Studies on Protein Contents and Enzyme Activities of Human Seminal Plasma (사람정장(精漿)의 단백질농도(蛋白質濃度)와 효소활성도(酵素活性度)에 관한 연구(硏究))

  • On the basis of the semen analysis in 66 subjects, they were divided into six different groups: Group I consisted of 16 normal subjects with sperm counts of over 40 ${\times}10^6$/ml and motility of over 40 percent, Group II, 7 subjects with normal sperm counts, but motility of under 40 percent, Group III, 15 oligospermic patients with under 40 ${\times}10^6$/ml, Group IV 14 azoospermic patients, Group V, 10 patients with vasectomy and Group VI, 4 abnormal patients with 2 cases of hypoplastic testis, 1 case of Klinefelter's syndrome and 1 case of testis tumor. After seperation of semen into sperm and seminal plasma by centrifugation, the protein contents and the activities of hyaluronidase, ${\beta}$-N acetylglucosaminidase, ${\beta}$-glucuronidase, arylsulfatase, acrosin and azocoll proteinase in seminal plasma were measured. Vasectomy group has 30 percent less of total protein than normal group. For the comparison of enzyme activities of seminal plasma, it could be assumed that the enzymes in seminal plasma were not contaminated with the enzymes of spermatozoa by testing the enzymes of the seminal plasma from the vasectomy and azoospermic groups. It had been reported that hyaluronidase was only released from spermatozoa, however, the result obtained in this investigation showed that azoospermic and vasectomy group had high specific activities of hyaluronidase. The results indicated that hyaluronidase was not only from the testis but also from the male accessory sexual glands. Oligospermic group (Group III) showed the lowest total activity of hyaluronidase among them. The specific activities of ${\beta}$ -N-acetylglucosaminidase was high in oligospermic group (Group III) and low in vasectomy group (Group V). These results were contradictory with the pattern of hyaluronidase activities. This indicated that the spermatozoa which were stayed in epididymis would increase the activity of this enzyme. The specific activity of ${\beta}$ - glucuronidase was low in oligospermic and vasectomy groups. Group VI including testis tumor had remarkably high arylsulfatase activity. Arylsulfatase, a typical lysosomal enzyme, has been known to be released unusually large amounts from certain tumor cells. Arylsulfatase was also released with high activities from azoospermic and vascetomy group. This result indicated that this enzyme was also released from the sources other than testis. Acrosin, a proteolytic enzyme locating in the sperm acrosome, was not found throughout all the samples of seminal plasma. The activities of azocoll proteinase, a non-specific neutral proteinase was nearly identical in all the groups. This enzyme must have been released from the sources other than testis.

Effects of Tropical Climate on Reproduction of Cross- and Purebred Friesian Cattle in Northern Thailand

  • In the first part of the study, rates of estrus occurrence and success of A.I. service in the Thai-native and Friesian crossbred, and purebred Friesian cows fed in the National Dairy Training and Applied Research Institute in Chiang Mai, Thailand were traced monthly throughout a year. An electric fan and a water sprinkler cooled the stall for the purebred cows during the hot season (March-September). Both rates in pure Friesians were at their highest in the cold-dry season (October- February), but they decreased steadily during the hot-dry season (March-May) and were at their lowest in the hot-wet season (June-September). Seasonal change of a similar pattern was observed in the incidence of estrus, but not in the success rate of insemination in the crossbred cows. By the use of reproductive data, compiled in the same institute, on the 75 % cross- and purebred Friesian cows, and climatological data in Chiang Mai district, effects of ambient temperature and humidity on the reproductive traits of cows were examined by regression analysis in the second half of the study. Significant relationships in the crossbred, expressed by positive-linear and parabola regressions, were found between reproductive parameters such as days to the first estrus (DTFE), A.I. service (DTFAI), and conception, the number of A.I. services required for conception and some climatic factors. However, regarding this, no consistent or intelligible results were obtained in purebred cows, perhaps because electric fans and water sprinklers were used for this breed in the hot season. Among climatic factors examined, the minimum temperature (MINT) in early lactation affected reproductive activity most conspicuously. As the temperature during one or two months prior to the first estrus and A.I. service rose, DTFE and DTFAI steadily became longer, although, when MINT depleted below $17-18^{\circ}C$, the reproductive interval tended to be prolonged again on some occasions. The maximum temperature also affected DTFE and DTFAI, but only in limited conditions. The effect of humidity was not clear, although the inverse relationship between DTFE and minimum humidity during 2 months before the first estrus in the crossbred seemed to be significant. Failure to detect any definite effect of climate on the reproductive traits of pure Friesians seemed to indicate that forced ventilation by electric fans and water sprinklers were effective enough to protect the reproductive ability of this breed from the adverse effects of a hot climate.

  • Gametic biotechnologies involve the procedures which are utilized for procuring reproductive success through the mimicry of in vivo events as in in vitro fertilization, embryo transfer etc. With the realization that the oviduct performs most of the procedures mimicked in vitro under normal in vivo situations, the need to master the oviduct therefore, becomes paramount. The oviduct being an exocrine gland (with its output of glycoproteins) and possibly an ecdocrine gland must be implicated in all the preimplantational procedures of reproduction, which include ovulation, oocyte maturation, sperm capacitation, gametic and embryonal nutrition, fertilization, and implantation. The evidences in the literature for the implication of the oviduct in these processes are examined. It is concluded that there is a need for the mastery of oviductual activity in order to maximize the successes of the procedures in vitro, and provide gametic manipulations which will have high success rates in implantation that is the ultimate after of in vitro fertilization for reproductive success.

Pathogenic effects of porcine reproductive and respiratory syndrome virus isolates in swine tracheal ring culture

  • Pathogenic effects of 29 different porcine reproductive and respiratory syndrome(PRRS) virus isolates were investigated in swine tracheal ring(STR) cultures by examining their effects on the ciliary activity of STR. Inhibition of ciliary movement and destruction of the tracheal epithelium were seen between 72 and 96 hours postinoculation(PI). Virus replication was demonstrated by examining viral infectivity of the supernatants from the STR cultures. PRRS virus antigen in macrophages was detected by a streptavidin-biotin complex(ABC) immunoperoxidase method. Of the 29 PRRS virus isolates, 8 isolates were classified into pathogenic, and the remaining 21 isolates were determined as mildly pathogenic or apathogenic viruses. These results suggest that STR examination may be used as a method for predicting pathogenic variability of PRRS virus isolates.

  • The recent knowledge about the effects and the biological roles of prostaglandins(PGs) in the female reproductive system were reviewed on the following view points in detail and the authors also argued on the efficacy and reliability for the induction of labour and therapeutic abortions in midtrimester pregnancy. 1. The actions of PGs on the myometrium in vivo and in vitro and the effects of ovarian hormones on the actions of PGs. 2. The actions of PGs on the fallopian tube. 3. The role of PGs in the initiation of menstrual bleeding and dysmenorrhea. 4. The biological action of PGs on the mechanism of labour and parturition. 5. The efficacy and reliability of PGs for therapeutic abortion in the first and midtrimester pregnancy. 6. The biological role of PGs in the activity and life-span of corpus luteum.

  • To estimate the penile blood flow changes during artificial with erection intracorporeal papaverine injection, we injected 99 m Technitium-Human Serum Albumin via antecubital vein simultaneously and calculate the radioisotope activity of the penis using computerized Gamma Camera. We classified the results into 3 groups according to the transit time, erection persistent time and venous index in 67 impotence patients. We defined the arteriogenic impotence with the delayed transit time (more than 600 seconds) in 13 patients and venogenic with the shortened erection persistant time (less than 200 seconds) and decreased venous index (less than 0.8) in 11 patients. The rest of them (43 patients) were regarded as equivocal impotence, probably nonvasculogenic or psychogenic.

  • Objective : The purpose of this report was to provide the information activity and safety of Palmul-tang by analyzing domestic/international papers and theses about Palmul-tang, Methods: Domestic/international papers and theses related to Palmul-tang were reviewed and analyzed, These papers were then classified by year, experimental method and subject, Results: The following results were obtained in this study. 1. The study of Palmul-tang started from 1985 and was continuously increased. The study was mainly forcused on experimental model rather than clinical study. 2. As these studies were classified by subject, papers related to immune intensification were most abundant by 20 papers, Besides there were several papers related to cardiovascular activity, reproductive activity, anti-apoptotic activity and cerebral hemodynamics. 3. Among the papers related to immune intensification. the studies on recovery of fatigue were mostabundant by 10 papers and the studies of on immune cell and cytokine express were six. In addition to. several studies were associated with anti-cancer activity and anti-allergic activity. Recovery of fatigue was determined by measurement of fatigue markers in a living body such as lactate. CPK, pyruvate and triglyceride and assessment of exercise capability of animals such as swimming test. slopped plate test. Rota-rod test, and activity cage test after Palmul-tang treatment. 4. According to experimental data. it is supported that Palmul-tang has been used as Qi and Blood intensifier with immune intensification and recovery of fatigue. 5. The paper related to safety of Palmul-tang was only one paper which is studied on acute toxicity of Palmul-tang with experiment with ICR mouse. There was no study on evaluating safety by observing liver and kidney functions after Palmul-tang treatment Conclusion: Palmul-tang is being used in various ways associating with immune intensification. cardiovascular activity and reproductive activity. However. studies on efficacy and mechanism of Palmul-tang should be conducted at the molecular biology level and studies on safety of Palmul-tang need to be completed at the clinical level.

Relation of Breast Cancer Risk with Alcohol Consumption and Physical Activity : A Case-Control Study (음주 및 육체적 활동과 유방암 발생의 관련성 : 환자-대조군 연구)

  • This study was conducted to investigate the relationship between alcohol consumption, physical activity and breast cancer risk in Korean women. Newly histologically identified cases (N = 108) were selected at Hanyang and Soonchunhyang University Hospitals in Seoul, from January 1998 to August 1999. Hospital-based controls (n = 121) were the patients who visited at the same hospital in the department of plastic surgery, general surgery and opthalmology. Subjects were asked informations on demographic, reproductive, and history of breast feeding as well as physical activity, exercise habit, alcohol consumption by individual interview. Odds ratio (OR) and 95% Confidence Intervals (CI) were calculated by using unconditional logistic regression after adjusted for confounding factors. Among postmenopausal women, the risk of breast cancer increased with increasing alcohol drinking: relative risk for the highest group ( $\geq$ 150 g per week) compared with the lowest group (never drinking) was 2.05 (CI=1.74-3.79). Association of exercise and high physical activity for breast cancer appeared to be limited to women with high physical index among postmenopausal women. We observed decreasing risk of breast cancer with increasing physical activity index among postmenopausal women (OR=0.49,95% CI=0.56-0.92). Results of this study support a protective effects of physical activity during adulthood for breast cancer. (Korean J Nutrition 36(1): 40-48, 2003)

Effects of dietary supplementation with Pediococcus acidilactici ZPA017 on reproductive performance, fecal microbial flora and serum indices in sows during late gestation and lactation

  • Objective: This study was conducted to determine the effects of dietary supplementation with Pediococcus acidilactici (P. acidilactici) ZPA017 as a probiotic on reproductive performance, fecal microbial flora and serum indices in sows during late gestation and lactation. Methods: A total of 94 sows (Large White×Yorkshire, average 4.50 parities) were randomly allotted to two dietary treatments: control diet and the diet supplemented with P. acidilactici ZPA017 (2.40×109 colony-forming unit/kg of diets). The study started at d 90 of gestation and conducted until d 28 of lactation. Results: Compared to sows fed the control diet, supplementation of P. acidilactici ZPA017 increased the number of weaning piglets, weaning weight of litter and piglets, survival rate of piglets at weaning (p<0.05), and decreased diarrhea rate of piglets in lactation (p<0.05). Dietary P. acidilactici ZPA017 increased fecal Lactobacillus populations (p = 0.030) and reduced fecal Escherichia coli and Staphylococcus aureus populations (p<0.05) of sows at weaning. Moreover, the supplementation of P. acidilactici ZPA017 increased serum concentrations of immunoglobulin G, immunoglobulin A and total protein (p<0.05), while decreased serum haptoglobin concentration and alanine aminotransferase activity (p<0.05) of sows at weaning. Conclusion: Administration of P. acidilactici ZPA017 in diets during late gestation and lactation had positive effects on the reproductive performance, intestinal microflora balance and immunity of sows.

The Control Mechanism of Gonadotropin-Releasing Hormone and Dopamine on Gonadotropin Release from Cultured Pituitary Cells of Rainbow Trout Oncorhynchus mykiss at Different Reproductive Stages

  • The mechanism by which gonadotropin-releasing hormone (GnRH) and dopamine (DA) control gonadotropin (GTH) release was studied in male and female rainbow trout using cultured pituitary cells obtained at different reproductive stages. The mechanisms of follicle-stimulating hormone (FSH) release by GnRH and DA could not be determined yet. However, basal and salmon-type GnRH (sGnRH)- or chicken-II-type GnRH (cGnRH-II)- induced luteinizing hormone (LH) release increased with gonadal maturation in both sexes. LH release activity was higher after sGnRH stimulation than cGnRH-II stimulation at maturing stages in both sexes. The GnRH antagonist ([Ac-3, 4-dehydro-$Pro^1$, D-p-F-$Phe^2$, D-$Trp^{3,6}$] GnRH) suppressed LH release by sGnRH stimulation in a dose-dependent manner, although the effect was weak in maturing fish. The role of DA as a GTH-release inhibitory factor differs during the reproductive cycle: the inhibition of sGnRH-stimulated LH release by DA was stronger in immature fish than in maturing, ovulating, or spermiated fish. DA did not completely inhibit sGnRH-stimulated LH release, and DA alone did not alter basal LH release. Relatively high doses ($10^{-6}$ or $10^{-5}M$) of domperidone (DOM, a DA D2 antagonist) increased LH release, which did not change with reproductive stage in either sex. The potency of DOM to enhance sGnRH-stimulated LH release was higher in maturing and ovulated fish than in immature fish. These data suggest that LH release from the pituitary gland is controlled by dual neuroendocrine mechanisms by GnRH and DA in rainbow trout, as has been reported in other teleosts. The mechanism of control of FSH release, however, remains unknown.
