  • Monoclonal antibody to progesterone was produced using the antigen $11{\alpha}$-hydroxyprogesterone hemisuccinate conjugated to bovine serum albumin. Hybridomas secreting antibody to progesterone were detected by radioimmunoassay and enzyme-linked immunosorbent assay and cloned in soft agar. Two stable monoclonal antibodies which were highly specific to progesterone were obtained, so it may be advantageously used to study on several physiological functions of progesterone including immunological research.

  • In order to estimate the human placental lactogen levels and its value as an indicator of placental function during the first half of normal pregnancy, we defermined HPL levels. in normal snbjects (No=40) from the 6th week to the 20th week of gestation For the determination of human placental lactogen in serum of normal pregnant women during the first half of prognancy, radioimmunoassay have been performed using Phadebas HPL Test kit. The range of 25ng to 800ng HPL/ml. were determined by the procedures using Phadbas HPL Test kit, which correspond to levels recorded throughout the first half of normal pregnancy. HPL was detected in the earliest pregnancy samples collected at 6 weeks but was generally present in minute amount during the first trimester of pregnancy, when the HPL level ranged from 25 ng/ml. to 0.86 ${\mu}g$/ml. The mean concentration of HPL increased gradually from only 0.03 ${\mu}g$/ml, at 6 weeks to 0.25 ${\mu}g$/ml, at 12 weeks of gestation, Subsequently the mean HPL value rose steeply from 0.25 ${\mu}g$/ml, at 12 weeks to 1.65 ${\mu}g$/ml, at 20 weeks of gestation.

  • Radioiodine labelled human follicle stimulating hormone has been prepared using chloramine-T, with the approximate labelling yield of 65%. The labelled product is purified by means of a starch gel electrophoresis, and a Sephadex gel filtration, and the separation efficiencies are assessed for the effective use in radioimmunoassay. The results indicate that the gel filtration is efficient in view of the separation time, simplicity and bindability of the labelled hormone to the antibody. In determining the ratio of the free to the antibody hound labelled hormone, a double antibody technique is applied in comparison with a chromatoelectrophoresis. The ratio could be obtained only in the case of applying the double antibody technique.

  • A study was conducted to improve breeding efficiency of dairy and beef cows on Cheju Island. Milk progesterone and plasma progesterone profiles were determined by using radioimmunoassay techniques during the oestrus cycle, onset on oestrus and during the gestation period. Progesterone connections in milk were very low during the oestrus with a means value of 2.4ng/ml and 4.6ng/ml at the onset of oestrus but gradually declined at 4 to 9 hr after oestrus. In the cyclic cow, milk progesterone concentraitons in the early part of the cycle, ranged from 3 to 5ng/ml and increased from 6.0 to 11.6ng/ml on day 13 to 21 of cycle. Milk progesterone concentrations of pregnancy cows ranged from 5.0 to 40ng/ml during the gestation and grom basal line to 3.5ng/ml for nonpregnant cows. Plasma progesterone concentrations of beef during pregnancy period ranged form 3.0 to 33ng/ml and basal levels ranged from 0 to less than 2.5ng/ml for non-pregnant beef cows.

The Clinical Significance of Serum Triiodothyronine Measured by Radioimmunoassay in Various Thyroid Diseases (각종(各種) 갑상선질환(甲狀腺疾患)에서의 혈중(血中) $T_3$방사면역측정(放射免疫測定)의 진단적(診斷的) 의의(意義))

  • In an attempt to establish the diagnostic value of serum triiodothyronine and to correlate it with pathophysiologic mechanisms of thyroid hormones in various thyroid disorders, the author measured the serum triiodothyronine levels by means of radioimmunoassay and compared them with other thyroid function tests. This study was carried out in 152 cases with various thyroid functions; 28 cases as control, 51 cases of hyperthyroidism, 50 cases of euthyroidism and 23 cases of hypothyroidism. The results obtained were as follows: 1. The serum $T_3$ level in normal control group ranged between $131{\pm}34ng/dl$. 2. The serum $T_3$ levels ranged between $306{\pm}97ng/dl$ in hyperthyroidism $138{\pm}32ng/dl$ in euthyroidism and $60{\pm}27ng/dl$ in hypothyroidism. The significant differences between these groups were noted in this study. 3. In 5(9.9%) out of 51 cases with hyperthyoidism and 9(39.1%) of 23 cases with hypothyroidism, the serum $T_3$ were measured to be in normal range. Accordingly, the diagnostic value of the measurement of serum $T_3$ with hyperthyroidism was justifiable, but with hypothyroidism, it was less creditable than that of serum thyrotropin. 4. There was little significant difference between the diagnostic value of serum thyroxine and triiodthyronine levels in various thyroid disorders. However, $T_4/T_3$ ratio was decreased in patients with untreated hyperthyroidism because of more elevation of $T_3\;than\;T_4$. Consequently, the serum $T_3$ was more sensitive than $T_3$ in some thyroid disorders. 5. The serum $T_3$ level was much more sensitive and showed prompt shift in its level during the course of treatment on the patient with various thyroid disorders. And the measurement of serum $T_3$ was a good index for the evaluation of the thyroid function. From these results obtained, the measurement of serum $T_3$ by means of radioimmunoassay is a good way to understand the status of thyroid function with various thyroid disorders and evaluate the effects of the treatment given on these patients.

The Effect of Hemolysis sample on the Result of Nuclear Medicine Blood test (용혈검체가 핵의학 검체검사 결과에 미치는 영향)

  • Purpose In nuclear medicine blood tests, hemolysis samples are considered as inappropriate sample and are recommended not to be used for blood test. So, the lab are required to collect the blood again in the blood collection room However, The effect of hemolyzed samples on radioimmunoassay has not studied yet. This study was designed to evaluate effects of hemolysis on radioimmunoassay. Materials and Methods The kit manuals of 23 test items were reviewed to confirm whether hemolyzed samples were used. The subjects were 19 general applicants(male : 9, female : 13) and the samples were collected by each two SST tubes, one tube was obtained by centrifugation normally, and the other was obtained hemolyzed sample by centrifugation after external shock. It has been known that highly hemolyzed samples can affect the test results, so the test was performed using the severe hemolyzed sample. The test was performed for each test item using 23 normal serum and hemolysis serum, and SPSS19 program was used for statistical comparison of the test result. Results There was no significant difference between normal serum and hemolysis serum in 21 of 23 test items, but the results of insulin and C-peptide were significantly different(P<0.05). Conclusion It has been known that hemolysis in blood samples can affect the results of biochemical and hematological test, However, hemolysis effect is relatively low. Similarly, this study showed that hemolysis had not much effect on most of immunological radioimmunoassay except for some tests. Therefore, it is thought that the demand for re-collection due to hemolysis will be reduced in the laboratory, which will improve the work process of the laboratory.

  • Progesterone level in the seminal plasma and serum were measured by radioimmunoassay. The average level of seminal plasma progesterone was 0.76$\pm$0.15 ng/ml, similar to that of serum: 0.64$\pm$0.08 ng/ml. Progesteron levels in the seminal plasma were higher in the breeding season than in the nonbreeding season.
