The Effect on the Marine Water and Sediment Quality by the Use of Nano-S 1. Result of Water Tank Experiment (Nano-S가 해양수질 및 저질에 미치는 영향에 관한 연구 1. 실내수조실험 결과)

  • The purpose of this study was to observe the effect on the marine environment by the use of Nano-S. Nano-S was made to apply to improve the red tide bloom. The experiment was performed at round tank with volume of 180 L. Each tank was filled with an aggravated sediment about $14{\pm}1cm$ hight and sea water. The water flow-rate of tank was established on the rate of 6.25 L/hr. Sea water level was fitted to 40 cm, therefore the filled water was about 150 L. The sediment was stabilized during one week. Then the Nano-S and the red mud were added into each tank 0 kg(control), 1 kg(tank A), 2 kg(tank B), 5 kg(tank C) and 10 kg(tank D) each other. The quantity was fulfilled with 0 kg(control), 2.75 kg(tank A), 5.51 kg(tank B), 13.77 kg(tank C) and 27.55 kg(tank D) per square meter of sediment. The experiment was performed during 30 days. Water and sediment samples were collected from each tanks on the before 1hour and after 1, 3, 6, 12 hour and 1st, 3th, 5th, 7th, 10th, 15th, 30th day of the experiment period. The change of water and sediment quality was analyzed before and after applying the Nano-S and the red mud.

Effect of Yellow Clay on the Oxygen Consumption Rate of Korean rockfish, Sebastes schlegelii

  • Yellow clay dispersion has been applied to minimize fisheries impact by the red tide Cochlodinium polykrikoides blooms in Korean coasts since 1995. The present preliminary study documents the effect of yellow clay on Korean rockfish, Sebastes schlegelii, in terms of oxygen consumption rate (OCR). The OCR in the low clay suspension (0.05 and 0.23 %, w/w) showed normal level compared to the control. In contrast, the OCR for each one of three replicates in the high clay suspension (1.16 and 5.58 %, w/w) was not returned to the previous level that clay was not treated, indicating that high clay suspension (${\geq}1.16%$, w/w) might give negative effect on Korean rockfish. Overall, this result suggests that field application of clay to control Harmful Algal Blooms (HABs) may not give impact on Korean rockfish once the clay is dispersed in a low concentration (${\leq}0.23%$). In order to understand the changes of OCR in the repeated exposure to clay, it is required to do further studies on the changes of OCR when the fish is exposed to clay repeatedly after recovery in the normal seawater.

Temporal changes in the abundance of the fish-killing dinoflagellate Karlodinium veneficum (Dinophyceae) in Tongyeong, Korea

  • The toxic dinoflagellate Karlodinium veneficum has been implicated in numerous fish kill events around the world. Since this species commonly co-occurs with other morphologically similar dinoflagellates, field monitoring of this species in natural waters via light microscopy only has been problematic. In this study, we investigated temporal changes in K. veneficum's abundance in the waters of Obido, Tongyeong, using a species-specific real-time polymerase chain reaction (PCR) assay. The field survey, from April to December 2010, revealed K. veneficum occurred at low densities (12 to 425 cells $L^{-1}$) during this time and that cell numbers peaked in June (early summer in Korea), indicating this species generally occurs in the warmer season (mostly at $16.9-22.3^{\circ}C$ and 33.4-34.5‰) in the Obido area.

Probe of Algal Growth Potential (AGP) by Physio-Biochemical Analysis of Microalgae in the Stagnant Watershed (정체 수역 내에서 미세조류의 생리생화학적 분석에 의한 수화발생 잠재력 탐색)

  • Algal growth potentials were probed by algal growth rates, maximal PSII quantum yields and ATP amount as well as dry weights of algae to evaluate the water fertility due to the algal growth in the stream (CT) and stagnant watershed (WW). Oscillatoria agardhii (CY) and Coelastrum reticulatum (CH) were cultured in nitrogen (N) and phosphorus (P) starvation media (CH-10 medium) and re-inoculated in CT and WW for 7 days. Cell division rates of CY were the highest (k = 7.5) in WW after N starvation, while those of CH were the hight (k = 2.97) in WW after P starvation. The growth of CY was limited by P, while that of CH was by N. Conversely, maximal PSII quantum yields of CH were generally higher than those of CY in CT and WW according to culture time. CY was much more sensibly adapted than CH according to the variations of nutrient amounts in WW. The water fertility was much higher in WW than in CT. The potential assessment tool for water fertility will be able to compensate for the limit of physio-chemical analyses and to be applied as a monitoring system to forecast red-tide.

Effects of solar UV radiation on photosynthetic performance of the diatom Skeletonema costatum grown under nitrate limited condition

  • Availability of nutrients is known to influence marine primary production; and it is of general interest to see how nutrient limitation mediates phytoplankton responses to solar ultraviolet radiation (UVR, 280-400 nm). The red tide diatom Skeletonema costatum was cultured under nitrate (N)-limited and N-replete conditions and exposed to different solar irradiation treatments with or without UV-A (315-400 nm) and UV-B (280-315 nm) radiation. Its photochemical quantum yield decreased by 13.6% in N-limited cells as compared to that in N-replete ones under photosynthetically active radiation (PAR)-alone treatment, and the presence of UV-A or UV-B decreased the yield further by 2.8 and 3.1%, respectively. The non-photochemical quenching (NPQ), when the cells were exposed to stressful light condition, was higher in N-limited than in N-replete grown cells by 180% under PAR alone, by 204% under PAR + UV-A and by 76% under PAR + UV-A + UV-B treatments. Our results indicate that the N limitation exacerbates the UVR effects on the S. costatum photosynthetic performance and stimulate its NPQ.

  • 우리나라 연근해 해역에서 발생하는 적조 현상은 매년 특정 시기에 끊임없이 나타나고, 그 피해액은 해를 거듭해갈 수록 증가하고 있다. 이러한 적조 발생을 조기에 탐지할 수 있는 것만으로도 그 피해는 상당히 줄어들 것이라고 여겨진다. 본 논문에서는 우리나라 연근해 해역에서 발생하는 적조의 우점종 플랑크톤인 Cochlodinium polykrikoides를 탐지하기 위한 방법으로 C. polykrikoides 의 개체수 변화에 따른 후방산란신호를 획득하여 그 상관관계에 관한 실험을 실시하였다. 실험에 사용된 주파수는 5MHz, 10MHz 이고, 적조 주의보와 적조 경보에 해당하는 개체수인 300cell/mL, 1000cell/mL 을 중심으로 개체수를 변화하였다. 필터링 후의 해수와 Cochlodinium 이 있는 해수의 체적산란 패턴 변화뿐만 아니라 Cochlodinium 의 개체수 변화에 따른 산란 패턴의 변화를 볼 수 있었다.

Removal of High Strength Nitrogen in Dyeing Wastewater by Decomposition-Air Stripping Process (분해탈기법에 의한 염색폐수 중의 고농도 질소 제거에 관한 연구)

  • Total nitrogen is a major pollutant which mostly causes eutrophication and red tide. Wastewater effluent from printing of cotton-viscose rayon containing high concentrations of total nitrogen can not be effectively treated with a typical biological treatment process. This paper provides a new treatment process and experimental results for the removal of high strength nitrogen from dyeing wastewater. The optimum conditions of air stripping for the removal of total nitrogen include around pH 12, temperature $60^{\circ}C$ with 60 minutes of stripping time. In case of a filtration-air stripping process, an initial level ($500mg/{\ell}$) of total nitrogen was significantly reduced to below $60mg/{\ell}$. Deconite was synthesised for further decomposition of organic nitrogen. Thus, a filtration-decomposition-air stripping process was possibly achieved, by which a high level ($900mg/{\ell}$) of total nitrogen was effectively removed to below $60mg/{\ell}$ P. Finally, a continuous new process for the removal of total nitrogen is proposed and confirmed, based on batch experimental results, and its process validity is further discussed throughout.

Water Quality and Phytoplankton in the Waters of Manripo and Daechon Beaches (만리포 및 대천 해수욕장 수질 및 식물플랑크톤)

  • In spite of high concentrations of SS(>30mg/l) in July 2010, the water condition of Manripo was fine for swimming as COD data lower than 2.0mg/l in summer continuously. The bloom of phytoplankton or red tide were not occurred in the waters. COD data from the waters of Daechon were lower than 2.0mg/l, except August. Relatively high SS concentrations were the characteristics of Daechon beach and were caused by the effects of mud resuspension. The dominant taxa of phytoplnkton communities were consist of diatoms as the normal groups of neritic sea waters.

  • Relative DNA contents in some harmful algae were measured using DAPI staining and image analysis system. This method was useful to identify some morphologically similar species and isolates from harmful algal blooms (HABs). In exponential phase, Prorocentrum micans had higher relative DNA content (RD) of $1.83\pm0.52$ than any other isolates, followed by Cochlodinium polykrikoides $(1.10\pm0.46)$ Alexandrium tamarense $(0.93\pm0.32)$ Gyrodinium impudicum $(0.56\pm0.17)$, Scrippsiella trochoidea $(0.41\pm0.26)$ and P. minimum$(0.05\pm0.01)$. When they were fixed with Lugol's solution, it was difficult to d,iscern C. polykrikoides from G. impudicum under the light microscope, but the DNA contents were quite different in two species. C. polykrikoides contained about twice as much RD as G. impudicum under the same culture conditions and exponential phase. DAPI­stained DNA feature in C. polykrikodes showed concentrated in the peripheral part of the cell, but in G. impudicum showed a compact structure in the central part. Although A. tamarense and S. trochoidea were morphologically similar under the light microscope, nuclear DNA content of A. tamarense was twice as much as that of S. trochoidea.

GLOBAL MONITORING OF PLANKTON BLOOMS USING MERIS MCI

  • The MERIS MCI (Maximum Chlorophyll Index), measuring the radiance peak at 709 nm in water-leaving radiance, indicates the presence of a high surface concentration of chlorophyll ${\underline{a}}$ against a scattering background. The index is high in 'red tide' conditions (intense, visible, surface, plankton blooms), and is also raised when aquatic vegetation is present. A bloom search based on MCI has resulted in detection of a variety of events in Canadian, Antarctic and other waters round the world, as well as detection of extensive areas of pelagic vegetation (Sargassum spp.), previously unreported in the scientific literature. Since June 1 2006, global MCI composite images, at a spatial resolution of 5 km, are being produced daily from all MERIS (daylight) passes of Reduced Resolution (RR) data. The global composites significantly increase the area now being searched for events, though the reduced spatial resolution may cause smaller events to be missed. This paper describes the composites and gives examples of plankton bloom events that they have detected. It also shows how the composites show the effect of the South Atlantic Anomaly, where cosmic rays affect the MERIS instrument.
