Expression of Porcine Acid-labile Subunit (pALS) of the 150-kilodalton Ternary Insulin-like Growth Factor Complex and Initial Characterization of Recombinant pALS Protein

  • Acid-labile subunit (ALS) is a component of the 150-kDa insulin-like growth factor-binding protein-3 (IGFBP-3) complex, which, by sequestering the majority of IGFs-I and -II and thereby prolonging the half-life of them in plasma, serves as a circulating reservoir of IGFs in mammalian species. A pGEX-2T plasmid and a baculovirus expression constructs harboring a coding sequence for glutathione-S transferase (GST)-porcine ALS (pALS) fusion protein were expressed in BL21(DE3) E. coli and Sf9 insect cells, respectively. The expressed protein was purified by glutathione or Ni-NTN affinity chromatography, followed by cleavage of the fusion protein using Factor Xa. In addition, pALS and hIGFBP-3 were also produced in small amounts in the Xenopus oocyte expression system which does not require any purification procedure. A 65-kDa pALS polypeptide was obtained following the prokaryotic expression and the enzymatic digestion, but biochemical characterization of this polypeptide was precluded because of an extremely low expression efficiency. The baculovirus-as well as Xenopus-expressed pALS exhibited the expected molecular mass of 85 kDa which was reduced into 75 and 65 kDa following deglycosylation of Asn-linked carbohydrates by Endo-F glycosidase, indicating that the expressed pALS was properly glycosylated. Moreover, irrespective of the source of pALS, the recombinant pALS and hIGFBP-3 formed a 130-kDa binary complex which could be immunoprecipitated by anti-hIGFBP-3 antibodies. Collectively, results indicate that an authentic pALS protein can be produced by the current expression systems.

  • This study contemplates Croatian traditional costumes with focus on its geographical characteristics and acculturation with neighbor countries as a member of old Yugoslavian Union. Therefore, it contains the information from field works based on Croatian language, native costumes and folk materials wildly. Men usually wear wide linen pants ($ga\'{c}$), shirt ($ko\v{s}ulje,\;rnbina)$, vest (lajbek), hat and jacket ($kro\v{z}et$). Also they sometimes wear scarves or aprons. Women put on blouse, long skirt, apron, headgear with various decoration and scarf ($pe\v{c}a$), shawl (tibetui robe) or necklace together. Croatian folk dresses fall into three different styles by provinces, such as Pannonia, Dinar and Adria, according to the basic environmental and racial characteristics of each province. People of Pannonia plain generally wear white linen clothing and often use flounce and lace, which are influenced by Serbia and Hungary. In the Dinar mountain province, aprons with tassel, knitted vests made of leather and fur are very popular. The clothing of Dinar province has much commonness with those of Bosnia - Herzegovina and Montenegro which have metal buttons and belts. And the clothing of Adria coast is influenced by Italy a lot. There are many splendid silk clothes in its folk dress code. As mentioned above, there are a lot of foreign influences in Croatian history. Although there were severe cultural invasions in many times through its history, Croatians didn't just surrender to these cultural influences. People in Croatia comply with these new coming culture and reconcile these into their own tradition and dress code in their own way.

  • This study aims at revealing important utilization planning by surveying and analyzing unused classrooms in primary school's facilities. That is various and positive utilization planning of unused classrooms in order to cope with the changes of educational environment focused on high and multi-functionization, rather qualitative aspect than quantitative. By field-survey and interview, physical and environmental factors such as, the change procedure of students, unused facilities in schools and improvement affairs and purpose stressed by schools were surveyed. Additionally, the way of educational facilities to future society linked with residents was examined by surveying about the relationship between educational facilities and regional communities. The curriculums and "School facilities standard ordinance" have been revised in part by several times individually and couldn't meet with educational facilities, educational and social change positively. In rural area, to apply the standard of the capacity by a class with 40 students like city is not desirable. To make educational environment better qualitatively, the capacity should be reduced by 30 students and the unit square per a student enlarged by $2.4m^2$. By room unit module "school institution and managerial regulations under high school" should be applied distinctively to break down like 6, 7~12, 13~24 and over 25 classes. So re-institution of the capacity by a class and the standard unit of the number of class by scale is needed.

Patient Safety Program and Safety Culture (투약간호 중심의 환자안전 프로그램 개발 및 평가)

  • Purpose: To determine the impact of the Patient Safety Program on the safety culture of nursing department. Methods: Patient Safety Program focused on medication safety was launched by QI team and patient safety committee. Patient Safety Program was composed of the establishment of improved reporting system as s way to learn from error, 'Patient Safety Guard' movement, and continuous education for medication safety. With one group pretest-posttest design, nurses' perception of the safety culture were measured with self-administered questionnaire. Subjects were all nurses and managers in nursing department of a tertiary teaching hospital in Seoul. Collected data from survey was statistically analyzed using t-test. Results: Patient Safety Program had been continued for 20 months in participation of all nurses and managers. Safety culture was improved (pretest=2.84, posttest=2.90, p<.001; 4 point scale). Conclusions: This study indicates that there has been a statistically significant increase in the nurses' perception of safety culture. These findings suggest that Patient Safety Program had made great contribution toward system wide safety culture in the hospital. To improve safety culture, leadership supports and flexibility to apply tailored interventions to the hospital were required necessarily.

A Study on the History and Characteristics of Jeongamsa Sumano Stupa (정암사 수마노탑의 유래와 양식적 특성)

  • Jeongamsa Sumano Stupa(Korean National Treasure No. 332) is a brick like stone stupa and it is considered that this stupa was built in the Later Silla period or in the early part of the Koryo period, considering its structure and style. This Buddhist stupa is highly significant in Korean Buddhism since it was built to enshrine Buddhist sarira reliquaries that 'Jajang' brought from China in the 7th century, according to the 『Overlooked Historical Records of the Three Korean Kingdoms, 『三國遺事』』. However, the stupa was positioned at the very unique location. It was positioned not at the center of Jeongamsa Temple, but at the separate edifice. The location of Jeongamsa Sumano Stupa indicates that this stupa embodies the nature of Mahaparinirvana treasure palaces and that this stupa was recognized as a solemn symbol, which reflected a sarira faith. Jeongamsa Sumano Stupa is the only brick stone stupa built in Gangwon province. It is worth noticing that this stupa was mostly made of dolostones, found around this region, and thereby regional characteristics were reflected in this stupa. The other interesting fact about Jeongamsa Sumano Stupa is that there are many records available which include information about stupa's reconstruction, considering that there aren't many records of other stupas. The most representative records are 5 pieces of memorial stones which include information about when and on what ground the stupa was reconstructed as well as information about originators of the stupa. In conclusion, Jeongamsa Sumano Stupar could be regarded as the academically valuable material since it is considered very important and helpful for understanding not only the history of Korean Buddhist stupas, but also so many other things such as construction techniques, styles of brick pagoda and brick stone stupa, and such.

A Study of Job Stress and Job Satisfaction of Medical Aid Client Managers (의료급여관리사의 직무스트레스와 직무만족도)

  • Purpose: The purpose of this study is to investigate the degree of job stress and job satisfaction and the relationship between job stress and job satisfaction of Medical Aid Client Managers. Method: Data were collected from 185 Medical Aid Client Managers in 234 areas between April 2-15, 2007. Collected data was analyzed for the frequency, t-test, ANOVA, Pearson's Correlation Coefficient etc. Results: The average score of job stress was 2.6. Job instability was highest as 2.9 and job autonomy was lowest as 2.2. in eight items. When the job stress of Medical Aid Client Managers was converted to 100 point, the average was 51.7 and ranked in upper 50% of Korean woman workers' job stress. Among the eight items, because the relationship satisfaction and the instability of duty regime ranked in upper 25% of Korean woman workers' job stress, those items were identified to have high stress. The average score of job satisfaction was 2.3 out 4.0 which could be interpreted as unsatisfactory. The correlation between the job stress and the job satisfaction demonstrated significant difference. Decrease of the job satisfaction was associated with increase of the job stress. In regard to the eight items, the inverse correlation was demonstrated to be significant in organization, inappropriate compensation, and workplace culture. Conclusion: Medical Aid Client Managers have high stress in carrying out their duties. They have experienced very severe instability in their duty regime. Also, they have felt alienated from their organization and experienced unfair rewards. They had low duty satisfaction but got a heavy workload and experienced overtime work. To relieve job overburden of Medical Aid Client Managers. it was necessary to improve business system and also was necessary to relieve a regional disparity of business charge.

Chemical Compositions and Thermal Characteristics of Rice Husk and Rice Husk Ash in Korea (왕겨 및 왕겨재의 화학적 조성 성분과 열적 특성)

  • For better and large utilization of rice husk, the production and consumption status, differences in chemical composition and heating value due to region and variety, and thermogravimetric characteristic of rice husk were studied. In addition, the differences in chemical composition due to region and variety and the crystallization characteristic of rice husk ash were also studied. Approximately 800,000 M/T of rice husk was produced per year in Korea, which is about $18\%$ of the paddy production by weight. Noticeable varietal and regional difference pattern in chemical composition was not found among the domestic rice husk samples. Their average ash content and higher heating value were $16.4\%$ and 16,660 kJ/kg by dry basis, respectively. A relation seemed to exist between the carbon content and higher heating value. Noticeable difference pattern in chemical composition was not found among the domestic rice husk ash samples. The $SiO_2$ contents were a little low, the maximum being $92.9\%,$ and the contents of major components such as CaO, MgO, and $K_2O$ were also lower compared with foreign rice husk ash due to the deficiency of compost matters in domestic soils. Thermogravimetry study showed the thermal decomposition of rice husk started at about $250^{\circ}C,$ followed by relatively fast combustion of combustible gas until the temperature rose to $350^{\circ}C.$ After $350^{\circ}C,$ combustion of the carbon component proceeded relatively slowly as the temperature increased. Therefore, the ignition temperature of the rice husk could be estimated around $300^{\circ}C$. Crystallization of $SiO_2$ in the rice husk ash was found from the combustion temperature of $750^{\circ}C$ and became distinctly when the combustion temperature exceeded $900^{\circ}C$. The ash became darker with $SiO_2$ crystallization.

Petrochemical Study on the Precambrian Granitic Rocks in the Basement Area of Hambaeg Basin (함백익지(咸白益地) 기반지역(基盤地域)에 분포(分布)하는 선(先)캠브리아 화강암질암류(花崗岩質岩類)의 암석화학적(岩石化學的) 연구(硏究))

  • The area of this study is located in the Sang dong district, Youngwol Gun, Kangwon Do, where the Ogcheon fold belt comes into contact with the Ryongnam massif. The area is covered by the Precambrian metasedimentary rocks of Yulri Group in the south from the line of Ungyosan-Maebongsan-Jansan-Taebaegsan Mountains and by the Cambro-Ordovician sedimentary rocks of Choseon Supergroup in the north. The Choseon Supergroup unconformably overlies the Yulri group. Several granitic intrusives occur in the Precambrian and Cambro-Ordovician terrain. The purpose of this study is to clarify the geochronology, mineralogical composition, geochemical characteristics, petrogenesis and tectonic settings of the Precambrian granitic rocks, and to evaluate the P.T. conditions of granitic intrusions. The K/Ar ages obtained from the muscovite of Nonggeori Granite, Naedeogri granite and pegmatite intruded into the Yulri Group are Early Proterozoic ($1805{\pm}18Ma$ to $1642{\pm}23Ma$), and those from the migmatitic pegmatite are Late Carboniferous ($305{\pm}4Ma$), respectively. The Precambrian granitic rocks are characterized by the presence of muscovite, tourmaline and grey feldspar with faint lineation of mafic minerals. In terms of mineralogical and chemical composition, the granitic rocks are felsic, calc-alkalic, peraluminous and S-type (ilmenite-series). The geochemical characteristics of major and trace elements indicate that the granitic rocks belong to syn-collision setting at the compressional plate margin. They were formed by progressive melting of relatively homogeneous crustal materials under 1~3kb and $670^{\circ}{\sim}720^{\circ}C$ in aqueous fluid conditions, and the Naedeogri granite was more fractionated than the Nonggeori granite. During the Taebaeg disturbance, Nonggeori granite, Naedeogri granite and pegmatite were intruded and emplaced into the Yulri Group. Migmatitic pegmatite occurring in the southwestern area, however, gave much younger muscovite age than the pegmatite intruded into the Yulri Group in rest of the area did, that might be due to the regional metamorphism of the Post-Choseon disturbance. The Geodo granitic mass and the Imog granite were intruded during the Bulgugsa disturbance.

Analysis on Visual Preference of Bridge Landscapes of Background and Shape in Rural Area - Focused on the Natural Landscape in Rural Area - (농촌지역의 교량의 배경경관 및 교량형태에 따른 시각적 선호도 분석 - 농촌지역 자연경관을 중심으로 -)

  • Due to the rapid economic development of Korea, the bridge have been built by government over the several years. Additionally, there are too many mountain and river and the bridge have been built in rural area. But bridge designers weren't considering the bridge landscape. And bridge was a negative factor in regional landscape. Because of this, this study surveyed the landscape preferences of rural bridge landscapes according to different bridge types. The results were summarized as follows: And this research include conducting a study on visual preference according to the bridge's type and background. And, the landscape of arch bridge in the river 1 is landscape of the highest preference. The the landscape of girder bridge in the river 2 is landscape of the lowest preference. In the river 1 and 2 landscape, high preference is observed in the arch bridge and low preference is noted in the girder bridge. In the mountain 1 and 2 landscape, high preference is observed in the cable-stayed bridge and low preference is noted in the girder bridge. In conclusion, the visual preference of bridge landscape depend on the background and bridge shape, the study said. Therefore, when bridge designer design the bridge, designer have to choose proper bridge shape according to the background. This research was conducted only in bridge landscape of rural area but the visual preference of bridge landscape can be changed according to the various background. And further research is needed to analyze visual preference of bridge landscape according to the various background.

Analysis of Precipitation Distribution in the region of Gangwon with Spatial Analysis (I): Classification of Precipitation Zones and Analysis for Seasonal and Annual Precipitation (공간분석을 이용한 강원도 지역의 강수분포 분석 (I): 강수지역 구분과 계절별 및 연평균 강수량 분석)

  • In this study, we separated the precipitation zones using the geographic location of stations and precipitation characteristics (monthly, seasonal, annual) in Gangwon province. Precipitation data of 66 weather stations (meterological office: 11 locations, auto weather system (AWS): 55 places) were used, and statistical method, K-means cluster method, was conducted for division of the precipitation regions. As the results of regional classification, the five zones of precipitation (Yongdong: 1 region, Youngseo: 4 regions) were separated. Seasonal average precipitation in spring is similar throughout Gangwon Province, seasonal average precipitation in summer has high values at Youngseo, and seasonal average precipitation in autumn and winter have high values at Youngdong. The some areas, the vicinity of Misiryeong and Daegwallyeong, happens the orographic precipitation in spatial analysis, but the orographic effects didn't occur for the whole Gangwon areas. However, to achieve more accurate results, the expansion of observatories per elevation and AWS data are demanded.
