  • Choline is an important membrane phospholipid constituent and a neurotransmitter precursor that is minimally synthesized in brain. The long-term maintenance of brain choline concentration is dependent on choline transport from plasma, which occurs via saturable transport system at the blood-brain barrier. In the present study, we examined to elucidate the characteristics of transport of cationic amines, especially choline which is one of cationic amines, to BBS using conditionally immortalized rat brain capillary endothelial cell line (TR-BBB) in vitro.   • Large conductance $Ca^{2+}$-activated $K^{+}$ channels (Maxi-K channel) have been implicated in many important physiological processes such as co-ordination of membrane excitability in neurons. Modulation of these channels are archived by the activity of various protein kinases. The most widely studied example of Maxi-K channel regulation by protein phosphorylation has been obtained using plasma membranes from the rat brain incorporated into lipid bilayers.(omitted)

The Effects of $Taegye(KI_3)$ Acupuncture and Acupuncture with Twirling Method on Blood Pressure in Hypertensive RAT Induced by Two Kidney One Clip (2K1C) (념전(捻轉) 수기(手技)에 따른 태계(太谿) 랄침(剌鍼)이 2K1C 고혈압(高血壓) 백서(白鼠)의 혈압(血壓)에 미치는 영향)

  • Objectives : The purpose of this study was to investigate the effects of $Taegye(KI_3)$ acupuncture and acupuncture with twirling method on the blood pressure, cardiomegalic index, plasma levels of renin and atrial natriuretic peptide in hypertensive RAT induced by two kidney one clip (2KIC). Methods : This study was conducted on one control group and four experimental groups. Control group that had no treatment, Tail group that took tail acupuncture, K-0 group that took $KI_3$ acupuncture, K-6 group that took $KI_3$ acupuncture with 6 turns of twirling method, K-9 group that took $KI_3$ acupuncture with 9 turns of twirling method. Results : Blood pressure was decreased significantly after $KI_3$ acupuncture. Cardiomegalic index decreased significantly after $KI_3$ acupuncture. Plasma levels of renin was decreased significantly after $KI_3$ acupuncture and acupuncture with 9 turns of twirling method Ⅳ. Plasma levels of atrial natriuretic peptide was increased significantly after $KI_3$ acupuncture. Conclusions : These results suggest that blood pressure was decreased significantly after $KI_3$ acupuncture in hypertensive RAT induced by two kidney one clip (2K1C).

  • The relative distribution ratio of barbiturates between hyarocarbon interior and surface region of outer monolayer of synaptosomal plasma membrane vesicles (RSPMV) isolated from rat whole brain was determined by employing the fluorescent probe technique. The two fluorescent probes N- octadecylnaphthyl-2-amine-6-sulfonic acid (ONS) and 12-(9-anthroyloxy) stearic acid (AS) were utilized as probes for hydrocarbon interior and surface of outer monolayer of RSPMV. respectively. The Stern-Volmer equation for fluorescent quenching was modified to calculate the relative distribution ratio. The analysis of preferential quenching of these probes by barbiturates indicates that pentobarbital, hexobarbital, amobarbital and phenobarbital are predominantly distributed on the surface region. whereas thiopental sodium has an accessibility to the hydrocarbon interior of the outer monolayer of the RSPMV. From these results, it is strongly suggested that the more effective penetration into the hydrocarbon interior of the outer monolayer of the membrane lipid bilayer could result in higher general anesthetic activity.

Effects of Intravenous Catecholamine on Gastric Acid, Gastrin and Secretin Secretion in Basal State of the Rat (정맥 주입한 Catecholamine이 흰쥐의 기초상태시 위산, Gastrin 및 Secretin 분비에 미치는 영향)

  • This study was conducted to investigate the effects of epinephrine and norepinephrine on basal gastric acid secretion and plasma gastrin and secretin concentration in the conscious rat. One hundred and eighty-four albino rats with gastric cannula were used after 18 hours or more of fast, with water ad libitum. In a restraint cage for collection of gastric juice, physiological saline (0.9% NaCl) was continuously infused into the jugular vein through a catheter for one hour at a rate of 1 ml/hr (control period). Immediately after the control period, epinephrine (1, 2, 4, 8 and $16{\mu}g/ml/hr)$, norepinephrine (1, 2, 4, 8 and $16{\mu}g/ml/hr)$ or physiological saline (1 ml/hr) was infused for another one hour. Gastric juice was collected at one hour interval for two hours infusion period. Adrenergic antagonists, phentolamine and propranolol were injected into the jugular vein 5 min prior to the infusion of epinephrine or norepinephrine at a dose of 0.2 mg/0.1 ml. Blood was sampled from the jugular vein for the radioimmunoassay of plasma gastrin and secretin after the collection of gastric juice. The results were as follows: 1) Both epinephrine and norephinephrine significantly increased gastric acid output in a dosedependent manner. 2) The effects of epinephrine and norepinephrine on the gastric acid secretion were antagonized by the pretreatment with phentolamine and propranolol. 3) Plasma gastrin and secretin concentrations were not significantly affected by the intravenous infusion of epinephrine and norepinephrine. It can be inferred from the above results that epinephrine and norepinephrine facilitate gastric acid secretion in conscious rats and the mechanism of which is attributed to ${\alpha}\;and\;{\beta}$ adrenergic receptors rather than gastrin and secretin.

  • Objectives : The purpose of this study is to compare Xingjian(LR2) Shaofu(HT8) with Dadun(LR1) Shaofu(HT8) on Blood Pressure in Hypertensive RAT induced by Two Kidney One Clip(2KIC). Methods : This experiments was to investigate the effects of LR2 HT8(originated from ${\ll}$Classic on Difficulty${\gg}$ Shi Ze Xie Qi Zi), LR1 HT8(originated from ${\ll}$Ling Shu${\gg}$ Sheng Ze Xie Zhi) acupuncture on the blood pressure, cardiomegalic index, and plasma levels of atrial natriuretic peptide in hypertensive rat induced by 2K1C. Results: 1. Blood pressure was decreased significantly after third acupuncture of LR2 HT8. 2. Blood pressure was decreased significantly after acupuncture of LR2 HT8, but was increased after LR2 HT8. 3. Cardiomegalic index was not changed after acupuncture of LR2-HT8 and LR1-HT8 4. Plasma levels of atrial natriuretic peptide was increased significantly after acupuncture of LR2 HT8 but LR1 HT8 was not changed.

Effects of Prunella vulgaris Pharmacopuncture on Lipopolysaccharide-Induced Acute Inflammatory Rat Model (하고초 약침이 LPS로 유발된 급성염증 백서 모델에 미치는 영향)

  • Objectives : To investigate the anti-inflammatory effects of Prunella vulgaris pharmacopuncture in lipopolysaccharide (LPS)-induced inflammatory rat model. Methods : Sprague-Dawley rats were divided into 5 groups; normal control (n=8), LPS control (n=8), LPS+Prunella vulgaris pharmacopuncture at CV4 (CV4, n=8), LPS+Prunella vulgaris pharmacopuncture at ST36 (ST36, n=8), and LPS+Prunella vulgaris pharmacopuncture at CV12 (CV12, n=8). Pharmacopuncture was given every two days for 4 weeks followed by inflammation induction by peritoneal LPS injection (5mg/kg). Proinflammatory cytokines including interleukin-$1{\beta}$ (IL-$1{\beta}$), interleukin-6 (IL-6), tumor necrosis factor-$\alpha$ (TNF-$\alpha$), interleukin-10 (IL-10), thiobarbituric acid reactive substance (TBARS) from blood and liver tissue were compared before and 5 hrs after inflammation induction. Results : In CV4 and CV12 groups, plasma IL-$1{\beta}$, IL-6 and TNF-$\alpha$ levels increased by LPS injection, significantly decreased 5 hrs after injection (p<0.05). For CV12 group, plasma IL-10 concentration significantly increased (p<0.05). Liver IL-$1{\beta}$ and IL-6 levles significantly decreased in CV4 and CV12 groups (P<0.05), while normal and LPS control groups were not significantly different in TNF-$\alpha$ and IL-10 levels. Plasma TBARS concentration was significantly decreased in CV12 group, while there was no significant difference among LPS control and pharmacopuncture groups for liver TBARS concentration. Conclusions : Based on the present findings, Prunella vulgaris pharmacopuncture at CV12 may have a potentially preventive anti-inflammatory effect in an LPS-induced inflammatory rat model.

Development and Validation of an LC-MS/MS Method for Determination of Damaurone D in Rat Plasma and its Application to Pharmacokinetic Study in Rats

  • Damaurone D belongs to the genus Rosa and is a traditional medicinal product used for the treatment of depression, inflammation, and infectious diseases. The purpose of this study was to develop a simple liquid chromatography-tandem mass spectrometry method for the detection of damaurone D in rat plasma and to demonstrate its application in pharmacokinetic studies. Damaurone D and berberine (internal standard) were extracted with acetonitrile using a protein precipitation method. Mass transition was monitored in multiple reaction monitoring mode at m/z $323.2{\rightarrow}267.0$ for damaurone D and m/z $336.1{\rightarrow}320.0$ for berberine in positive ion mode. Analytical validation was conducted by evaluating the specificity, linearity, accuracy, precision, matrix effect, extraction recovery, and stability. The calibration curves were linear over 2-1000 ng/mL. The intra- and inter-day precision and accuracy of quality control samples were 4.79-13.33% and 86.23-102.75%, respectively. The matrix effect and extraction recovery were 96.11-98.47% and 96.11-102.25%, respectively. In the pharmacokinetic study after intravenous administration of damaurone D at a dose of 3 mg/kg in rats, the area under the curve and clearance of damaurone D in rat plasma were $16750.26{\pm}2676.10min{\cdot}ng/mL$ and $182.44{\pm}31.36mL/min/kg$, respectively.
