Comparative Evaluation of 25-OH-VitD and 25-OH-VitD3 by Radioimmunoassay (방사면역 측정법에 의한 25-OH-VitD와 25-OH-VitD3의 비교 평가)

  • Purpose Vitamin D is essential for maintaining bone health, controling cell proliferation or differentiation, strengthening immune function by controlling calcium metabolism in the body. Vitamin D deficiency can lead to increase the risk of rickets, osteoporosis, cardiovascular disease, diabetes and cancer. Especially, South Korea is one of the highest population proportion of vitamin D deficiency. Accurate determination of levels of 25-OH-VitD or 25-OH-VitD3 in blood serum is required for the diagnosis and treatment of vitamin D deficiency. In this study, radioimmunoassay of 25-OH-VitD and 25-OH-VitD3 was performed and compared to evaluate the effectiveness of Vitamin D radioimmunoassay. Materials and Methods Serum 25-OH-VitD and 25-OH-VitD3 levels were measured using radioimmunoassay. The interrelationship, reproducibility and population distribution rate were evaluated. In addition, the internal quality control was performed at Asan Medical Center from April 2017 to June 2019 and the result of external quality control (Interagency proficiency evaluation) of first and second half of 2018 hosted by the Korean Society of Nuclear Medicine Technology (KSNMT). Both tests were measured by same manufacturer's reagent. Results 25-OH-VitD showed a strong positive correlation on 97 samples, as 25-OH-VitD3 x 0.9 + 0.3 (R>0.9). In repeated measurement, the average Diff(%) value of the reproducibility evaluation of 25-OH-VitD and 25-OH-VitD3 were 7.7% and 7.4%, respectively. Population distribution results showed no statistically significant differences(p>0.05). The resultant value of internal quality control, which measured from April, 2017 to June 2019 in Blood test room of Nuclear Medicine at Asan Medical Center, showed average (CV%) 6.2% and 6.8%, respectively. As a result of the external quality control (interagency proficiency evaluation) Z value obtained under 2.0, as shown below; Conclusion The interrelationship, reproducibility, population distribution rate, internal quality control and external quality control between 25-OH-VitD and 25-OH-VitD3 radioimmunoassay shows superior outcome. Radioimmunoassay, which can be alone measured in the blood as 25-OH-VitD or 25-OH-VitD3, is considered suitable screening tests for the diagnosis of vitamin D deficiency.

An Analysis of Performance Error of High Precision Measuring Instrument (진원도 측정기의 오차특성에 관한 연구)

  • A phase evil method and spectrum analysis were instrument error which is originated from measurement system and the form error of standard specimens. An instrument with a rotating table supported by an air bearing is calibrated using standard specimens. The phase of standard specimens was measured 12 times on the rotating table with rotating 30 in turn and its measurement magnification was set by 100000 times. As a result of data analysis of all the observations, read out at each of 144 orientations(per 2.5) from recorded datafiles, the error of the performance of the instrument and those of the standard specimens are evaluated and a systematic deviation of the instrument is determined. In the particular instrument used in the present experiment, the deviation of the instrument is determined with the accuracy of 15nm and those of standard specimens with the accuracy of 23, 13 n, respectively. The reproducibility of the instrument is investigated, too. If the instrument is calibrated by using the above standard specimens, then the accuracy of the measurement of roundness error can be improved to about 15nm.

Validation of Customized Cancer Panel for Detecting Somatic Mutations and Copy Number Alterations

  • Accurate detection of genomic alterations, especially druggable hotspot mutations in tumors, has become an essential part of precision medicine. With targeted sequencing, we can obtain deeper coverage of reads and handle data more easily with a relatively lower cost and less time than whole-exome or whole-genome sequencing. Recently, we designed a customized gene panel for targeted sequencing of major solid cancers. In this study, we aimed to validate its performance. The cancer panel targets 95 cancer-related genes. In terms of the limit of detection, more than 86% of target mutations with a mutant allele frequency (MAF) <1% can be identified, and any mutation with >3% MAF can be detected. When we applied this system for the analysis of Acrometrix Oncology Hotspot Control DNA, which contains more than 500 COSMIC mutations across 53 genes, 99% of the expected mutations were robustly detected. We also confirmed the high reproducibility of the detection of mutations in multiple independent analyses. When we explored copy number alterations (CNAs), the expected CNAs were successfully detected, and this result was confirmed by target-specific genomic quantitative polymerase chain reaction. Taken together, these results support the reliability and accuracy of our cancer panel in detecting mutations. This panel could be useful for key mutation profiling research in solid tumors and clinical translation.

  • Not only the emission regulation of on-road vehicle engine, but also emission regulation of off-road engine have been reinforced. It is the reason of wide application of emission reduction technology for off-road engines. In this study, optimization of engine parameters (Injector hole number, Injection timing and EGR rate) for reduction of NOx and smoke emissions were conducted by using the analysis of sensitivity and S/N ratio of Taguchi method(DOE). As results, this paper shows optimum value of the parameters for NOx and smoke emission reduction. From the result of reproducibility verification, it is final that the prediction value of NOx and smoke has the error of below 10%. Consequently, the method and results of this study will be used for quantitative reference to EGR control mapping in next study.

Prediction of rock fragmentation and design of blasting pattern based on 3-D spatial distribution of rock factor

  • The optimum blasting pattern to excavate a quarry efficiently and economically can be determined based on the minimum production cost, which is generally estimated according to rock fragmentation. Therefore, it is a critical problem to predict fragment size distribution of blasted rocks over an entire quarry. By comparing various prediction models, it can be ascertained that the result obtained from Kuz-Ram model relatively coincides with that of field measurements. Kuz-Ram model uses the concept of rock factor to signify conditions of rock mass such as block size, rock jointing, strength and others. For the evaluation of total production cost, it is imperative to estimate 3-D spatial distribution of rock factor for the entire quarry. In this study, a sequential indicator simulation technique is adopted for estimation of spatial distribution of rock factor due to its higher reproducibility of spatial variability and distribution models than Kriging methods. Further, this can reduce the uncertainty of predictor using distribution information of sample data. The entire quarry is classified into three types of rock mass and optimum blasting pattern is proposed for each type based on 3-D spatial distribution of rock factor. In addition, plane maps of rock factor distribution for each ground level are provided to estimate production costs for each process and to make a plan for an optimum blasting pattern.

Sources of Variability of Multiple Self-reported 24-hour Recalls in Dietary Survey (식이조사방법중 자기 기록식 24시간 회상 반복 조사의 변이원에 관한 연구)

  • This study was designed to study sources of variability(subjects, day by day, season, day of week, sequence, number of repeated days, compliance etc.) in multiple 24-hour dietary recall method. Dietary intakes of college women in Seoul were obtained using 24-hour dietary recalls, consecutive 7 days, 4 times for 1 year. The result of this study shows that 1) the reproducibility of multiple 24-hour recalls is low(intraclass correlation coefficient<0.3), 2) the intraindividual variations of all nutrients are more than 80%, 3) the major components of variation are interindividual vriability and intraindividual variability(within person variability and methodologic error). There were small but statistically significant season and compliance effects. 4) the ratio within-person varation to between-person variation for absolute and transformed nutrient intakes except some nutrients such as vitamin A, vitamin C, energy from protein and fat etc, was less than 1.7 the minimum numbers of days needed in 24-hour dietary recalls to estimate usual intake for this group were 1 to 21 days. And the numbers of days needed to get the usual intake of an individual varied among individuals and within individuals for different nutrients, ranged from 1 days for log carbohydrate(% energy) to 470 days for Na. 6) There were greater than 0.7 correlation coefficients between the average nutrient intakes of 12 days and intakes of 28 days. Therefore, it was desirable to take the 24-hour recall more than 12 times, repeating 3 days every season.

Development of the Automatic Control System for the Advanced Phosphorus Treatment in Sewage Treatment Plant (하수처리시설에서 인 고도처리를 위한 자동제어시스템 개발에 관한 연구)

  • It has a limitation to satisfy the phosphorus effluent criteria of 0.2 mg/L which will be reinforced from 2012 with the Biological Nutrient Removal (BNR) process. The chemical coagulation process has been operated in parallel with the biological treatment process for advanced treatment of phosphorous in the developed countries including Europe. However, the coagulation process has some disadvantages such as the desired goal may not be achieved without injecting the optimum dosage of the coagulant. This study developed the automatic control system to inject the optimum dosage of phosphorous coagulant into the coagulation process. The adopted coagulant was 10% Poly Aluminum Chloride (PAC) in this study. The automatic control system developed in this study was adopted for the treatment of the phosphorus from the effluent in SBR process. The automatic control system was composed of the data receiving part, the optimum coagulant dosage control part and the data transmit part. The result of the phosphorous advanced treatment of the SBR effluent using the automatic control system showed the removing efficiency over 95% consistently with the phosphorous concentration under 0.02 ~ 0.15 mg/L. The reproducibility analysis for checking the safety of automatic control system showed more than 95% correlation.

Change of Pulmonary Function after Pulmonary Resection (폐절제술후 폐기능 변화에 관한 연구)

  • Pulmonary function studies today are generally accepted as an integral part of the evaluation of poor-risk patients who are to have pulmonary surgery. The effect of various pulmonary surgery on lung function was investigated in 54 patients in whom comprehensive lung function test were performed before and between 2 months and 14 months after operation at the Department of Thoracic Surgery, Seoul National University Hospital. According to the result of analysis, the effect of pulmonary resection on forced flow rate was keeping with the change of lung volume, and the preoperative level of ventilatory function plays a major role in determining postoperative loss of functioning lung. Although all measures of expiratory flow [FVC, FEV1, FEFO.2-1.2, MEF50, FEF25-75] have the same percentage of reproducibility, but FEV1 shows most sensitive, reliable linear correlation with the functioning pulmonary tissue loss than other parameters. The linear regression lines derived from the correlation between preoperative [X] and postoperative [Y] FEV1 on various surgical procedures were as follows: 1. Y = 0.57X 0.03. in pneumonectomy group of lung cancer[r=0.84]. 2. Y = 0.56X + 0.33. in lobectomy group of lung cancer[r=0.79]. 3. Y = 0.69X + 0.25. in lobectomy group of pulmonary infection[r=0.91].

Studies on the Sterol of Sesame Oil Sold In Markets and Used in Restaurants (시중(市中) 참기름의 Sterol에 관(關)한 연구(硏究))

  • Methods for the determination of sterols in sesame oils were studied. The sesame oils were saponified and the sterols isolated from the unsaponifiable matter by Florisil column chromatography, and the individual components were determined by means of gas chromatography. Campesterol, ${\beta}-sitosterol$, stigmasterol were found in sesame oil including unknown Ⅰ and Ⅱ. The use of SE-30 gas chromatographic column allows the slow elution, duplication of peaks and relatively low reproducibility, therefore, 3% OV-17 was suitable for the sterol analysis. The result of this study showed that contents of sterols in sesame oil were campesterol 8.4%, stigmasterol 4.5%, ${\beta}-sitosterol$ 33.9% and others 53.0% involving 8.8% of unknown I and 44.3% of unknown Ⅱ. There has been no specific test available for identifying the sesame oil among common edible oils. But the ratio of sterols in sesame oils allowed the estimation of genuiness. The ratio of sterols vs. campesterol in genuine sesame oils were stigmasterol 0.3- 0.6, ${\beta}-sitosterol$ 3.0-3.8 and unknown Ⅱ 3.0, respectively. The 65 samples were composed of genuine sesame oil 40%, mixed rape seed oil 3%, cotton seed oil 1. 5% others were reused soybean oil or re-extracted oil.

Large Scale Rainfall-runoff Analysis Using SWAT Model: Case Study: Mekong River Basin (SWAT 모형을 이용한 대유역 강우-유출해석: 메콩강 유역을 중심으로)

  • This study implemented the rainfall-runoff analysis of the Mekong River basin using the SWAT (Soil and Water Assessment Tool). The runoff analysis was simulated for 2000~2007, and 11 parameters were calibrated using the SUFI-2 (Sequential Uncertainty Fitting-version 2) algorithm of SWAT-CUP (Calibration and Uncertainty Program). As a result of analyzing optimal parameters and sensitivity analysis for 6 cases, the parameter ALPHA_BF was found to be the most sensitive. The reproducibility of the rainfall-runoff results decreased with increasing number of stations used for parameter calibration. The rainfall-runoff simulation results of Case 6 showed that the RMSE of Nong Khai and Kratie stations were 0.97 and 0.9, respectively, and the runoff patterns were relatively accurately simulated. The runoff patterns of Mukdahan and Khong Chaim stations were underestimated during the flood season from 2004 to 2005 but it was acceptable in terms of the overall runoff pattern. These results suggest that the combination of SWAT and SWAT-CUP models is applicable to very large watersheds such as the Mekong for rainfall-runoff simulation, but further studies are needed to reduce the range of modeling uncertainty.
