Effect of Lecithin Intake on Lipid Metabolism and Antioxidative Capacity in Rats Fed High Fat Diet (레시틴섭취가 고지방 식이를 섭취한 흰쥐의 지방대사와 항산화능에 미치는 영향)

  • This study was performed to investigate the effect of lecithin on lipid metabolism and antixidative capacity in 9-week-old rats. Forty-five male Sprague-Dawley rats weighing 249.8 g were blocked into three groups according to their body weight and raised for 8 weeks with experimental diets containing 1% (LM) or 5% lecithin (LH) and control (C) diet. Plasma and liver total lipids, triglyceride, total cholesterol and plasma HDL-cholesterol concenterations, and fecal total lipids, triglyceride, total cholesterol and bile acid excretions were measured. Malondialdehyde (MDA) levels in plasma, liver, superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) activities in red blood cell and liver, xanthine oxidase (XO) activities in plasma and liver, and total antioxidant status (TAS) in plasma were also measured. Effect of lecithin intake on antioxidative capacity was not significantly different among all the groups. Plasma total lipids, triglyceride and total cholesterol levels were lower in lecithin groups compared to control group, and these three lipid levels of lecithin groups were lowered dose-dependently as dietary lecithin level increased. But liver total lipids, triglyceride and total cholesterol levels were not different among all the groups. Also fecal total lipids, triglyceride and total cholesterol excretions were highest in high lecithin groups compared to two other groups. Thus it is plausible that lecithin intake decreases plasma lipid levels through increasing fecal lipid excretions, and may be beneficial for treatment and prevention of hyperlipidemia, but has no effect on antioxidative capacity.

Regulation of HIF-1α stability by lysine methylation

  • The level and activity of critical regulatory proteins in cells are tightly controlled by several tiers of post-translational modifications. HIF-1α is maintained at low levels under normoxia conditions by the collaboration between PHD proteins and the VHL-containing E3 ubiquitin ligase complex. We recently identified a new physiologically relevant mechanism that regulates HIF-1α stability in the nucleus in response to cellular oxygen levels. This mechanism is based on the collaboration between the SET7/9 methyltransferase and the LSD1 demethylase. SET7/9 adds a methyl group to HIF-1α, which triggers degradation of the protein by the ubiquitin-proteasome system, whereas LSD1 removes the methyl group, leading to stabilization of HIF-1α under hypoxia conditions. In cells from knock-in mice with a mutation preventing HIF-1α methylation (Hif1αKA/KA), HIF-1α levels were increased in both normoxic and hypoxic conditions. Hif1αKA/KA knock-in mice displayed increased hematological parameters, such as red blood cell count and hemoglobin concentration. They also displayed pathological phenotypes; retinal and tumor-associated angiogenesis as well as tumor growth were increased in Hif1αKA/KA knock-in mice. Certain human cancer cells exhibit mutations that cause defects in HIF-1α methylation. In summary, this newly identified methylation-based regulation of HIF-1α stability constitutes another layer of regulation that is independent of previously identified mechanisms.

Effects of Dietary Combinations of Vitamin A, E and Methionine on Growth Performance, Meat Quality and Immunity in Commercial Broilers

  • The experiment was conducted to study the effect of dietary combinations of vitamin A (VA), vitamin E (VE) and methionine (Met) on growth performance, meat quality and immunity in commercial broilers. Ross chicks (n=3,630) were allocated to five experimental treatments with three replicates per diet. The dietary treatments were: VA 8,000 IU, VE 10 IU (diet 1); VA 12,000 IU, VE 10 IU (diet 2); VA 8,000 IU, VE 100 IU (diet 3); VA 12,000 IU, VE 100 IU (diet 4) and; VA 12,000 IU, VE 100 IU/kg diet and 20% Met higher than other groups (diet 5). The Met content in diet 1 to diet 4 were as per the requirement suggested by NRC. Separate vitamin premixes were prepared for each treatment diet as per the requirement of study. The 35 d study revealed significantly (p<0.0001) higher weight gains in broilers fed diet 3 and diet 5, than in the rest of the groups during starter phase (0-3 weeks) only. The feed intake did not vary significantly at all phases of study, but feed efficiency was significantly (p<0.05) lower in diet 1 during starter and overall phase (4-5 weeks). The bone strength and bone composition, except bone calcium, remained unaffected due to experimental diets studied after 35 d of experimental feeding. The thio-barbituric acid reactive substances (TBARS) were significantly (p=0.0013) lower in the breast meat in group 5, followed by group 3, than in the rest of the groups. The immune studies conducted, antibody titers to sheep red blood cells, thickness index to phytohaemagglutinin-P, and heterophil: lymphocyte ratio, did not show any significant difference among treatments. It could be concluded that supplementation of VA, VE and Met at higher levels could be beneficial to broilers only during the starter phase.

Preparation and characterization of Ga-68-deferoxamine to test the feasibility as a bifunctional chelating agent or a renal imaging radiopharmaceutical

  • Chelating agents 1,4,7-triazacyclononanetriacetic acid (NOTA), 1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid (DOTA) and 30-amino-3,14,25-trihydroxy-3,9,14,20,25-penta-azatriacontane-2,10,13,21,24-pentaone (desferrioxamine, DFO) were labeled with $^{68}Ga$ and tested in vitro properties to check the feasibility of using DFO as a bifunctional chelating agent or renal imaging agent. The chelating agents of concentration $2{\mu}M$ were labeled with $^{68}Ga$ in 0.1 M HCl at pH 1.7-10.3 at room temperature and $80^{\circ}C$ and the optimal pH for labeling each chelating agent was found. And then, the chelating agents were labeled with $^{68}Ga$ in various concentration of chelating agents at optimal pH. The labeled chelating agents were subject to stability test in human serum and to binding studies to human red blood cell (RBC) and plasma protein. The optimal pH's of NOTA, DOTA and DFO for $^{68}Ga$-labeling were 4.4, 3.6 and 5.6, respectively. DFO ($10{\mu}M$) showed high labeling efficiency (>97%) at pH 5.6. All the labeled chelating agents showed high stability in human serum. $^{68}Ga$-DFO showed low RBC binding but significant amount was bound to plasma protein. The results demonstrated that $^{68}Ga$-DFO can be used as a bifunctional chelating agent but not as a renal imaging agent.

Estimating Infection Distribution and Prevalence of Malaria in South Korea Using a Back-calculation Formula (후향연산식을 활용한 국내 삼일열 말라리아의 감염분포와 유병자수 추정)

  • Incidence of Plasmodium vivax malaria in South Korea have been reemerged from mid-1990 and infected around 1600 patients annually recent years. The authors calculated the distribution of malaria infection and prevalence in South Korea using incidence (2001-2006) and incubation period distributions by a back-calculation formula and the least squares estimation method. The estimated infection has a normal distribution with a mean 207 and a standard deviation 30.7 days. In addition, the authors found the estimated daily average prevalence is 628.8 patients.

The Experimental Study on the Anti-allergic Effects of Sipjeondaebo-tang (십전대보탕이 알레르기반응에 미치는 영향)

  • This experimental research has been done to study the effects of Sipjeondaebo-tang(SOT) on the anti-allergic reaction. We found the several important results from the research which has been performed by two experiments toward immediately type and delayed type in order to study the effects of SDT on hypersensitivity response to mice. The results obtained from our research are as following. The survival rate of one group to which we injected only the compound 48/80 is almost 0% according to its density and timing test. In the other hand, the survival rates of the other group to which we injected both of the compound 48/80 and SDT are 20%, 10%, 30%, 10%, 40% and 70% according to 0.025, 0.05, 0.1, 0.25 0.5 and 1 (mg/g) of compound 48/80. Time dependency test also shows the 30% and 20% survival rates in 5 and 10 minutes. SDT revealed the significantly inhibitory effect on Compound 48/80 induced Mast cell degranulation. SDT showed the significantly inhibitory effect in the delayed type hypersensitivity response to picry1 chloride. SDT showed the significantly inhibitory effect in the delayed type hypersensitivity response to sheep red blood cell. Our research provides the important evidence that SDT is benificial to the prevention and treatment of allergy related diseases.

Safety Pharmacology of CJ-11555 (CJ-11555의 안전성 약리실험)

  • Safety pharmacological properties of CJ-11555, an anti-cirrhotic agent, were investigated in experimental animals and in vitro test system. CJ-11555 had no effects on normal body temperature in rats, motor coordination, chemoshock induced by pentetrazol, electric shock induced by electric shocker and writhing syndromes in mice at dose levels of 100, 300 and 1,000 mg/kg. CJ-11555 inhibited intestinal activity and prolonged hexobarbital-induced sleeping time in mice at the dose level of 1,000 mg/kg. CJ-11555 affected on general activity and behaviour tests in SD rats, such as lacrimation, ptosis, piloerection, decreased body tone, abnormal dispersion within the cage, diarrhoea, red colored faeces, slight hypothermia and decreased grooming, at the dose level of 1,000 mg/kg in rats. CJ-11555 was effected on cardiovascular and respiratory system in anesthetized beagle dogs, such as tachycardia, increase of mean blood pressure and decrease of PR interval, decrease of respiratory rate and minute volume, at dose levels of 10 and 30 mg/kg. However, these effects were also observed in vehicle treated anesthetized beagle dogs. In in vitro experiments, CJ-11555 inhibited agonists (histamine, acetyl-choline or $BaCl_2$) induced contraction of isolated guinea-pig at the concentration of 30$\times$$10^6$ M. CJ-11555 was weekly inhibited hERG channel current at concentrations of 10 and 30$\times$$10^6$ M, and $IC_{50}$ was estimated to be higher than 30${\times}$$10^6$M. Based on these results, it was concluded that CJ-11555 affected on cardiovascular and respiratory system, general activity and behaviour and hexobarbital-induced sleeping time at the dose level of 1,000 mg/kg and contraction of the smooth muscle and hERG channel current at the concentration of 30$\times$$10^6$ M.

Evaluation of Two ELISA and Two Indirect Hemagglutination Tests for Serodiagnosis of Pulmonary Hydatid Disease

  • To establish a definite diagnosis for pulmonary hydatid disease, combination of radiology and serology is useful. In this study, 19 preoperative sera from patients with surgically confirmed pulmonary hydatidosis, 40 sera from patients with other parasitosis and pulmonary diseases, and 20 sera from healthy donors were evaluated using 4 different serological tests, i.e., the commercial ELISA (ELISA-kit) test, the ELISA (ELISA-lab) test prepared in our laboratory, the commercial indirect hemagglutination assay kit (IHA-kit) test, and the IHA test using sensitized sheep red blood cells with tannic acid (IHA-TA). The ELISA-kit was the most sensitive (84.2%) and the most specific test (100.0%). The ELISA-kit also demonstrated the highest positive (100.0%) and negative (95.2%) predictive values. The sensitivity of the ELISA-lab test, that we prepared, was found to be 73.6%, whereas the IHA-kit test and the IHA-TA test were found to be 73.6% and 68.4%, respectively. The specificity of these tests was 96.6%, 98.3%, and 83.3%, respectively. When all 4 tests were assessed together, it was found that the sensitivity had risen to 94.7%. When the ELISA-kit was assessed with the IHA-kit and IHA-TA together, it was found that the sensitivity was 89.5% and 84.2%, respectively. Likewise, the combination of the ELISA-lab and IHA-kit or IHA-TA allowed us to achieve a sensitivity of 84.2% in cases of pulmonary echinococcosis. In conclusion, the diagnosis would be imminent if least 2 tests were applied together.

Effect of Terminalia chebula on Physiological Activity in Mice (가자(Terminalia chebula) 추출물이 마우스의 생리활성에 미치는 영향)

  • In this study, we investigated the effect of water, extract of Terminalia Chebula (TC) on physiological activity in mice. TC water extract showed hemagglutination against several different types of red blood cells. $LD_{50}$ of TC extract was 390 mg/kg (po). Treatment of TC water extract orally administered 200, 300 mg/kg daily for one week. Hepatic cytosolic enzymes, xanthine oxidase and aldehyde oxidase activities were significantly increased comparison with normal group. Treatment of TC water extract increased hepatic malondialdehyde (MDA) formation, and reduced glutathione content. We also found that the decreased activities of glutathione S-transferase and glutathione reductase but was not affected activities of $\gamma$-glutamylcysteine synthetase after treatment of TC water extract. These results suggested that increase of the hepatic lipid peroxide is caused by glutathione reduction.

Intestinal T cell lymphoma in a cat, Korea (고양이 장에서 발생한 T 세포 림프종)

  • An 11 year-old male Korean short-haired cat was presented to local animal hospital due to weight loss, vomiting, and intestinal hypomotility. After the cat was euthanized by poor clinical outcomes, necropsy was performed at Animal and Plant Quarantine Agency. At necropsy, the stomach was enlarged and had some nearly complete pellet food and the yellow mucous contents. The lumen of the middle and lower parts of the jejunum became narrow. Histopathologically, medium-sized lymphoid cells with hyperchromatic nuclei enclosed by scant cytoplasm were diffusely proliferated from mucosa to serosa of the small intestine. These findings were mainly observed in the jejunum and slightly in the duodenum and ileum. The monomorphous lymphocytes were 1 to 1.5 times larger than red blood cells and had few mitotic figures. Metastasis of the tumor cells to other organs was not observed. In the result of immunohistochemical analysis for identifying the origin of tumor cells, CD3 was expressed, but $CD79{\alpha}$ was not detected in the infiltrated cells. This case was diagnosed as T cell intestinal lymphoma in a Korean short-haired cat based on the clinical signs, gross findings, histopathology, and immunohistochemistry.
