Genomic RNA was extracted from Cy strain of odontoglossum ringspot tobamovirus (ORSV-Cy) isolated from infected leaves of tobacco cv. Samsun. Size of the genomic RNA was about 6.6 kb in length. The genomic RNA was fractionated using Sephadex G-50 column chromatography into 2 fractions. They were polyadenylated at their 3'-end using E. coli poly(A) polymerase. Polyadenylated viral RNA was recovered by oligo (dT) primer adapter containing NotI restriction site and Moloney murine leukemia virus SuperScript reverse transcriptase (RNase H-). Second-strand cDNA was synthesized by using E. coli DNA ligase, E. coli DNA polymerase I and E. coli RNase H. Recombinant plasmids containing cDNAs for ORSV-Cy RNA ranged from about 800 bp to 3,000 bp. Among the selected 238 recombinants, pORCY-124 clone was the largest one covering 3'-terminal half of the viral RNA. This clone contained two restriction sites for EcoRI and XbaI and one site for AccI, AvaI, BglII, BstXI, HindIII, PstI, and TthIII 1. respectively. The clone contained partial viral replicase, a full-length movement protein and a complete coat protein genes followed by a 3' untranslated region of 414 nucleotides based on restriction mapping and nucleotide sequencing analyses. Clones pORCY-028, -068, -072, -187 and -224 were overlapped with the pORCY-124. Clones pORCY-014 and -095 covered 5' half upstream from the middle region of the viral RNA, which was estimated based on restriction mapping and partial sequence analysis. Constructed cDNA library covered more than 90% of the viral genome.
Metallothionein은 세포내의 중금속의 농도을 조절하는 주요한 단백질로서 bacteria에서 척추동물에 이르기까지 모든 생명체에서 나타나는 공통된 단백질이다. 비록 metallothionein의 정확한 기능은 알려져 있지 않으나 독성을 나타내는 중금속에 대하여 세포내 방어기작에 관여할 뿐만 아니라 여러다른 유전자의 총괄적 조절기작 및 matalloprotein의 발현에 관여할 것으로 보고있다. 본 연구에서는 Channel Catfish의 metallothionein cDNA 유전자를 poly(A)를 갖는 mRNA로 부터 Reverse Transcriptase-Polymerase Chain Reaction(RT-PCR)에 의하여 cloning하였다. 증폭된 PCR products는 pBluescript SK+의 EcoRV site 및 pUC19의 Smal site에 dT tailing을 하여 cloning하였으며, PCR products는 multicloning site에 있는 EcoRI 및 HindIII 로 절단하여 확인하거나 신속한 PCR screening에 의하여 확인하였다. 여러 PCR clone 중 하나인 pMT150에 대한 DNA 염기서열을 조사한 결과 다른 어류의 metallothionein cDNA 유전자와 높은 유사성을 보였다.
Bacillus thurintensis subsp. kurstaki HD1 살충성 단백질 ICP 유전자가 있는 NdeI 단편 3.856 kb를 클로닝하여 제조한 pHLN2-80(-) 클론이 pHLN1-80(-)에 비해서 대장균에서 ICP발현량이 과다발현되는 현상을 규명하고자 하였다. 본 연구에서는 상기의 pHLN2-80(+) 클론의 발현량을 조절하는 원인을 규명하기 위하여 ICP의 아미노산 서열은 변화되지 않는 범위 내에서 pHLN1-80(+) 클론에 있는 Plac프로모터와 ICP유전자 프로모터의 일부인 -80 bp프로모터의 염기서열, 전사 개시점과 종결부위의 변이가 ICP유전자발현에 미치는 영향을 조사하였다. pHLN1-80(+)에 5'-말단에 존재하는 -80 bp 프로모터만을 보유한 pHLNK-80 클론은 ICP 생산은 매우 저조하였다. Plac프로모터와 -80 bp 프로모터의 구조 골격을 일부 변이 시킨 pHLNF1-80클론의 ICP생산량은 pHLN2-80(-)가 생산한 양보다는 낮아서 과다발현이 안되었다. Plac프로모터 상류를 약 350bp을 제거하여 만든 클론 pHLND2-80의 ICP 생산량은 모클론인 pHLN2-80(-) 보다 매우 높게 과다발현 되었다. ICP 유전자의 과다발현 현상에 대한 전사 개시점과 전사종결 부위의 역할을 알아보기 위해서 -72bp ICP유전자프로모터를 갖는 클론 pHLD1-72는 재조합 클론 pHLN2-80(-)가 생산한 양보다 적은 양의 ICP을 생성하였고, 클론 pHLD2-72는 재조합 클론 pHLN2-80(-)보다 적은 ICP을 발현하여 과다 발현되었으며, 클론 pHLN2-72는 모클론인 pHLN2-80(-)보다 약간 높은 ICP 생산량을 보여 과다발현되었다. 클론 pHLN2-72를증식하여 파쇄액을 만든 후에 Bombyx mori유충에 대한 살충력 검사에서 클론 pHLN2-72이 생산한 ICP는 pHLN1-80(+)이 생산한 ICP보다 약 90배의 살충력을 보였다. SDS-PAGE와 Western blot 분석에서도 클론 pHLN2-72는 재조합 클론 pHLN2-80(-)보다 약간 높게 ICP가 생성이 되었었다. 이상의 결과는 과다발현에 Plac프로모터와 종결부위가 반드시 필요하며, -72 bp ICP 프로모터가 -80 bp 프로모터보다 과다발현률이 높았으며, ICP 유전자는 반드시 Plac프로모터의 전사 방향에 역방향으로 삽입이 되어야 하는 것으로 나타났다.
Porphyromonas gingivalis, a Gram negative, anaerobic, asaccharolytic rod, is one of the most frequently implicated pathogens in human periodontal disease and has a requirement for hemin for growth. A 30 kDa (heated 24 kDa) hemin-binding protein whose expression is both hemin and iron regulated has recently been purified and characterized in this oral pathogen. This study has identified a hemin-binding P. gingivalis protein by expression of a P. gingivalis genomic library in Escherichia coli, a bacterium which does not require or transport exogenous hemin. A library of genomic DNA fragments from P. gingivalis was constructed in plasmid pUC18, transformed into Escherichia coli strain $DH5{\alpha}$ , and screened for recombinant clones with hemin-binding activity by plating onto hemin-containing agar. Of approximately 10,000 recombinant E. coli colonies screened on LB-amp-hemin agar, 10 exhibited a clearly pigmented phenotype. Each clone contained various insert DNA. The Hind III fragment transferred to the T7 RNA polymerase/promoter expression vector system produced a sligltly smaller (21 kDa) protein, a precursor form, immunoreactive to the antibody against the 24 kDa protein, suggesting that the cloned DNA fragment probably carried an entire gene for the 24 kDa hemin-binding protein.
The number of sites at which a protein can be readily cleaved by a proteolytic enzyme is greatly influenced by its three-dimensional structure. For native, properly-folded proteins both the rate of cleavage and number of sites at which cleavage takes place are usually much less than for the denatured protein. In order to compare the tertiary structure of recombinant HepG2 type glucose transporter with that of its native counterpart in the erythrocyte, the pattern of tryptic cleavage of the protein expressed in insect cell membranes was therefore examined. After 30 minutes digestion, a fragment of approximate Mr 19,000-21,000 was generated. In addition to this, there were two less intensely stained fragments of apparent Mr 28,000 and 17,000. The pattern of labelling was similar up to 2 hours of digestion. However, the fragments of Mr 19,000-21,000 and Mr 17,000 were no longer detectable after 4 hours digestion. The observation of a very similar pattern of fragments yielded by tryptic digestion of the HepG2 type transporter expressed in insect cells suggests that the recombinant protein exhibits a tertiary structure similar if not identical to that of its human counterpart. Also, the endogenous sugar transporter(s) present in Sf21 cells did not bind cytochalasin B, the potent transporter inhibitor. Therefore, the baculovirus/Spodoptera frugiperda (Sf) cell expression system could be very useful for production of large amounts of human glucose transporters, heterologously.
We have got several clones from Serratia marcescens which stimulated the cells to use maltose as a carbon source in E. coli TP2139 (${\Delta}$lac, ${\Delta}$crp). One of the cloned genes, pCKB13, was further analyzed. In order to find whether the increased expression of the gene under the direction of maltose metabolism, we constructed several recombinant subclones. We have confirmed that the clone, pCKB13 codes Coenzyme A transferase gene by partial nucleotide sequencing in the terminal region. The enzyme activity of Coenzyme A transferase increased after introduction of the multicopy of the cloned gene in E. coli. The recombinant proteins expressed by multicopy and induction with IPTG, two polypeptide of 26-and 28-kDa, were confirmed by SDS-PAGE. Southern hybridization analysis confirmed that the cloned DNA fragment was originated from S. marcescens chromosomal DNA.
Genetic chracterstics of the structural gene of guamerin (a novel elastase inhibitor from Korean leech), integrated into the HIS4 locus of chromosomal DNA of Pichia pastoris along with the $\alpha$-factor leader sequence, were investigated. In the selected clone from candidates, two copies of the integration cassette including the structural gene copies of the integration cassette including the structural gene of guamerin were found in the integration site of the chromosomal DNA of P.pastoris. It was demonstrated that the integrated structural gene of guamerin was stable up to about 70 generations in the relay flask culture. Then, a high-cell-density culture could be fulfilled easily by DO-stat fed-batch culture, in which the cell growth and the recombinant guamerin production reached about 250 of OD600nm and 260 mg/l, respectively. Finally, it was revealed that the DNA sequence of the integrated structural gene of guamerin in P. pastoris was maintained correctly in the end of production cells of relay flask culture and high-cell-density culture.

Application of recombinant protein production and particularly their isotopic enrichment has stimulated development of a range of novel multidimensional heteronuclear NMR techniques. Peptides in most cases are amenable to assignment and structure determination without the need for isotopic labeling. However, there are many cases where the availability of $^{15}N$ and/or $^{13}C$ labeled peptides is useful to study the structure of peptides with more than 30 residues and the interaction between peptides and membrane. CRAMP (Cathelicidin-Related AntiMicrobial Peptide) was identified from a cDNA clone derived from mouse femoral marrow cells as a member of cathelicidin-derived antimicrobial peptides. CRAMP was successfully expressed as a GST-fused form in E. coli and purified using affinity chromatography and reverse-phase chromatography. The yield of the CRAMP was 1.5 mg/l 1. According to CD spectra, CRAMP adopted ${\alpha}$-helical conformation in membrane-mimetic environments. Isotope labeling of CRAMP is expected to make it possible to study the structure and dynamic properties of CRAMP in various membrane systems.
Glycosyltransferase (GT)-specific degenerate PCR screening followed by in silico sequence analyses of the target clone was used to isolate a member of family1 GT-encoding genes from the established fosmid libraries of soil actinomycetes Micromonospora echinospora ATCC 27932. A recombinant MeUGT1 was heterologously expressed as a His-tagged protein in E. coli, and its enzymatic reaction with semi-synthetic phenoxodiol isoflavene (as a glycosyl acceptor) and uridine diphosphate-glucose (as a glycosyl donor) created two different glycol-attached products, thus revealing that MeUGT1 functions as an isoflavonoid glycosyltransferase with regional flexibility. Chromatographic separation of product glycosides followed by the instrumental analyses, clearly confirmed these previously unprecedented glycosides as phenoxodiol-4'-α-O-glucoside and phenoxodiol-7-α-O-glucoside, respectively. The antioxidant activities of the above glycosides are almost the same as that of parental phenoxodiol, whereas their anti-proliferative activities are all superior to that of cisplatin (the most common platinum chemotherapy drug) against two human carcinoma cells, ovarian SKOV-3 and prostate DU-145. In addition, they are more water-soluble than their parental aglycone, as well as remaining intractable to the simulated in vitro digestion test, hence demonstrating the pharmacological potential for the enhanced bio-accessibility of phenoxodiol glycosides. This is the first report on the microbial enzymatic biosynthesis of phenoxodiol glucosides.
한국 마늘 바이러스의 유전자 구조와 병 발생 메카니즘을 연구하기 위하여, 바이러스가 감염된 마늘잎으로부터 바이러스 입자를 분리하고 RNA를 추출하였다. 그 virus RNA를 이용하여 마늘 바이러스 cDNA 유전자 은행을 만들어 일부 clone의 염기 서열을 결정하였다. 여기에서 얻은 cDNA clones 중에서 poly(A) tail을 갖는 clone S81를 분리하고 873 bp의 전체 염기서열을 결정하였다. Clone S81의 염기서열을 다른 식물 바이러스와 비교한 결과 potexvirus의 껍질단백질 부분의 염기서열과 $30{\sim}40%$의 유사성을 보여주었다. Clone S81은 바이러스 RNA의 3' 말단 부위에 해당하고, 껍질단백질의 N-terminal 3개 아미노산이 빠진 open reading frame (ORF) 및 3'-noncoding region을 포함하고 있다. 3' 말단 부분에는 바이러스 복제과정에서 cis-acting element로 작용한다고 여겨지는 hexamer motif와 polyadenylation signal이 존재한다. 이 clone을 probe로 하여 Northern blot을 실시한 결과 genome의 크기는 7.5 knt라는 것을 알 수 있었고 clone S81은 potexvirus의 cDNA clone이라는 결론을 얻었다. 한국 마늘에서 이 바이러스의 분포 양상을 알아보기 위해 껍질단백질에 대한 항체를 만들었다. 먼저 발현벡터를 이용하여 대장균에서 대량으로 발현시키고 affinity chromatography로 껍질단백질을 정제하였다. 그 단백질을 토끼에 주사하여 껍질단백질에 대한 항체를 얻었다. 이 항체를 사용하여 다양한 지역에서 재배되는 마늘잎의 추출액에 대해 immunoblot을 실시하였다. 그 결과 분자량 29,000과 27,000 위치에서 signal을 보였다. 분자량 27,000 단백질이 29,000이 분해되어 생긴 산물인지 알아보기 위하여 그 추출액을 $37^{\circ}C$에서 시간을 달리하여 incubation한 후 immunoblotting하였다. 그 결과도 마찬가지로 같은 위치에서 signal을 보여줬다. 따라서 한국 마늘에는 재배되는 지역에 따라 다소 다르기는 하지만 대체로 두 종류의 potexvirus로 감염되어 있다고 추정된다.
