HMG-CoA Reductase의 저해제 탐색과 가금의 콜레스테를 저하 효과 (Screening of 3-Hydroxy-3-Methylglutaryl-Coenzyme A Reductase Inhibitors In Vitro and Its Application to Pullets)

  • 체내 cholesterol 수치를 낮추기 위하여 HMG-CoA reductase 활성을 저해하는 물질들을 닭에게 급여하여 in vitro상에서 검토한 결과, HMG-CoA reductase 활성에 대한 마늘, lovastatin과 copper의 순으로 저해능은 51.3%, 87.5%, 82.0%이었다. 또한 혈장, 가슴조직, 간장의 cholesterol 함량에 대한 마늘(3% in diet), lovastatin(300mg/Kg of diet), copper(200mg/Kg of diet)의 효과를 검토하였다. 혈장 중의 cholesterol 함량과 HDL, LDL cholesterol 함량은 3% 마늘의 투여로 감소되어졌으나 copper의 투여는 오히려 증가하였고, lovastatin 투여에 의한 차이는 나타나지 않았다. 그리고 간장과 가슴조직의 cholesterol과 triglyceride는 사료에 cholestrol의 수치를 저하시키는 물질을 첨가하였으나 별다른 차이가 없었다.

곡류 및 두류 추출물로 부터 HMG-CoA reductase 저해활성 검색 (Screening of HMG-CoA Reductase Inhibitory Activity of Ethanol and Methanol Extracts from Cereals and Regumes)

  • 국내산 곡류의 에탄올 및 메탄올 추출물로부터 콜레스테롤 생합성계에서 가장 중요한 율속효소로 알려져 있는 HMG-CoA reductase 저해활성을 검색하였다. HMG-CoA reductase activity 측정용 효소원은 cholestyramin을 급여하여 사육 한 후 밤 12시경에 간율 적출하여 얻어진 microsome을 freeze-thaw방법으로 조제하였고 효소활성은 spectrophotometric assay로 측정하였다. 그 결과 각 시료 추출물을 $100{\;}{\mu}g/assay$ 되게 첨가한 경우 HMG-CoA reductase 억제활성은 에탄올 추출물에서는 수수가 가장 높은 활성을 보였고 다음은 흑임자였으며 나머지는 활성이 미미하거나 전혀 없는 것으로 나타났다. 메탄올 추출물에서는 기장이 가장 높은 활성을 나타내었고 다음은 수수였으며 그 다음은 메밀, 흑미의 순으로 나타났다. 또한 추출물에서 높은 억제활성을 나타내었던 수수와 기장 메탄올 추출물을 각종 용매로 분획하여 활성을 검토한 결과 수수와 기장 모두 헥산 획분에서 가장 높은 활성을 나타내었다.

Esculetin, a Coumarin Derivative, Inhibits Aldose Reductase Activity in vitro and Cataractogenesis in Galactose-Fed Rats

  • Naturally occurring coumarin compounds have received substantial attention due to their pharmaceutical effects. Esculetin is a coumarin derivative and a polyphenol compound that is used in a variety of therapeutic and pharmacological strategies. However, its effect on aldose reductase activity remains poorly understood. In this study, the potential beneficial effects of esculetin on lenticular aldose reductase were investigated in galactose-fed (GAL) rats, an animal model of sugar cataracts. Cataracts were induced in Sprague-Dawley (SD) rats via a 50% galactose diet for 2 weeks, and groups of GAL rats were orally treated with esculetin (10 or 50 mg/kg body weight). In vehicle-treated GAL rats, lens opacification was observed, and swelling and membrane rupture of the lens fiber cells were increased. Additionally, aldose reductase was highly expressed in the lens epithelium and superficial cortical fibers during cataract development in the GAL rats. Esculetin reduced rat lens aldose reductase (RLAR) activity in vitro, and esculetin treatment significantly inhibited lens opacity, as well as morphological alterations, such as swelling, vacuolation and liquefaction of lens fibers, via the inhibition of aldose reductase in the GAL rats. These results indicate that esculetin is a useful treatment for galactose-induced cataracts.

수종 생약에 의한 Lens Aldose Reductase의 억제 (Inhibition of Lens Aldose Reductase by Medicinal Plants)

  • To search for inhibitory activities for lens aldose reductase, twenty two medicinal plants were tested by the method of bovine lens aldose reductase inhibition test. In this test Rubia akane(68%), Artemisia selengensis(61%), Lespedeza cuneata (58%), Ligustrum lucidum(58%), Viola patrinii(58%) extracts were shown to have aldose reductase inhibitory activity. From the radix of Rubia akane, $2-methyl-1,\;3,\;6-trihydroxy-9,\;10-anthraquinone-3-O-(6'-O-acetyl)-{\alpha}-rhamnosyl-(1{\rightarrow}2)-{\beta}-D-glucoside$ was isolated and the $IC_{50}$ of this compound in the aldose reductase inhibition test was about $1.2{\times}10^{-6}M$.

천연물로부터 $5{\alpha}-Reductase$ 활성억제 및 면포용해효과의 검색 (Screening of $5{\alpha}-Reductase$ Inhibition and Comedolytic Effects from Natural Products)

  • The antibacterial activity against Propionibacterium acnes (P. acnes), $5{\alpha}-reductase$ inhibition and comedolytic effects are the important pharmacological target sites of antiacne drughs. We previously reported on the antibacterial activities against P. acnes by natural products. In the present study the screening of $5{\alpha}-reductase$ inhibition and comedolytic effects from natural products were performed. Seven natural products such as Angelica koreana, Sophora flavescens, Prunus persica, Bombyx mori, Areca catechu, Galla rhois and Gleditschia koraiensis perfectly inhibited the activity of $5{\alpha}-reductase$ at the concentration of 0.01% (w/v). Sixteen natural products which were shown to have the potent antibacterial activities against P.acnes or $5{\alpha}-reductase$ inhibition activities were assayed for the comedolytic test. In the results of comedolytic effects on experimentally-induced comedones (EIC), Sophora flavescens showed the strongest comedolytic effect on EIC, and Polygonum cuspidatum and Angelica koreana showed stronger comedolytic effects on EIC than azelaic acid used for a positive control at the concentration of 3% (w/v). These results suggest that several natural products including Sophora flavescens can be developed as noble antiacne agents.

시험관법에의한 식물열수추출물의 3-Hydroxy-3-Methylglutaryl CoA Reductase 및 Acyl-CoA:Cholesterol Acyltransferase 활성 저해도 측정 (Measurement of Inhibitory Activities on 3-Hydorxy-3-Methylglutaryl CoA Reductase and Acyl-CoA:Cholesterol Acyltransferase by Various Plant Extracts in vitro)

  • Hydroxy methylglutaryl CoA(HMG CoA) reductase and acyl CoA:cholesterol acyltransferase(ACAT) are two important enzymes that are associated with regulation of cholesterol metabolism. The inhibitors of HMG CoA reductase and ACAT are very effective in lowering serum cholesterol in most animal species. In present study, various plant extracts with hot water were used to examine the inhibitory activities against HMG CoA reductase and ACAT that are involved in cholesterol biosynthesis and cholesterol esterification in tissues, respectively. The extracts of Fagophyrum rotundatum, Rosa multiflora, Rosa rugosa and Alisma orientalis exhibited significant inhibitory activities against the ACAT, 29%, 24%, 19%, and 18%, respectively. However the extracts of Typha augustifolia, Polygonum cuspidatum, Crataegus pinnatifida, Polygonum multiflorum inhibited the HMG CoA reductase activity by 53%, 42%, 37%, and 33% respectively. Results suggest that these plant extracts might play important roles in the regulation of the cholesterol metabolism in vivo.

식용 Azo색소가 Hepatic Microsomal Mixed Function Oxidase System에 미치는 영향 (The Effects of Food Azo Dyes on the Hepatic Microsomal Mixed Function Oxidase System in Rats)

  • The four azo dyes such as Amaranth (FD & C Red No. 2), Tartrazine (FD & C Yellow No. 4), sunset Yellow (FD & C Yellow No. 5) and Allura red (FD & C Red No. 40) are currently employed as a food additives in Korea. In this study, the effects of these azo dyes on the hepatic microsomal mixed function oxidase systems in Rats. (i.e., Cyt. P-450, Cyt. b$_5$, NADPH cyt. c-reductase and azo reductase) were investigated. Furthermore, to determine the relationship among the electron transport systems, each level of azo reductase, Cyt. P-450 and NADPH cyt. c-reductase was measured upon the administration of phenobarbital (known as an inducer of Cyt. P-450), 3-methylcholanthrene (Known as an inducer of Cyt. P-448), CoCl$_2$ (inhibitor on Cyt. P-450) or $CCl_4$ (inhibitor on Cyt. P-450). The results of these studies are as follows; (1) The levels of Cyt. P-450 and Cyt. b$_5$ were decreased upon the administration of these azo dyes. (2) When the level of Cyt. P-450 was decreased, the azo reductase activity was also decreased. (3) These azo dyes did not show any significant effect on the level of NADPH cyt. c-reductase. (4) The administration of 3-methylcholanthrene resulted in the elevation of azo reductase activity. The 3-methylcholanthrene may be responsible for the induction of CO-insensitive electron transport system.

Aldose Reductase Inhibitory Alkaloids from Corydalis ternata

  • A methanolic extract of Corydalis ternata having aldose reductase inhibitory activity was examined as a possible aldose reductase (ALR2) inhibitor, a key enzyme involved in diabetic complications. Seven alkaloids, tetrahydrocoptisine (1), corydaline (2), tetrahydropalmatine (3), isocorybulbine (4), corybulbine (5), dehydrocorydaline (6), and N-methyltetrahydroberbinium (7) were isolated from $CHCl_3$ fraction of C. ternata methanol extract. Among them, compounds 1, 5, and 7 exhibited $5.04{\pm}1.97%$, $5.00{\pm}1.26%$, and $1.80{\pm}2.33%$ inhibitions, respectively at $40{\mu}M$. The activities of the single compounds were not comparable to that of the whole extract, suggesting that the whole combination of each single compound was responsible for the activity of the extract as shown in many cases of natural medicines. Even though this is the second report on aldose reductase inhibition activity of C. ternata, recombinant human aldose reductase was employed in this study unlike in the previous report. Furthermore, the aldose reductase inhibitory activities of isocorybulbine, corybulbine, and N-methyltetrahydroberbinium, to the best of our knowledge, were evaluated for the first time in this study. These results suggest a use of the extract of C. ternata for ameliorating diabetic complications.

Isolation and Structural Determination of Aldose Reductase Inhibitor from Korean Fermented Soybean Paste

  • Aldose reductase catalyzes the conversion of glucose into sorbitol. Inhibiting this enzyme in diabetes mellitus can delay or prevent pathogenic process. Aldose reductase inhibitor was screened from Korean fermented soybean pastes (Doen-jang) and purified via sequential processes of ethanol extraction, HP-20 column chromatography, ethyl acetate extraction, silica gel column chromatography, and crystallization. Aldose reductase inhibitor was identified as genistein with molecular weight of 270 Da and molecular formula of $C_{15}H_{10}O_5$ based on UV spectrometry, $^1H$ and $^{13}C\;NMRs$, and mass spectrometry. Genistein inhibited aldose reductase of pig lens with $IC_{50}$ level of $20\;{\mu}M$. Because genistein was effective against aldose reductase of animal source, it may be a potential therapeutic agent for diabetic complications.

저온 처리한 상추 잎에서 monodehydroascorbate 환원효소의 반응 (Response of Monodehydroascorbate Reductase in Lettuce Leaves Subjected to Low Temperature Stress)

  • 식물의 저온 적응 메카니즘에서 아스코브산과 관련된 효소 중 MDHA 환원효소의 활성도와 과산화수소, 아스코브산의 함량, mRNA의 발현수준과의 연관성을 연구한 결과는 다음과 같다. MDHA 환원효소의 활성도 변화는 저온에 노출되는 시간이 길어질수록 증가하였으며 6시간 이후에 엽록체분획과 세포질분획에서 급격하게 증가하는 경향을 보였으나 실온으로 회복시켰을 때 효소의 활성도가 상대적으로 감소하는 경향을 보였다. 저온에 노출된 동안 아스코브산의 함량은 비교적 일정한 경향을 보이다가 실온으로 회복시키면 그 이후에는 급격하게 증가하는 경향을 보였다. 반면 저온에 노출되는 동안 급격히 dehydroascorbate 함량이 감소하였다가 실온으로 회복되면 약간 증가하는 경향을 보였다. 아스코브산의 함량과 엽록체분획과 세포질분획의 MDHA 환원효소의 활성도와의 상관관계는 각각 정의 상관($R^2$=0.9240, 0.9108)을 나타내었으나 디하이드로아스코브산의 함량과 MDHA 환원효소의 활성도 사이에는 각각 부의 상관($R^2$=0.8638, 0.8980)을 나타내었다. MDHA 환원효소 활성도와 과산화수소의 함량과의 상관관계를 과산화수소의 생성량이 증가하면 MDHA 환원효소의 활성도가 증가하는 정의 상관($R^2$=0.9443, 0.9647)을 나타내었다. 저온스트레스 처리 시간이 증가할수록 MDHA 환원효소의 mRNA의 발현 수준과 총 MDHA 환원효소의 활성도가 증가하는 경향을 나타내었다.
