A Study on Inactivation of Pathogenic Bacteria for Nutrient Solution Recycling Using Advanced Oxidation Processes

  • This study was conducted to test the applicability of hydroxy radical reactor system, which applied advanced oxidation processes, to sterilize pathogenic bacteria for nutrient solution recycling in closed hydroponics. Removal efficiency was tested on 25 L of nutrient solution maxed with 10 mL culture solution of bacteria, E. coli, and R. solanacearum in a pilot tank. The testing conditions included various levels of hydroxy radicals resulting from air flow rates of 40, 80, and $120L\;min^{-1}$, and 12 hours processing time. The removal of bacteria, E. coli, and R. solanacearum by hydroxy radical in nutrient solution was significantly increased with an increase in the flow rate of the air from $40L\;min^{-1}$ to $120L\;min^{-1}$. The optimum removal efficiency was achieved at an air flow rate of $120L\;min^{-1}$ for 2 hours treatment. There were no significant differences in removal efficiency among bacteria, E. coli, and R. solanacearum for tested level and time of hydroxy radical. These results verified the efficiency of hydroxy radical in removing the pathogenic bacteria and the applicability of hydroxy radical reactor system in the field.

농축 블루베리 퓌레 열처리 시 pH 조절제의 품질 저하 억제 효과 (Quality Deterioration Suppressing Effects of pH Regulators during Heat Treatment of Concentrated Blueberry Puree)

  • Purpose: In this study, we aimed to minimize quality degradation of concentrated blueberry puree by thermal treatment at $90^{\circ}C$ for 5 min, with citric acid (CA) and acidic sodium metaphosphate (ASM) as pH modifiers. Methods: The color values, redness a, anthocyanin contents, DPPH radical scavenging, elastase inhibition and tyrosinase inhibition activities were evaluated for quality parameters of concentrated blueberry puree. Results: The highest values of color value, redness a, anthocyanin contents, DPPH radical scavenging, elastase inhibition and tyrosinase inhibition activities were 1% CA+0.5% ASM followed by 1% CA and control. The quality parameters of concentrated blueberry puree were significantly different between control and 1% CA+0.5% ASM treatment (p<0.05). Conclusion: During thermal treatment, quality degradation of concentrated blueberry puree was reduced by 1% CA+0.5% ASM treatment, which is expected for new acidulants.

Volatile Compound, Physicochemical, and Antioxidant Properties of Beany Flavor-Removed Soy Protein Isolate Hydrolyzates Obtained from Combined High Temperature Pre-Treatment and Enzymatic Hydrolysis

  • The present study investigated the volatile compound, physicochemical, and antioxidant properties of beany flavor-removed soy protein isolate (SPI) hydrolyzates produced by combined high temperature pre-treatment and enzymatic hydrolysis. Without remarkable changes in amino acid composition, reductions of residual lipoxygenase activity and beany flavor-causing volatile compounds such as hexanol, hexanal, and pentanol in SPI were observed after combined heating and enzymatic treatments. The degree of hydrolysis, emulsion capacity and stability, 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity, and superoxide radical scavenging activity of SPI were significantly increased, but the magnitudes of apparent viscosity, consistency index, and dynamic moduli (G', G") of SPI were significantly decreased after the combined heating and enzymatic treatments. Based on these results, it was suggested that the enzymatic hydrolysis in combination with high temperature pre-treatment may allow for the production of beany flavor-removed SPI hydrolyzates with superior emulsifying and antioxidant functionalities.

Protective Effect of Brazilin on Cisplatin Nephrotoxicity

  • Cisplatin is one of the most effective antitumor agents currently available for cancer therapy. However, its clinical use has been limited by its severe side effects, especially nephrotoxicity. Therefore, brazilin, which has a radical scavenging effect, was given intraperitoneally to evaluate the effect on cisplatin nephrotoxicity in rats. Remarkable protective effects against nephrotoxicity of cisplatin were observed when brazilin was administered to rats simultaneously with cisplatin. Hepatotoxicity induced by combination treatment of cisplatin and brazilin was evaluated by measuring serum glutamic pyruvate transaminase and serum glutamic oxalate transaminase. Combination treatment did not affect the levels of sGPT and SGOT, and any combination treatment did not induce metallothionein in kidney. Brazilin which has radical scavenging effect directly reduced nephrotoxicity of wisplatin in vivo. Thus, it seems that nephrotoxicity of cisplatin was caused by free radicals. The present results Indicate that brazilin, when it is given with cisplatin, may provide protection against cisplatin nephrotoxicity in rats.

Barley Malt Treated with Enzymes Increases Polyphenol Content and Antioxidant Activity

  • The purpose of this study was to improve the functionality of a healthy drink with examining the possibility of manufacturing different enzymes (alpha-, beta-, glucose-amylase) in barley malts (BM) produced in various malting periods. The study showed that enzyme treatment increased significantly total polyphenol content (TPC), DPPH radical scavenging activity and hydroxly radical scavenging activity in malted liquid samples (MLS) which obtained from various malting periods. The highest of TPC were found in Gluco-24M with 1.981 mgTAE/ml, followed by Beta-24M and Alpha-72M with 1.878 mgTAE/ml and 1.845 mgTAE/ml, respectively. The DPPH result revealed that percent of inhibition increased by 71-75% compared to the control. No statistical difference was found between MLS obtained by 24 hr of malting (24 M) and 72 hr of malting (72 M) after enzyme treatment. In addition, an increasing of hydroxyl radical was in the same trend to the TPC and DPPH. The hydroxyl radical scavenging activity of enzyme treated samples was 1,5 times higher than the control. These results suggest the possibility of enzyme application to barley malts obtained in various germination periods for improving quality and functionality of barley malts.

Cu-Zn 금속 합금의 산화.환원 반응에 의한 난분해성 COD처리에 관한 연구 (A Study on the Treatment of Refractory Organics by Redox Reaction of Cu-Zn Metal Alloy)

  • 본 연구는 압연공정에서 발생하는 폐수 중에 함유되어있는 난분해성 COD 물질을 $80{\mu}m$ 두께의 극세사 형태로 제조된 Cu-Zn 금속합금의 산화 작용으로 인하여 발생하는 OH 라디칼을 이용하여 처리하는 방법에 관한 기초 연구이다. OH 라디칼은 유기화합물(RH) 속에 포함된 수소를 수소추출반응(H Abstraction) 또는 탄소와 탄소(C-C)의 불포화 결합에 첨가됨으로써 빠르고 비 선택적인 반응을 수행하는 것으로 알려진 것처럼 난 분해성 유기화합물의 처리에 효과적인 것으로 나타났다. 금속합금 반응 물질은 극세사 형태로 표면적이 넓어서 1회 처리만으로도 수용액의 pH를 평형에 도달하게 하여서 반응 효율성이 높은 것으로 나타났다. COD처리 효율은 중성 pH에 가까운 pH 7, pH 6에서 최고치를 보였으며 산성분위기인 pH 5이하 및 알칼리성 분위기인 pH 8이상에서는 낮은 효율을 보였다. 실제 압연 폐수의 응집 침전을 이용한 COD 처리에서도 redox 반응장치의 유무에 따라 2배 이상의 처리효율의 차이를 보였다.

모형 가스터빈 연소기의 연소특성 -라디칼 자발광강도와 국소당량비계측에 대하여- (Combustion Characteristics of Model Gas Turbine Combustor -Radical Luminous Intensity and Local Equivalence Ratio Measurement-)

  • There are three active radicals which become to the scale of flame diagnostics at the flame front. They are OH, CH and $C_2$ radical. For this, optical measurement system which could monitor simultaneously the luminous waves of three radicals, was constructed. These were analyzed statistically into the cross correlation, coherence and phase. Through such an statistical treatment, combustion characteristics was investigated at the primary zone of gas turbine combustor. The local equivalence ratio was predicted with the ratio of luminuous intensity between CH and $C_2$ radical. This result was matched up to the equivalence ratio calculated from gas composition within 5% error. In general, equivalence ratio was said to be 1.0 at flame front, but it could be increased up to about 1.2 depending on the degree of swirl intensity in case of changing properly the air amount of primary zone.

오수목과탕(吳茱木瓜湯)의 자유(自由)라디칼 소거활성(演去活性) 및 肝(간) 세포(細胞) 보호효과(保護效果) (Free Radical Scavenging and Hepatoprotective Effects of Osumokkya-tang)

  • In order to find the new hepatoprotective agents preliminary screening of twelve prescriptions which have been used for the treatment of hepatic disease in Oriental traditional medicine has been carried out. Both DPPH free radical scavenging and hepatoprotective effect on tacrine-induced cytotoxicity in Hep G2 cells are performed. Osumokkya-tang which is one of the prescription belongs to interior-warming properties showed the DPPH free radical scavenging effect as well as hepatoprotective activity significantly. It is also suggested that its hepatoprotective effect is derived from Chaenomelis Fructus.

Toxicological Studies on the Essential Oil of Eugenia caryophyllata Buds

  • The essential oil (EC-oil) obtained from the buds of Eugenia caryophyllata (Myrtaceae) was examined for its free radical-scavenging activity, cytotoxicity, and in vivo toxicity. To find the xenobiotic properties of EC-oil, serum thiobarbituric acid reactive substances (TBARS) level and hepatic drug-metabolizing enzyme activities were measured. It was found that EC-oil displayed xenobiotic properties like bromobenzene. The cytotoxicities of eugenol and of the EC-oil were greatly attenuated by the sulfhydryl-containing N-acetyl-L-cysteine (NAC), suggesting that eugenol was susceptible to nucleophilic sulfhydryl. In addition, eugenol also showed potent free radical-scavenging activity in the 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay. Moreover, methyleugenol considerably exhibited less cytotoxicity and less potent free radical-scavenging activity than eugenol, and the cell viability of the methyleugenol was more increased with NAC treatment than the eugenol. These results indicate that the phenolic OH in eugenol may play a crucial role in both cytotoxicity and free radical-scavenging activity. The fashion on oxidative stress and hepatic drug-metabolizing enzyme activities of eugenol resembled those of bromobenznene.

오공(蜈蚣)의 항산화효과에 관한 연구 (Antioxidative Effects of Scolopendra subspinipes)

  • Objective: The purpose of this study was to evaluate the antioxidative effects of the extract of Scolopendra subspinipes which has been used mainly for detoxication in the oriental medicine and reported to have sedative action, antiinflammatory effect, antihypertensive property and immunity enhancing activity. Method: Inhibitory activities on oxygen radical generating enzymes (aldehyde oxidase and xanthine oxidase) and increasing activities on oxygen radical scavenging enzymes (superoxide dismutase, glutathione peroxidase, glutathione-S-transferase) were investigated. Furthermore, the content of glutathione in the mouse brain, DPPH radical scavenging activity and also anti-lipid peroxidative effects in vivo and in vitro were estimated. Results: The extract showed weak inhibitory effects on the activities of aldehyde oxidase and xanthine oxidase which are oxygen radical generating enzymes. The extract inhibited lipid peroxidation with 26.1% against control group at 500 mg/kg in vivo and with 11.2% against control group at 10 mg/kg in vitro in a dose-dependent manner, which means this drug may protect radical-induced cell damages. The extract showed dose-dependently the scavenging effect on DPPH radical with 24.8% activity at 10 mg/ml in vitro. The extract enhanced the activities of superoxide dismutase, glutathione peroxidase and glutathione-S-transferase, which are oxygen radical scavenging enzymes, with 28.9%, 22.3% and 23.1%, respectively at 500mg/kg in vivo. Finally, this extract strongly increased the glutathione content in the mouse barin. Conclusion: Above results indicated that Scolopendra subspinipes can be useful for the protection or treatment of some diseases caused by reactive oxygen species.
