Objectives : The purpose of this study was to investigate the effects of modified fasting therapy on body composition, blood cell count and blood chemistry in inpatients. Methods : The modified fasting therapy program consists of three phases, a period of reducing food intakes, fasting and refeeding. Body weight, body composition, and the degree of obesity were estimated in before fasting therapy and after refeeding. Blood cell count and blood chemistry were estimated in before fasting therapy, before refeeding and after refeeding. Results : After modified fasting therapy, body weight, body fat mass, percent body fat, body mass index(BMI), body matabolic rate(BMR) and muscle mass was decreased significantly. WBC count and platelet count decreased significantly, RBC count and hemoglobin increased significantly and hematocrit was not changed significantly but within normal limit. AST and ALT increased significantly, especially patients group over BMI 25 within normal liver function. This study was not founded correlation between decreased muscle mass and increased liver function test. ALP, BUN, Total-cholesterol, and triglyceride decreased significantly. Creatinine increased significantly, but within normal limit. Sodium was not changed significantly, potassium decreased significantly but within normal limit. Conclusions : These results suggest that modified fasting therapy will be beneficial if several measures complement.
Objectives: The purpose of this study was to investigate the effects of modified fasting therapy on the changes of body compositions. Methods: We analyzed the medical records of 33 patients, who carried out modified fasting therapy at Dunsan Korean Medicine Hospital of Daejeon University from January 1st, 2011 to December 30th, 2015. They went through reducing food intakes period (7 days), fasting period (14 days) and refeeding period (14 days). Body compositions (weight, body mass index, skeletal muscle mass, percent body fat, basal metabolic rate, waist-hip ratio, visceral fat area) were reviewed at each state. And then the data was analyzed. Results: The body composition values (weight, body mass index [BMI], skeletal muscle mass, percent body fat, basal metabolic rate, waist-hip ratio, visceral fat area) decreased during the fasting therapy period, as a whole. The weight, BMI, percent body fat and visceral fat area decreased during the reducing food intakes period, the fasting period and the refeeding period. The skeletal muscle mass and basal metabolic rate significantly decreased during the reducing food intakes period and the fasting period, and insignificantly increased during the refeeding period. Conclusions: Results from this investigation showed that modified fasting therapy using fermented herbal medicine have positive effects on changes of body compositions.
Effect of fasting and refeeding on growth and blood chemistry of juvenile olive flounder Paralichthys olivaceus L. was investigated when fish achieved compensatory growth. Fish were fed the experimental diet for 6 days a week. Five treatments in triplicate were prepared: C, S1, S2, S3 and S4. Fish in the control group (C) were hand-fed to apparent satiation twice a day. Fish in treatments of S1, S2, S3 and S4 experienced 1, 2, 3 and 4 weeks of starvation and were then hand-fed to satiation twice daily during the remaining 7, 6, 5 and 4 weeks of the experiment, respectively. Weight gain of fish in C, S1 and S2 were higher than those of fish in S3 and S4. A significant difference in plasma total protein, glucose, triglyceride, $T_3$ and $T_4$ was observed in between starved and refed fish for the rest periods of the feeding trial. Plasma total protein and $T_3$ of flounder decreased with week of fasting and following correlationships were obtained; Y (Total protein) = -0.13X (week of fasting) + 1.54, $R^2=0.9792$ and $Y(T_3)=-11.48X$ (week of fasting) + 79.57, $R^2=0.8822$, respectively.
Melatonin plasma in Swallows exhibited circadian rhythmical secretions in the LD (Light and dark, 12:12) period with and without feeding. But their average difference between at CT6 (Circadian Time) and CT18 was 3.53 ng/$\textrm{m}{\ell}$ in LD period with feeding. on the other side 1.60 ng/$m\ell$ during without feeding. Melatonin concentration at CT6 without feeding incresed from 0.22 ng/$\textrm{m}{\ell}$ to 0.93 ng/$\textrm{m}{\ell}$. It is demonstrated that decresing melatonin secretion may reduce digestive function in order to ready for the migration. While the birds with feeding exhibited circadian rhythmical activity, their activity without feeding was durable. The concentrations of melatonin plasma by refeeding were 1.53 ng/$\textrm{m}{\ell}$ at CT6 and 6.07 ng/$\textrm{m}{\ell}$ at CT18. Melatonin plasma concentration in the night increased by more than ca. quadruple at day. This results suggest that melatonin regulates metabolism for the return to the normal metabolism condition after migration. After 3 days refeeding melatonin was secreted circadian rhythmically same as the secretions with feeding at beginning.
Effects of starvation, ligation, refeeding and methoprene treatment in the feeding phase of the fifth instar larvae of Bombyx mori on the regulation of juvenile hormone esterase(JHE) activity were investigated. Starvation and ligation contributed to the reduction of JHE activity, however, JHE levels in starved larvae were slightly higher than in legated larvae. Haemolymph JEH activity of starved larvae was increased by refeeding, and duration of increasing time of JHE activity after starvation was related to duration of starvation. When starved larvae were applied methoprene topically, JHE activity were not changed at day 0 and 1, but were increased by 1.3-1.4 times between day 2 and 5. When ligated larvae were applied methoprene topically, JHE activity was not changed at day 0, but were increased by 1.9-2.3 times between day 1 and 5. These results suggest that head factor, juvenile hormone(JH) and nutrient are major factors in the regulation of JHE in the feeding phase of the fifth instar larvae of Bombyx mori. Especially JHE might be regulated by the co-operative action of head factor and JH. However, head factor plays important role in the early stage, while JH plays important role thereafter.

The effect of refeeding with various single essential amino acids on the recovery of plasma insulin-like growth factor-I (IGF-I) concentration in fasted young chickens was examined. Young chickens (29 days of age) were divided into 15 experimental groups. Chickens in one group were fed on the commercial diet ad libitum for 4 days. The remaining 56 chickens in 14 experimental groups were fasted. After 2 days of fasting, 52 chicks in 13 fasted groups were refed with one of the following experimental diets for 2 days. Eleven experimental diets were protein-free diets supplemented with one of 11 essential amino acids (Arg, Gly, His, Ileu, Leu, Met, Phe, Lys, Thr, Trp, Val). The remaining 2 experimental diets were a protein-free diet containing 11 essential amino acids and a protein-free diet not supplemented with amino acids. Birds in the remaining fasted group continued to be fasted for 2 days. Fasting for 2 days markedly reduced plasma IGF-I concentration. When fasted chickens were refed the protein-free diet containing either Gly alone or all essential amino acids, plasma IGF-I concentration was recovered to the level similar to that of fed chickens. Protein-free diet alone, however, failed to restore the reduced IGF-I concentration in plasma. Body weight loss modulated by feeding with protein-free diets supplemented with various single essential amino acids was associated with changes in plasma IGF-I concentrations. We concluded that body weight loss by feeding with a protein-free diet was lower than that of fasted chickens and that body weight loss associated with the decrease in plasma IGF-I concentration was modulated by feeding with protein-free diets containing various single essential amino acids.
PAI-1 (plasminogen activator inhibitor-1), leptin, and resistin are synthesized and secreted by Int cells of rodents and have recently been postulated to be an important link to obesity. This study was conducted to identify the nutritional regulation of PAI-1, leptin, and resistin gene expression in 0b/ob mice. The mice were divided into four groups according to nutritional status: control, 48 hour fasting, 48 hour-fasting/12 hour-refeeding, and 48 hour-fasting/24 hour-refeeding. The mRNA levels of each peptide were measured by semi-quantitative RT-PCR. In visceral fat tissue, the level of PAI-1 mRNA increased markedly when 48h-fasted animals were refed with a high carbohydrate-low fat diet. However, lasting/refeeding did not appreciably change PAI-1 mRNA levels in subcutaneous fat tissue. Similar results were obtained for resistin mRNA levels in both types of fat tissues. These findings suggest that visceral adipose tissue might be more sensitively involved in the nutritional regulation of PAI-1 and resistin gene expression compared to subcutaneous fat tissue. The level of leptin mRNA decreased markedly in the 48h-fasted animals, and increased markedly when 48h-fasted animals were refed with a high carbohydrate-low fat diet. The nutritional regulation of leptin mRNA showed similar patterns in both types of fat tissues. In conclusion, the nutritional regulation of gene expression encoding PAI-1, resistin, and leptin from adipocytes may vary according to the type of adipose tissue.
SDS gel electrophoresis has been employed to examine the changes in distribution of three major classes of nuclear proteins extracted from isolated liver nuclei in response to refeeding of starved rats with a fat-free high carbohydrate diet and following insulin injection into streptozotocin-diabetic rats. The relative quantity of electrophoretically separated proteins in the fraction showed marked changes with 0.14 NaCl extracts, but not with histones and phenol soluble non-histone proteins. During 48h starvation at least five proteins ranging in molecular weight from 50,000 to 180,000 daltons decreased relative to normal controls while a protein with 36,000 daltons was increased. Refeeding the starved rats with a high carbohydrate diet reversed these changes over 24 h. Insulin injection into streptozotocin-diabetic rats increased levels of the set of five 0.14 M NaCl soluble proteins identified from refeeding experiment of starved rats. The 36,000 daltons protein was also diminished. These results indicate that changes in distribution of certain nuclear proteins in 0.14M NaCl extracts are associated with the control of nuclear activity ralated to known insulin-signalled modulation and induction of cytosolic lipogenic enzymes.
Nutritional regulation of gene expression associated with growth and feeding behavior in avian species can become an important technique to improve poultry production according to the supply of nutrients in the diet. Insulin-like growth factor-I (IGF-I) found in chickens has been characterized to be a 70 amino acid polypeptide and plays an important role in growth and metabolism. Although it is been well known that IGF-I is highly associated with embryonic development and post-hatching growth, changes in the distribution of IGF-I gene expression throughout early- to late-embryogenesis have not been studied so far. We revealed that the developmental pattern of IGF-I gene expression during embryogenesis differed among various tissues. No bands of IGF-I mRNA were detected in embryonic liver at 7 days of incubation, and thereafter the amount of hepatic IGF-I mRNA was increased from 14 to 20 days of incubation. In eyes, a peak in IGF-I mRNA levels occurred at mid-embryogenesis, but by contrast, IGF-I mRNA was barely detectable in the heart throughout all incubation periods. In the muscle, no significant difference in IGF-I gene expression was observed during different stages of embryogenesis. After hatching, hepatic IGF-I gene expression as well as plasma IGF-I concentration increases rapidly with age, reaches a peak before sexual maturity, and then declines. The IGF-I gene expression is very sensitive to changes in nutritional conditions. Food-restriction and fasting decreased hepatic IGF-I gene expression and refeeding restored IGF-I gene expression to the level of fed chickens. Dietary protein is also a very strong factor in changing hepatic IGF-I gene expression. Refeeding with dietary protein alone successfully restored hepatic IGF-I gene expression of fasted chickens to the level of fed controls. In most circumstances, IGF-I makes a complex with specific high-affinity IGF-binding proteins (IGFBPs). So far, four different IGFBPs have been identified in avian species and the major IGFBP in chicken plasma has been reported to be IGFBP-2. We studied the relationship between nutritional status and IGFBP-2 gene expression in various tissues of young chickens. In the liver of fed chickens, almost no IGFBP-2 mRNA was detected. However, fasting markedly increased hepatic IGFBP-2 gene expression, and the level was reduced after refeeding. In the gizzard of well-fed young chickens, IGFBP-2 gene expression was detected and fasting significantly elevated gizzard IGFBP-2 mRNA levels to about double that of fed controls. After refeeding, gizzard IGFBP-2 gene expression decreased similar to hepatic IGFBP-2 gene expression. In the brain, IGFBP-2 mRNA was observed in fed chickens and had significantly decreased by fasting. In the kidney, IGFBP-2 gene expression was observed but not influenced by fasting and refeeding. Recently, we have demonstrated in vivo that gizzard and hepatic IGFBP-2 gene expression in fasted chickens was rapidly reduced by intravenous administration of insulin, as indicated that in young chickens the reduction in gizzard and hepatic IGFBP-2 gene expression in vivo stimulated by malnutrition may be, in part, regulated by means of the increase in plasma insulin concentration via an insulin-response element. The influence of dietary protein source (isolated soybean protein vs. casein) and the supplementation of essential amino acids on gizzard IGFBP-2 gene expression was examined. In both soybean protein and casein diet groups, the deficiency of essential amino acids stimulated chickens to increase gizzard IGFBP-2 gene expression. Although amino acid supplementation of a soybean protein diet significantly decreased gizzard IGFBP-2 mRNA levels, a similar reduction was not observed in chickens fed a casein diet supplemented with amino acids. This overview of nutritional regulation of IGF-I and IGFBP-2 gene expression in young chickens would serve for the establishment of the supply of nutrients to diets to improve poultry production.

본 연구는 절식 및 재급여가 Sprague-Dawley 계통 수컷 흰쥐(55마리)의 지방조직내 LPL 활성과 지방합성량에 미치는 영향을 구명하였다. 아울러 재급여하는 동안 다양한 식이섭취 제한이 LPLghkf성과 지방합성에 어떻게 영향을 미치는 지도 조사하였다. 대조군(5마리)은 무제한 식이를 급여하였으며 실험 첫날 식이섭취 상태에서 바로 희생시켰으며, 그 외의 모든 쥐는 2일간 절식시켰다. 2일 절식 후 그중 5마리의 쥐를 희생하여 절식군으로 두었으며, 나머지 쥐(45마리)는 절식 후 식이를 재급여하였는데, 이때 재급여하는 동안 식이섭취 제한 효과를 보기 위해 ad libitum group, 20% food restricted group(ad libitum group의 80%) 및 40% food restricted group(ad libitum group의 60%)로 나누어 실시하였다. 재급여 후 7일, 14일 및 21일 째 되는 날 각각 희생하였다. 지방조직의 지방합성은 포도당이 총 지방으로 변한 양으로 측정하였다. 체중과 지방조직의 무게는 재급여 후 ad libitum group은 5일만에, 20% food restricted group은 14일만에 대조군 수준에 도달하였다. 재급여시 40% food restricted group의 체중과 지방조직의 무게는 재급여 후 21일째가 되도록 대조군 수준에 도달하지 못했다. 식이 재급여 후 21일 때 ad libitum group의 혈청 총 콜레스테롤함량은 대조군에 비해 높았다. (p<0.05). HDL-콜레스테롤함량은 재급여시 40% food restricted group가 대조군에 비해 높았다. (p<0.05). 그러나 HDL-C/TC비율과 중성지방함량은 처리군간에 유의적인 변화를 나타내지 않았다. 절식 후 지방조직내 지방합성량과 LPL(HR-LPL과 TE-LPL)활성은 대조군에 비해 현저희 저하되었다. (p<0.05). 재급여 후 21일째 ad libitum group의 지방합성량은 대조군에 비해 낮았다.(p<0.05). 그러나 40% food restricted group의 지방합성량은 대조군에 비해 높았다. (p<0.05). HR-LPL과 TE-LPLghkf성은 재급여시 ad libitum group과 20% food re-stricted group은 대조군 수준에 도달하였으나, 40% food restricted group은 재급여 후 21일째가 되도록 대조군 수준에 도달하지 못했다. 이러한 결과를 통해 절식 후 식이를 재급여할 경우 식이섭취 제한은 LPLghkf성이 증가되는 것을 부분적으로 혹은 완전히 예방할 수 있으며, 지방 조직내 지방축적에 영향을 미친다는 것을 말해 준다.
