifies to be incorporated as a part of proposed operator support systems of the referenced nuclear power plant.

Evaluation of Nutritional Status of Cancer Patients during Treatment by Patient-Generated Subjective Global Assessment: a Hospital-Based Study

  • Cancer patients frequently experience malnutrition. Cancer and cancer therapy effects nutritional status through alterations in the metabolic system and reduction in food intake. In the present study, fifty seven cancer patients were selected as subjects from the oncology ward of Cachar Cancer Hospital and Research Centre, Silchar, India. Evaluation of nutritional status of cancer patients during treatment was carried out by scored Patient-Generated Subjective Global Assessment (PG-SGA). The findings of PG-SGA showed that 15.8% (9) were well nourished, 31.6% (18) were moderately or suspected of being malnourished and 52.6% (30) were severely malnourished. The prevalence of malnutrition was highest in lip/oral (33.33%) cancer patients. The study showed that the prevalence of malnutrition (84.2%) was high in cancer patients during treatment.

  • In this study, actual status of safety management for the objects of 173 university, government research institution, and corporate affiliated research center was carried out through questionnaires. For unexpected accident status according to kinds, scale, and types of laboratories, frequency analysis using cross analysis and multiple replies analysis was used. The result of this study is judged to be used for laboratory safety education prevention programs, laboratory environment improvement, investment or improvement of laboratories through the actual safety management status of laboratories such as vulnerable works for generation of unexpected sudden accidents according to kind, scale, and type of laboratories, period of occurrence, prevention education, necessity for preliminary test, and cause of laboratory accidents.

The Impact of Conspicuous Consumption and Perceived Value on New Product Adoption Intention

  • Unidimensional construct of conspicuous consumption which focuses on the ostentation of the economic assets are limited in describing today's consumers. We propose that both ostentation of social status and demonstration of uniqueness through overt consumption should be considered in the realm of conspicuous consumption. This research aims to examine the two dimensions of conspicuous consumption tendencies and their impact on new product adoption intention, which is mediated through the perceived value of the new products. This research empirically validates the theoretical conjecture by conducting an online survey (N=272). Our empirical findings reveal that ostentation of social status influences new product adoption intention both directly and indirectly through perceived social value, while demonstration of uniqueness improves new product adoption intention only indirectly through perceived utilitarian and hedonic value. This research identifies the ostentation of social status and demonstration of uniqueness as two dimensions of conspicuous consumption and refines scales to measure them. In addition, this research identifies conspicuous consumption as a driver of new product adoption intention and recognizes the mediating role of perceived value.

Effect of Diet Regimen of Sasang Constitution on Health Status (사상체질별 식이 섭생이 건강에 미치는 영향 -한방건강증진센터 시범운영을 위한 기초연구-)

  • This study, as a basic research to manage a Chinese Medicine Health Promotion Center by way of showing an example, is a before and after experiment research for simple group to verify a difference with cholesterol, health status and perception of health in order to confirm a effectiveness of diet and regimen according to the 4th status of physical constitution. Research object was chosen of 42 persons who operate a physical constitutional dietary regimen among them after selecting professors and clinical nurses (55 persons) majoring in the science of nursing who participated in Chinese Medicine-oriented Nurse Training Course from Aug. of 2001 to Feb. of 2002 all over the country. Diagnostic tools for physical constitution was used of the questionary that is currently consisted of physical constitution grouping test in Eastern & Western Diagnose Center of K Medical Center, and rating of health status was used of the tool that standardized CMI(Cornell Medical Index) to be available for Korean, and perception measurement for health status was used of a visual analogue scale for the health status that each one perceive personally, and physiological status was measured of cholesterol in blood. Analysis for the collected data was carried out by percentage, $X^2$ test, paired t-test according to research object by using SPSS, and the results of this study are as follows. 1) There was no difference with cholesterol before or after the experiment for objects. As a result of estimation about difference with health status by areas before or after the experiment, there are more improved result in eyes, ears, digestive organs, bones and sinews organs, frequency of a disorder, habit, adaptation status, angry, healthy status than before the experiment. As a whole, after the experiment the health was more improved than before the experiment. As the result to inspect a difference of health perception between before and after experiment, after the experiment the health perception level was improved than before, however there was no meaningful differences. 2) As the result to inspect a difference of cholesterol between before and after experiment according to object's physical constitution, in the case of So-yang-in(a person with the minimum male: according to the male and female principles(the sun and the moon)) among the 4th status of physical constitution there was only meaningful difference statistically, however, after the experiment their cholesterol's value was increased. As the result to inspect the difference of health status between before and after the experiment according to physical constitution, all of Ta-um-in(a person with maximum the female), So-yang-in(with the minimum male), So-um-in(with the minimum female) had a meaningful difference before and after the experiment, which means that in all case by physical constitutional groups, after the experiment their health status was more improved than before the experiment. As the result to inspect a difference of health perception between before and after according to physical constitutions, in the case of Tae-um-in and So-um-in, average score after the experiment was risen than before the experiment so that it means that the level of health perception was improved, however, there was no meaning statistically. According to the above results, if continuous diet and regimen by each physical constitutions could be implemented, it is certain that the health could be maintained and promoted. And, what we are healthy is for oneself to feel it subjectively. However, I think that cholesterol score in blood that we can view objectively could be changed distinctly if we can implement a strict diet and regimen. Accordingly, it is necessary for a method and period of experiment to be more strict and longer. According to the above results, I would like to suggest as follows. 1) In order to understand health status by Korean's physical constitutions and to generalize it, these research will be repeated against much more objects that could be selected by proper grouping method to consider a representative. 2) It is necessary for a research to inspect health status by physical constitution by developing a health status measurement tool that has higher confidence and propriety based on physical constitutional theory.

Community Structure and Health Assessment of Macrobenthic Assemblages at Spring and Summer in Cheonsu Bay, West Coast of Korea (천수만에 서식하는 대형저서동물의 춘계와 하계의 군집구조 및 건강도 평가)

  • This study was performed to investigate the community structure and health assessment of macrobenthic assemblages in Cheonsu Bay, West Coast of Korea. Total species number was 224 and mean density was $1,940ind./m^2$, both of which were dominated by polychaete worms (111 species and $1,213ind./m^2$). In terms of density, the major dominant species were the bivalve Theora fragilis ($280{\pm}213ind./m^2$), the polychaetes Heteromastus filiformis ($254{\pm}218ind./m^2$), Lumbrineris longifolia ($216{\pm}41ind./m^2$), Mediomastus californiensis ($158{\pm}97ind./m^2$) and the amphipod Eriopisella sechellensis ($167{\pm}100ind./m^2$). As a result of community statistics (cluster analysis and nMDS ordination), the macrobenthic community structure was classified two major groups (inner and mouth of bay) and two stations (station 2 near Ganwol dyke in spring and summer). The Benthic Pollution Index (BPI) and AZTI's Marine Biotic Index (AMBI) were applied to assess the benthic ecological status. During the sampling periods, the BPI Index values varied between 11 (bad status) and 92 (high status). On the other hand, the AMBI Index values was ranged between 1.34 (good status) and 4.39 (poor status). In conclusion, in terms of the mean BPI and AMBI Index values, the ecological status of Cheonsu Bay were moderate status (ecotone stage) and good status, respectively.
