Apparent digestibility of dry matter, organic matter, protein and energy of native Peruvian feedstuffs in juvenile rainbow trout (Oncorhynchus mykiss)

  • Trout production is a growing activity in recent years but requires new alternative sources of feed to be sustainable over time. The objective of this research was to determine the apparent digestibility coefficient (ADC) of dry matter (DM), organic matter (OM), crude protein (CP) and digestible energy (DE) of $ka{\tilde{n}}iwa$ (Chenopodium pallidicaule Aellen), kiwicha (Amaranthus caudatus L), quinoa (Chenopodium quinoa Willd), beans (Phaseolus vulgaris L.), sacha inchi, (Plukenetia volubilis L) and jumbo squid (Dosidicus gigas) meal in juvenile rainbow trout. The experimental diets were composed of a 70% basal diet and 30% of any raw materials. The ADC was determined by the indirect method using insoluble ash as a non-digestible marker. Jumbo squid, sacha inchi and quinoa showed the highest values of ADC (%) of DM (84.5, 73.5 and 69.7), OM (89.1, 78.4 and 72.9), CP (93.2, 98.0 and 90.3), and DE (4.57, 4.15 and 2.95 Mcal/kg DM), respectively. The ADC values for $ka{\tilde{n}}iwa$, kiwicha and bean were significantly lower. In conclusion, quinoa meal and jumbo squid meal have an acceptable digestibility but sacha inchi meal is a potential alternative for rainbow trout feeding in the future.

  • Five strains of gram-negative and yellow-pigmented bacteria were recently isolated from diseased freshwater fishes in Korea. All isolates were confirmed as a known fish pathogen of columnaris disease, Cytophaga columnaris based on their colonial and cellular morphology, and on physiological, biochemical and antigenic characteristics. Although the isolates were from different fish species, their characteristics of them were very similar to those of the reference strains of C columnaris (NCMB $2248^T$ and EK 28). Also, profiles of OMPs of Korean isolates were similar to those of the reference strain, C columnaris NCMB $2248^T$ when analyzed by SDS-PAGE.

Determination of Valine requirements by usging plasma free amino acid concentrations in rainbow trout(Oncorhynchus mykiss) with dorsal aorta cannulation

  • Dorsal aorta cannulated rainbow trout averaging 505 $\pm$ 6.5g (Mean $\pm$ SD) were divided into 7 groups (5 fish per group). Twenty four hour post feeding, cannulated fish were intubated with one of seven L-amino acid diets containing graded levels of Valine (Val. : 0.45, 0.95, 1.20, 1.45, 1.70, 1.95 or 2.45%) at 1% of body weight. Blood samples were taken at 0, 5 and 24hr after feeding the experimental diets. Post-prandial (5h after feeding) plasma-free Valine concentrations (PPval.) increased linearly from fish fed diets containing Valine between 0.45 and 1.45%, but the concentration remained constant from fish fed diets containing valine between 1.45 and 2.45%. Post-absorptive (24h after feeding) plasma free valine concentrations (PAval) increased linearly from fish fed diets containing valine between 0.45 and 1.45%, but the concentration remained constant from fish fed diets containing valine between 1.45 and 1.95%. Using the broken-line model analysis, the dietary valine requirement by PPval and PAval could be 1.44 and 1.50% in rainbow trout, respectively. These results supported that the estimated dietary valine requirement by PP(val.) and PA(val.) could be in close agreement with the values obtained from the previous conventional feeding method. Thus, the use of PFAA concentrations for determining essential amino acid requirements could be possible in rainbow trout with dorsal aorta cannulation.

  • Effect of 2,4-Dichlorophenoxy acetic acid (2,4-D) on vitellogenin (VTG) production and estrogen ($E_2$)-estrogen receptor (ER) binding affinity were examined in primary hepatocyte cultures of rainbow trout, Oncorhynchus mykiss. Hepatocytes were pre-cultured for 2 days; subsequently, $E_2$( 2$\times$$10^{-6}$/ M) and 2,4-D ($10^{-9}~10^{-6}/M$) were simultaneously added to the incubation medium. They were cultured for more than 5 days. VTG and $E_2$-ER binding affinities were analyzed by SDS-PAGE and ELISA, respectively. 2,4-D concentration used had no appreciable effect on the morphology, viability, and DNA content of hepatocytes in culture. It had also no effect on VTG production. However, it interfered with $E_2$-ER binding affinity, which was reduced with increasing concentration of 2,4-D. The affinity was inhibited by 25 and 30% at $10^{-7}$ M and $10^{-6}$ M of 2,4-D, respectively. This result suggested that although 2,4-D had no effect on VTG production, it acted as reno-estrogenic contaminant in ER.

Effects of Dietary Inclusion of Astaxanthin on Growth, Muscle Pigmentation and Antioxidant Capacity of Juvenile Rainbow Trout (Oncorhynchus mykiss)

  • This study was designed to investigate the effects of dietary astaxanthin levels on growth performance, feed utilization, muscle pigmentation, and antioxidant capacity in juvenile rainbow trout. Four experimental diets were formulated to contain 0, 50, 75, and 100 mg/kg astaxanthin (designed as AX0, AX50, AX75, and AX100). Each diet was fed to triplicate groups of fish (18.5 g/fish) for 10 weeks. Growth performance and muscle composition of fish were not affected by dietary astaxanthin levels. Total carotenoid concentration in the muscle of fish fed the AX50 diet was higher than that of fish fed the AX0 diet, but no significant differences were observed between these fish and those fed the AX75 and AX100 diets. Muscle astaxanthin content increased with increased astaxanthin in the diet. Deposition of astaxanthin in the flesh resulted in a decrease in lightness and an increase in redness and yellowness. The fillets from trout fed the AX75 diet had significantly lower lightness than trout fed the AX50 and AX100 diets. Fish fed the AX50 and AX75 diets showed significantly lower catalase activity than those fed the control diet. Total antioxidant status increased significantly in all astaxanthin supplemented groups when compared to the control group. Superoxide dismutase activity was significantly decreased in fish fed the AX50 diet compared to fish fed the AX0 diet. These findings suggest that while fillet pigmentation increased with increasing dietary astaxanthin concentration, indices of fish antioxidant capacity may not be affected in a dose dependent manner.

Study of pathogenicity and severity of Lactococcus garvieae isolated from rainbow trout (Oncorhynchus mykiss) farms in Kohkilooieh and Boyerahmad province

  • Background: Lactococcus garvieae is one of the most important risk factors in the rainbow trout culture. Therefore, the purpose of this study was to identify and detect strains isolated from rainbow trout suspected of having Lactococcus garvieae using biochemical characteristics and PCR and determination of the degree of severity of isolated strains. Methods: In this study, the cause of lactococcosis in selected rainbow trout farms in Kohkilooieh and Boyerahmad province was assayed. Gram-positive and catalase-negative bacterial isolates were first obtained from selected trout fish farms using conventional biochemical tests and PCR assay. The 10-day LD50 method (concentration causing 50% mortality in 10 days) was used to determine the severity of the isolated bacteria. Results: One bacterial isolate was detected from all sampled fish which confirmed as Lactococcus garvieae using a specific PCR assay based on the 16S rDNA gene by producing a single band of 1107 bp. Analysis of the rate of mortality showed that the 10-day LD50 was 4.6 × 105 CFU/fish. The results of this study showed that isolated bacteria had high severity for rainbow trout. The presence of bacteria in internal organs of suspected fish showed a severe systemic infection in challenged fish. Antibiogram assay also indicated that the isolated Lactococcus garvieae were resistant to some mostly used antibiotics in rainbow trout. Conclusions: According to current research, it can be concluded that the condition of lactococcosis in the studied area is not suitable, and despite the presence of disease, there is no proper action to control and prevent the disease. Unfortunately, isolated bacteria from the studied area have a very high severity compared to bacteria isolated from other regions of the country or other countries. Therefore, further investigation is needed to determine the cause of this difference and possibly in the design of the vaccine.

  • Effects of A23187 on estradiol$-17^{\beta}$-induced vitellogenin (VTG) induction were electrophore-tically examined in primary hepatocyte cultures in rainbow trout. hepatocytes were predultured for 2 days and then estradiol-$17^{\beta}$(E2, $2{\times}10^{-6}$M) and calcium ionophore (A23187, $10^{-7)$~$10^{-5}$ M) were added to the incubation medium. The hepatocytes were cultured for 7 more days. In addition, effects of A23187 on $E_2$-primed VTG production were investigated for 7 days. The addition of A23187 ($10^{-7)$~$10^{-5}$M) to the incubation medium specifically reduced VTG production by hepatocytes in a concentration-dependent way. The addition of A23187 significantly reduced the rate of $E_2$-primed VTG production to 18% of the control (E2 only) on Day 7. However, $E_2$-primed VTG production was reduced to 47% of the control by withdrawal of $E_2$ from the incubation medium. Therefore, these results suggest that intracellualr sequestered calcium could regulate VTG synthesis at the translational and/or post-translational stage.

Electron Microscopic Obsenrations on Micropvle after Sperm Penetration in Rainbow Trout, Oncorhynchus mykiss (정자 침입전후 무지개 송어의 난문에 대한 미세구조적 변화)

  • The time-course process by which spermatozoa penetrates through the micropvle apparatus into the egg cytoplasm of rainbow trout, Oncorhvnchus mvkiss, was examined with transmission and scanning electron microscopy. In the unfertilized egg, the ess surface beneath the inner opening of the micropylar canal did not differ distinctly from the rest of the animal pole area. A spermatozoon attached to the micropvle opening 20 seconds after insemination. In the initial stases of penetration, the spermatozoon still within the micropvlar canal attached perpendicularly at its apical tip to the ess surface, then the sperm head was rapidly engulfed by the folded egg surface with its manly microvilli. A large fertilization cone with microvillus-free surface appeared on the esS surface sutra-rounding the penetrating spermatozoon. The head portion of the penetrating spermatozoon was completely wrapped by the ess surface with only the tail portion visible externally 30 seconds after insemination. The fertilization cone displayed the tail portion of the penetrating spermatozoon on the central portion of its surface 60 seconds after insemination. 150 seconds after insemination, breakdown of the cortical granules elevation were initiated at the animal pole, then completed at the vegetable pole area. The spermatozoon disappeared from the outer surface of the ess before the fertilization cone completely retracted 250 seconds after insemination. In result, the block to polvspermv to permit entry of a sin81e sperm is considered to be mechanical by the rnorpholoSical design of the micropvle and fertilization cone.

Prediction and analysis of acute fish toxicity of pesticides to the rainbow trout using 2D-QSAR (2D-QSAR방법을 이용한 농약류의 무지개 송어 급성 어독성 분석 및 예측)

  • The acute toxicity in the rainbow trout (Oncorhynchus mykiss) was analyzed and predicted using quantitative structure-activity relationships (QSAR). The aquatic toxicity, 96h $LC_{50}$ (median lethal concentration) of 275 organic pesticides, was obtained from EU-funded project DEMETRA. Prediction models were derived from 558 2D molecular descriptors, calculated in PreADMET. The linear (multiple linear regression) and nonlinear (support vector machine and artificial neural network) learning methods were optimized by taking into account the statistical parameters between the experimental and predicted p$LC_{50}$. After preprocessing, population based forward selection were used to select the best subsets of descriptors in the learning methods including 5-fold cross-validation procedure. The support vector machine model was used as the best model ($R^2_{CV}$=0.677, RMSECV=0.887, MSECV=0.674) and also correctly classified 87% for the training set according to EU regulation criteria. The MLR model could describe the structural characteristics of toxic chemicals and interaction with lipid membrane of fish. All the developed models were validated by 5 fold cross-validation and Y-scrambling test.

  • This study was designed to determine the optimum pigment concentration supplemented in diet and feeding periods on the pigmentation of rainbow trout(Oncorhynchus mykiss) by using acetone-extracts of ascidian tunic as a natural pigment source. The eight pigmented diets contained carotenoid of ascidian tunic extracts at concentrations of 0, 200, 400, 800, 1,600 and 3,200mg/kg of diet, carophyll pink at concentration of 800mg/kg and commercial diet. No difference in pigment concentration was found between the ascidian extracts group and the control group until 4 weeks, but the redness of muscle and integument in the 1,600, 3,200mg/kg diet and carophyll pink was increased in the dorsal and caudal areas of fish from 6 weeks of age. In the sensory panel test, fish fed the ascidian tunic extracts diet were similar to those fed the carophyll pink diet. The optimum concentration and feeding periods for pigmentation of rainbow trout was found to be ascidian tunic extracts of 1,600mg/kg diet for 8 weeks.
