Skin Radioprotector (Diethone) Modifying Dermal Response of Radiation on Rats (방사선 보호제(Diethone)의 랫드 피부반응에 대한 수식작용)

  • Investigations were carried out into the time-and dose-related changes in acute skin reaction following graded single dose (20,30 and 40 Gy) of x-ray irradiation in Wistar rats, in order to evaluate the radioprotective effect of Diethon on skin. For the duration of skin response over 1. 5 score in dose of 40 Gy, the Diethone group of 24.7 days was significantly different (p<0.02) from that of control (29.8 days) and vaseline (29.2 days) groups, it was $17.1\%$ diminution of skin response period compared with that of control group. By the averaging daily scores for 10 days during peak skin reaction the mean scores were obtained. Mean score of Diethone group $(2.43\pm0.22)$ was significantly different (p<0.01) from that of control $(2.91\pm0.23)$ and vaseline $(2.81\pm0.18)$ groups of 40Gy dose. By iso-effect dose obtained at level of 2.5 score the dose reduction factor (DRF) was 1.41 which reduced radiation dose of $41\%$ by radioprotective effect of Diethone. From this experimental data, it may be possible to give higer radiation dose to large and/or radioresistant tumor mass rather than conventional treatment doses for improving therapeutic ratio by using topical application of skin radioprotector.

Effect of Si-Wu-Tang and Si-Jun-Zi-Tang on the Survival of Jejunal Crypt Cells and Hematopoietic Cells in Irradiated Mice (방사선조사 마우스에서 소장움세포 및 조혈세포 생존에 미치는 사물탕 및 사군자탕의 영향)

  • In order to investigate the radioprotective effect of Si-Wu-Tang (Korean name: Sa-Mul-Tang), a kind of traditional Oriental medicine as a blood-building decoction (Oriental medical concept: Bu-Xie), and Si-Jun-Zi-Tang (Korean name: Sa-Gun-Ja-Tang), one of the widely used Oriental herbal medicines as an energy tonic (Chinese medical concept: Bu-Qi). the jejunal crypt survival, endogenous spleen colony formation, and apoptosis in jejunal crypt cells were observed in irradiated mice. Jejunal crypts were protected by Si-Wu-Tang pretreated both per os (2 mg/mL of drinking water for 7 days, p<0.05) and intraperitoneally (1 mg/head, single injection at 24 hours before irradiation). Si-Wu-Tang adminstration before irradiation(1 mg/head, single injection at 24 hours before irradiation) resulted in an increase of the formation of endogenous spleen colony (p<0.005). The frequency of radiation-induced apoptosis in intestinal crypt cells was also reduced by pretreatment of Si-Wu-Tang (p<0.01). However, the radioprotective effect of Si-Jun-Zi-Tang was not as significant as that of Si-Wu-Tang. These results suggest that Si-Wu-Tang may be a useful radioprotective food, especially since it is a relatively nontoxic natural product.

  • Radioprotective effects of a red ginseng extracts on antioxidant enzymes(Superoxide dismutase, catalase and peroxidase) activities relationship to lipid peroxidation were studied in the cytosol fraction of mice liver. The experiments were carried out on Irradiated (5.5 Gy, $^{\60}Co$) and non-irradiated ICR mice after treatment of red ginseng extracts (5.5mg/mouse ; ip), In wholebody irradiated mice, irradiation caused a decrease in the activity of all these enzymes(on Day 21) The activities of SOD, Catalase and Peroxidase of red ginseng extracts treated mice were enhanced by $35.4\%,\;20.2\%$ and $20.1\%$, compared with non-treated mice. The red ginseng extracts led to inhibited increase of malondialdehyde product by ionizing radiation. The enhanced activity of enzymes that removed free radicals generated by radiation and thereby indicate that ginseng probably plays on important role in radioprotective effect.

  • In case of occuring the atomic energy accidents the proper medical treatments should be necessary. As the aim of the basic data for protective actions, the present studies were carried out to evaluate the decontamination of radioiodine by the administration of the antithyroid drugs (KI, NaI) and isotonic saline. Some recommended methods of decorporating radioiodine were investigated using 450, NIH-GP mice, each injected intraperitoneally with $1{\mu}Ci$ of $NaI^{131}$ as the internal contamination and treated with 2mg/0.2ml-saline of NaI and 2.6mg/0.2ml-saline of KI as the antithyroid drugs. Accordingly, effects of antithyroid drugs for internal contamination were: 1. Administration of NaI and KI caused to rapidly excrete internal radioiodine as the antithyroid drugs and decrease in whole body retention was reduced than in the saline group. 2. After internal contamination NaI and KI were to be administered for radioprotective effects as quickly as possible. 3. Decrease in body-retention made temporary shifts with enough fluids (water), however, as far as radioprotective effects is concerned, saline was not more significant than in the other group (NaI and KI). 4. Regarding to thyroid protective effects NaI, KI and saline were significant in effectively order.
