Effects of Pretreatment Time and pH low set value on Continuous Mesophilic Hydrogen Fermentation of Food Waste (열처리 시간과 pH 하한값이 음식물쓰레기 연속 중온 수소 발효에 미치는 영향)

  • Since 2005, food waste has been separately collected and recycled to animal feed or aerobic compost in South Korea. However, the conventional recycling methods discharge process wastewater, which contain pollutant equivalent to more than 50% of food waste. Therefore, anaerobic digestion is considered as an alternative recycling method of food waste to reduce pollutant and recover renewable energy. Recent studies showed that hydrogen can be produced at acidogenic stage in two-stage anaerobic digestion. In this study, the authors investigated the effects of pretreatment time and pH low set value on continuous mesophilic hydrogen fermentation of food waste. Food waste was successfully converted to $H_2$ when heat-treated at $70^{\circ}C$ for 60 min, which was milder than previous studies using pH 12 for 1 day or $90^{\circ}C$. Organic acid production dropped operational pH below 5.0 and caused a metabolic shift from $H_2/butyrate$ fermentation to lactate fermentation. Therefore, alkaline addition for operational pH at or over 5.0 was necessary. At pH 5.3, the result showed that the maximum hydrogen productivity and yield of 1.32 $m^3/m^3$.d and 0.71 mol/mol $carbohydrate_{added}$. Hydrogen production from food waste would be an effective technology for resource recovery as well as waste treatment.

Experimental Study of Isolated Canine Heart Preservation for 24 Hours at 4$^{\circ}C$ - A Portable Continuous Hypothermic Perfusion System - (적출심장의 장시간 [24시간] 보존에 관한 실험적 연구 -4$^{\circ}C$ 관류 보존법-)

  • After 24 hours of preservation under 15 mmHg perfusion pressure the recovery rates of isolated canine hearts were determined. Preservation was performed in a cold room maintained at 4*C with 4 different types of perfusates bubbled with a mixture of 95% 0y and 5% CO~ using a modified perfusion unit designed in our institute. The perfusates used were as follows; Group 1: Krebs-Henseleit solution, Group 2: Krebs solution added by albumin and PGE1. Group 3: Modified Wicomb*s solution, Group 4: Modified Collin*s solution. The extent of myocardial recovery was evaluated using a modified isolated carmine perfusion model by measuring heart rate, systolic arterial pressure, left atrial pressure[LAP] and cardiac output. In addition to the above hemodynamic parameters, biochemical and enzymatic assays from perfusates and electron microscopic changes of the myocardium were also studied. The results were as follows; 1] The heart recovery rates were 41.6%, 53.4% and 108.9% in groups 1, 2 and 3, respectively, and group 3 elicited the best result[p< 0.001]. The heart beat was never recovered in group 4. 2] Recovered systolic arterial pressures[mmHg] were 63.3% in group 1, 94.9% in group 2 and 94.3% in group 3. 3] LAPs[mmHg] were 20 in group 1, 13.5 in group 2 and 11.2 in group 3, which suggested that the best myocardial preservation was elicited in group 3[p< 0.05]. 4] Cardiac output, the sum of aortic stroke volume and coronary leakage, were 69.1% in group 2, and 90.7% in group 3, but these were not statistically significant[p=0.24]. No aortic stroke output was measured in group 1 and 4. 5] The degree of myocardial edema increase was 17.5` in group 1, 24.6% in group 2, 20.9% in group 3 and 55.3% in group 4. But there were no statistical differences in each group[p= 0.08]. 6] CPK-MB[U/L] levels were increased 750% and 332%[p< 0.05], glucose levels[mg/dl] 60.5% and 78.2% and SGOT[U/L] levels 523% and 333%, in groups 2 and 3, respectively. Biochemical and enzymatic assays could not be performed in group 1 and group 4, because of poor recovery of heart beat. 7] Electron microscopic findings in the myocardium of most groups revealed slight to moderate muscle cell and mitochondrial edema. But all these findings were within the limits of reversible change. From these above results, it is suggested that modified Wicomb*s solution seems to be the most useful physiologic salt solution for preservation of the heart. We propose that after further study and improvement, our portable continuous hypothermic perfusion system will contribute to the development of a better preservation method for donor hearts for human heart transplantation.

Bioproduction of trans-10,cis-12-Conjugated Linoleic Acid by a Highly Soluble and Conveniently Extracted Linoleic Acid Isomerase and an Extracellularly Expressed Lipase from Recombinant Escherichia coli Strains

  • The low solubility and high-cost recovery of Propionibacterium acnes polyunsaturated fatty acid isomerase (PAI) are key problems in the bioproduction of high value-added conjugated linoleic acid (CLA). To improve the solubility of recombinant PAI, six chaperone proteins were coexpressed with PAI. Introduction of GroELS proteins dramatically improved the PAI solubility from 29% to 97%, with increased activity by 57.8%. Combined expression of DnaKJ-GrpE and GroELS proteins increased the activity by 11.9%. In contrast, coexpression of DnaKJ-GrpE proteins significantly reduced the activity by 57.4%. Plasmids pTf16 harboring the tig gene and pG-Tf2 containing the tig and groEL-groES genes had no visible impact on PAI expression. The lytic protein E was then introduced into the recombinant Escherichia coli to develop a cell autolysis system. A 35% activity of total intracellular PAI was released from the cytoplasm by suspending the lysed cells in distilled water. The PAI recovery was further improved to 81% by optimizing the release conditions. The lipase from Rhizopus oryzae was also expressed in E. coli, with an extracellular activity of 110.9 U/ml. By using the free PAI and lipase as catalysts, a joint system was established for producing CLA from sunflower oil. Under the optimized conditions, the maximum titer of t-10,c-12-CLA reached 9.4 g/l. This work provides an effective and low-cost strategy to improve the solubility and recovery of the recombinant intracellular PAI for further large-scale production of CLA.

Recycling of Waste Bittern from Salt Farm (I) : Recovery of Magnesium (염전 폐간수의 재활용(I) : 마그네슘 회수)

  • The purpose of the research was to examine the utilization of waste bittern from salt farm as a source for producing magnesium (Mg). In this work, a precipitation process for recovering Mg, where Mg is precipitated as $Mg(OH)_2$ by the addition of NaOH solution, was investigated. At the NaOH/Mg molar ratios of 2.70 : 1 to 2.75 : 1 and pH 9.5-10, > 99% of Mg could be precipitated from the bittern. The molar concentration of NaOH solution added as an alkaline reagent had no significant influence on the recovery efficiency of Mg precipitate. The particle size of Mg precipitate was strongly affected by the flow rate of caustic addition. The faster the flow rate of caustic addition, the smaller particles were formed. The Mg precipitate recovered was 100-120 g per 1 L of bittern and contained 94% $Mg(OH)_2$ after washing with water. Our results showed that the bittern can be used as a potential resource for Mg production.

Comparative Analysis of Dissolution and Refolding Processes for Inclusion Body Protein Renaturation (내포체 단백질 재생을 위한 용해 및 재접힘공정의 비교분석)

  • Using rlFN-$\alpha$ and rhGH as the model proteins, the refolding performances of the published processes were evaluated and compared. Key engineering parameters such as the type of denaturant and this concentration, protein concentration in the refolding buffer, and pH and ionic strength of the buffer were experimentally investigated. Furthermore, the role of a co-solvent of surfactant type in aggregation reduction was also studied. Of the denaturants tested (8M urea, 6M guanidine HCI, 0.5% SDS), SDS at alkaline pH (9.5) and ambient temperature gave the highest recovery yield. The SDS process was effective in the refolding of observed where dissolution proceeded better under lower strength (10 mM) but aggregation was suppressed under higher strength (>50 mM.) When PEG-4000 and/or Tween were added as co-solvent or refolding-enhancing additive, 1.6-2 times higher yield was realized. The‘masking’of the hyrophobic patches located on the surface of the protein with the surfactant molecules was believed to be responsible for the considerable reduction in aggregation during refolding.

Purification and Characterization of an Extracellular Alkaline Protease from Aspergillus niger C-15

  • An alkaline protease produced by Aspergillus niger C-15 was purified and characterized. The enzyme was purified 19.41-fold with a specific activity of 74150 U/mg and a recovery of 34.4% by gel filtration and ion exchange chromatography. The molecular weight of the enzyme was estimated to be 34 kDa. The optimum pH and temperature for the protease activity were pH 8.0 and $60^{\circ}C$, respectively. The enzyme activity inhibited by EDTA suggests that the preparation contains a metalloprotease. The enzyme activity of the metalloprotease was completely inhibited by 5 mM $HgCl_2$ and $FeCl_3$, while partially inhibited by $CuSO_4$, and $MnCl_2$. When polyols such as glycerol, mannitol, sorbitol and xylitol, were added to the reaction medium, most polyols tested enhanced protease activity. Especially, glycerol showed the highest effect. The alkaline metalloprotease was stable at high temperature and retained more than 90% of the initial activity at $60^{\circ}C$ and 86.4% under addition of glycerol.

Atomic Absorption Spectrophotometric Determination of Trace Cadimuim after Preconcentration by Extracting Its 8-Hydroxyquinoline Complex into Molten Benzophenone

  • A sensitive method for the determination of trace cadmium after the preconcentration by extracting its 8-hydroxyquinoline complex into a molten benzophenone was developed. Several experimental conditions such as the pH of solution, the amounts of 8-hydroxyquinoline and benzophenone, stirring time, and standing time were optimized. Trace cadmium in 100 mL water sample was chelated with 2.5 mL of 0.001 M 8-hydroxyquinoline at pH 8.0. After 0.07 g benzophenone was added, the solution was heated to about 70 ℃ and stirred vigorously for 1 minute to dissolve the complex quantitatively in a molten benzophenone, and stood for 30 minutes to reproduce the microcrystalline benzophenone. The benzophenone containing Cd-8-hydroxyquinoline complex was filtered and dissolved in acetone. Cadmium was determined by a flame atomic absorption spectrophotometry. The interfering effects of diverse concomitant ions were investigated and eliminated. This method could be applied to natural water samples and the recovery of more than 90% was obtained in the real samples.

Precipitation of Magnesium Sulfate from Concentrated Magnesium Solution for Recovery of Magnesium in Seawater (해수 중 마그네슘 회수를 위한 마그네슘 농축액으로부터 황산마그네슘의 석출)

  • The precipitation test, which is the last step of magnesium recovery process consisting of three processes (pre-precipitation, selective dissolution of magnesium, precipitation) is performed to obtain magnesium sulfate powder from seawater. In the study, we succeed in precipitating the magnesium sulfate by adding acetone into the solution of magnesium over 4 times concentrated from seawater. The yield efficiency of magnesium sulfate increases with increasing pH and the ratio of added acetone. More than 99% of magnesium is obtained as magnesium sulfate hydrate ($MgSO_4{\cdot}6H_2O$) under the following conditions; pH 1.0 ~ 1.5, and the ratio of solution and acetone 1 : 1.5 (v:v). The acetone used in the precipitation process is recovered by the fractional distillation.

TiO2 Reuse and Recovery from the Photocatalytic Oxidation of Cu(II)-EDTA using TiO2/UV-A System (TiO2/UV-A 시스템을 이용한 Cu(II)-EDTA의 광촉매 산화반응에서 TiO2 재사용 및 회수)

  • $TiO_2-catalyst$ suspensions work efficiently in Photocatalytic oxidation (PCO) for wastewater treatment. Nevertheless, once photocatalysis is completed, separation of the catalyst from solution becomes the main problem. The PCO of Cu(II)-EDTA was studied to determine the reusability of the titanium dioxide catalyst. Aqueous solutions of $10^{-4}M$ Cu(II)-EDTA were treated using illuminated $TiO_2$ particles at pH 6 in a circulating reactor. $TiO_2$ was reused in PCO system for treatment of Cu(II)-EDTA comparing two procedures: reuse of water and $TiO_2$ and reuse of the entire suspension after PCO of Cu(II)-EDTA. The results are as follows; (i) Photocatalytic efficiency worsens with successive runs when catalyst and water are reused without separation and filtration, whereas, when $TiO_2$ is separated from water, the reused $TiO_2$ is not deactivated. (ii) The $TiO_2$ mean recovery (%) with reused $TiO_2$ was 86.4%(1.73g/L). Although the mean initial degradation rate of Cu(II)-EDTA and Cu(II) was lower than that using fresh $TiO_2$, there was no significant change in the rate during the course of the three-trial experiment. It is suggested that Cu(II)-EDTA could be effectively treated using an recycling procedure of PCO and catalyst recovery. (iii) However, without $TiO_2$ separation, the loss of efficiency of the PCO in the use of water and $TiO_2$ due to Cu(II), DOC remained from previous degradation and Cu(II)-EDTA added to the same suspension was observed after 2 trials, and resulted in the inhibition of the Cu(II)-EDTA, Cu(II) and DOC destruction.

A Prospective Study on the Changes of Serum 3,5,3'-Triiodothyronine Levels after Cardiopulmonary bypass in Pediatric Age (소아환자에서 개심술후 혈중 3,5,3'- 삼옥소타이로닌 [triiodothyronine] 농도 변화에 관한 연구)

  • Triiodothyronine[T3] is an important regulator of the tissue metabolism, and may have potential use as an inotropic agent. The change of serum T3 level was studied in the pediatric age patients after cardiopulmonary by pass. Thyroid function was tested pre-operatively in 33 patients and total triiodothyronine[TT3] levels were serially measured during and after cardiopulmonary bypass[CPB]. After correction of dilutional effect, effects of various factors on the TT3 levels were analyzed. Abrupt fall of TT3 level was demonstrated at 15 minutes after CPB[80.1$\pm$5.9ng/dL] from the initial level of 133.6$\pm$5.3ng/dL, with some recovery at 6 hours[115.4$\pm$6.7ng/dL]. After then, gradual decrease occured reaching to the level of 77.2$\pm$4.2ng /dL at 24 hours. These falls of the TT3 after CPB were statistically significant. [p<0.01 ANOVA] Statistically significant correlation were found between the degree of hemodilution and TT3 concentration at 15 and 30 minutes after CPB[p<0.05]. But, other factors were analyzed to have no effect on TT3 levels. As the degree of the hemodilution increases, TT3 decreased less. This observation probably supports the fact that decrease of TT3 during CPB may be a result of sequestration of T3 into peripheral tissue. Although it was not statistically significant[p=0.08], the fall of TT3 was greater in the group to which plasmanate was added, than those not added. This finding seemed to be due to the increase of albumin and other thyroid-hormone-binding-proteins in the serum. Increase of these binding proteins might potentiate the sequestration of T3 into the liver and the kidney from serum, and as a consequence, decrese the serum TT3 level further.
