Objective : The aim of this study is to determine the antimutagenic effect and genetic safety of Buthus martensi Karsch aqua-acupuncture solution(BMKAS) against various chemical carcinogens. Method : Ames(Salmonella typhimurium) test and Rec assay(Bacillus subtilis) were used as indicators for DNA damage and antimutagenesis. Furthermore, the levels of umu operon expression by measuring the ${\beta}$-galactosidase activity wete monitored with the SOS umu test using S. typhimurium 1535 containing plasmid pSK1002. And the host-mediated assay was used to investigate the mutagenicity and antimutagenicity of BMKAS inducing various chemical carcinogens after the activation with in vivo metabolic systems. Results : From the results, BMKAS did not atfect DNA of S. typhimurium and B. subtilis strains and showed no mutagenicity at the all concentrations of tested solution. Furthermore BMKAS dose-dependently protected the mutagenecity by AF-2, 2-AA and B[a]P. These phenomena was also similar to that after metabolic activation of BMKAS in in vivo system. Conclusion : These results suggested that BMKAS did not show the mutagenicity and protected the mutagenesis against various chemical carcinogens by four different methods used in this study.

The mutagenic and antimutagenic activities of juices from 20 common edible wild herbs found in Korea were investigated using the spore-rec assay and Ames test. The juices of Hemerocallis fulva and Capsella bursapastoris exhibited a little induction or inhibition of mutagenesis in the presence of selected metal ions, but juices of most edible wild herbs did not affect on the mutagenesis in the spore-rec assay. In the other hand, all of the juices strongly inhibited the mutagenesis induced by benzo[a]pyrene, 2-amino-fluorene, and 3-amino-1,4-dimethyl-5H-pyridol tested on Salmonella typhimurium TA98 or TA100 in the presence of S-9 mix. The antimutagenic effects increased as the concentration of the mutagens increase. The results suggest that concentration of samples or types of various mutagen interact to affect the antimutagenic potential of the juices in the TA98 and TA100 strain.
We have previoosly demonstrated that the RecA-like protein of Schizosaccharomyces pombe (S. pombe) is immunologically related to Escherichia coil (E. coil) RecA protein and that the cellular level of the protein is significantly increased by inhibitors of nucleotide pool-forming enzymes such as hydroxyurea (HU) and methotrexate (MTX) (lee and Park, 1994; lee et al., 1994). In this study, we report that the ribonudeotide redudase activity of S. pombe is inhibited by E. coil RecA antibody, as determined by thin layer chromatography using [5-$^3$H]CDP as a substrate. The relative activity of ribonucleotide reductase was dramatically inhibited by 100 mM of flu (26.4% reduction) in in vitro assay, compared to that of non-treated control. The ribonucleotide reductase activity was also inhibited by immunoprecipitation with E. coil RecA antibody (43.3% reduction). These results indicate that the strudure of S. pombe ribonucleotide reductase is in part similar to that of E. coil RecA protein.
Antaeum(ATE) has been used as a prescription for threatened abortion, associated with pregnancy in traditional medicine. Because gravida could be administered ATE for a long period, its administration might cause a harmful effect on fetus and gravida during the pregnancy. This study aimed to determine whether exposure to ATE caused mutagenicity or hepatotoxicity during the pregnant period. For mutagenicity test of ATE, Salmonella typhimurium and Bacillus subtilis were used as indications for DNA damage. In the Ames test, Samonella typhimurium TA98 and TA100 were used for mutagenicity testing, and the number of histidine revertants was measured. In Rec-assay, Bacillus subtilis H $17(Rec^+)$ and $M-45(Rec^-)$ strains were used to clarify the DNA damage property. In the SOS umu test, Salmonella typhimurium TA15335 containing plasmid pSK1002 was used as a tester strain, and we monitored the levels of umu operon expression by measuring the ${\beta}-galactosidase$ activity. From the tested results, ATE did not show DNA damage and mutagenicity. On the other hand, hepatotoxicity of ATE to female ICR mice was monitored by the measurements of s-GOT, s-GPT and LDH activities after oral feeding for 15 days. ATE did not show significant change of s-GOT, s-GPT and LDH activities in mice sera.

In order to obtain mutagenic data of enzymatic browning reaction products, we investigated their mutagenicity. The rec-assay with Bacillus subtilis strains $H17(rec^+)$ and $M45(rec^-)$ were carried out with their spores. Detection of double strand breakage in calf-thymus DNA was investigated into sample solution with and without $Cu^{2+}$ by the agarose slab gel electrophoresis. In the rec-assay, catechol, pyrocatechol, DL-dopa, 3,4-dihydroxytoluene, hydroquinone, hydroxyhydroquinone with and without $Cu^{2+}$ in 0.05M ana 0.1M at the enzymatic browning reaction products stowed mutagenic action. And also browning solution of 0.05M hydroxyhydroquinone and catechol with $Cu^{2+}$, hydroquinone with and without $Cu^{2+}$ of 0.1M at the enzymatic browning reaction products were strong mutagenic action. The DNA breakability of the enzymatic browning reaction products of 0.1M pyrogallol was stronger than that of 0.05M pyrogallol browning solution with $Cu^{2+}$ and 3,4-dihydroxytoluene browning solution was stronger than that of 0.01M 3,4-dihydroxytoluene browning solution.
This study was carried out to investigate the antimutagenic effects of five kinds of apple enzymatic browning reaction products(AEBRP) on mitomycin C (MMC), N-methyl-N'-nitro-N-nitrosoguanidine (MNNG), 4-nitroquinoline-1-oxide(4NQO), benzo(${alpha}$)pyrene(B(${alpha}$)P) and 3-amino-1,4-dimethyl-5H-pyri-do [4,3-b]indol (Trp-P-1). In spore rec-assay using B. subtilis Hl7($rec^+$) and M45($rec^-$), homocatechol-AEBRP and hydroquinone-AEBRP showed strong antimutagenic effects on MMC and MNNG as the concentration of AEBRP increased. In the Ames test using S. typhimurium TA98 and TA100, hydroxyhydroquinone-AEBRP and pyrogallol-AEBRP showed strong antimutagenic effects on Trp-p-1 and B(${alpha}$)p in TA98 and TA100 in the presence of S-9 mix. Most of AEBRPs suppressed about 50% to 80% the mutagenesis in S. typhimurium TA98 induced by MNNG, however, AEBRPs except hydroxyhydroquinone-AEBRP showed antimutagenic effects of about 94% in TA100. Antimutagenic effects of the five kinds of AEBRPs on 4-NQO were more or less weak, in particular homocatechol-AEBRP exhibited the inhibitory effect of about 48% in TA98, and homocatechol-AEBRP and hydroquinone-AEBRP showed inhibitory effects of about 46% to 58% in TA 100.
Potential mutagenicity of ten heated edible mountain herbs were examined with spore recassay, Ames test and DNA breaking test. Samples of edible mountain herbs were prepared with water extraction at $100^{\circ}C$ for 20 minutes. With the rec-assay, no significant mutagengic activity could be obtained from all of the samples, but among the eight of metal ions added to sample solution, $Pb^{2+}$ to R. crispus heated juice, $Zn^{2+}$ to L. fischeri and S. bracycarpa heated juice increased mutagenic activity of the samples. With the Ames test and DNA breaking test, all of the samples did not show mutagenicity. However, breaking action was activated on heated L. fischeri, P. japonicus. A. triphylla and A. tataricus juices in the presence of 25mM $Cu^{2+}$. But heated A. elata, H. aurantiaca, A. triphylla, S. bracycarpa and A. scaber juices were inactivated in the presence of 25mM $Fe^{2+}$. Desmutagenic activities against benzo$({\alpha})$pyrene significantly increased as increasing concentration of the heated edible mountain herb juices.
Various lactic acid bacteria were isolated from Korean Kimchi in order to study their antimutagenic substances. Ames test using Salmonella typhimurium TA98 and TA100 showed the strain KLAB21 to have the highest antimutagenic activity among the 230 isolated strains against MNNG (N-methyl-N'-nitro-N-nitrosoguanidine), NPD (4-nitro-O-phenylenediamine), NQO (4-nitrosoquinoline-1-oxide) and AFB1 (aflatoxin B1). The strain was identified as Lactobacillus plantarum based on its morphological, cultural and physiological characteristics. Antimutagenic activity of L. plantarum KLAB21 was found in culture supernatant suggesting the bacterium secrete antimutagenic substance in the media. No mutagenic activity was found in the culture supernatant. The isolated strain L. plantarum KLAB21 showed much higher antimutagenic activity than L. plantarum IAM1261 which is being used industrially for fermented milk production. The antimutagenic activity of L.plantarum KLAB21 was reconfirmed by the spore-rec assay using spores of Bacillus subtilis H17($Rec^+$) and M45($Rec^-$).
The mutagenicity and desmutagenicity on enzymatic browning reaction products which obtained from prunes salicina (yellow) enzyme and polyphenol compounds were carried out. In the rec-assay on Bacillus subtilis strains H17 and M45, the enzymatic browning reaction products of pyrogallol, hydroxyhydroquinone, 3,4-dihydroxytoluene and catechol of $10^{-2}M$ did not showed mutagenicity. In the effects of various metal ions on the rec-assay, the enzymatic browning reaction products of pyrogallol showed mutagenic activity by $Fe^{3+},\;Mn^{2+},\;Zn^{2+},\;Ni^{2+}$ and $Al^{3+}$. In the enzymatic browning reaction products of hydroxyhydroquinone, $Cu^{2+},\;Mn^{2+}$ and $Pb^{2+}$ were effected in mutagenic action and the enzymatic browning reaction products of catechol was effected in mutagenic action by $Mn^{2+}$. In the DNA-breaking action of enzymatic browning reaction products of pyrogallol, hydroxyhydroquinone, 3,4-dihyroxytoluene and catechol did not show, DNA-breaking action. In the effects of various metal ions on the DNA-breaking action of enzymatic browning reaction products, $Cu^{2+}$ showed DNA-breaking action. In the mutagenicity test on Sal. typhimurium strains TA98 and TA 100 with S-9 mix, 4 kinds of browned substances did sot shove muragenicity, all the browned substances showed strong desmutagenic activity in the presence of benzo $({\alpha})-pyrene$ with S-9 mix.
In the screening process of anti- or desmutagenic substance from the various microbial metabolites with the method of Ames and Rec-assay, a desmutagenic substance producing bacterial strain which inactivates the mitomycin C-induced mutagenicity was isolated and identified as Psudomonas sp. AM-10.
