To investigate the adsorption-desorption reaction of Cd by sewage sludge, the adsorption of Cd from $Cd(NO_3)_2$ solutions of concentrations ranging from 5 to $50{\mu}g\;Cd\;mL^{-1}$ by sewage sludge was examined for reaction periods up to 48 hours. The amount of Cd adsorbed as a function of time was measured. The adsorption between Cd in solution and the solid phase could be described by two stages. The initial adsorption of Cd was very rapid, that is, approximately 95% of the added Cd was removed from solution within the first 30 minutes. Further, the greater the concentration of Cd added, the greater was the amount of Cd adsorbed. After the rapid initial decrease of Cd, a slower decline in the Cd concentration resulted which followed first order reversible kinetics. The equilibrium concentrations for the reactions, as well as the time period for the equilibrium reactions were dependent on the initial Cd concentrations. If equilibrium is reached, the amount of Cd remaining in solution is greater when the amount adsorbed is higher, although the percentage of Cd in solution is constant relative to the initial concentration of Cd. Some of the adsorbed Cd was released back to solution since the concentration of Cd after 48 hours was higher than the equilibrium concentration of Cd. However, despite the increased amount of Cd measured, the overall net reaction was a significant adsorption of Cd from solution by sewage sludge.

In this work, effects of the blend composition composed of 0, 10, 20, 30 and 40 wt% of nylon 6 to epoxy (diglycidylether of bisphenol-A, DGEBA) resin were investigated in terms of cure kinetics and thermal stability by differential scanning calorimeter (DSC) and thermogravimetric analysis (TGA). As the content of the nylon 6 increased, the maximum exothermic temperature ($T_{max}$) and the value of cure activation energy ($E_a$) decreased. The maximum exothermic temperature of the blending samples decreased with increasing in nylon 6 content, resulting in the decrease in curing activation energy of them due to the rapid curing reaction with epoxy resin in this system. From TGA analysis results of the DGEBA/nylon 6, the thermal stability based on the thermal stability index ($A^*{\cdot}K^*$) and integral procedure decomposition temperature (IPDT) increased with increase in the nylon 6 content. This was because of the combination of DGEBA and nylon 6 having good heat resistance, resulting in improving thermal stability of the DGEBA/nylon 6.
Experimental leaching of tailings was performed as a function of times (1, 2, 4, 7, 14, 21 and 30 days) in the laboratory using reaction solutions equilibrated to three different pH set-points (pHs 1,3 and 5). The initial pHs of 5 and 3 stabilized at either 4.6-6.1 or 2.8-3.5 in 2 days and decrease gradually with time afterwards. The results of the leaching tests indicate that the significant increase in the sulfate concentrations and in acidity after 7 days of leaching results from the oxidation of sulfide minerals. There were no significant variations in the extractable Pb found in the leach solutions of pH 5 and 3 within the reaction time (1-30 days), while Zn, Cd and Cu concentrations tend to significantly increase with time. In tailings leaching at an initial pH=1, two trends were observed: i) The 'Zn-type' (Zn, Cd and Cu), with increasing concentrations between days 1 and 30, corresponding to the expected trend when continuous dissolution is the dominant process, ii) the 'Pb-type' (Pb), with decreasing concentrations over time, suggesting rapid dissolution of a Pb source followed by the precipitation of 'anglesite' in relation to the large increase in dissolved sulfates. The high sulfate concentrations were coupled with high concentrations of released Fe, Zn and Cd. Release of Zn and Cd and acidity from these leaching experiments can potentially pose adverse impact to surface and groundwater qualities in the surrounding environment. The kinetic problems could be the important factor which leads to increasing concentrations of trace metals in the runoff water.
In this study, we described a paper-based neuraminidase assay sensor (PNAS) which can be applied to detect the infection by influenza viruses. The PNAS was designed and manufactured to quantitatively identify the levels of neuraminidase in the sample, which is based on colorimetric analysis using the X-Neu5Ac substrate. The limit of detection of the PNAS was determined as 0.004 U/mL of neuraminidase. According to the amount of neuraminidase in human serum, the PNAS could monitor the enzyme activity with a good linearity ($R^2$ > 0.99). In addition, the initial performance of the PNAS has been maintained up to 70 days in the $4^{\circ}C$. Finally, we demonstrated whether the Michaelis-Menten kinetics is applied to the PNAS, which can show the reliability of the enzyme reactions. The kinetic studies indicated that the PNAS provides the good condition for enzyme reactions ($K_m=8.327{\times}10^{-3}M$), but they were performed on paper chip nonetheless. The paper-based neuraminidase assay sensor may be useful in a wide range of rapid and safe detection of influenza virus.

Recently a B cell surface molecule, CD40, has emerged as a receptor mediating a co-stimulatory signal for B cell proliferation and differentiation. To investigate the mechanism of synergy between interleukin-4 (IL-4) and CD40 ligation in B cell activation, we have examined the effect of CE40 cross-linking on the IL-4 receptor expression in human B cells using anti-CE40 antibody. We observed that IL-4 and anti-CD40 both induce IL-4 receptor gene expression with a rapid kinetics resulting in a noticeable accumulation of IL-4 receptor mRNA within 4 h. While IL-4 caused a dose-dependent induction of surface IL-4 receptor expression, the inclusion of anti-CD40 in the IL-4-treated culture, further up-regulated the IL-4-induced IL-4 receptor expression as analyzed by flow cytometry. Pretreatment of B cells with inhibitors of protein tyrosine kinase (PTK) resulted in a significant inhibition of both the IL-4- and anti-CD40-induced IL-4 receptor mRNA levels, while protein kinase C (PKC) inhibitors had no effects. These results suggest that IL-4 and CD40 ligation generate B cell signals, which via PTK-dependent pathways, lead to the synergistic induction of IL-4 receptor gene expression. The rapid induction of IL-4 receptor gene expression through the tyrosine kinase-mediated signal transduction by B cell activating stimuli, would provide cells capacity for an efficient response to IL-4 in the early phase of IL-4 action, and may in part constitute the molecular basis of the reported anti-CD40 co-stimulatory effect on the IL-4-induced response.

Adsorption behaviors of Ni(II) ions were investigated using amorphous alumina as adsorbent. In the adsorption kinetic study, it was observed that Ni(II) ions were bound to the alumina surface in two adsorption stages, the rapid and slow adsorption stages. The rapid adsorption proceeded within 1 hr, thereafter the slow adsorption occurred. The results of adsorption isotherm experiments showed that the Ni(II) adsorption obeyed the Langmuir-Freundlich isotherm, and that the adsorption enhanced with increasing pH. The change in ionic strength did not exhibit a conspicuous trend in Ni(II) adsorption, thereby suggesting that the adsorption occurs through surface complexation rather than electrostatic interactions. The amounts of adsorption were measured with varying pH at three different Ni(II) concentrations under the condition of constant ionic strength, showing that with increasing Ni(II) concentration, the percentage of adsorption decreased and the adsorption edge was shifted to a higher pH value.
To study the feasibility of transmucosal delivery of leucine enkephalin (Leu-Enk) and $[D-ala^2]$-leucine enkephalinamide (YAGFL), their degradation extents and pathways in various rabbit mucosa extracts were investigated by high performance liquid chromatography. The degradation of Leu-Enk and YAGFL was observed to follow the first-order kinetics. The degradation half-lives of Leu-Enk in the nasal, rectal and vaginal mucosal extracts were 1.62, 0.37 and 1.12 hrs and those of YAGFL were 30.55, 9.70 and 6.82 hrs, respectively, indicating Leu-Enk was degraded in a more extensive and rapid manner than YAGFL. But the mucosal and serosal extracts of the same mucosa showed the similar degradation rates for both pentapeptides. The degradation was most rapid in the neutral pH and increasing concentrations of substrates retarded the degradation rates. The maior hydrolytic fragments of Leu-Enk were Des-Tyr-Leu-Enk and tyrosine, indicating the enzymatic hydrolysis by aminopeptidases. However, the data also suggested endopeptidases such as dipeptidyl carboxypeptidase and dipeptidyl aminopeptidase could play some role in the degradation of Leu-Enk. On the other hand, the hydrolytic fragments of YAGFL in all the mucosa extracts were mainly Tyr-D-Ala-Gly and Phe-Leu-Amide, demonstrating the hydrolytic breakdown by endopeptidases. The degradation pathways were further explored by concomitantly determining the formation of smaller metabolites of primary hydrolytic fragments of Leu-Enk and YAGFL in the mucosa extracts.

Bacteriophages are considered excellent sensing elements for platforms detecting bacteria. However, their lytic cycle has restricted their efficacy. Here, we used the minor coat protein pIII domain (N1N2) of phage CTXφ to construct a novel surface plasmon resonance (SPR) biosensor that could detect Vibrio cholerae. N1N2 harboring the domains required for phage adsorption and entry was obtained from Escherichia coli using recombinant protein expression and purification. SDS-PAGE revealed an approximate size of 30 kDa for N1N2. Dot blot and transmission electron microscopy analyses revealed that the protein bound to the host V. cholerae but not to non-host E. coli K-12 cells. Next, we used amine-coupling to develop a novel recombinant N1N2 (rN1N2)-functionalized SPR biosensor by immobilizing rN1N2 proteins on gold substrates and using SPR to monitor the binding kinetics of the proteins with target bacteria. We observed rapid detection of V. cholerae in the range of approximately 103 to 109 CFU/ml but not of E. coli at any tested concentration, thereby confirming that the biosensor exhibited differential recognition and binding. The results indicate that the novel biosensor can rapidly monitor a target pathogenic microorganism in the environment and is very useful for monitoring food safety and facilitating early disease prevention.
The kinetics of the loss of leaf fresh weight during incubation of barley and rice leaves in 9% or 15% NaCl solutions were biphasic, indicating the existence of a controlling mechanism for water transport. The first rapid phases reached their plateaus within 1 and 2 h in the case of rice and barley leaves, respectively. When barley leaves were fed with sodium fluoride, an inhibitor of phosphatase inhibitor, through their epicotyls for 3 h in darkness, prior to the treatment of NaCl, the biphasic pattern shown during NaCl treatment was disappeared resulting in linear decreases in the relative fresh weights. The results suggest that NaF accelerates salt-induced water efflux from plant cells, possibly by inhibiting the protection mechanism that may act in NaF-untreated leaves. The linear water loss can be explained in terms of phosphorylation of aquaporin by blocking its dephosphorylation in the presence of the phosphatase inhibitor to keep aquaporin in a phosphorylated form. However, the effect of NaF shown in barley leaves were not observed in rice. These results suggest that the regulation of water transport depends on plant species, and the mechanism for the controlling water transport in rice is different from that of barley.
Uptake kinetics of Cd and Zn by leaves and rhizome of the seagrass Zostera marina were examined in controlled laboratory radiotracer experiments. Subsequently, acute toxicity of Cd, Cu and TBT on photosynthetic quantum yield (ΔF/Fm’ of Z. marina were determined, and the differential sensitivities of rapid light curve (RLC) to those harmful substances were also compared. All measurements on photosynthetic activity were determined by chlorophyll a fluorescence method using pulse amplitude modulation (PAM). Metal uptake by Z. marina was saturated with increasing exposure time in leaves and rhizomes. Uptake of Zn by Z. marina was faster than that of Cd. Metal uptake rates in Z. marina decreased with the increase of dissolved metal concentrations and also with the increase of biomass. The adverse effect of TBT on effective quantum yield was stronger than other pollutants. Average acute toxicity on the RLC of the seagrass exposed to TBT and two heavy metals (Cd and Cu) was going to decrease as follows: TBT > Cd > Cu. Our preliminary results in this study suggested that Z. marina potentially can be used as a biomonitor of harmful substances contamination in coastal waters.
