Apparent Dominance of Regenerated Primary Production in the Yellow Sea

  • The Yellow Sea is known to be a very productive region in terms of fisheries. However, its trophic status seems to be highly variable, ranging from oligotrophic to eutrophic, based on new production (NP) values. The NP and regenerated production (RP) values estimated from $^{15}N$-labelled nitrate and ammonium uptake in spring (April 1996) and winter (February 1997) during this study ranged from 0.05 to 19.8 mg $N m^{-2} d^{-1}$ and from 0.1 to 22.8 mg $N m^{-2}d^{-1}$, respectively. Our measurements and earlier observations suggested that NP in the Yellow Sea varied over the four orders of magnitude (range 0.05-180.9 mg $N m^{-2} d^{-1}$) temporally and spatially, and that RP (range 0.1-507.5 mg $N m^{-2}d^{-l}$) based on ammonium predominated during most period of the year, except in winter when both productions were low. The significant nitrogen uptake by phytoplankton below the euphotic zone and episodic entrainment of phytoplankton from below the euphotic zone into the euphotic zone, and nitrite excretion and dissolved organic nitrogen release during nitrate uptake might explain the apparent dominance of RP in the Yellow Sea.

  • Chemical production by anaerobic bacterial fermentations was an important microbiological topic in the past due to both fundamental and applied aspects related to acetone-butanol production prior to its replacement by chemical synthetic routes from petroleum. Presently, the depletion and price-escalation of petroleum has regenerated a great interest in the potential of anaerobic bacteria to transform the renewable resouces such as biomass and wastes into chemical feed-stocks and fuels.

  • The reactive printing properties of regular viscose rayon and a new regenerated cellulosic fiber (en Vix^{\textregistered}$) which was prepared from cellulose acetate fiber was investigated in a comparative manner. From the results, it was found that en Vix exhibited better printing properties than regular viscose rayon. It showed stable final color yields, irrespective of the amount of thickener, hence reproducibility of printing of en Vix is expected to be excellent. In addition, urea requirements were less for the printings on en Vix than for the corresponding printing on viscose rayon. Therefore, en Vix is also expected to reduce the amount of the urea which causes environmental problems in dyehouse effluent.

  • Nitrogenous new production and regenerated production were measured in the southern part of the Yellow Sea (Hwanghae) using a stable isotope /SUP 15/N nitrate and ammonia between April 25∼30, 1993. Nitrogen production varied between 155 and 688 mg N m/SUP -2/ d/SUP -1/, which belongs to meso to eutrophic area values. This is equivalent to 881∼3909 mg C m/SUP -2/ d/SUP -1/, assuming the Redfield ratio for C:N of 5.7:1 (by weight). the f0ratio which is the fraction of new production from primary production, varied between 0.12 and 0.26, indicating that 74 to 88% of primary production was supported by the regeneration of nutrients within the euphoric zone. This low f0ratio is the characteristics of the oligo- to mesotrophic area. Contrary to the expected, the ambient nutrient concentration was not an important factor for controlling productivity in this area during the study period. The difference in productivity among stations was mainly due to the variations in phytoplankton biomass in different water masses.

Triterpenoid production and phenotypic changes in hairy roots of Codonopsis lanceolata and the plants regenerated from them

  • Codonopsis lanceolata (Campanulaceae) has been used in traditional medicines, as its roots contain several kinds of triterpenoid saponin with high medicinal values. In this work, we induced transgenic hairy roots of C. lanceolata and analyzed triterpenoid saponins from the hairy roots and hairy root-derived transgenic plants. Hairy roots were obtained from leaf explants by the transformation of Agrobacterium rhizogenes R1000. Transgenic hairy root lines were confirmed by the transcriptional activities of rolA, B, C, and D genes by RT-PCR. Transgenic root lines actively proliferated on hormone-free medium but not in nontransformed roots. Hairy roots contained richer triterpenoids (lancemaside A, foetidissimoside A, and aster saponin Hb) than nontransformed roots. Transgenic plants were regenerated from the hairy roots via somatic embryogenesis. They showed phenotypic alterations such as shortened shoots and an increased number of axillary buds and adventitious roots. The transgenic plants also contained higher triterpenoid levels than wild-type plants. These results suggest that hairy roots and transgenic plants of C. lanceolata could be used as medicinal materials for the production of triterpene saponins.

Effect of Plant Growth Regulators on Clonal Production through Basal Stem Explant Cultures of a Phalaenopsis Hybrid

  • This study was conducted to develop the clonal propagation technique through in vitro culture using basal stem explants in Phalaenopsis hybrid grown in vitro. The highest frequency of protocorm-like body (PLB) formation was obtained when basal stem explants were cultured on VW medium containing 30g/L sucrose, 500 mg/L activated charcoal, 150 ml/L coconut water, 1 mg/L NAA, 5 mg/L 2iP and 2.5g/L gel rite. PLBs transferred to Hyponex medium were regenerated to plantlets. Plantlets transferred to plastic pots containing spagnum moss were developed and successfully acclimatized under greenhouse. The flower was bloomingly opened in plants regenerated from basal stem explants. The flower was not different from both mother plant and plant induced through clonal propagation of Phalaenopsis hybrid.

  • Changes of annual seed production related to climate change were studied for 12 years in Piagol, a riparian valley in Mt. Jiri. Sixty-four seed traps (sized 0.5 ${\times}$ 0.5 $m^{2}$) were set up on the forest floor of surveyed area. Seeds were collected from these traps at an interval of 15 days from September to November since 1984. Vegetation of the study area was mainly consisted of the naturally regenerated Carpinus tschonoskii in the tree layer. Acer mono, Quercus serrata, Carpinus laxiflora and Symplocos chinensis also appeared in the same layer. Maximum production occurred in 1984 and 1994. As a result of comparing seed production with local climate factors for 12 years, seed productivity and the year of maximum production of Carpinus forest were merely related with precipitation, air temperature and duration of sunshine among local climate factors. Duration of sunshine was, however, not contributed to periodically high productivity of seed of riparian valley carpinus forest.

Breeding of the native vegetables using the biotechnology

  • For breeding of a new rootstock for eggplant production, somatic hybrids between two species, Solanum integrifolium and S. sanitwongsei were obtained through protoplast fusion. The former species has been commonly used for rootstock for eggplant production in Japan. Eggplants on these rootstocks are more productive than ungrafted plants, but are susceptible to bacterial wilt caused Ralstonia solanacearum. While the latter species is resistant, the growth of eggplants on this rootstock is rather slow and low yield. Protoplast of both species were isolated from cotyledons, and inactivated with iodoacetamide or UV-irradiation, then fused electrically. The fused products were then cultured. Regenerated plantlets were then transplanted on soil then maintained in a green house. The plants were classified into four groups. Those in the first group showed morphological characters intermediate of the parentalspecies. The plants bore fruit with viable seeds. The plants showed a chromosome number of 2n=48, the sum of those of the parental species, and are suggested to be symmetric fusion products. While plants in the other groupswas less vigorous and showed chromosome number 2n= 68 to 72 suggesting asymmetric fusion products by genomic in situ hybridization(GISH). Isozyme pattern of shikimate dehydrogenase (SKDH; EC 1.1.1.25), isocitrate dehydrogenase (IDH; EC 1.1.1.41) and phosphoglucomutase (PGM; EC 2.7.5.1) showed that 24 regenerated plants in three groups were somatic hybrids. Analysis of random amplified polymorphic DNA (RAPD) showed that 43 S. integrifolium-specific and 57 S. sanitwongsei-specific bands were all found in 24 plants. Both somatic hybrids and its S1 plants were found to be resistant to bacterial wilt, and eggplant grafted these plants using for rootstocks were more productive than grafted mother plants. Now, S1 progenies are used for commercial eggplant production in Osaka Prefecture.

Development of Doxorubin Overproducing Streptomyces Using Protoplast Regeneration

  • To establish an effective and reliable technique of mutation by protoplast regeneration in doxorubicin overproducing industrial strain, it is essential to optimize the conditions for protoplast regeneration. $CaCO_3$ as buffer, the negative effect of glucose was still evident without significant changes in pH, ruling out acidity as responsible for the suppression of anthracycline production and suggesting a direct effect of glucose on antibiotic biosynthesis. Production of doxorubicin was improved in doxorubicin overproducing industrial strain (BR-Dox) when protoplast regenerated. BR-Dox4 and BR-Dox6 of BR-Dox derivatives improved doxorubicin production by 25.2 % and 12.2 %, respectively.
