Study on DNase activity specific from Haemonchus contortus reproductive tissue (염전위충(Haemonchus contortus) 생식기관의 DNase활성에 관한 연구)

  • DNase activity in Haemonchus contortus reproductive tissue was characterized and compared to that in whole worm. DNase activity in reproductive tissue was detected throughout pHs 4-10 with high activity under acidic conditions. The activity was not inhibited by 10 mM EDTA at pH 5.0, but largely inhibited by pH 7.0. The activity produced DNA fragments with mixtures of 3'-hydroxyls (OH) and 3'- phosphates (P) at each pH. Three distinct DNase activities were identified and had $M_rs$ of 34, 36 and 38.5 kDa in zymograms, which were distinguished according to pH requirement and sensitivity to EDTA. Among them, the 36 kDa reproductive tissue DNase had predominant activity at pH 5.0, but very weak at pH 7.0, and this activity was not inhibited by EDTA at pH 5.0. These characteristics of the 36 kDa reproductive tissue DNase resemble those of classic acidic DNases. In contrast, 36 kDa whole worm DNase activity had high activity at both pH 5.0 and 7.0. While the 36 kDa DNase activity at pH 5.0 was similar in both reproductive tissue and whole worm samples, the activity at pH 7.0 was predominantly detected in whole worm sample. This suggests that the 36 kDa whole worm DNase at pH 5.0 differs from that at pH 7.0. Thus, results indicate that the EDTA-insensitive 36 kDa DNase at pH 5.0 is specific for H. contortus reproductive tissue.

  • Plasminogen activators (PAs) are serine proteases that convert plasminogen to plasmin. Two type of PAs are urokinase-type PA (uPA) and tissue-type PA (tPA). Plasminogen is present in most extracellular fluids. PAs play in various reproductive processes including implantation, ovulation and fertilization. In the spermatozoa, PAs and PAIs play a role in sperm motility and fertilization. PAs in the sertoli cell are stimulated spermatozoa maturation and sperm activation through the phospholipase A2. The oocyte maturation is the process for fertilization and implantation. PAs in cumulus-oocyte complexes (COCs) are related to oocyte maturation by protein kinase A and C. In the ovulatory process, PAs activity are changed and it are related to reducing the tensile strength of ovarian follicle wall. The uterine environment is important for reproduction and the uterus undergo tissue remodeling. In the uterus and oviduct of mammals, expression and activity of PAs are changed during estrous cycle. Thus, expression and activity of PAs are concerned to many reproductive functions. Therefore, PAs seem to important factor of regulator in reproductive events.

Evaluation of some indigenous plant extracts for antiimplantation activity in albino rats

  • In the present investigation twelve indigenous medicinal plants have been screened for their antiimplantation activity in albino rats. The plant material was subjected for soxhlation successively and separately from non-polar solvents to polar solvents i.e., petroleum ether benzene and ethanol. Out of these three extracts the petroleum ether extract of seeds of Citrus medica, aerial part of Oxalis corniculata and Tinospora cardifolia have showed maximum antiimplantation activity. Ethanol extract of leaves of Cardiospermum helicacabum, roots of Echinops echinatus, leaves of Melia azedarach, seeds of Momordica charantia and bark of Terminalia bellirica have shown maximum antiimplantation activity amongst the three extracts of each plant material screened. Though all the three extracts of seeds of Annona squamosa and leaves of Zizyphus jujube screened for antiimplantation activity, no extract has showed any loss in implantation. The details of the results obtained are discussed.

Gender Differences and Relationships among Lifestyle and Reproductive Health in University Students (성별에 따른 대학생의 생활습관과 생식건강과의 관계)

  • Purpose: University students happen to be in a transitional period at the beginning of one's adult life and thereby establish the basis for their health care. The negative lifestyles followed by students during this period can also affect their reproductive health. The purpose of this study was to identify lifestyle, reproductive health, gender differences and relationships between lifestyle and reproductive health in university students. Methods: We used a descriptive cross-sectional design. A total of 300 subjects were enrolled. Data were collected using structured questionnaires between October 11 and 25, 2017 and analyzed using SPSS 25.0. Subjects agreed to undergo a face-to-face interview, including administration of the Health Promotion Lifestyle Profile II (HPLP-II) and reproductive health (knowledge, attitude, and behaviors). Results: The mean age of the subjects was 21.4 years. HPLP-II and reproductive health behaviors were significantly different between the genders. The scores of physical activity and nutrition in females were significantly lower than males. The scores of safe sex and sexual responsibility in females were significantly higher than males, and the score of genital health management was significantly lower in females than males. High HPLP-II score was observed to be in correlation with high reproductive health attitudes and behaviors. Conclusion: The result revealed differences in lifestyle and reproductive health between both the genders. For improvement of reproductive health of university students, provision of lifestyle intervention including healthy nutritional habits and physical activity is imperative.

Protein Expression of Matrix Metalloproteinases of Mouse Reproductive Organs During Estrous Cycle (생식주기에 따른 자성 생쥐의 생식기관의 Matrix Metalloproteinase의 단백질 발현)

  • Protein expression patterns of matrix metalloproteinases (MMPs) were examined in mouse reproductive organs during estrous cycle. Estrous cycle was classified into diestrus, proestrus, estrus or metestus and MMP expression was analyzed by zymography using gelatin as a substrate. Uterine fluid (UF) obtained both at diestrus and proestrus exhibited 4 major MMPs including 106kDa, 64kDa, 62kDa and 59kDa gelatinases. However, in UF at estrus, the gelatinolytic activity of 64kDa MMP disappeared and that of 106kDa and 62kDa MMPs dramatically decreased. At metestrus, 64kDa MMP activity reappeared and 106kDa and 62kDa MMP exhibited increased activities such that the band intensity of 106kDa was comparable to that in UF at diestrus. Gelatinolytic activity of 59kDa MMP was not changed throughout the cycle. Both ovarian and oviductal tissue homogenate revealed 4 MMPs which corresponded to the 4 MMPs of UF. However, unlike UF MMPs, gelatinolytic activity of these MMPs did not show distinct changes throughout the cycle. Either an inhibitor of MMP, 1,10-phenanthroline, or a metal chelator, EDTA, abolished the appearance of the above MMP activities in gelatinated gel whereas a serine proteinase inhibitor, phcnylmethylsulfonyl fluoride, failed to inhibit the appearance of MMP activities, proving that gelatinolytic activity of the above reproductive tissues were due to the enzymatic activity of MMP. When gclatinolytic activity of mouse serum was examined, it revealed 5 MMPs (131kDa, 106kDa, 89kDa, 64kDa and 62kDa bands) and one gelatinase (84kDa) band. From these results, it is concluded that the protein expression of MMPs of mouse reproductive organs, particularly uterus, is temporally regulated during estrous cycle and uterine 106kDa, 64kDa and 62kDa MMPs are suggested to play an important role in cyclic tissue remodeling of mouse uterus.

The Potential Regressive Role of Syzygium aromaticum on the Reproduction of Male Golden Hamsters

  • The flower buds of Syzygium aromaticum (clove) have been used as traditional medicine for the treatment of male sexual disorders in Asian countries. Recently, there are some reports about the effects of the clove on reproductive activities in mammals. Therefore, its effect on testicular function was examined in male golden hamsters whose reproductive activity is inhibited by photoperiod such as winter climate. The male animals were given by daily oral administrations (56 consecutive days) in three doses (4 mg, 20 mg, and 100 mg/kg BW) of the alcoholic extract of the clove. Generally lower dose (4 mg) of the extract continued to keep the reproductive activities of testes. The both middle and high doses (20 mg and 100 mg) of the extract completely inhibited the testicular activity in some animals. Taken together, these results suggest a possible biphasic action of alcoholic extract of Syzygium aromaticum flower bud on testicular function.

Investigation of trunk conformation and reproductive activity in Donggyeongi (동경이의 체형 및 번식 능력에 대한 실태조사)

  • The trunk conformations of the Gyeongju Donggyeongi (GD), Korean native dog was measured in the present study. Also, the information of reproductive activity was investigated in GD dogs. A total of 73 dogs were randomly selected out of 121 dogs in the Gyeongju County in July 2010, which were registered by inserting electronic chips in the withers. The hair colors of the GD were white, yellow, black and tiger brindle, the mean withers heights were $45.46{\pm}4.17cm$ and $49.28{\pm}3.56cm$ for female and male dogs, respectively The mean body length to withers height ratios were 117.63% and 116.4% for the female and male dogs. Concerning the reproductive capability of the GD, the mean litter number was $4.6{\pm}1.7$, the optimal mating time was $12.5{\pm}2.3$ days after the rutting season and the mean gestation period was $60.2{\pm}2.8$ days. The results of this study would be useful for compiling the official standard for the adult GD.

Effect of Crotalaria juncea seed extracts on the estrous cycle and ovarian activity in albino mice

  • Petroleum ether, benzene and alcohol extracts of seeds of C. juncea administered orally at the dose level of 25mg/100g body weight to adult female mice for 30 days, resulted in irregular estrous cycle with prolonged estrus and metaestrus and reduced diestrus and proestrus during the experimental period. Histological studies of the ovary indicate increases in the number of atretic follicles but decreases in the number of developing follicles, Graafian follicles and corpora lutea. The total cholesterol content of the ovary is increased, whereas ascorbic acid content is decreased. The weight of the uterus and its micrometric measurement in all experimental mice are increased significantly. The alcoholic extracts showed estrogenic activity in immature mice by early opening of the vagina, premature cornification of the vaginal epithilium and increases in uterine weight. However, alcohol extract of seeds of C. juncea was more effective in causing these changes compared to other extracts. After subjecting to preliminary phytochemical screenings alcohol extract showed positive; test for alkaloids, steroids, glycosides, flavones, phenols and tannins.

Studies on the serochemical status in dairy Holstein cows of reproductive disorder (불임유우의 혈청화학치에 관한 연구)

  • To make index on therapy or releasing from dairy holstein cows of reproductive disorder, the levels of serological sex hormones and total ALP, GOT, GPT, and serum protein were measured. The results obtained through the experiments are summarized as follows ; 1. The levels of FSH and estradiol in reproductive disorder group were significantly higher and lower than those in pregnant group and control group, respectively. (p<0.05) 2. The level of prolactin was showed mild difference in three groups and that of progesterone was reasonably higher in pregnant group with significance (p<0.05). 3. The activity of GPT was significantly higher and that of GOT was showed higher In reproductive disorder group, respectively ( p<0.05) 4. The level of total ALP was higher in reproductive disorder group, however no difference was showed between pregnant and control group. 5. The level of serum protein was followed as control, pregnant, and reproductive disorder with insignificance.
