In use of pesticides in golf courses has been increased steadily. Environmental concern as well as decrease in efficiency led the turfgrass management into an alternate approach of using beneficial microorganism to deal with turfgrass pests. This study was focused on the use of such microorganisms for improving cultural environment and minimizing the use of pesticides. Microorganisms antagonistic to turfgrass diseases were applied to zoysiagrass fairways and creeping bentgrass greens in Yusung country club. Tharch accumulation, disease occurrence, and other cultural environments were compared among the combinations of microorganisms and suppliemental N applications. The application of microorganisms antagonistic to turfgrass diseases improved turf resiliency. Thatch thickness was 3.03cm in the control plot but it was 2.11cm in plots treated by microorganisms, indicating significant effects of microorganism application on reduction of thatch accumulation. Number of microorganism that can decompose of cellulose was higher at the plots treated with useful microbial products and it was considered that existence of higher population of microorganisms resulted in reduction of thatch accumulation. In the evaluation of relationship between thatch accumulation and disease occurrence, greater thatch accumulation was observed at the golf courses which have been frequently infested by large patch. However, the rate of thatch accumulation varied among surveyed golf courses regardless of the year of turf establishment. Therefore, management practice which can be effective for reduction of thatch could result in large patch suppression. The application of microorganisms on the established turfgrasses reduced the occurrence rate of pythium blight and yellow path diseases, whereas occurrence of brown patch and dollar spot increased.

To test the potential effects of extracellular electron shuttles (EES) on the rate and extent of heavy metal release from contaminated soils during microbial iron reduction, we created anaerobic batch systems with anthraquinone-2,6-disulfonate (AQDS) as a surrogate of EES, and with contaminated soils as mixed iron (hydr)oxides and microbial sources. Two types of soils were tested: Zn-contaminated soil A and As/Pb-contaminated soil B. In soil A, the rate of iron reduction was fastest in the presence of AQDS and > 3500 mg/L of total Fe(II) was produced within 2 d. This suggests that indigenous microorganisms can utilize AQDS as EES to stimulate iron reduction. In the incubations with soil B, the rate and extent of iron reduction did not increase in the presence of AQDS likely because of the low pH (< 5.5). In addition, less than 2000 mg/L of total Fe(II) was produced in soil B within 52 d suggesting that iron reduction by subsurface microorganisms in soil B was not as effective as that in soil A. Relatively high amount of As (~500 mg/L) was released to the aqueous phase during microbial iron reduction in soil B. The release of As might be due to the reduction of As-associated iron (hydr)oxides and/or direct enzymatic reduction of As(V) to As(III) by As-reducing microorganisms. However, given that Pb in liquid phase was < 0.3 mg/L for the entire experiment, the microbial reduction As(V) to As(III) by As-reducing microorganisms has most likely occurred in this system. This study suggests that heavy metal release from contaminated soils can be strongly controlled by subsurface microorganisms, soil pH, presence of EES, and/or nature of heavy metals.
Hexavalent chromium in aqueous solutions was successfully removed via sorption and reduction in the presence of waste leaves. Cr(VI) removal followed a first-order reaction, and removal rates were proportional to the amount of waste leaves used in the tests. Most of Cr(VI) were removed via sorption in early stages of the tests, but the reduction reaction played a significant role in Cr(VI) removal later. Solution pHs were continuously decreased due to the microbial activity, which was induced from the microorganisms attached on waste leaves. The decreased solution pHs further enhanced the sorption and reduction of Cr(VI). To characterize the microorganisms found in the tests, a denaturing gradient gel electrophoresis (DGGE) method was used. The majority of microorganisms were composed of Bacillus sp. which can reduce Cr(VI). Thus, waste leaves can be effective reactive media for the treatment of Cr(VI) in the subsurface.
To better understand dissimilatory iron and sulfate reduction (DIR and DSR) by subsurface microorganisms, we investigated the effects of sulfate and electron donors on the microbial goethite (${\alpha}$-FeOOH) reduction. Batch systems were created 1) with acetate or glucose (donor), 2) with goethite and sulfate (acceptor), and 3) with aquifer sediment (microbial source). With 0.2 mM sulfate, goethite reduction coupled with acetate oxidation was limited. However, with 10 mM sulfate, 8 mM goethite reduction occurred with complete sulfate reduction and x-ray absorption fine-structure analysis indicated the formation of iron sulfide. This suggests that goethite reduction was due to the sulfide species produced by DSR bacteria rather than direct microbial reaction by DIR bacteria. Both acetate and glucose promoted goethite reduction. The rate of goethite reduction was faster with glucose, while the extent of goethite reduction was higher with acetate. Sulfate reduction (10 mM) occurred only with acetate. The results suggest that glucose-fermenting bacteria rapidly stimulated goethite reduction, but acetate-oxidizing DSR bacteria reduced goethite indirectly by producing sulfides. This study suggests that the availability of specific electron donor and sulfate significantly influence microbial community activities as well as goethite transformation, which should be considered for the bioremediation of contaminated environments.
Diesel-contaminated soils were ozonated for different times (0 - 900 min) and incubated for 9 wk to monitor petroleum hydrocarbons (PH)-degradative potential of indigenous microorganisms in the soils. Increased ozonation time decreased not only concentration of PH but also number of microorganisms in the soils. Microorganisms in the ozonated soils increased during 9-wk incubation as monitored by culture- and nonculture-based methods. Higher (1-2 orders of magnitude) cell number was observed by quantitative analysis of soil DNA using probes detecting genes encoding 165 rRNA(rrn), naphthalene dioxygenase (nahA), toluene dioxygenase (todC), and alkane hydroxylase (alkB) than microbial abundance estimated by culture-based methods. Such PH-degraders were relatively a few or under detection limit in 900-min ozonated soil. Further PH-removal observed during the incubation period supported the presence of PH-degraders in ozonated soils. Highest reduction (25.4%) of total PH (TPH) was observed in 180-min ozonated soil white negligible reduction was shown in 900-min ozonated soil during the period, resulting in lowest TPH-concentration in 180-min ozonated soil among the ozonated soils. Microbial community composition in 9-wk incubated soils revealed slight difference between 900-min ozonated and unozonated soils as analyzed by whole cell hybridization using group-specific rRNA-targeted oligonucleotides. Results of this study suggest that appropriate ozonation and subsequent biodegradation by indigenous microorganisms may be a cost-effective and successful remediation strategy for PH-contaminated soils.
This study was conducted to determine the effect of hygienic processing (HP) on the reduction of microorganisms during manufacturing of saengshik with two vegetables (carrots and cabbage) and two grains (barely and glutinous rice) compared to general processing (GP). For GP, distilled water was used for washing raw materials and equipment. For HP, aqueous ozone (3 ppm) in combination with 1% citric acid and 70% alcohol were used for washing raw materials and the equipment, respectively. In carrots, after cutting, total aerobic bacteria (TAB), yeast and mold (YM) and coliforms were significantly increased to 5.19, 8.04 and 2.08 ($log_{10}$ CFU/g), respectively (p<0.05). Washing effectively reduced the increased microorganisms from cross contamination during cutting, but cross contamination increased with subsequent GP drying and milling procedures to 8.56, 8.27 and 3.71 ($log_{10}$ CFU/g) for TAB, YM and coliforms, respectively (p<0.05). On the other hand, HP washing of carrots with 3 ppm ozone in combination with 1% citric acid showed higher antimicrobial effect than GP washing, significantly decreasing the number of microorganisms (p<0.05). Further cross contamination did not occur through drying and milling due to cleaning the equipments with 70% alcohol prior to processing. After milling, the number of TAB, YM and coliforms were significantly decreased to 3.89, 4.47 and not detectable level ($log_{10}$ CFU/g), respectively (p<0.05). Similar results were observed in cabbage and grains. During storage for two months at different temperatures (22 or $4^{\circ}C$), there were no changes in numbers of spoilage microorganisms in the packaged saengsik after either processing. This suggests the importance of HP for the reduction of microorganisms during saengsik production, and demonstrates the effectiveness of disinfection at each processing stage in minimizing contamination levels to enhance microbial safety of saengshik products.
침출수 미생물에 의한 크롬 6가이온의 3가이온으로의 환원을 회분식 배양 및 실험실 규모의 연속배양체제에서 연구하였다. 회분식 배양에 있어서 다양한 범위는 침출수 (10~60%)와 glucose (50~200 mM)를 첨가한 최소배지에서 초기 농도 $20mg\;L^{-1}$의 크롬(VI)을 첨가한 경우 72시간내에 최대 90%의 크롬(VI)이 침출수 미생물에 의하여 환원되었다. 이때 glucose의 첨가는 크롬환원을 촉진시켰으나, 배지내 침출수 비율의 증가는 오히려 크롬환원을 저해하였다. 후자의 경우는 침출수에 존재하는 황산이온이 크롬이온과 경쟁적 저해를 일으키기 때문인 것으로 추정되었다. 연속배양 실험은 124일간 상온에서 크롬(VI)의 농도를 $5{\sim}65mg\;L^{-1}$로 증가시키면서 수행하였으며, 수리학적 체류시간이 36시간일 때 $5{\sim}50mg\;L^{-1}$의 크롬농도 범위에서의 환원은 거의 100%에 달하는 것으로 나타났다. 평균 81.2%의 환원효율을 보인 101~124일 구간에서 계산된 specific reduction rate는 $3.492mg\;g^{-1}\;protein\;h^{-1}$였다. 이상의 결과는 침출수에 존재하는 미생물이 크롬은 함유한 다양한 침출수 또는 피혁폐수의 크롬 무독화과정에 유용하게 쓰일 수 있다는 가능성을 제시해 주는 것이다.
The food wastes from a refectory and an eating house were heated in domestic microwave oven(700W) equipped with a fan and the drying rates and destruction of microorganisms were investigated. The drying rate was decreased with the size of food waste and the food wastes in polypropylene basket were dried faster than that on glass dish. The rate was increased with lower initial moisture content. Death rate of microorganisms was also decreased with the size of food waste. Ninety eight percent of reduction in viable cell numbers for the 400g of food waste could be achieved in 240sec of microwave irradiation. The growth of microorganisms in food wastes after microwave irradiated was observed at $32^{\circ}C$ and 95% relative humidity after 7days and the cell numbers in microwave irradiated food wastes were found to be 1/2 ~ 1/20 of the numbers in untreated wastes in accordance with the mass and the length of exposed time to microwave. To minimize the moisture and microorganisms in food wastes, the use of microwave oven are recommended.
Fe (III) impurities in clay could be microbially removed by inhabitant dissimilatory Fe (III) reducing microorganisms. Insoluble Fe (III) in clay particles was leached out as soluble reductive form, Fe (II). The microorganisms removed from 10 to 45% of the initial Fe (III) when each sugar was supplemented to be in ranges of 1 - 5 % (w/w; sugar/clay). The microorganisms reduced 2.1 - 12.8 mol of Fe (III) per 100 mol of carbon in sugars metabolized when sugars such as glucose, maltose, and sucrose were used as sole carbon source. Bacillus sp. IRB-W and Pseudomonas sp. IRB-Y were isolated from the enrichment culture of the clay. The isolates were considered to participate in metabolizing organic compounds to fermentative intermediates with relatively little Fe (III) reduction at initial Fe (III) reduction process. By the microbial treatment, the whiteness of the clay was increased form 63.20 to 79.64, whereas the redness was obviously decreased form 13.47 to 3.55. This treatment did not cause any unfavorable modifications in mineralogical compositions of the clay.
