The ability of orexin-A to modify pain-induced cyclooxygenase-2 and brain-derived neurotrophic factor expression is associated with its ability to inhibit capsaicin-induced pulpal nociception in rats

  • Background: The rostral ventromedial medulla (RVM) is a critical region for the management of nociception. The RVM is also involved in learning and memory processes due to its relationship with the hippocampus. The purpose of the present study was to investigate the molecular mechanisms behind orexin-A signaling in the RVM and hippocampus's effects on capsaicin-induced pulpal nociception and cognitive impairments in rats. Methods: Capsaicin (100 g) was applied intradentally to male Wistar rats to induce inflammatory pulpal nociception. Orexin-A and an orexin-1 receptor antagonist (SB-334867) were then microinjected into the RVM. Immunoblotting and immunofluorescence staining were used to check the levels of cyclooxygenase-2 (COX-2) and brain-derived neurotrophic factor (BDNF) in the RVM and hippocampus. Results: Interdental capsaicin treatment resulted in nociceptive responses as well as a reduction in spatial learning and memory. Additionally, it resulted in decreased BDNF and increased COX-2 expression levels. Orexin-A administration (50 pmol/1 µL/rat) could reverse such molecular changes. SB-334867 microinjection (80 nM/1 µL/rat) suppressed orexin's effects. Conclusions: Orexin-A signaling in the RVM and hippocampus modulates capsaicin-induced pulpal nociception in male rats by increasing BDNF expression and decreasing COX-2 expression.

Influence of Prenatal Noise and Music on the Expressions of c-Fos and Nitric Oxide Synthase in the Hippocampus of Rat Pups

  • The expressions of c-Fos and nitric oxide synthase (NOS) represent neuronal activity and play' a crucial role in the shaping of the development of brain. During the late pregnancy, stresses may influence neuronal activity of prenatal rats. In the present study, the effects of prenatal noise and music on the expressions of c-Fos and NOS in the hippocampus of rat pups were investigated. Exposure to the noise during pregnancy decreased c-Fos and NOS expressions in the hippocampus of rat pups, whereas exposure to music during pregnancy increased c-Fos and NOS expressions in the hippocampus of rat pups. The present results show that prenatal music stimulation may increase neuronal activity of rat offspring, whereas exposure to noise during pregnancy may reduce the neuronal activity of offspring. The present study suggests that prenatal stimuli including noise and music could affect the fetal brain development.

Carbofuran이 쥐의 조직에 미치는 형태적 변화와 Phenobarbital Sodium 및 3- Methylcholanthrene에 의한 억제효과 (Textural Changes in Rat Tissues by Carbofuran and Its Suppression by Phenobarbital Sodium and 3-Methylcholanthrene)

  • 본 연구는 carbofuran이 흰쥐의 신장, 간 및 뇌조직내 각종 세포의 미세구조에 미치는 독성을 비교${\cdot}$관찰하고, 더불어 phenobarbital sodium(PB) 및 3-methylcholanthrene(3-MC)이 carbofuran의 독성에 어떠한 보상효과를 갖고 있는지를 밝히고자 각 실험동물에 carbofuran과 PB 및 3-MC 단독 또는 carbofuran과 PB 및 carbofura과 3-MC를 조합 투여한 후 경시적으로 각 조직을 적출하여 광학 현미경으로 관찰을 하였다. 신장에서의 carbofuran단독투여균의 경우 투여후 48시간까지 신장소체의 많은 세뇨관들이 폐쇄되는 독성이 나타났으나 Carbofuran과 PB 또는 3-MC의 조합처리군의 경우 각각 투여후 6시간부터 다소 재생되기 시작하여 48시간 후에는 현저히 회복되는 경향이었다. 간에서의 carbofuran 단독투여군의 경우 투여후 48시간까지 간세포에 퇴행성변화인 괴사 및 심한 변성이 관찰되었으나 Carbofuran과 PB 또는 6시간후부터 간세포들의 재생이 뚜렷하였다. 뇌에서의 carbofuran 단독투여군의 경우 투여후 48시간까지 퇴행성변화 및 대뇌피질의 모세혈관 확장이 나타났으나 Carbofuran과 PB 또는 3-MC의 조합처리군의 경우 투여후 6시간후 이후에는 대조군과 유사하였다. 이상의 결과로 보아 PB와 3-MC 모두 carbofuran에 의한 쥐의 조직독성을 감소시킬수 있는 물질임을 알 수 있었다.
