  • Polymerase chain reaction (PCR) is well established as an indispensable tool of molecular biology; and yet a limitation for cloning applications continues to be that products often require subsequent restriction to be that products often require subsequent restriction digests, blunt-end ligation, or the use of special linear vectors. Here a rapid, PCR-based system is described for the simple, restriction enzyme-free generation of synthetic, 'restriction-like' DNA fragments with staggered ends. Any 3'- or 5'-protruding terminus, but also non-palindromic overhangs with an unrestricted single strand length are specifically created. With longer overhangs, "Restriction-PCR" does not even require a ligation step prior to transformation. Thereby the technique presents a powerful tool e.g. for a successive, authentic reconstitution of sub-fragments of long genes with no need to manipulate the sequence or to introduce restriction sites. Since restriction enzyme-free and thereby devoid the limitations of partial DNA digests, "Restriction-PCR" allows a straight one-step generation and cloning of difficult DNA fragments that internally carry additional sites for specific sequence insertions or deletions can be precisely engineered into genes of interest. With these properties "Restriction-PCR" has the potential to add significant speed and versatility to a wide variety of DNA cloning applications.

  • A nicotinic acetylcholine receptor (nAchR) isolated from the electric tissues of Torpedo californica has been reconstituted into a vesicle comprising a bifunctional azo-ligand (Bae 1) compound, and a liposome containing phospholipids and cholesterol (1 : 1, w/w). The liposome-mediated reconstituted receptor showed a concentration-dependent response to cholinergic drugs in a lithium ion flux assay. This liposome-mediated reconstituted nAchR was immobilized onto an electrode using various synthetic polymers which were tested for their response to the cholinergic ligands. The immobilized nAchR not only exhibited a linear response to a wide range of cholinergic ligand concentrations but also retained an operational stability which lasted for longer than 6 days. Thus, this result provides a basis for application of the immobilized nAchR-based biosensor in detecting cholinergic ligands in vitro.

  • The population of the elderly is rapidly increased because maturation of the social welfare system and development of medical technology. However, welfare environments for the elderly are poor either in its quality or its quantity. The aims of this study is to confirm the space improvement possibility to change the Gyung Ro Dang to the senior life support center as a community welfare service network. This study is done by the comparative analysis between existing Gyung Ro Dang in korea and former cases in Japan based on the elderly's facility preference. The facility's space reprogramming alternative from that analysis is applied to change the Gyung Ro Dang to a network facility for the regional senior life support. Afterward, it is proposed that first, facilities should be expanded scale by the new education and health improvement facilities functional room that the modern elderly need. second, Facility space will be reconstitution because the Gyung Ro Dang have to be extended it's role as a community welfare network.

  • This study is aimed to specify the meaning of the body and the design trends in contemporary architecture. Architecture is based on the human life of various meaning, events, experiences, images, senses and interactions through the body. Thoughts, behaviors, and senses of human are interrelated in architectural experiences. Individuals experience the built environments and space, not through the ideas but through the senses and movements of the body. So, bodies make the real space of architecture. Contemporary architecture accepts the theory of phenomenology and places on the thoughts of Maurice Merleau-Ponty, Heidegger, Nroberg Schulz and so on. Such researches effect on the architectural trends to make design processes works on the programs in views of the expansion and the structuralization of human body. In detail, the aim of this study is to analyse the architecture as the fields of the subject with body as the center, design processes and principles changed form metaphysical thought to phenomenological discourse, and the design trends in contemporary architecture at last. In process of movements, vision centered architecture moves into the bodily experienced architecture and changed the trends from absolute form design to design of relative processes. In conclusion, architectural formation-dissolution-reconstitution of body creates the architectural thoughts such as human proportions, perspective space, ergonomics, modular, organic architecture, experience space, synesthesia, event architecture, fashion-invoked architecture, interactive surfaces, metamorphosis, and others.

  • Recently, various types of image contents are being produced, and interest in copyright protection technology is increasing. In this paper, we propose an image encryption technology for color images. This technique divides the image into RGB color components and then performs quad-tree decomposition based on the edge of image. After the quad-tree partitioning, encryption is performed on the selected blocks. Encryption is performed on color components to measure encryption efficiency, and encryption efficiency is measured even after reconstitution into a color image. The encryption efficiency uses a visual measurement method and an objective image quality evaluation method. The PSNR values were measured as 7~10 dB for color difference components and 16~19 dB for color images. The proposed image encryption technology will be used to protect copyright of various digital image contents in the future.

  • In this study, horse spleen apoferritins were induced to form biominerals using up to 3000 Fe atoms per protein molecule. The morphology and crystallinity of the nanometer-sized biominerals formed in the ferritins were then analyzed using field emission-energy filtering-transmission electron microscopy (FE-TEM). The ferritins were found to have reconstitution yields of 60-70% in the experiments. The mean core size of the ferritins varied somewhat with protein concentrations, indicating that crystal growth in ferritins could be controlled via protein concentrations. The core mineral size increased with the amount of Fe used. Lattice fringes of the core, associated with good crystallinity, were found in all samples. The lattice fringe images of a single domain ferrihydrite mineral appeared frequently in the (011) planes (d-spacing of 0.246 nm) under [100] zone axis in all samples of this study. In addition, the lattice image occasionally revealed fringes corresponding to the (100) planes (d = 0.254 nm) from the [001] zone axis, indicating the characteristic pattern of hexagonal crystal lattice. Diffraction patterns in the minerals identified as ferrihydrite were fitted well into the space group of $P3_{1c}$.

  • The purpose of this study was to examine elementary teachers' perception on birds and problems on teaching it. For this study, the science textbooks from 3rd grade to 6th grade according to the 7th national curriculum were analyzed. Results revealed that, firstly, there were 34 bird species recorded and these were described 55 times in the surveyed textbooks. Secondly, the survey showed that $20\%$ of the teachers were interested in birds despite the fact that $98\%$ of them had no special knowledge about birds nor had studied them in college. Additionally, it was revealed that $41\%$ of the teachers felt lacking in specialist knowledge about birds and this lead to difficulties in teaching on this subject. Thirdly, academic backgrounds had an affect teachers' perceptions on teaching this area. The teachers' levels of interest in birds and the teachers' ratio of teaching on this topic tended to increase as their academic careers developed, and the proportion of male teachers in this category were higher than female teachers. To overcome difficulty in teaching on this area, it is suggested that a reconstitution of subject materials, as well as a revision of the teachers' guidebooks be undertaken.

  • First half of my article focused on analyzing the current state of historical materials regarding weather and climate, and established a list of weather-related historical literature collection of Korea with which to make a lexical approach to the situations of all kinds of weather literature. It also put emphasis on gathering information and data of weather logs from journal-type historical records which were contained in 48 weather-related journals of Choseon period. The results of this research are expected to be useful for the activation of study in historical meteorology. The latter half of my research focused on analyzing various meteorological states of sunny, cloudy, rainy, snowy and frosty weather which were recorded in the official Annals of King Kojong (1864~1907). And it re-verified historical rainfall data of preceding researches of Wada Yuji (1917), Jung-Lim (1994), Jhun-Moon (1997). In result, different records were found between data of theirs and mine. It means that we have to analyze and reconstruct newly the meteorological data of the Annals of King Gojong and the Daily Records of Royal Sungjungwon (1623~1910) during the late Choseon period.

  • In this study, we evaluated the stability of amoxicillin (AMX) and clavulanic acid (CLA) in aqueous solution, and compared the stability of AMX and CLA in commercial combination products. In aqueous solutions, the degradation of AMX ($t_{90}=8.88\;day\;at\;10^{circ}C$) and CLA ($t_{90}=3.53\;day\;at\;10^{circ}C$) occurred rapidly. From the pH-rate profile, AMX and CLA were the most stable at the range of pH 5.5 and 6.0. After reconstitution of commercial dry syrups, the contents of AMX and CLA in suspensions were gradually decreased for 7 days. However, AMX and CLA in dispersible tablet were not changed at all. The contents of CLA in the dispersible tablet ($87.92\%$) and dry syrups (2.16 and $3.91\%$) were remained in the accelerated stability test ($75\%\;RH,\;at\;40^{circ}C$) after 10 hours. And the colors of the dry syrups were rapidly changed from white to yellow. From these results, we concluded that the dispersible tablet could overcome the stability problems of dry syrups.

  • Heterotrimeric G Proteins transduce ligand binding to a wide variety of seven transmembrane cell surface receptors into intracellular signals. The currently accepted model for the activation of G protein suggests that ligand-activated receptor accelerates GDP-GTP exchange reactions on the ${\alpha}$ subunit of the heterotrimeric G protein. At least seventeen distinct isoforms of the G${\alpha}$ subunit protein have been identified in mammalian organisms. Among them, the G${\alpha}$q family consists of five members whose ${\alpha}$ subunits show different expression patterns. G${\alpha}$q and G${\alpha}$11 seem to be almost ubiquitously expressed, whereas G${\alpha}$14 is predominantly expressed in spleen, lung, kidney and testis. G${\alpha}$16 and its murine counterpart G${\alpha}$15 are expressed in hematopoietic cells and has been shown to couple a wide variety of receptors to phosphoinositide-specific phospholipase C activity. Beta-isoforms of phospholipase C were shown to be activated by all members of G${\alpha}$q family, i.e., G${\alpha}$q, G${\alpha}$11, G${\alpha}$l4 and G${\alpha}$16 subunits either in reconstitution system. or in experiments using cDNA transfection with intact Cos-7 cells.
