Effects of Ginkgo biloba Leaf Extracts on the bevels of Neurotransmitters in the Spontaneously Hypertensive Rat Brain (은행잎 추출성분이 고혈압쥐의 뇌중 신경전달물질 함량에 미치는 영향)

  • Effects of Ginkgo biloba leaf extract (GBE) and its components kaempferol-coumaroyl glucosyl rhamnoside (Kc), quercetin-coumaroyl glucosyl rhamnoside (Qc), ginkolide (G) and bilobalide (B) on blood pressure and on the levels of neurotransmitters in the spontaneously hypertensive rat brain were examined. Blood pressure decreased about 10 mmHg after administration of GBE but without significance. The concentration of norepinephrine increased (3 times) and that of DOPAC was decreased (38%) after administration of Qc. And 3-MT concentration was increased (36%) by Kc administration in striatum. While Qc administration increased the concentration of Ach (260%) and Ch(29%) significantly, B administration increased Ch concentration in cerebral cortex.

Prenatal Development of Tvrosine Hlrdroxvlase-Containing Neurons in the Rat Brain (흰쥐 태아 뇌에서 TH 신경세포의 초기발생과정)

  • The present study attempts to explore the first appearance 8nd subsequent development of %cosine hvdroxvlase (TH)-containing neurons in the rat brain from embryonic day (E) 10.5 to the neonate. To increase the senti노ivity and resolution power, a double bridge peroxidase-anti-peroxidase technique was employed for cellular localization of TH. In situ hybridization histochemistw with synthetic TH oligomer (30-mer) codinB TH was also used to detect TH mRNA. TH-containing neurons were first detected at E11.5 in the intermediate zone of prosencepha1on and mesencephalon. At this stage, TH-immunoreactive neurons were small, ovoid bee and emitted their fibres into their immediate surroundings. From this stage, TH-immunoreactive neurons increased in their number and underwent migration and cell differentiation. At E15.5, the distribution pattern of the maior groups of TH neurons was similar to that of adult catecholaminergic groups, and at E19.5 the external laver of median eminence showed TH-immunoreactive processes.

Dual Action of d-Tubocurarine on Large-Conductance $Ca^{2+}-activated$ $K^+$ Channels from Rat Brain Reconstituted into Planar Lipid Bilayer

  • Using the planar lipid bilayer method, we investigated the effect of d-tubocurarine (dTC) on the extracellular side of large-conductance $Ca^{2+}-activated\;K^+$ channel from rat brain. When the initial open probability (Po) of the channel was relatively high, dTC decreased channel activity in a concentration dependent manner. In contrast, when the initial Po was lower, sub-micro molar dTC increased channel activity by destabilizing the closed states of the channel. Further addition of dTC up to micro molar range decreased channel activity. This dual effect of dTC implicates that there exist at least two different binding sites for dTC.

  • The characteristics of steroid acyl transferase were studied in the rat brain with (4-14C)-dehydroepiandrosterone(DHEA). The results could be summarized as followings: The enzyme system responsible for the biosynthesis was localized at the microsome fraction. The optimum pH of this enzyme was 4.6 When DHEA was utilized as substrate, $\Delta$5-pregnenolone was proved to be a competitive inhibitor. However testosterone was a noncompetitive inhibitor. The acylation at 3${\beta}$-hydroxyl group was favored when the hydrophilicity at Cl7 position increased. However, this acylation at C3 was very low when A ring was aromatic. The acylation at Cl7 hydroxyl group reguired an absolute 17${\beta}$-conformation.

Essential Role for c-jun N-terminal Kinase on tPA-induced Matrix Metalloproteinase-9 Regulation in Rat Astrocytes

  • Tissue plasminogen activator (tPA) is used to lyse clots and reperfuse brain in ischemic stroke. However, sideeffects of intracerebral hemorrhage (ICH) and edema limit their clinical application. In part, these phenomena has been linked with elevations in matrix metalloproteinase-9 (MMP-9) in neurovascular unit. However little is known about their regulatory signaling pathways in brain cells. Here, I examine the role of MAP kinase pathways in tPA-induced MMP-9 regulation in rat cortical astrocytes. tPA $(1-10\;{\mu}g/ml)$ induced dose-dependent elevations in MMP-9 and MMP-2 in conditioned media. Although tPA increased phosphorylation in two MAP kinases (ERK, JNK), only inhibition of the JNK pathway by the JNK inhibitor SP600126 significantly reduced MMP-9 upregulation. Neither ERK inhibition with U0126 nor p38 inhibition with SB203580 had any significant effects. Taken together, these results suggest that c-jun N-terminal kinase (JNK) plays an essential role for tPA-induced MMP-9 upregulation.

  • We cloned the cDNA encoding the neuron-specific $\beta$-subunit ($\beta$4) of large-conductance calcium-activated potassium channels from rat brain and determined the DNA sequences of the entire coding region (GenBank accession; AY028605). The deduced amino acid sequences of r$\beta$4, 210 amino acids in length, are closely related to the $BK_{Ca}$ $\beta$4 subunits of other species but show only limited sequence homology to other $\beta$-subunits, $\beta$1-$\beta$3.(omitted)d)

Prenatal Development of Gonadotropin Releasing Hormone (GnRH) Neurons in the Rat Brain (흰쥐 태아 뇌에서 GnRH 신경세포의 초기발생과정)

  • The present experiment was carried out 1) to study the developmental topography of GnRH neuronal system and 2) to characterize the cellular localization of GnRH neurons in the prenatal brain development of the rat. At embryonic day (I) 14.5, immunoreactive cell bodies of GnRH were first seen in the nasal septum and in the ganglion terminate located in the ventral protion of the caudal olfactory bulb. Two days later (E 16.5), GnRH-containing neurons were observed at the level of olfactory tubercle and diagonal band of Broca, which is the first appearance in the intracerebral region. From 118.5, the topographic pattern of immunoreactive GnRH perikarya was similar to that of adult rats. The present data suggest that GnRH neurons were originated from the nasal septum and gradually extended to the hvpothalamic regions with increasing fetal age.

  • The activity of Mg and Na-K activated ATPase of homogenate from rat brain cortex was measured in vitro under the variety of conditions. The effects of various hypnotic and tranquilizer such as phenobarbital, amobarbital, diazepam, promazine and chlorpromazine on the activities of both ATPase was investigated and the results was summarized as follows. 1. Na-K ATPase was slightly inhibited by phenobarbital and amobarbital while Mg ATPase was moderately activated by these drugs. 2. Both Mg and Na-K ATPase activities were markedly inhibited by diazepam. 3. Promazine and chlorpromazine markedly inhibited both Mg and Na-K ATPase activities. These findings indicate that remarkable correlation between hypnotic or tranquilizing potency and ATPase inhibition could be observed.

Swimming During Pregnancy Increases the Expression c-Fos and c-Jun in the Hippocampus of Rat Offspring

  • The expression of c-Fos and c-Jun represents neuronal activity and plays a crucial role in the shaping of the development of brain. During the late pregnancy, exercise is known to influence neuronal activity of offspring. In the present study, the effect of swimming during pregnancy on the expression of c-Fos and c-Jun in the CA1, CA2, CA3 regions, and the dentate gyrus of the hippocampus of rat offspring was investigated using immunohistochemistry. Pregnant rats in the swimming group were forced to swim for 10 min once a day from 15 days after pregnancy until delivery. The expression of c-Fos and c-Jun in the CA1, CA2, CA3 regions, and the dentate gyrus of the hippocampus of pups was significantly increased by maternal swimming during late pregnant period. The present results show that prenatal swimming may enhance the neuronal activity of pups and affect the neonatal brain development.

  • Neuroprotective strategies have been appeared to be effective in a variety of stroke models. One of the major focuses has been related to the activities of estrogen. $17\beta$-estradiol valerate(EV) has been reported to exert neuroprotective effects when administered before an ischemic insult. The purpose of this study was to determine whether EV can protect against brain injury via estrogen receptor. Chronic and acute pretreatment can reduce the ischemic damage of focal cerebral ischemia in OVX rat, indicating that EV may be a new therapeutic class of drugs to prevent neuronal damage associated with cerebral ischemia. RNAs were extracted from the hippocampus of ovariectomized female rat with or without EV. Differential gene expression profiles were revealed(Bone morphogenetic protein type 1A receptor, Protein disulphide isomerase, cytochrome bc-1 complex core P, thiol-specific antioxidant protein). RT-PCR and in situ hybridization were used to validate the relative expression pattern obtained by the cDNA array.
