  • Hyaluronic acid (HA) is one of the most abundant glycosaminoglycans (GAGs) in the female reproductive tract such as uterine, oviductal and follicular fluids in mouse, pig, cattle and human. CD44 is the principal cell membrane receptor for HA, expressed from the 1-to 8-cell stage in human embryos, during post-implantation mouse and bovine embryogenesis and on the surface of differentiated embryonic stem cells. Effect of Ginsenoside Rd on Nitric Oxide System Induced by Lipopolysaccharide Plus $TNF-{\alpha}$ in C6 Rat Glioma Cells

  • Effects of ginsenosides on nitric oxide (NO) production induced by lipopolysaccharide plus TNF-$\alpha$ (LNT) were examined in C6 rat glioma cells. Among several ginsenosides, ginsenoside Rd showed a complete inhibition against LNT-induced NO production. Ginsenoside Rd attenuated LNT-induced increased phosphorylation of ERK. Among several immediate early gene products, only Jun Band Fra-1 protein levels were increased by LNT, and ginsenoside Rd attenuated Jun Band Fra-1 protein levels induced by LNT. Furthermore, LNT increased AP-1 DNA binding activities, which were partially inhibited by ginsenoside Rd. Our results suggest that ginsenoside Rd exerts an inhibitory action against NO production via blocking phosphorylation of ERK, in turn, suppressing immediate early gene products such as Jun Band Fra-1 in C6 glioma cells.

  • Retinal prosthesis is regarded as the most feasible method for the blind caused by retinal diseases such as retinitis pigmentosa or age-related macular degeneration. One of the prerequisites for the success of retinal prosthesis is the optimization of the electrical stimuli applied through the prosthesis. Since electrical characteristics of degenerate retina are expected to differ from those of normal retina, we investigated differences of the retinal waveforms in normal and degenerate retina to provide a guideline for the optimization of electrical stimulation for the upcoming prosthesis. After isolation of retina, retinal patch was attached with the ganglion cell side facing the surface of microelectrode arrays (MEA). $8{\times}8$ grid layout MEA (electrode diameter: $30{\mu}m$, electrode spacing: $200{\mu}m$, and impedance: 50 $k{\Omega}$ at 1 kHz) was used to record in-vitro retinal ganglion cell activity. In normal mice (C57BL/6J strain) of postnatal day 28, only short duration (<2 ms) retinal spikes were recorded. In rd/rd mice (C3H/HeJ strain), besides normal spikes, waveform with longer duration (~100 ms), the slow wave component was recorded. We attempted to understand the mechanism of this slow wave component in degenerate retina using various synaptic blockers. We suggest that stronger glutamatergic input from bipolar cell to the ganglion cell in rd/rd mouse than normal mouse contributes the most to this slow wave component. Out of many degenerative changes, we favor elimination of the inhibitory horizontal input to bipolar cells as a main contributor for a relatively stronger input from bipolar cell to ganglion cell in rd/rd mouse.

  • RD-14은 CANDU형 발전소를 묘사하기 위한 실험장치로서 여러 초기조건에서 자연순환 실험이 수행되었다. 본 연구에서는 RD-14 실험장치의 자연순환 실험을 RELAP5 코드로 모사하여 결과를 비교·평가하였다. 본 연구는 CANDU형 발전소 사고해석의 검증용 코드로서 RELAP5 코드의 적용 타당성을 평가하기 위한 과정으로 수행되었다. 계산결과, RELAP5 코드는 RD-14 실험장치의 자연순환 실험을 잘 예측하고 있음을 보여주고 있어 CANDU형 발전소의 자연순환 평가에 유용하다는 결론을 얻었다.

  • In order to realize the simple and economic 3rd order LPF using switched-capacitor(SC), we proposed the 3rd order RC LPF using a OP amplifier and realized the 3rd order SC LPF by replacing all the resistors in the RC LPF to SC. The frequency response of SC LPF was compared with the that of RC LPF. As a result, it was found that the response of realized SC LPF is almost similar to that of RC LPF.

Morphological Characteristics of the Rice (Oryza sativa L.) with Red Pigmentation (적색종피 돌연변이 벼의 형질특성조사)

  • Seven rice germplasms with red pigmentation within the pericarp were isolated from a large mutant collection. These red pericarp phenotypes resulted from the functions of the Rc, Rd and RdRc genes. Among them, two brown pericarp of the Rc type, four red pericarp of the RdRc type, and one white pericarp of the Rd type were identified. Morphological and agronomic characteristics of those rice germplasms were studied. The Rc type germplasms have the faint red or brown color pericarp and the Rd types produce the white pericarp, whereas the RdRc type germplasms have the dark red pericarp. Most of the important agronomic characteristics including plant stature, tillering ability, spikelet fertility, and total grain yield were lower in the colored rice than those of the wild-type control. All of the studied colored rice germplasms had a tendency of easy seed-shattering in comparison to the control. These characteristics of newly identified germplasms will be useful for identifying the genes responsible for pericarp color phenotype determination.

  • It is expected that synaptic construction and electrical characteristics In degenerate retina might be different from those In normal retina. Therefore, we analyzed the retinal waveform recorded with multielectrode array in normal and degenerate retina using principal component analysis (PCA) and Independent component analysis (ICA) and compared the results. PCA Is a well established method for retinal waveform while ICA has not tried for retinal waveform analysis. We programmed ICA toolbox for spatiotemporal analysis of retinal waveform. In normal mouse, the MEA spatial map shows a single hot spot perfectly matched with PCA-derived ON or OFF ganglion cell response. However In rd/rd mouse, the MEA spatial map shows numerous hot and cold spots whose underlying interactions and mechanisms need further Investigation for better understanding.

Effects of the Antioxidative Components to Ginsenoside in the Liver of 40-week-old Mice (40주령의 생쥐 간에 미치는 ginsenoside의 항산화효과)

  • To study on antioxidant effects of saponin fractions, we investigated effects in the liver of 40-week-old mice to which were pretreated with 5 mg/kg per body weight of saponins for 5 days. The ability of saponins to protect against oxidative damage to the mouse liver was examined by determining the level malondialdehyde (MDA),hydrogen peroxide and the activity of superoxide dismutase (SOD) and catalase (CAT). The only panaxadiol (PD)among the ginseng saponin fractions significantly increased the hepatic SOD activities (p<0.01), Whereas PD, panaxatriol (PT), ginsenoside Rd (G-Rd) (p<0.01) and ginsenoside Re (G-Re) (p<0.05) significantly decreased the contents of hydrogen peroxide. It was only G-Rd that significantly increased CAT activities (p<0.05). The level of MDA was significantly decreased by G-Rd and PD. In conclusion, PD and G-Rd among the saponin fractions were especially increased in the activity of hepatic antioxidative enzyme and decreased the lipid peroxidation that was expressed in term of MDA formation.

Anti-lipid Peroxdation Effect of Ginsenoside Rd and Its Metabolite Compound K (진세노사이드 Rd와 사포닌 대사물인 compound K의 항지질과산화 효과)

  • To study on antioxidant effects in the liver of 40-week-old mouse, the sample were orally pretreated 5mg/kg/day for 5 days with red ginseng saponin components(total saponin, protopanaxadiol saponin, protopanaxatriol saponin, ginsenoside-Rd, ginsenoside-Re, compound-K) for 5 days. The ability of saponin to protect the mouse liver from oxidative damage was examined by determining the activity of superoxide dismutase(SOD), glutathione peroxidase(GPx) and the contents of glutathione, the level of malondialdehyde, The only protopanaxadiol among the ginseng saponin fractions was significantly increased the hepatic SOD activity(p<0.01). The red ginseng saponin induced a slight increase of GPx activity, especially ginsenoside Rd, compound K and protopanaxatriol treatments significantly increased its activity. The content of glutathione was significantly increased by total saponin, protopanaxadiol and ginsenoside Rd(p<0.01), but the oxidized glutathione level was lowered in all the red ginseng saponin. Finally, the level of malondialdehyde was significantly decreased by ginsenoside Rd and protopanaxadiol. In conclusion, protopanaxadiol and ginsenoside Rd among the saponin fraction were especially increased in the activity of hepatic antioxidative enzyme and decreased the lipid peroxidation that was expressed in term of MDA formation. This comprehensive antioxidant effects of red ginseng saponin seems to be by a certain action of saponin other than a direct antioxidant action.

  • Immunoglobulin G4-related disease (IgG4-RD) is a chronic inflammatory condition involving multiple organs, including the salivary or lacrimal glands, orbit, pancreas, bile duct, liver, kidney, retroperitoneum, aorta, lung, and lymph nodes. It is histologically characterized by tissue infiltration with lymphocytes and IgG4-secreting plasma cells, storiform fibrosis, and obliterative phlebitis. In the thoracic involvement of IgG4-RD, mediastinal lymphadenopathy and perilymphangitic interstitial thickening of the lung are the most common findings. Peribronchovascular and septal thickening and paravertebral band-like soft tissue are characteristic findings of IgG4-RD. Other findings include pulmonary nodules or masses, ground-glass opacity, alveolar interstitial thickening, pleural effusion or thickening, mass in the chest wall or mediastinum, and arteritis involving the aorta and coronary artery. Radiologic differential diagnosis of various malignancies, infections, and inflammatory conditions is needed. In this review, we describe the imaging findings of IgG4-RD and the radiologic differential diagnoses in the thorax.
