EFFECTS OF GINSENOSIDE $Rb_2$ ON THE ANTIOXIDANTS IN SENESCENCE - ACCELERATED MICE(SAM - R/1)

  • In biological system, there are enzymes such as superoxide dismutase(SOD), catalase and glutathione(GSH) peroxidase which scavenge reactive oxygen species as well as antioxidants such as ceruloplasmin, albumin and nonprotein-bound SH including GSH related to defense mechanism. In the present study, the protective effects of ginsenoside $Rb_2$ against oxidative stress were investigated in the SAM-R/1 mice. Treatment with ginsenoside $Rb_2$ significantly increased Cu, Zn-SOD and Mn-SOD in the liver. Ginsenoside $Rb_2$ tended to increase hepatic catalase activity and significantly increased serum albumin and nonprotein-bound SH levels in the liver. But treatment with ginsenoside $Rb_2$ showed a significant decrease in hepatic malondialdehyde(MDA) levels compared to control group. Furthermore, we compared the effects in the hepatic SOD, MDA and serum albumin. These findings suggest that the increase of antioxidants by ginsenoside $Rb_2$ results in the protective effects against reactive oxygen species.

Effects of Dietary Inclusion of Red Ginseng Byproduct on Growth, Body Composition, Serum Chemistry, and Lysozyme Activity in Juvenile Olive Flounder (Paralichthys olivaceus)

  • This study examined the effects of the dietary inclusion of various concentrations of red ginseng byproduct (RB) and a mixture containing red ginseng byproduct, garlic extract, yeast and filler (CR) on the growth, body composition, serum chemistry, and lysozyme activity of juvenile olive flounder (Paralichthys olivaceus). Juvenile fish (n= 630) weighing 5.0 g were randomly distributed into 21 180 L flow-through tanks (30 fish/tank). Seven experimental diets were prepared in triplicate: a control diet without additive, and diets containing 0.5, 1 and 2% concentrations of RB (RB-0.5, RB-1, RB-2) and CR (CR-0.5, CR-1, CR-2) at the expense of wheat flour. After an 8-week feeding trial, serum chemistry and lysozyme activity of fish were measured. Mean weight gain was significantly higher in fish fed the control diet than in fish fed the RB and CR diets. The dietary inclusion of RB and CR reduced feed utilization. Mean serum glucose and triglyceride (TG) levels were higher in fish fed the control diet than in fish fed the other diets. Mean glutamate pyruvate transaminase (GPT) levels of fish fed the control and RB-2 diets were higher than those of fish fed the RB-0.5, RB-1, CR-1, and CR-2 diets. Mean lysozyme activity levels of fish fed the RB-0.5 and RB-1 diets were higher than those of fish fed the control and CR diets. The results of this study indicate that red ginseng byproduct may be utilized as an immunostimulant rather than as a growth promoter for juvenile olive flounder. Dietary inclusion of 0.5% red ginseng byproduct effectively improved serum glucose, GPT, TG, and lysozyme activity of the fish in this study.

  • The NMR spectra of $Li_3Rb(SO_4)_2$ crystals and their relaxation processes were investigated by using $^7Li$ and $^{87}Rb$ NMR. The relaxation rates of the $^7Li$ and $^{87}Rb$ nuclei in the crystals were found to increase with increasing temperature, and can be described by the relation $T_1^{-1}{\propto}AT^2$. The dominant relaxation mechanism for these nuclei with electric quadrupole moments is provided by the coupling of these moments to the thermal fluctuations of the local electric field gradient via Raman spin-phonon processes.

Effect of Microwave Treatment on Korean Ginseng (고려인삼의 마이크로파 처리 효과)

  • The effect of microwave treatment on Korean ginseng was studied by measuring the changes in moisture, crude lipid, crude ash, crude protein, total dietary fiber and saponin contents, as well as changes in density, color and microstructure. Korean ginseng was treated with 100 or 200 watts of microwaves for 1 or 3 hrs, respectively, followed by drying using an oven at $60^{\circ}C$ for 96 hrs. The moisture contents decreased to 13.12~10.77% from an initial 76.26%. The amounts of lipid and ash were reduced in proportion to the time of microwave treatment and level of microwave power. The amount of protein in ginseng after microwave treatment did not significantly change. The amount of total dietary fiber increased after microwave treatment and the color of dried ginseng became dark. The amounts of ginsenoside-$Rb_1$, $Rb_2+Rb_3$, Rc, Rd, Re, Rf, $Rg_1$, $Rg_2+Rh_1$ and $Rg_3$ were reduced after treatment with 100 watts of microwave radiation for 1 and 3. The amounts of ginsenoside-$Rb_1$, Rd, Re, Rf, $Rg_1$, $Rg_2+Rh_1$ and $Rg_3$ after treatment with 200 watts of microwave radiation for 1 and 3 hr also reduced. On the other hand, the amounts of ginsenoside-$Rb_2+Rb_3$ and Rc after treatment of ginseng with 200 watts of microwave radiation for 1 and 3 hrs were increased.

Changes of Protopanaxadiol Ginsenosides in Ginseng Leaves by Far Infrared and Steaming Heat Treatments (원적외선 및 증숙 처리에 따른 인삼 잎의 Protopanaxadiol Ginsenosides 변화)

  • PPD ginsenosides in ginseng leaf were analyzed to determine effects of either FIR heat or steaming heat treatment. Among the PPD ginsenosides, Rb1, Rc and Rb3 forming four glycoside-attached aglycons were increased as FIR heat temperatures were increased from 60 to $120^{\circ}C$, while Rb3 was decreased. In addition, FIR heat treatment was effective to increase Rd forming a three glycoside-attached aglycon. Rg3 and Rh2 were not increased by the FIR heat treatment. In steaming heat treatment, Rb1 was significantly decreased, while Rb2 was increased. Rd was also increased by increased steaming temperature, yet its content was lower than in the FIR heat treatment. However, the steaming heat treatment increased yields of Rg3 and Rh2, which were not observed in the FIR heat treatment. Thus, FIR heat treatment was beneficial to efficient products of Rb1, Rc, Rb3 and Rd. Steaming heat treatment was effective to higher collection of Rb2, Rg3 and Rh2.

Comparative Analysis of Metabolites in Roots of Panax ginseng Obtained from Different Sowing Methods (파종 방법에 따른 고려인삼의 대사체 비교)

  • Ginsenosides of roots in Panax ginseng were analyzed by metabolic-targeting HPLC using the partial least squares discriminant analysis (PLS-DA) and compared depending on sowing methods between direct seeding and transplanting method. Score plots derived from PLS-DA could identify the sowing method between the direct seeding and transplanting method in P. ginseng roots. The ginsenoside compounds were assigned as Rg1, Re, Rf, Rg2, Rb1, Rc, Rb2, Rb3, and Rd. Contents of Re, Rf, Rg2, Rb1, Rc, Rb3, and Rd of main roots produced from the transplanting method were relatively higher than those of samples produced from direct seeding method. Also, contents of Rg1, Re, Rf, Rg2, Rb1, Rc, Rb2, Rb3, and Rd of lateral roots from the transplanted samples were relatively higher than those of samples produced from direct seeding method. Therefore, HPLC with PLS-DA analysis can be a straightforward tool for identification of ginsenosides in main or lateral roots of P. ginseng obtained from two different seeding methods between direct and transplanting methods.

Tolerability and pharmacokinetics of ginsenosides Rb1, Rb2, Rc, Rd, and compound K after single or multiple administration of red ginseng extract in human beings

  • Background: We investigated the tolerability and pharmacokinetic properties of various ginsenosides, including Rb1, Rb2, Rc, Rd, and compound K, after single or multiple administrations of red ginseng extract in human beings. Methods: Red ginseng extract (dried ginseng > 60%) was administered once and repeatedly for 15 days to 15 healthy Korean people. After single and repeated administration of red ginsengextract, blood sample collection, measurement of blood pressure and body temperature, and routine laboratory test were conducted over 48-h test periods. Results: Repeated administration of high-dose red ginseng for 15 days was well tolerated and did not produce significant changes in body temperature or blood pressure. The plasma concentrations of Rb1, Rb2, and Rc were stable and showed similar area under the plasma concentration-time curve (AUC) values after 15 days of repeated administration. Their AUC values after repeated administration of red ginseng extract for 15 days accumulated 4.5- to 6.7-fold compared with single-dose AUC. However, the plasma concentrations of Rd and compound K showed large interindividual variations but correlated well between AUC of Rd and compound K. Compound K did not accumulate after 15 days of repeated administration of red ginseng extract. Conclusion: A good correlation between the AUC values of Rd and compound K might be the result of intestinal biotransformation of Rb1, Rb2, and Rc to Rd and subsequently to compound K, rather than the intestinal permeability of these ginsenosides. A strategy to increase biotransformation or reduce metabolic intersubject variability may increase the plasma concentrations of Rd and compound K.

  • Relatively large quantity of the major components of saponin, $ginsenoside-Rb_1,\;-Rb_2,\;-Rc,\;-Rd,\;-Re\;and\;-Rg_1$ from Panax ginseng C.A. Meyer were isolated using preparative and semipreparative high performance liquid chromatography, and analyzed by analytical HPLC. The application of HPLC for isolation of ginsenosides was not only very effective for rapid analysis but also reduced the isolation time. The isolation capacity of pure ginsenosides was $30{\sim}50mg/hr$.

STUDIES ON THE ABSORPTION, DISTRIBUTION, EXCRETION AND METABOLISM OF GINSENOSIDE $Rg_1\;AND\;Rb_1$ IN RATS (진세노사이드 $Rg_1$$Rb_1$의 흡수$\cdot$분포$\cdot$배설 및 대사에 관한 연구)

  • 인삼의 주된 사포닌으로서 Rg,과 Rb,을 흰쥐에 투여하였을 경우에 이들 물질이 흰쥐의 장기에 흡수 또는 분포되는 상태와 배설에 대하여 연구하였다. 진세노사이드 $Rg_{1}$은 경구 투여량의 약 $1.9{\%}$가 소화관의 상부에서 흡수되었으며, 투여한지 30분 후에 최고 혈중 농도에 이르렀고 조직에서는 1.5시간 걸렸다. 그러나 뇌에서는 확인되지 않았으며 뇨와 당즙에는 2 : 5의 비로 배설되었다. $Rb_{1}$을 100mg/kg 경구투여한 결과, 소화관에서는 거의 흡수가 되지 않았으며, 한편 정맥주사(5mg/kg)의 경우는 혈중 $Rb_{1}$의 농도가 지수적으로 감소하였으며, B-phase의 반감기는 14.5시간이었다. 정맥주사후 혈청과 조직에 장시간 잔존은 활성을 나탄내는 혈청단백과의 결합과 관련이 있는 것으로 사료되며 시간에 따라 뇨로 배설되나 담즙에서는 확인되지 않았다. $Rg_{1}$$Rb_{1}$을 경구투여한 후 TLC와 $^{13}C$-NMR을 이용하여 위와 대장에서의 분해 상태를 연구한 결과 위에서 $Rg_{1}$의 일부가 분해, 6종류의 분해 산물이 r-everse phase TLC상에서 관찰되었고 이들 분해 산물은 약산성 (0.1N HCl, $37^{\circ}C$) 조건하에서 $Rg_{1}$의 가수분해산물과 동일하였다. 한편, $Rb_{1}$ 경구투여후 위장에서 얻은 시료중에서 미확인 분해산물이 관찰되었으며, 이 분해산물은 약산성 조건하에서 $Rb_{1}$의 가수분해산물과는 상이하다는 사실을 확인하였다. 대장에서, $Rg_{1}$은 미생물 tetracycline-susceptible bacteria와 tetracycline-resist bacteria에 의해 $Rb_{1}$$F_{1}$으로 분해되었으며, $Rb_{1}$은 장내의 효소와 tetracycline-resistantant bacteria에 의해 Rd와 2 종류의 미확인 물질로 분해되었다.
