Transcriptome Analysis of Induced Systemic Drought Tolerance Elicited by Pseudomonas chlororaphis O6 in Arabidopsis thaliana

  • Root colonization by Pseudomonas chlororaphis O6 induces systemic drought tolerance in Arabidopsis thaliana. Microarray analysis was performed using the 22,800-gene Affymetrix GeneChips to identify differentially-expressed genes from plants colonized with or without P. chlororaphis O6 under drought stressed conditions or normal growth conditions. Root colonization in plants grown under regular irrigation condition increased transcript accumulation from genes associated with defense, response to reactive oxygen species, and auxin- and jasmonic acid-responsive genes, but decreased transcription factors associated with ethylene and abscisic acid signaling. The cluster of genes involved in plant disease resistance were up-regulated, but the set of drought signaling response genes were down-regulated in the P. chlororaphis O6-colonized under drought stress plants compared to those of the drought stressed plants without bacterial treatment. Transcripts of the jasmonic acid-marker genes, VSP1 and pdf-1.2, the salicylic acid regulated gene, PR-1, and the ethylene-response gene, HEL, also were up-regulated in plants colonized by P. chlororaphis O6, but differed in their responsiveness to drought stress. These data show how gene expression in plants lacking adequate water can be remarkably influenced by microbial colonization leading to plant protection, and the activation of the plant defense signal pathway induced by root colonization of P. chlororaphis O6 might be a key element for induced systemic tolerance by microbes.

Genistein Combined with Exercise Improves Lipid Profiles and Leptin Levels in C57BL/6J Mice Fed a High Fat Diet

  • The aim of this study is to determine the anti-obesity effects of genistein and exercise, separately and in combination, in mice. Fifty male C57BL/6J mice were divided into 5 treatment groups: normal diet (ND), high fat diet (HD), high fat diet with exercise (HD+Ex), high fat diet with 0.2% genistein (HD+G), high fat diet with 0.2% genistein, and exercise (HD+G+Ex). They were allowed free access to feed and water, and exercised mice engaged in swimming on a regular basis for 12 weeks. Genistein supplemented mice gained less weight, had lower energy intake, better lipid profiles, and lower leptin than the HD mice. Furthermore, when genistein was combined with exercise (HD+G+Ex) the effects were even greater. HD, HD+Ex, and HD+G mice exhibited increased hepatic CPT-1 mRNA expression. Therefore, genistein and exercise has anti-obesity effects, as shown by changes in body weight, fat accumulation, energy intake, and leptin levels.

  • Emotion recognition and regular controls are concentrated interest in computer studies to emotional changes. Thus, the quantified by objective assessment methods are essential for application of color sensibility computing situations. In this paper, it is applied to a digital color image emotion emotional computing calculations numbered recognized as one representation. Emotional computing research approach consists of a color attribute to the image recognition focused sensibility and emotional attributes of color is the color, brightness and saturation separated by. Computes the sensitivity weighted according to the score and the percentage increase or decrease in the sensitivity property tone applied to emotional expression. Sensitivity calculation is free-degree (X), and calculates the tension (Y-axis). And free-level (X-axis) coordinate of emotion, which is located the intersection of the tension (Y-axis) as a sensitivity point. The emotional effect of the Russell coordinates are utilizing the core (Core Affect). Tue numbers represent the size and sensitivity in the emotional relationship between emotional point location and quantified by computing the color sensibility.

Myotube differentiation in clustered regularly interspaced short palindromic repeat/Cas9-mediated MyoD knockout quail myoblast cells

  • Objective: In the livestock industry, the regulatory mechanisms of muscle proliferation and differentiation can be applied to improve traits such as growth and meat production. We investigated the regulatory pathway of MyoD and its role in muscle differentiation in quail myoblast cells. Methods: The MyoD gene was mutated by the clustered regularly interspaced short palindromic repeat (CRISPR)/Cas9 technology and single cell-derived MyoD mutant sublines were identified to investigate the global regulatory mechanism responsible for muscle differentiation. Results: The mutation efficiency was 73.3% in the mixed population, and from this population we were able to establish two QM7 MyoD knockout subline (MyoD KO QM7#4) through single cell pick-up and expansion. In the undifferentiated condition, paired box 7 expression in MyoD KO QM7#4 cells was not significantly different from regular QM7 (rQM7) cells. During differentiation, however, myotube formation was dramatically repressed in MyoD KO QM7#4 cells. Moreover, myogenic differentiation-specific transcripts and proteins were not expressed in MyoD KO QM7#4 cells even after an extended differentiation period. These results indicate that MyoD is critical for muscle differentiation. Furthermore, we analyzed the global regulatory interactions by RNA sequencing during muscle differentiation. Conclusion: With CRISPR/Cas9-mediated genomic editing, single cell-derived sublines with a specific knockout gene can be adapted to various aspects of basic research as well as in functional genomics studies.

  • This study was started to study the artistic aesthetic value of the antique artwork which is gradually declining in the field of metal crafts. The aesthetic interpretation of the lines and faces that have been identified through previous researches has been defined as a vitality and repetitive beauty. The researchers here have tried to express the formative works which can be repeated and change. As a result, it was found that the object of inquiry which can express the aesthetic beauty felt in the simple change of the line and the face is possible without the advanced technique of the difficult technique or the molding method through the natural and gradual change of the line and the face rather than the regular repetition. Through this study, it was found that the use of future lines and surfaces could be utilized as a variety of aesthetic expression methods as molding elements.

  • An XML document has a structure which may be irregular. It is difficult for end-users to comprehend the irregular document structure exactly. For these XML documents, an end-user has a difficulty in using structured query. Therefore, an end-user formulates no structured query or a query which has a little structure information. In this context, we propose new retrieval models which use the structured information for ranking and compensate the difference between user query structure and document structure. To ease with querying, we assume the independence among querying paths which represent structural constraints. Since this assumption makes degradation of the expression power of a query language, we also propose a model which overcome this problem. As there had been no test collections for XML documents, we made a small test collection from TIPSTER of the RTEC and experimented on this collection without a structured query, From this experiment, we showed that our models improve average precision about 67% over conventional Vector-Space model.

  • The electrical properties of neurons are produced by the coordinated activity of ion channels (Hille, 1992). $K^{+}$ channels play a key role in shaping action potentials and in determining neural firing patterns. Small conductance $Ca^{2+}$-activated $K^{+}$ (S $K_{Ca}$ ) channels are involved in modulating the slow component of afterhyperpolarization (AHP) (Kohler et al., 1996). Here we examine whether rat type 2 S $K_{Ca}$ (rSK2) channels can affect the shape of the action potential and the neural firing pattern, by overexpressing rat SK2 channels in Aplysia neuron R15. Our results show that rSK2 overexpression decreased the intraburst frequency and changed the regular bursting activity of neurons to an irregular bursting or beating pattern in R15, Furthermore, the overexpression of rSK2 channels increased AHP and reduced the duration of the action potential. Thus, our results suggest that ectopic S $K_{Ca}$ channels play an important role in regulating the filing pattern and the shape of the action potential.ntial.

Dietary Aloe Improves Insulin Sensitivity via the Suppression of Obesity-induced Inflammation in Obese Mice

  • Background: Insulin resistance is an integral feature of metabolic syndromes, including obesity, hyperglycemia, and hyperlipidemia. In this study, we evaluated whether the aloe component could reduce obesity-induced inflammation and the occurrence of metabolic disorders such as blood glucose and insulin resistance. Methods: Male C57BL/6 obese mice fed a high-fat diet for 54 days received a supplement of aloe formula (PAG, ALS, Aloe QDM, and Aloe QDM complex) or pioglitazone (PGZ) and were compared with unsupplemented controls (high-fat diet; HFD) or mice fed a regular diet (RD). RT-PCR and western blot analysis were used to quantify the expression of obesity-induced inflammation. Results: Aloe QDM lowered fasting blood glucose and plasma insulin compared with HFD. Obesity-induced inflammatory cytokine (IL-$1{\beta}$, -6, -12, TNF-${\alpha}$) and chemokine (CX3CL1, CCL5) mRNA and protein were decreased markedly, as was macrophage infiltration and hepatic triglycerides by Aloe QDM. At the same time, Aloe QDM decreased the mRNA and protein of $PPAR{\gamma}/LXR{\alpha}$ and $11{\beta}$-HSD1 both in the liver and WAT. Conclusion: Dietary aloe formula reduces obesity-induced glucose tolerance not only by suppressing inflammatory responses but also by inducing anti-inflammatory cytokines in the WAT and liver, both of which are important peripheral tissues affecting insulin resistance. The effect of Aloe QDM complex in the WAT and liver are related to its dual action on $PPAR{\gamma}$ and $11{\beta}$-HSD1 ression and its use as a nutritional intervention against T2D and obesity-related inflammation is suggested.

  • In researching the ontological status of fashion, a good grasp of fashion can decide the direction of fashion study. Fashion is sometimes considered an area of industry far from art from a point of view of purity of art, in spite of its aesthetic value and expression. However, art properties can be differentiated from the purity of art in modern aesthetics, and fashion properties which were the reason for that fashion to be considered as non-art can be affirmed as the same with properties of contemporary art properties. Therefore, the purpose of this study was to suggest the possibility that general properties of fashion can be accepted in the art arena through justifying art properties deviating from the purity of art. It can provide a boost to fashion's cultural status. For the research method, a literature review and case analysis were carried out through specialty publications related to art history, aesthetics, and fashion, regular publications and websites specializing in fashion, and art museums. Through the research, art properties deviated from the purity of art, which are; tactile sense, impermanence, dailiness, and commercial viability, were justified as being the same as fashion properties. These art properties were not general properties of fine art in modern aesthetics, but the ones occurring in contemporary arts. These properties, now present in contemporary art, can no longer disqualify fashion as a non-art.

  • Natural forms could be ultimate visual expression power that affects on the industrial design. The rule of power in nature as nature formal characteristics have a direct influence and can be also applied to productional and artificial form. All organic forms and structures in nature have unique form and shape to be self controlled and good in order. Such an order in nature comes from regular and ratio principles which has aesthetical order by mathematics. The specialty of beauty in nature can be revealed not only visual form but also the ratio, balance and rhythm of structural principles. As we examine the aesthetic source, embodied some object can be developed into basic principles. Furthermore, through this study we can find out that the form theory in nature forms share the quality attribute with geometrical form to be shown in industrial product design. In this context, this study aims to make dear visual quality of organic form and structure that is immanent in nature about a form as formative principles of industrial product design, the significancy of this study is to prove the application into functional and structural form of product design.
