The Expression and the Subcellular Localization of Regulatory Subunits of Class IA Phosphoinositide 3-Kinase in L6 Skeletal Muscle Cell

  • PI3-kinase activity through p85, the regulatory subunit of class IA PI3-kinase, is indispensable for the growth, differentiation, and survival of skeletal muscle cells, but little is known about the function of other regulatory subunits such as p55 and p50. We examined the subcellular localization and the expression of the regulatory subunits of class IA PI3-kinase in L6 myoblasts. Both p55 and p50 as well as p85 were expressed in L6 myoblasts. Whereas p85 was localized at both cytosolic and nuclear tractions, p55 and p50 were localized at only the nuclear traction. During the differentiation of L6 myoblasts, the protein concentrations of both p55 and p50 were decreased but that of p85 was not significantly changed. Menadione-induced oxidative stress induced the translocation of p85 from cytosol to nucleus and the increase of p55 expression. These results suggest that the regulatory subunits of class IA PI3-kinase play an important role in L6 myoblasts.

Enhanced Clavulanic Acid Production in Streptomyces clavuligerus NRRL3585 by Overexpression of Regulatory Genes

  • We constructed four recombinant plasm ids to enhance the production of clavulanic acid (CA) in Streptomyces clavuligerus NRRL3585: (1) pIBRHL1, which includes ccaR, a pathway-specific regulatory gene involved in cephamycin C and CA biosynthesis; (2) pIBRHL2, containing claR, again a regulatory gene, which controls the late steps of CA biosynthesis; (3) pGIBR containing afsR-p, a global regulatory gene from Streptomyces peucetius; and (4) pKS, which harbors all of the genes (ccaR/ claR/ afsR-p). The plasmids were expressed in S. clavuligerus NRRL3585 along with the $ermE^*$ promoter. All of them enhanced the production of CA; 2.5-fold overproduction for pIBRHL1, 1.5-fold for pIBRHL2, 1.6-fold for pGIBR, and 1.5-fold for pKS compared to the wild type.

  • When the Internet phone service, generally PC-to-Phone, first entered into the telecommunications service market, no regulatory issues were arisen to manage the service within the regulatory framework because of it's low quality, no phone number assignment and etc. However, almost the same quality, more applications and cheaper price compared with the fixed telecommunications service, have changed the regulator's policy allowing Internet phone service within market mechanism. While incumbent fixed telecommunications service enterprises had given with tremendous duties to continue the job categorized 'facilities-based telecommunication service provider', and which can be unreasonable and unfair if Internet phone service enterprises are allowed to enter into the fixed telecommunications market without any regulation. Thus, new regulatory framework has worked for the Internet phone service named the VoIP service generally in the fixed telecommunications market. Recently, VoIP is provided not only in the fixed telecommunications market but also in the mobile market as Wi-Fi phone is feasible in the wireless LAN environment. Furthermore, bundled service of Wi-Fi and cellular phone service will be launched soon, and the next version will be the mobile VoIP service operating like a cellular phone service. Hence, we will meet similar situation when the VoIP sewice enters into the fixed telecommunications service market. And, in this paper, we prospect the regulatory issues arising when the wireless or mobile VoIP service enterprises enter the mobile market, by analyzing possible scenarios.

  • Regulatory systems for decommissioning of nuclear power plants in several countries, such as Japan, United States of America, Germany, United Kingdom, France, and Republic of Korea, are surveyed. In the survey, regulatory policies, legislations, licensing process, inspection and public involvements for decommissioning are identified and compared. Afterwards, the survey results will be utilized as a reference to establish the improvement directions of domestic regulatory system.

  • After the Fukushima Daiichi nuclear power plant (NPP) accident, new regulatory requirements were enforced in July 2013 and a backfit was required for all existing nuclear power plants. It is required to take measures to prevent severe accidents and mitigate their radiological consequences. The Regulatory Standard and Research Department, Secretariat of Nuclear Regulation Authority (S/NRA/R) has been conducting numerical studies and experimental studies on relevant severe accident phenomena and countermeasures. This article highlights fission product (FP) release and hydrogen risk as two major areas. Relevant activities in the S/NRA/R are briefly introduced, as follows: 1. For FP release: Identifying the source terms and leak mechanisms is a key issue from the viewpoint of understanding the progression of accident phenomena and planning effective countermeasures that take into account vulnerabilities of containment under severe accident conditions. To resolve these issues, the activities focus on wet well venting, pool scrubbing, iodine chemistry (in-vessel and ex-vessel), containment failure mode, and treatment of radioactive liquid effluent. 2. For hydrogen risk: because of three incidents of hydrogen explosion in reactor buildings, a comprehensive reinforcement of the hydrogen risk management has been a high priority topic. Therefore, the activities in evaluation methods focus on hydrogen generation, hydrogen distribution, and hydrogen combustion.

The Construction of Regulatory Network for Insulin-Mediated Genes by Integrating Methods Based on Transcription Factor Binding Motifs and Gene Expression Variations

  • Type 2 diabetes mellitus is a complex metabolic disorder associated with multiple genetic, developmental and environmental factors. The recent advances in gene expression microarray technologies as well as network-based analysis methodologies provide groundbreaking opportunities to study type 2 diabetes mellitus. In the present study, we used previously published gene expression microarray datasets of human skeletal muscle samples collected from 20 insulin sensitive individuals before and after insulin treatment in order to construct insulin-mediated regulatory network. Based on a motif discovery method implemented by iRegulon, a Cytoscape app, we identified 25 candidate regulons, motifs of which were enriched among the promoters of 478 up-regulated genes and 82 down-regulated genes. We then looked for a hierarchical network of the candidate regulators, in such a way that the conditional combination of their expression changes may explain those of their target genes. Using Genomica, a software tool for regulatory network construction, we obtained a hierarchical network of eight regulons that were used to map insulin downstream signaling network. Taken together, the results illustrate the benefits of combining completely different methods such as motif-based regulatory factor discovery and expression level-based construction of regulatory network of their target genes in understanding insulin induced biological processes and signaling pathways.

Regulatory Requirements on the Safety and Efficacy Evaluation for the Development of Stereoisomeric Drugs (광학이성질체 의약품 개발에서 안전성과 유효성 평가에 대한 규제 요건)

  • This study was performed to investigate the current regulatory guidances of safety and efficacy evaluation for the development and approval of stereoisomeric drugs in US, EU, Canada and Japan. The important categories for the development of stereoisomeric drugs are classified as 1) development of a single enantiomer as a new active substance 2) development of a racemate as a new active substance 3) development of a new single enantiomer from an approved racemate. The regulatory documents adopted in major countries for the safety and efficacy evaluation of stereoisomeric drugs were investigated with the focus on three major categories mentioned above. For the regulatory approval of stereoisomeric drugs in Korea, it is expected that the investigated results obtained in this study will be useful for the basic materials to ensure the safety and efficacy of stereoisomeric drugs as well as the stereochemical issues in chiral drug development in domestic pharmaceutical company.

A Study on the improvement plan of regulatory standard for construction noise and vibration (공사장 소음진동 규제기준 개선방안에 관한 연구)

  • Noise and vibration problems at the construction site, expansion of construction scale and frequency of urban construction is increased, has given the damage to local residents. Also, the result of civil complaint about noise and vibration that occurred in 2011 was 56,244; it is showing an increasing trend of more than 4% annually. In particular, the construction noise and vibration, the civil complaints of around construction site accounts for 64.6%(36,353), are harmful to the tranquility of living environment. As a result, the government has managed to strengthen the regulatory standard of construction noise (65dB(A), Day-time) from 2011. However, the regulatory standard of construction noise and vibration does not meet and also complaints related the construction noise and vibration not decreased. Because the management system can be applied to the construction site is insufficient and a shortage of manpower. In this study, investigated the status and problems of the regulatory standards related to construction noise and vibration, we propose an efficient management plan of construction noise and vibration.

The Regulatory Region of Muscle-Specific Alpha Actin 1 Drives Fluorescent Protein Expression in Olive Flounder Paralichthys olivaceus

  • To develop a promoter capable of driving transgene expression in non-model fish, we identified and characterized the muscle-specific alpha-actin gene in olive flounder, Paralichthys olivaceus (PoACTC1). The regulatory region of PoACTC1 includes putative regulatory elements such as a TATA box, two MyoD binding sites, three CArG boxes, and a CCAAT box. Microinjection experiments demonstrated that the regulatory region of PoACTC1, covering from -2,126 bp to +751 bp, just prior to the start codon, drove the expression of red fluorescent protein in developing zebrafish embryos and hatching olive flounder. These results suggest that the regulatory region of PoACTC1 may be useful in developing a promoter for biotechnological applications such as transgene expression in olive flounder.

Buying Virtual Fashion Items in the Metaverse - Focusing on Self-Regulatory Focus - (메타버스 가상 패션아이템 구매 - 자기조절초점을 중심으로 -)

  • The metaverse is emerging as the next digital environment for people not only to interact and collaborate with others, but also to have virtual consumption experiences. In this study, virtual fashion items for the consumer's digital self are deemed significant products with consumption value. Drawing from the regulatory focus theory, this study examines how consumers' promotion and prevention focus influence value perception and buying behaviors of virtual fashion items in the metaverse context. The data were collected through an online survey. A total of 546 consumers in their twenties who are aware of the metaverse responded to a self-administered questionnaire. The results showed that promotion focus influenced all the perceived consumption values of virtual fashion items such as the economic, visual authority, hedonic, and social value, whereas prevention focus influenced only the visual authority value. Visual authority value negatively affected both purchase intention and willingness to pay premium price, while others had a positive effect. The findings provide theoretical evidence that consumers' regulatory focus is critical in buying virtual fashion items and suggest that marketers devise effective strategies to stimulate consumers' regulatory focus and to emphasize the economic, hedonic, and social value of the items in the metaverse context.
