The effect of curcumin on blood pressure and cognitive impairment in spontaneously hypertensive rats

  • BACKGROUND/OBJECTIVES: It is known that the renin-angiotensin system (RAS) in the brain could regulate cognitive functions as well as blood pressure. Inhibition of RAS for the improvement of cognitive function may be a new strategy, but studies so far have mostly reported on the effects of RAS inhibition by drugs, and there is no research on cognitive improvement through RAS inhibition of food ingredients. Therefore, this study investigated the effect of curcumin on blood pressure and cognitive function and its related mechanism in spontaneously hypertensive rat/Izm (SHR/Izm). MATERIALS/METHODS: Six-week-old SHR/Izm rats were divided into 5 groups: control group (CON), scopolamine group (SCO, drug for inducing cognitive deficits), positive control (SCO and tacrine [TAC]), curcumin 100 group (CUR100, SCO + Cur 100 mg/kg), and curcumin 200 group (CUR200, SCO + Cur 200 mg/kg). Changes in blood pressure, RAS, cholinergic system, and cognitive function were compared before and after cognitive impairment. RESULTS: The SCO group showed increased blood pressure and significantly reduced cognitive function based on the y-maze and passive avoidance test. Curcumin treatments significantly improved blood pressure and cognitive function compared with the SCO group. In both the CUR100 and CUR200 groups, the mRNA expressions of angiotensin-converting enzyme (ACE) and angiotensin II receptor type1 (AT1), as well as the concentrations of angiotensin II (Ang II) in brain tissue were significantly decreased. The mRNA expression of the muscarinic acetylcholine receptors (mAChRs) and acetylcholine (ACh) content was significantly increased, compared with the SCO group. CONCLUSIONS: The administration of curcumin improved blood pressure and cognitive function in SCO-induced hypertensive mice, indicating that the cholinergic system was improved by suppressing RAS and AT1 receptor expression and increasing the mAChR expression.

  • 하천법에 의거하여 국내 하천들에는 상당한 국가예산으로 하천정비기본계획이 5-10년 주기로 수립되고 있으며, 홍수위 계산을 위한 HEC-RAS 모의에 필요한 하천단면 등 다양한 하천측량이 실시되고 있다. 그러나, 하천측량자료들은 하천관리지리정보시스템(RIMGIS)에 pdf 보고서 형태로만 제공되고, 원자료는 CAD 형식으로 하천정비계획을 수행한 설계사 등이 분산 소유하고 있어 관리부재로 망실의 우려도 있어, 다른 용도로의 활용성이 상당히 저하되어 있는 실정이다. 그리고, 측량된 CAD 형식의 단면자료 등을 HEC-RAS에 활용할 때, 'Dream'과 같은 툴을 활용하나 거의 수작업에 가까운 시간과 비용이 소요되는 현실에 있다. 본 연구에서는 이러한 문제들을 해결할 수 있는 툴인 RAUT(River information Auto Upload Tool)를 개발하였다., RAUT 툴은 첫째, 실무에서 하천기본계획 수립 시 활용되는 HEC-RAS 1차원 모형의 입력자료를 CAD 측량자료를 직접수기로 입력 및 모의를 실시하는 복잡한 단계를 자동화시키고자 하였다. 둘째, 하천공간정보인 CAD측량 자료를 직접 읽어 표준 데이터 모델 (Arc River)기반 하천공간정보 DB에 자동 업도드하여 전국단위의 하천정비계획의 하천측량자료 관리가 가능하게 할 수 있다. 즉, 만약 RIMGIS가 RAUT와 같은 툴을 사용하면 하천단면과 같은 전국단위 하천측량 자료를 체계적으로 관리할 수 있게 된다는 의미이다. 개발한 RAUT는 제주도 한천유역을 대상으로 하천정비기본계획의 하천공간정보 CAD자료를 읽어들여 mySQL기반 공간 DB로 구축하고, 구축된 DB로부터 HEC-RAS 1차원 모의 실시하기 위한 지형자료를 자동으로 생성시키는 과정을 시범적으로 구현하였다.

p53 and K-ras Expression in Interstitial Lung Disease (간질성 폐질환에서 p53 및 K-ras 암표지자의 발현)

  • Background : Approximately 10-13% of patients with interstitial lung disease(ILD) die of lung cancer, and patients with ILD have been reported to have a 7 fold higher incidence of lung cancer compared to the normal population. Recently, overexpression of the p53 and p21 proteins were observed in the epithelial cells from pathologic specimens of ILD. Overexpression of these proteins may result from chronic or recurrent DNA damage by unknown causes of inflammation. However, these proteins may also contribute to oncogenesis if other genetic alterations such as K-ras are superimposed. Methods : Immunohistochemical stains for p53 and K-ras proteins were performed with pathologic specimens from 38 cases with ILD(M/F : 27/11, mean agea : $54{\pm}10$ years) and from 10 control subjects. Results : The p53 protein was expressed in 21.1% (8/38 ILD cases) and K-ras protein expression was observed in 65.8% (25/38 ILD cases). However, neither p53 nor the K-ras protein staining was observed in the control subjects. Conclusion : A significant proportion of cases with ILD expressed the p53 and K-ras proteins in their bronchial epithelial cells. These proteins may be potentially oncogenic with the addition of further genetic alterations. However, to clarify the significance of these findings, further studies looking for correlations with the incidence of lung cancer and other genetic changes are needed.

Expression of Oncogene Product in the Colorectal Carcinoma (결장암 및 직장암에서 암유전자 산물의 발현)

  • The expression of $p62^{c-myc}$ and $p21^{ras}$ can be seen in many solid tumor, but the pattern and incidence of expression were different according to organ, countries, and examiners, thus it is not definitely defined. Total 67 colorectal carcinoma in paraffin sections are analysed by immunohistochemically for evaluation of the $p62^{c-myc}$ and $p21^{ras}$ expression according to the age, sex, chief complaints, location, differentiation, modified Dukes stage, using the specific monoclonal antibodies. The results were summarized as follows : The age of patients ranged from 32 years to 82 years. The mean age was 57.6 years. The expression of $p62^{c-myc}$ and $p21^{ras}$ was not correlated with age. Male was 29 cases(43.3%) and female was 38 cases(56.7%). The male to female ratio was 1:1.31. The expression of $p21^{ras}$ was increased in female(p<0.05). Abdominal pain(43.7%) was the most frequent chief complaint. The most frequent tumor location was rectum(44.8%). The expression of $p62^{c-myc}$ was increased in the rectum(p<0.05). The 65 cases(97.0%) out of 67 cases showed positive reaction of $p62^{c-myc}$ in the nucleus, cytoplasmic membrane, and cytoplasm. The 62 cases(92.5%) out of 67 cases showed positive reaction of $p21^{ras}$ in the cytoplasmic membrane and cytoplasm. The positive rate of $p62^{c-myc}$ and $p21^{ras}$ was 97.0% and 91.4% in well differentiated adenocarcinoma, 100.0% and 95.0% in moderately differentiated adenocarcinoma, 90.0% and 90.0% in poorly differentiated adenocarcinoma, 100.0% and 100.0% in mucinous carcinoma. The positive rate of $p62^{c-myc}$ and $p21^{ras}$ was 94.1% and 88.2% in Dukes stage $B_1$, 96.0% and 96.0% in Dukes stage $B_2$, 100.0% and 100.0% in Dukes stage $C_1$, 100.0% and 88.9% in Dukes stage $C_2$, and 100.0% and 100.0% in Dukes stage D. The expression of $p62^{c-myc}$ in metastatic colorectal carcinoma showed diffuse and strongly positive reaction than primary colorectal carcinoma. The expression of $p21^{ras}$ in primary colorectal carcinoma showed diffuse and strongly positive reaction than metastatic colorectal carcinoma.

Feasibility Analysis of HEC-RAS for Unsteady Flow Simulation in the Stream Channel with a Side-Weir Detention Basin (강변저류지가 있는 하도에서의 부정류 흐름 모의를 위한 HEC-RAS의 적용성 검토)

  • It is necessary to perform the precise analysis of unsteady flow for effective design of the side-weir detention basin installed in the river. Generally, the HEC-RAS program, which is a 1D unsteady numerical model, is mostly used to simulate the unsteady flow for rivers. However, it is difficult to have confidence of unsteady flow results simulated by HEC-RAS due to the lack of experimental data and field monitoring data for the channel with a side-weir detention basin. Therefore, the purpose of this study is to validate or verify the simulation results calculated by HEC-RAS through the experiments for the open channel with a side-weir detention basin using specially-designed unsteady discharge-supply system. The experimental cases included unsteady flows in the straight channel with and without a side-weir detention basin. Especially, for the case with a detention basin, the experiment was performed to consider only the free flow condition over the side-weir. The study results showed that values of water level and discharge obtained from HEC-RAS coincided reasonably with experimental results with the maximum error of 3% for water level and 1% for discharge in the case of the flow without the side-weir detention basin and 4% for water level and 2% for discharge with the side-weir detention basin.

Significance of HPV Infection and Genic Mutation of APC and K-ras in Patients with Rectal Cancer

  • Background: Significance of HPV infection and genic mutation of APC and K-ras in rectal cancer has been investigated but not clarified. The objective of our study was to investigate these parameters in patients with rectal cancer to analyze correlations with biological behaviour, to determine relationships among the three, and also to demonstrate survival prognosis effects. Methods: From December 2007 to September 2008, 75 rectal cancer cases confirmed by histopathology in the Tumor Hospital of Xinjiang Medical University were enrolled. The control group consisted of normal rectal mucous membrane taken simultaneously, a least 10 cm distant from the carcinoma fringe. HPV DNA, the MCR of APC and exon-1 of K-ras were detected by PCR and PCR-SSCP. All results were analyzed in relation to clinical pathological material, using chi-square and correlation analysis via SPSS.13 and Fisher's Exact Probability via STATA. 9.0. All 75 patients were followed up for survival analysis using Kaplan-Meier and Log-rank tests. Results: 55 out of 75 cases demonstrated gene HPV L1 while it was notdetected in normal rectal mucosa tissue. HPV infection was correlated with age and lymphatic metastasis (P<0.05) but not other characteristics, such as ethnicity, tumor size, histological type, tumor type, Duke's stage and infiltration depth. Some 43 cases exhibited APC genic mutation (57.3%) and 34 K-ras genic mutation (45.3%). APC genic mutation was correlated with gender(P<0.05), but not age, histological type, infiltration depth, lymphatic metastasis and Duke's stage. In 55 cases of rectal cancer with HPV infection, there were 31 cases with genic mutation of APC (56.4%) and 24 with genic mutation of K-ras (43.6%). For the 20 cases of rectal cancer with non-HPV infection, the figures were 12 cases (60%) and 10 (50.0%), respectively, with no significant relation. Survival analysis showed no statistical significance for K-ras genic mutation, APC genic mutation or HPV infection (P>0.05). However, the survival time of the patients with HPV infection was a little shorter than in cases without HPV infection. Conclusions: Our results suggest that HPV infection might be an important factor to bring about malignant phenotype of rectal cancer and influence prognosis. Genic mutation of APC and K-ras might be common early molecular events of rectal cancer, but without prognostic effects on medium-term or early stage patients with rectal cancer.

Accuracy Analysis of HEC-RAS for Unsteady Flow Simulation considering the Flow Pattern Variations over the Side-weir of Side-Weir Detention Basin (강변저류지 횡월류부의 흐름 형태 변화를 고려한 HEC-RAS의 하도 내 부정류 모의 정확도 분석)

  • Accurate quantitative assessment of flood control effect of side-weir detention basin as a flood countermeasure was highly required, in which one-dimensional HEC-RAS model has been widely utilized in practice. When the submerged overflow occurred particularly driven by limited storage capacity of a given detention basin, HEC-RAS model could not be sufficiently applicable by guaranteeing acceptable accuracy without reliable benchmark dataset. From this perspective, a dedicated unsteady experiment was planned and carried out to physically realize such submerged overflow for accommodating better accuracy. Subsequently, the experimental results were applied to validate and calibrate HEC-RAS unsteady modeling to provide flood control effect of the detention basin for various inflow scenarios. After following this procedure, the modelled results indicated that there appeared within -5% of difference in stage height and maximum 2.4% accuracy to assess the flood control effect, thereby ensuring the calibrated HEC-RAS unsteady model to be accurate with practically acceptable error range.

Evaluating the Properties and Commercializing Potential Of Rape Stalk-based Pellets Produced with a Pilot-scaled Flat-die Pellet Mill (파일럿 규모의 평다이 성형기로 제조한 유채대 펠릿의 연료적 특성 및 상용화 가능성 평가)

  • This study was conducted to evaluate the potential of rape stalk (RAS) as a raw material for the production of solid bio-fuels. RAS was immersed in an aqueous solution with acetic acid concentration of 1 percent, The content of reducing sugars separated from the RAS was analyzed. Glucose showed the highest content followed by xylose, galactose, arabinose and mannose. The immersed and non-immersed RAS were used for producing pellets with a pilot-scaled flat-die pellet mill. Bulk density and calorific values of the pellets improved with the use of the immersed RAS and the addition of wood particles. The values exceeded the minimum requirements for the A-grade of non-woody pellets (≧600 kg/m3 & ≧ 14.5 MJ/kg) designated by the ISO. Ash content of the pellets reduced with the immersion of RAS and the value satisfied the A-grade level (≦6.0%) of the ISO standard. The durability of the immersed RAS-based pellets was much higher than that of non-immersed IRS-based pellets, and the values were increased with the addition of wood particles. However, the durability did not meet the acceptance level for the B-grade of non-woody pellets (≧96.0%) designated by the ISO. These results suggested that the addition of binders in the production of non-woody pellets using an RAS immersed in acetic acid-based aqueous solution is required for the commercialization of the pellets.
