Cellular DNA Repair of Oxidative Deoxyribose Damage by Mammalian Long-Patch Base Excision Repair

  • 2-Deoxyribonolactone (dL) arises as a major DNA damage induced by a variety of agents, involving free radical attack and oxidation of C1'-deoxyribose in DNA. We investigated whether dL lesions can be repaired in mammalian cells and the mechanisms underlying the role of DNA polymerase $\beta$ in processing of dL lesions. Pol $\beta$ appeared to be trapped by dL residues, resulting in stable DNA-protein cross-links. However, repair DNA synthesis at site-specific dL sites occurred effectively in cell-free extracts, but predominantly accompanied by long-patch base excision repair (BER) pathway. Reconstitution of long-patch BER demonstrated that FEN1 was capable of removing the displaced flap DNA containing a 5'-dL residue. Cellular repair of dL lesions was largely dependent on the DNA polymerase activity of Pol $\beta$. Our observations reveal repair mechanisms of dL and define how mammalian cells prevent cytotoxic effects of oxidative DNA lesions that may threaten the genetic integrity of DNA.

올라퍼 엘리아슨의 작품에 나타난 블러 색채 특성 연구 (A Study on the Characteristics of Blur Color in Olafur Eliasson's Works)

  • This thesis has purpose of explaining that blur color is not only appreciated but also undecided spacial color in contemporary space design and examining effects of space experience through the light. In keeping such developments, blur color has been perceived as the object of another tool in contemporary space and has provided us with new views by providing various program, sensor and screen from digital media. With the expansion of blur color, light has changed a spacial color structural that is not simple meaning factor into the essential concept. So this research aims to understand the principle of blur color in Olafur Eliasson's work, which of the fine artist in diverse fields from installation art to media art. The principle of blur color such as deconstruction of vanishing point, dimensional transfer, color gradation, and fiction of visual perception makes it possible to extract the expressed element and method of blur color. Meanwhile, the features of blur color such as sense of depth from viewpoint changes, hybrid from collapse of order, media metamorphosis from reconstitution and interaction from uncanny can be inferred from case analysis.

  • A straightforward and effective method is presented for immobilizing enzymes on a microchip platform without chemically modifying a micro-channel or technically microfabricating a column reactor and fluid channel network. The proposed method consists of three steps: the reconstitution of a nitrocellulose (NC) membrane on a plane substrate without a channel network, enzyme immobilization on the NC membrane, and the assembly of another substrate with a fabricated channel network. As a result, enzymes can be stably and efficiently immobilized on a microchip. To evaluate the proposed method, two kinds of enzymatic reaction are applied: a sequential two-step reaction by one enzyme, alkaline phosphatase, and a coupled reaction by two enzymes, glucose oxidase and peroxidase, for a glucose assay.

  • In the past several years the popularity of the motor cycle has produced an increasing incidence of the injuries to the larynx and trachea. Most of all on accidents come to death and survivors to the hospital are rare. Early diagnosis and to keep air way are necessary to initiate proper treatment in injury of upper air way. Meticulous apposition of mucous membrane and reconstitution of laryngeal skeleton are important. We experienced a rare case of 26 year old men with cricothyroidal transection after trauma. On Oct. 17, 1985, the patient struck his neck on baggage frame of truck when dropping from his motor cycle on sudden stop. Emergency tracheal intubation on distal segment of trachea was accomplished by otolaryngologist in a local clinic. He was transferred to our hospital. Exploration 2 hours later revealed complete separation of cricoid cartilage from thyroid cartilage. The recurrent laryngeal nerve could not be identified. Anastomosis of thyroid and cricoid was accomplished and Portex endotracheal tube was inserted as splint for 10 days. No stenosis developed. The air way appeared adequate for moderate physical activity though paramedian fixation of vocal cord paralysis. Postoperative follow-up course has been good after he discharged on POD 14 days.

Leucostim, A Human Granulocyte-Colony Stimulating Factor, Facilitates Granulopoiesis After Bone Marrow Transplantation In Mice

  • In vivo administration of Leucostim, a human recombinant granulocyte colony-stimulating factor (G-CSF), was evaluated for the effects on survival, hematologic recovery, and colony forming unit- spleen (CFU-5) in murine bone marrow transplantation (BMT) model. Sublethally irradiated (9 Gy) mice received bone marrow cells from untreated mice, and then were treated with G-CSF subcutaneously at doses of 2.5,5, or $10\mu\textrm{g}$/kg or vehicle solution (control) for 14 days from one day after BMT. There was no effect of irradiation and BMT on mortality. The repeated subcutaneous injections of Leucostim for 14 days post- BMT significantly facilitated hematologic recovery compared with vehicle control in a dose-dependent manner. Moreover, mice treated with Leucostim had significantly increased numbers of CFU-s colonies on day 10 post-BMT. These results suggest that Leucostim, a new G-CSF, has beneficial effects on hematologic reconstitution after BMT.

Non-Permanent Transcatheter Proximal Renal Artery Embolization for a Grade 5 Renal Injury with Delayed Recanalization and Preserved Renal Parenchymal Enhancement

  • Super-selective renal artery embolization is an increasingly popular technique for the management of traumatic, low-grade renal trauma. When performed in distal arterial branches, this intervention enables tissue preservation and arrest of hemorrhage, but it may not be practical in cases of multifocal, high-grade renal injuries. In such cases, surgical nephrectomy remains the more common treatment modality to ensure hemodynamic control. We present the unique case of a patient who presented in hemorrhagic shock following a major trauma that resulted in a grade 5 renal injury treated with complete renal artery embolization using Gelfoam, resulting in hemodynamic stabilization. Interestingly, imaging 1 month after embolization revealed residual enhancement of the inferior pole of the kidney, suggesting reconstitution of flow and partial renal salvage. Ultimately, transcatheter "nephrectomy" with careful selection of a temporary embolic agent may serve as a safe and efficient alternative to surgical nephrectomy with the added possibility of preserving partial renal perfusion and function in the emergent setting.

A Simple Method for Determining Residual p-arsanilic Acid in Aquatic Products using EDTA-Assisted Solvent Extraction and LC-MRM

  • A simple method was developed to determine residual p-arsanilic acid (ASA), an organo-arsenic compound used as a feed additive, in aquatic products (eel, halibut, and shrimp) using EDTA-assisted solvent extraction and LC-MRM. The method was successfully validated in terms of specificity, linearity (coefficient of determination ≥ 0.995), accuracy (recovery or R, 72.72-78.73%), precision (the relative standard deviation of R, 2.08-6.98%), and sensitivity (the lower limit of quantitation, 5 ppb) according to CODEX guidelines (CAC-GL 71-2009). The use of EDTA in the extraction solvent and water with a suitable pH modifier as the reconstitution solvent may be the key factors for successful results. This is the first method that can be used for monitoring residual ASA in aquatic products using LC-MRM and could contribute to establishing a better aquatic product safety management system.

정보보호 사전점검 활성화를 위한 점검항목 개선 연구 (A Study on Improvement of Inspection Items for Activation of the Information Security Pre-inspection)

  • IT환경은 사물인터넷, SNS, 빅데이터, 클라우드 컴퓨팅 등으로 급변하고 있다. 이 기술들은 기존 기술에 새로운 기술이 추가되어 정보시스템 간의 복잡도를 증가시킨다. 그러므로 신규 정보통신서비스의 보안 기능 강화가 필요하다. 정보보호 사전점검은 신규 정보통신서비스 설계 단계부터 보안을 고려한 개발 단계를 제시하여 신규 정보통신서비스 제공자와 이용자에게 안정성 및 신뢰성 보장을 목표로 한다. 기존 '정보보호 사전점검(점검영역 22개, 통제항목 74개/세부항목 129개)'은 6단계(요구사항, 설계, 교육, 구현, 시험/테스트, 대응/사후)로 구성되어 있으며 실효성 검토를 위해 실제 정보시스템 개발 업체에서 파일럿 테스트를 수행하였다. 그 결과로 점검항목에 대한 개선 요구사항과 일부 점검항목의 재구성이 필요한 것으로 나타났다. 본 논문은 정보시스템의 사전예방이 목적인 '정보보호 사전 점검'의 활성화를 위한 연구를 하였다. 그 결과로 요구사항과 보완사항이 반영된 '정보보호 사전점검(점검영역 16개, 점검항목 54개/세부항목 76개)'을 도출하였다.

  • 염증 질환의 원인이 되는 사람 중성구 elastases는 혈액에 존재하는 ${\alpha}_1-PI$${\alpha}_2-macroglobulin$과 같은 단백질 분해효소 억제제에 의하여 조절되어진다. 그러나 특수한 병리적 상황에서 과다하게 분비되는 효소나 또는 단백질 분해효소 억제제의 비정상적 작용으로 말미암아 다양한 염증질환이 유발된다. 비스테로이드성 항염증제는 염증 질환을 치료하기 위하여 이미 임상에 적용 하고 있으며, prostaglandin 합성하는 효소인 cyclooxygenases의 활성도를 억제하는 것이 그 작용 기전으로 잘 알려져 왔다. 사람 중성구 elastase의 활성도는 naproxen, phenylbutazone, oxyphenbutazone 등에 의하여 억제되었으나, ibuprofen, ketoprofen, aspirin, salicylic acid, tolmetin 등에 의하여서는 억제되지 않았다. 또한 사람 중성구 elastase의 활성도는 EDTA, EGTA, tetracycline 등에 의하여서도 억제되었다. EDTA에 의하여 2가 이온 $Ca^{++}$$Zn^{++}$등을 elastase 분자로부터 일부 제거함으로 효소 활성도가 억제되었고 Raman spectra의 변화도 강하게 일어났으며, 금속이온 $Zn^{++}$를 새로 충진함으로 그 활성도는 원래대로 회복되고 Raman spectrum도 원래 상태와 유사한 상태로 회복되었다. 이런 현상은 chelator나 chelator-like agents가 효소분자안에 존재하는 $Zn^{++}$ 이온을 제거하거나 chelation함으로 활성 부위나 그 인접 부위의 4차원 구조의 변화를 일으켰음에 기인하며 특히 -C=O나 -COOH기의 관여에 의한 것으로 생각된다.

  • 환경설계를 통한 범죄예방이론(Crime Prevention Through Environmental Design : CPTED)은 어떻게 지속 가능한 발전의 수요에 따라 융합할 것인가라는 문제에 있어서 아직 현단계 제3세대 이론의 발전 과정을 지속하고 있다. 게다가 국제적으로도 논쟁과 토론을 많이 일으키고 있다. 더욱 주목할 만한 것은 그 논쟁과 토론하는 관점이 이론 모델 및 이론 모델 속의 원칙 간의 상호 작용 관계에 무시 못 할 영향을 일으켰다는 것이다. 본 연구는 이론 발전의 전체적 맥락을 파악한 후 CPTED이론 발전의 정체성이 환경과 환경 사용자 상호 간에 다양한 이해의 차원과 교류의 방식을 제공한다. CPTED 이론 발전의 정체성을 기초로 해서 CPTED 범죄 예방 디자인 패턴은 지속 가능성의 다양한 차원을 연결시켜야 하며, 거주적합성을 디자인 목표의 출발점으로 삼고 Top-down 패턴에 의거하여 환경의 안전성에 대해 조정해야 한다는 관점을 명확히 했다. 그러나 단지 기존 물리적 설계 방법으로 상향식(Bottom-up)의 패턴을 사용해 환경을 개선해서는 안 된다. 그래서 본 연구는 정체성을 핵심으로 하향식의 범죄 예방 디자인 패턴을 결합해, 이를 이론 모델의 재구성으로 시각화 제안을 제기했다. 이는 어떤 단계의 성과나 원칙의 응용에 보충하는 것이 아니며, 따라서 CPTED의 이론적 모델과 지속 가능한 발전에 참고와 자료를 제공할 것으로 기대된다.
