Effect of Kyungohkgo(瓊玉膏) on Antioxidant Capacity in D-galactose Induced Aging Rats (경옥고(瓊玉膏)가 노화유발 흰쥐의 항산화능에 미치는 영향)

  • Objectives : In order to examine the antioxidant activities of Kyungohkgo(瓊玉膏), the study was done through measurement of parameters such as Thiobarbituric acid reactive substance(TBARS), Superoxide dismutase(SOD), Catalase(CAT), Glutathione peroxidase(GSH-px), Plasma total lipid, Plasma total triglyceride, Plasma total cholesterol, HDL-cholesterol concentrations in rat erythrocytes and plasma. Methods : Sprague-Dawley rats divided into 3 groups, Normal group(l2 weeks old), pathologically induced group(injected D-galatose 50mg/kg, 1time/day for 6 weeks, CONTROL) and Kyungohkgo(瓊玉膏) administered group(D-galactose 50mg/kg and Kyungohkgo extracts 1125.0mg/kg 1time/day for 6 weeks, KOG). Rats were sacrificed and TBARS, SOD, CAT, Plasma total lipid, Plasma triglyceride, Plasma total cholesterol, Plasma HDL-cholesterol concentrations and GSH-px were measured in rat erythrocytes and plasma. Results : Plasma TBARS concentrations of KOG group were significantly lower than those of control. Red blood cell(RBC) SOD activities of KOG group was increased(F=3.619, p=0.052, ANOVA test), and RBC catalase activities of all experimental group were not significantly different. RBC GSH-px activities of KOG group was increased(F=6.844, p=0.008, ANOVA test). The changes of Plasma triglyceride was not significantly different. Plasma total lipid of KOG group showed significant decrease compared to the control group(F=19.337, p=0.0001, ANOVA test). Plasma total cholesterol and HDL-cholesterol concentrations of all experimental groups were not significantly different. Conclusions : According to the above results, it is considered that Kyungohkgo(瓊玉膏) is effective in inhibiting lipid peroxidation and increasing antioxidative enzyme activities in D-galactose induced aging rat.

  • The nested polymerase chain reaction (PCR) assay was used to determine the sequences of reverse transcriptase (RT) codons 41, 67, 70, 210, 215 and 219 of human immunodeficiency virus-1 (HIV-1) pol gene. Template DNA was obtained from uncultured peripheral blood mononuclear cells from 27 Korean HIV-1 infected patients treated with ZDV and Korean red ginseng. The second PCRs were done for 2 separated regions (RT codons $13{\sim}98$ and $152{\sim}259$) with $5\;{\mu}l$ of the first PCR productNucleotide sequences were determined by direct sequencing. In the 27 patients, CD4+ cell count decreased from $230{\pm}117/{\mu}l$ to $152{\pm}162/{\mu}l$ for $46{\pm}26$ months (Mo), and actual duration of ZDV intake was $72{\pm}16$ Mo. In the 16 patients who had been treated with ZDV therapy ${\ge}25$ Mo, the incidences of 70R, 215F/Y, and 41L were 61%, 28% and 22%, respectively and those of 67N, 210W and 219Q were 17%. The incidences of 215F/Y were 6.7% for group ${\le}12$ Mo treatment, 22.7% for group with 13 to 24 Mo, and 27.8% for group ${\ge}25$ Mo. There was no mutation in 9 patients. It might be associated with the interruption of ZDV therapy for more than 6 months in 6 patients. This study shows that the detection of mutation could be useful prognostic marker with other clinical and virological data, and very low mutation rate is dectected compared to overseas reports.

Identification of Immunodominant B-cell Epitope Regions of Reticulocyte Binding Proteins in Plasmodium vivax by Protein Microarray Based Immunoscreening

  • Plasmodium falciparum can invade all stages of red blood cells, while Plasmodium vivax can invade only reticulocytes. Although many P. vivax proteins have been discovered, their functions are largely unknown. Among them, P. vivax reticulocyte binding proteins (PvRBP1 and PvRBP2) recognize and bind to reticulocytes. Both proteins possess a C-terminal hydrophobic transmembrane domain, which drives adhesion to reticulocytes. PvRBP1 and PvRBP2 are large (>326 kDa), which hinders identification of the functional domains. In this study, the complete genome information of the P. vivax RBP family was thoroughly analyzed using a prediction server with bioinformatics data to predict B-cell epitope domains. Eleven pvrbp family genes that included 2 pseudogenes and 9 full or partial length genes were selected and used to express recombinant proteins in a wheat germ cell-free system. The expressed proteins were used to evaluate the humoral immune response with vivax malaria patients and healthy individual serum samples by protein microarray. The recombinant fragments of 9 PvRBP proteins were successfully expressed; the soluble proteins ranged in molecular weight from 16 to 34 kDa. Evaluation of the humoral immune response to each recombinant PvRBP protein indicated a high antigenicity, with 38-88% sensitivity and 100% specificity. Of them, N-terminal parts of PvRBP2c (PVX_090325-1) and PvRBP2 like partial A (PVX_090330-1) elicited high antigenicity. In addition, the PvRBP2-like homologue B (PVX_116930) fragment was newly identified as high antigenicity and may be exploited as a potential antigenic candidate among the PvRBP family. The functional activity of the PvRBP family on merozoite invasion remains unknown.

Anti-oxidant, Anti-coagulation, and Anti-platelet Aggregation Activities of Black Currant (Ribes nigrum L.) (블랙커런트의 항산화, 항응고 및 혈소판 응집저해 활성)

  • The black currant (Ribes nigrum L.) is belong to the Grossulariaceae family, and has piquant berries, which can be eaten as raw or as processed foods, such as jams, jelly, juice and syrups. In this study, the fresh juice of black currant (FJBC) from Austria and its subsequent organic solvent fractions, such as hexane fraction, ethylacetate (EA) fraction, butanol fraction and water residue, were prepared and their in-vitro anti-oxidant, anti-coagulation and anti-platelet aggregation activities were evaluated. The FJBC and EA fraction, which has concentrated polyphenol and flavonoid, showed strong radical scavenging activities and reducing power. The $RC_{50}s$ of EA fraction against DPPH anion, ABTS cation, nitrite were 136.3, 66.2 and $115.5{\mu}g/ml$, respectively, those are 1/10, 1/16, and 1/7.7 of $RC_{50}s$ of vitamin C. In anti-coagulation assay, the FJBC, EA and butanol fraction showed significant inhibitory activities against thrombin, prothrombin and coagulation factors. Furthermore, the anti-platelet aggregation activities of EA and butanol fraction were the stronger than that of aspirin. The concentrations required for 50% platelet aggregation inhibition of aspirin, EA and butanol fraction were 0.395, 0.192 and 0.261 mg/ml, respectively. The EA and butanol fraction have no hemolysis activities up to 0.5 mg/ml against human red blood cells. The results suggest that the FJBC and its EA and butanol fraction have high potentials as novel anti-thrombosis agents. This report provides the first evidence of anti-thrombosis activity of black currant.

Effect of Mercury Chloride on Humoral and Cell-mediated Immune Responses in Mice (수은이 마우스의 체액성 및 세포성 면역반응에 미치는 영향)

  • The in vivo and in vitro humoral and cell-mediated immune responses of lymphocytes of BALB/c mouse exposed to mercury chloride$(HgCl_2)$ were investigated. In vitro exposure of the splenocytes to mercury chloride produced overt cytotoxicity in 3 hours period. The $IC_{50}$(the concentration required to inhibit a splenocyte viability by 50%) for mercury chloride was >0.1mM for cytotoxicity. In vivo mercury chloride exposed mice were significantly depressed delayed type hypersensitivity(DTH) response to sheep red blood cells(SRBC) in a dose-dependent manner compared with control group. Mercury chloride inhibited the proliferative responses of splenocytes to lipopolysaccharide, pokeweed mitogen, concanavalin A and phytohemagglutinin in a dose-dependent manner. Hemagglutinin response to SRBC in mercury chloride exposed mice was significantly depressed in a dose-dependent manner compared with control group. After 7 weeks of mercury chloride exposure in vivo, mercury chloride induced an increase of nonspecific serum $IgG_1$ and IgE levels in BALB/c mice.

Epidemiological surveys of an outbreak of false positive classical swine fever in Gyeongbuk province (의사 돼지콜레라 발생농장 역학조사)

  • This study was conducted to survey the farm which suffered from disease similar to classical swine fever(CSF) in Gyeongbuk province. Clinical signs appeared first in a few number of growing pigs which showed specific signs of diarrhea, depression, sleepiness, and reluctance to get up or to eat. Younger piglets may have appeared chilled, shiver and huddle together. As the disease progresses the affected pig's skin went red and purple. In histopathological signs, there were many haemorrhages throughout the body and larger haemorrhages in some organs such as lymph nodes. And there is a precipitous fall in the number of circulating leukocytes in the blood. In spite of insisting of farmer which did not vaccinate to classical swine fever, significant antibody production was detected in these affected pigs at enzyme-linked immuonsorbent assay. According to the above results at first glance, these affected pig suspected with CSF in clinical signs and histopathological lesions only. Because the symptoms and post-mortem picture were very similar to CSF, these false positive results would have been dangerous to diagnostician. But by reverse transcriptase polymerase chain reaction(RT-PCR) and comparative nucleotide sequence analysis, the disease was correctly diagnosed with post-weaning multisystemic wasting syndrome(PMWS) and porcine reproductive and respiratory syndrome(PRRS) compoundly. And the antigen which were detected the lesion similar to CSF virus, was confirmed with LOM vaccine strain of CSF. In most national CSF eradication program and in countries which are free of the CSF virus, vaccination against CSF is not practiced and generally is not allowed. But now in Korea, routine vaccination is practiced because of outbreaking the CSF repeatedly. When CSF is diagnosed the whole herd and other in contact animal are slaughtered continuously.

Investigation of diseases incident to pre- and pos-weaning piglets (포유 및 이유자돈의 질병발생 동향)

  • This experiment was carried out to investigate the tendency of incident diseases in pre- and post- weaning piglets which ages were 1 to 7 weeks old by laboratory diagnosis and in order to minimize death in preweaning piglets and of stunted growth in postweaning piglets. The result of this experiment used as the basic data for the preventive programs in pre- and post- weaning piglets and were as follows: 23 different diseases diagnosed in 331 cases were studied in relation to age, season, and etiology. The most prevelent diseases of pre- and post- weaning piglet were Colibacillosis(79 case, 23.9%) and the major diseases were Salmonellosis(44 cases, 13.3%), Anemia(37 cases, 11.2%). Unknown viral disease(20 case, 6.1%), Rota viral infection(19 case, 5.8%), Porcine reproductive & respiratory syndrome(PRRS; 15 case 4.5%), Transmissible gastroenteritis(TGE; 12 case, 3.6%). The gastrointestinal disease, such as Colibacillosis, Salmonellosis, Swine dysentery, Clostridial infection, Rotaviral infection, TGE, Porcine epidemic diarrhea(PED) and Ballantidiosis occured pro- dominently in the period of pre- and post- weaning, which were 178 cases(53.8%) and not related to occurrence according to age and season. The respiratory diseases were Atrophic rhinitis(AR), Swine enzootic pneumonia, Pneumonic pasteurellosis, Pleuropneumonia, Branchopneumonia, PRRS and which were 48 cases(14.5%) and higher prevalent in spring and summer. The viral diseases was 73 cases(22.1%) that occurred in the period of 5 weeks piglet and prevalent mainly in spring. The bacterial diseases were 188 cases(56.8%) that were not related to occurrence according to age and season. Salmonellosis was prevalent in 3 to 5 weeks piglet and mainly occurred in summer. Viral septicemia and rotaviral infection occurred after 5 weeks piglets intensively and 3 to 5 weeks, respectively. And the both occurred without relation with season. PRRS occurred after 4 weeks piglet and prevalent in summer. TGE occurred 1 to 7 days old piglets and prevalent in spring and winter. Hematologic values of anemia was decrease in number of Red Blood Cell, concentration of Hemoglobin and Hematocrit. Amikacin, cephalothin, colistin, norfloxacin were effective to E coli, and amikacin, cephalothin, nortloxacin, neomycin were effective to Salmonellra spp. but clindamycin, erythromycin, penicillin, sulfonamides were resistant to E coli and Salmonella spp.

The Studies of the low power Wavelengths of He-Ne IR laser on the changes of the serum biochemical components in burn rats (저 출력 레이저 파장에 따른 화상 쥐 혈청 생화학 성분 변화에 대한 연구)

  • This studies were to investigate the effects of low power wavelengths Helium-Neon Infra-Red(He-Ne IR)laser on the changes of the blood biochemical components in burn rats. The thirty Spraque-Dewely adult male rats were assigned to the 5 groups; the experimental groups(3), the burn control group(1) and the control group(1). There was made three degree burn by the 250mW IR on the back of each rats, from 3 days after being, burned, the experimental laser groups were irradiated low power wavelengths (292Hz, 1168Hz, 4672Hz) He-Ne IR laser for 5 minutes every day during the 7days. The results wee as follows: The activities of aspirate aminotransferase(AST), alanine aminotransferase(ALT), and lactate dehydrogenase(LDH), on treated with the wavelengths ((292Hz, 1168Hz, 4672Hz) laser groups during 5 min. for 7 days were significantly increased to the control group respectively, but those of total cholesterol(CHOL) and alkaline phosphatase(ALP) were significantly decreased to the a control group (p<0.05). there were significantly decreased on the activity of alanine aminotransferase and alkaline phosphatase on the all experimental groups to the burn control group and those of lactate dehydronase on treated with 292Hz and 4672Hz wavelength9 groups to the burn control group but, were significantly increased on those of urea nitrogen (292Hz and 1168Hz)(p<0.05). The activity of AST, LDH, on treated with 1168Hz wavelengths were significantly decreased of the 292Hz and 4672Hz wavelengths groups that of ALT on the treated with 1168Hz group was significantly increased to the 4672 Hz wavelength group but those of CHOL and LDH on the 1168Hz and 4672Hz wavelength groups were dignificatly decreased to the MHz wavelengths group. As above results, the changes of the activities of biochemical conponents in the serum levels on the healing process have meaningful effected of the role of low power wavelengths He-Ne IR laser.

  • Nonspecific suppressions of the immune responses have been reported to occur in the cancer-bearing patients. The mechanisms responsible for these immunosuppressions remain unclear but some investigators suggested that the immunosuppressions may result from immunosuppressive factors exudated from tumor masses. This study was undertaken to evaluate the in vivo effects of ascitic and pleural fluids from patients with cancers metastatic to peritoneum or pleura on the immune responses of mice. Non-cancerous ascitic and pleural effusions were used as controls. The ascitic fluids from hepatomap atients and pleural fluids from pulmonary carcinoma patients decreased both the delayed-type hypersensitivity and the antibody formation only when these were injected before antigenic stimulation. Control effusions exerted no effect on the immune responses to mice. These results suggested that cancerous fluids suppressed the immune responses by affecting the afferent arc of the immune system.

  • The role of macrophages in the resistance of ICR mice to Candida albicans and Salmonella typhimurium was assessed using silica, agent which selectively inactivates macrophages or poly-2-vinylpyridine-N-oxide(PVNO), a lysosomal stabilizing agent. In addition, effect of silica on humoral and cellular immune responses to sheep red blood cells(SRBC) or polyvinylpyroridone(PVP) was examind. Colonyforming units(CFU) of C. albicans or S. typhimurium in the spleen, livers and kidneys of silica-treated or diluent-treated mice were enumerated at various times after infection. Silica was given i. v. to mice at 4 days or 1 day before infection. Although there was no apparent differences in the number of CFU of C. albicans cultured from the spleens or livers of silica-treated and control mice at every assay period, significant differences in the number of CFU of C. albicans in the kidneys of silica-treated and control mice. Namely, silica given to mice 1 day before infection significantly increased the number of CFU of C. albicans in the kidneys at 2, 4 and 6 days after infection, but did not change the number of CFU at 8 days after infection. Silica given to mice at 4 days before infection significantly increased the number CFU in the kidneys at 2 and 4 days after infection, but rather decreased the number of CFU at 8 days after infection. The number of CFU of C. albians cultured from the kidneys of splenectomized which were experimentally infected mice was similar to the number recovered from sham-operated mice at 4 and 8 days postinfection irrespective of time of infection relative to operation. The pretreatment of mice with PVNO appeared to abrogate the silica-induced susceptibility of mice to C. albicans. PVNO alone showed somewhat protective effect against challenge with C. albicans. In contrast, silica treatment did not alter the number of CFU of S. typhimurium recovered from the spleens and kidneys of mice. The administration of silica to mice at 4 days or 1 day before SRBC immunization significantly suppressed delayed-type hypersensitivity(DTH) reactions to SRBC and antibody production to SRBC, a thymus-dependent antigen and PVP, a thymus-independent antigen. These results provide evidence that macrophages play an important role in susceptibility to Candida infection and strongly demonstrated that macrophages play an essential role in the induction of immune responses in mice.
