수중에서 E. coli 소독을 위한 전기-UV-초음파 복합 공정에 대해 실험계획법과 반응표면분석법(RSM)을 적용하였다. 2차반응표면 모형식을 추정할 수 있는 Box-Behnken법을 이용하여 전기-UV-초음파 복합 공정의 소독 반응에서 전기분해($X_1$), UV ($X_2$), 및 초음파 공정($X_3$)의 전력을 독립변수로 선정하여 수학적으로 모형화하였다. 소독 후 잔류 E. coli 수와 독립변수 사이의 실험에서 독립변수에 대해 다음의 모형식이 얻어졌다. 잔류 E. coli number (Ln CFU) = 23.69 - 3.75 Electrolysis - 0.67 UV - 0.26 Ultrasonic - 0.16 Electrolysis UV + 0.05 Electrolysis Ultrasonic + 0.27 $Electrolysis^2$ + 0.14 $UV^2$ - 0.01 $Ultrasonic^2$). 예측된 모형식은 실험 자료와 잘 일치하였다($R^2$ = 0.983). 2차원 등고선도와 3차원 반응표면도가 잔류 E. coli 수에 대한 최적 범위를 구하기 위하여 사용되었다. Design-Expert 소프트웨어의 '수치 최적화'를 이용하여 잔류 E. coli 수에 대한 최적 값을 찾은 결과 1.47 Ln CFU/L이었고, 최적 조건은 전기분해 6.94 W, UV 6.72 W 및 초음파 공정 14.23 W로 나타났다. 본 연구는 반응표면분석법이 복합 소독 공정에서 잔류 E. coli 수를 최소화하고 운전 조건을 최적화하기 위한 적절한 방법 중의 하나라는 것을 보여주었다.
Aluminum alloy-based additive manufacturing (AM) has emerged as a popular manufacturing process for the fabrication of complex parts in the automotive and aerospace industries. The addition of an inoculant to aluminum alloy powder has been demonstrated to effectively reduce cracking by promoting the formation of equiaxed grains. However, the optimization of the AM process parameters remains challenging owing to their variability. In this study, the response surface methodology (RSM) was used to predict the crack density of AM-processed Al alloy samples. RSM was performed by setting the process parameters and equiaxed grain ratio, which influence crack propagation, as independent variables and designating crack density as a response variable. The RSM-based quadratic polynomial models for crack-density prediction were found to be highly accurate. The relationship among the process parameters, crack density, and equiaxed grain fraction was also investigated using RSM. The findings of this study highlight the efficacy of RSM as a reliable approach for optimizing the properties of AM-processed parts with limited experimental data. These results can contribute to the development of robust AM processing strategies for the fabrication of high-quality Al alloy components for various applications.
본 실험에서는 Aspergillus niger NRRL 567의 고체배양을 이용한 구연산 생산 시, 물리/화학적 발효 조건인 배양 온도, 배지 pH, 접종 농도 및 수분 함량이 구연산 생산에 미치는 영향을 단일변수(one-factor-at-a-time)와 반응표면 분석법(surface response methodology)을 이용하여 순차적 최적화를 수행하였다. 단일변수 최적화의 경우, A. niger에 의한 구연산 생산은 물리/화학적 발효 조건에 의해 영향을 받으며, 발효 온도 $30^{\circ}C$, 영양 배지 pH 7.1, 수분 함량 75%와 접종 농도 $4.0{\times}10^6$ spores/ml에서 최대 구연산 생산인 98.2 g/kg DPM (dry peat moss)을 보였다. 단일변수 최적화에 근거하여 반응표면 분석법을 도입하여 2차 최적화를 수행했을 경우, 배지 pH와 수분 함량이 구연산 생산에 유의한 영향을 주었으며 온도 $26.5^{\circ}C$, 영양 배지 pH 9.9, 수분 함량 75.1%와 접종 농도 $6.0{\times}10^6$ spores/ml에서 최대 구연산 생산인 118.8 g/kg DPM가 얻어졌다. 이는 최적화 이전의 대조군에 비해 구연산 생산이 1.6배 증가한 결과이다.
본 논문에서는 반응표면분석법을 이용한 Arthrobacter sp. PAMC 25486의 carotenoids 생산 배지의 최적화를 수행하였다. Placket-Burman 방법을 이용하여 yeast extract, $MgSO_4$, dextrose가 carotenoids의 생산에 영향을 미치는 주요인자인 것을 확인하였다. 반응표면분석 방법을 이용하여 최대 carotenoids 생산 농도를 갖는 yeast extract, $MgSO_4$, dextrose의 농도를 계산한 결과 1 g/L yeast extract, 0.0879 g/L $MgSO_4$ and 1 g/L dextrose의 농도에서 최대 307 mg/L의 carotenoids 농도가 예측됐으며, 실제 배양 결과 288 mg/L carotenoids가 얻어졌다. 얻어진 농도 값은 최적화 이전의 값에 비하여 200% 이상 증가하였다. 이러한 결과로부터 미생물 배양에 의한 carotenoids 생산을 증가시키기 위한 배지최적화 방법으로서 반응표면분석법의 유용성을 확인할 수 있었다.
The automation technology for overlay welding is needed due to the occurrence of severe corrosion and abrasion on the surface of internal contact in different shape of fittings. In Korea, different shapes of fittings have been manufactured by using the imported equipment of overlay welding automation at some companies. Thus the research on the development of overlay welding automation system (in short, OWAS) for a large L-type tube is urgently needed. In this paper, the investigation is focused on the optimal design of a supporting base for the (currently developing) OWAS of large L-type tube. Specifically we assume that the base which supports the equipment during the process of overlay welding is loaded as self-weight in the direction of gravity through static analysis especially when it is rotated 180 degree on the OWAS. For optimal design of a supporting base for OWAS of large L-type tube, Solidworks(R) (for 3-dimensional modelling) and ANASYS Workbench(R) (for structural analysis) are incorporated so as to proceed an optimization routines based on Response Surface Method (RSM) and Design of Experiment (DOE). In more specific, DOE finds out major factors (or dimensions) of the supporting base by using MINITAB(R). Then the regression equations between design variables (the major factors of supporting base) and response variables (deformation, stress and safety factor for the supporting base), which will be resulted in by RSM, verify the major factors of DOE. In the next step, Central Composite Design (CCD) plans 20 simulations of ANASYS Workbench(R) and then figures out the optimal values of design variables which will be reflected on the manufacturing of supporting base. Finally welding experiment is conducted to figure out the influence of overlay welding quality in applying the optimized design values of supporting base to the actual OWAS.
Response surface methodology (RSM) was applied to determine the optimum drying conditions for pork jerky. The physicochemical properties of pork jerky, such as final moisture content, water activity (Aw), pH, and shear force were investigated. In addition, sensory characteristics of pork jerky were evaluated and were used as a parameter for determining the optimum condition. Pork jerky samples were dried at different temperatures between 40 to $80^{\circ}C$ for the time ranged from 0 to 10 hr. The predicted values for moisture content, Aw, and shear force of dried pork samples were in good agreement with the experimental values with correlation coefficients ($R^2$) of 0.95, 0.96, and 0.97, respectively. Both drying temperature and time significantly (p<0.01) affected moisture content, Aw, pH, and shear force and their interactions were also significant at p<0.01 except for Aw. RSM showed the optimum drying conditions for pork jerky, based on moisture content, shear force, and sensory evaluation to be oC$65-70^{\circ}C$ for 7-8 hr.
식물체의 플라보노이드 성분 함량 분석에 필요한 적정 가수분해 조건을 구명하기 위하여 은행잎을 이용하여 중심합성법으로 설계한 산 가수분해 시간과 HCl 농도에 따라 myricetin, quercetin, kaempferol 함량을 HPLC로 분석하고, SAS의 반응표면분석법으로 산 가수분해 조건의 최적화를 시도하였다. HCl 0.5~2.5 M, 0.5~2.5시간 범위 내에서 10개의 구간을 잡아서 최적화한 결과 1.5~1.9 M의 HCl 농도와 1.4~2.0시간의 가수분해 조건에서 myricetin, quercetin, kaempferol 성분이 가장 높았다. Superimposing하여 구한 식물체의 플라보노이드의 적정 가수분해 조건은 HCl 1.5 M, 1.5시간이었으며, 이 조건에서 구한 플라보노이드 함량의 예측치와 측정치는 95% 이상 일치하였다. 본 연구에서 구한 적정 가수분해 조건을 이용하여 녹차잎, 무화과잎, 상동나무잎의 플라보노이드 함량을 분석하여 보았다.
식품공업과 의학, 의료, 화장품 등 많은 분야에서 널리 이용되고 있는 단백질 분해 효소의 하나인 papain을 papaya 유액으로부터 추출해내기 위한 최적 조건을 반응표면 분석법(RSM)을 이용하여 결정하였다. 중심 합성법에 의한 실험 계획을 설정하여 부분요인 실험법에 의하여 crude papain을 추출 및 분리하고 선정된 독립 인자의 반응 표면에 대한 영향을 분석하여 다중 회귀 분석으로 다음과 같은 모델식(Y)을 얻었으며, EDTA는 다중 회귀 분석에 의하여 제거되었다. $Y=29.0630-2.5812X_3+149.3208X_4+{13.1705X_1}^{2}+{0.0208X_3}^2-{9.8494X_4}^2$. 따라서 이 결과 papain 추출은 $NaHSO_3$, 농도가 4%, 추출 시간은 120분, pH는 7.6일때 최적 조건이 된다. 이러한 최적 추출 조건으로 얻은 효소 단백질 추출 양의 실험치는 793.16 mg prot./g latex로, 이는 효소 단백질 추출 예측치인 795.60 mg prot./g latex와 매우 근접한 값을 나타냄으로써 r=0.9951의 높은 상관 관계를 입증하였다.
