A Study on Combustion Characteristics and Evaluating of RDFs(Refused Derived Fuels) from Mixture of Petrochemical Wastewater Sludge and Organic Matters (석유화학폐수슬러지와 유기성 폐기물 혼합에 의한 연소특성 및 고형연료 폐기물화 재활용에 관한 연구)

  • This objectives of research are to figure out combustion characteristics with increasing temperature with petrochemical sludge by adding wasted organic matters which are waste electric wire, anthracite coal and sawdust, and to exam heating value and ignition temperature for using refused derived fuels(RDFs). After analyzing TGA/DTG, petrochemical sludge shows a rapid weight reduction by vaporing of inner moisture after $170^{\circ}C$. Gross weight reduction rate, ignition temperature and combustion rates represent 68.6%, $221.9^{\circ}C$ and 54.1%, respectively. In order to assess the validity of the RDFs, the petrochemical sludge by adding wasted organic matters which are waste electric wire, anthracite coal and waste sawdust. The materials are mixed with 7:3(petrochemical sludge : organic matters)(wt%), and it analyzes after below 10% of moisture content. The ignition temperatures and combustion rates of the waste electric wire, anthracite coal and waste sawdust are $410.6^{\circ}C$, $596.1^{\circ}C$ and $284.1^{\circ}C$, and 85.6%, 30.7% and 88.8% respectively. In heating values, petrochemical sludge is 3,600 kcal/kg. And the heating values of mixed sludge (adding 30% of the waste electric wire, anthracite coal and waste sawdust) each increase up to 4,600 kcal/kg, 4,100 kcal/kg and 4,300 kcal/kg. It improves the ignition temperatures and combustion rates by mixing petrochemical sludge and organic matters. It is considered that the production of RDFs is sufficiently possible by using of petrochemical sludge by mixing wasted organic matters.

Anti-inflammatory Effects of Resveratrol, (-)-Epigallocatechin-3-gallate and Curcumin by the Modulation of Toll-like Receptor Signaling Pathways (Toll-like receptors 신호전달체계 조절을 통한 resveratrol, (-)-epigallocatechin-3-gallate, curcumin의 항염증 효과)

  • Toll-like receptors (TLRs) induce innate immune responses that are essential for host defenses against invading microbial pathogens, thus leading to the activation of adaptive immune responses. In general, TLRs have two major downstream signaling pathways: the MyD88- and TRIF-dependent pathways, which lead to the activation of $NF-{\kappa}B$ and IRF3. Numerous studies have demonstrated that certain phytochemicals possessing anti-inflammatory effects inhibit $NF-{\kappa}B$ activation induced by pro-inflammatory stimuli, including lipopolysaccharides and $TNF{\alpha}$. However, the direct molecular targets for such anti-inflammatory phytochemicals have not been fully identified. Identifying the direct targets of phytochemicals within the TLR pathways is important because the activation of TLRs by pro-inflammatory stimuli can induce inflammatory responses that are the key etiological conditions in the development of many chronic inflammatory diseases. In this paper we discuss the molecular targets of resveratrol, (-)-epigallocatechin-3-gallate (EGCG), and curcumin in the TLR signaling pathways. Resveratrol specifically inhibited the TRIF pathway in TLR3 and TLR4 signaling, by targetting TBK1 and RIP1 in the TRIF complex. Furthermore, EGCG suppressed the activation of IRF3 by targetting TBK1 in the TRIF-dependent signaling pathways. In contrast, the molecular target of curcumin within the TLR signaling pathways is the receptor itself, in addition to $IKK{\beta}$. Together, certain dietary phytochemicals can modulate TLR-derived signaling and inflammatory target gene expression, and in turn, alter susceptibility to microbial infection and chronic inflammatory diseases.

A Study on the Official Uniform of Najang from the Late - Joseon Dynasty, with Focus on the Relic Collections in Leipzig Grassi Museum, Germany - (조선 말기 나장복에 관한 연구 - 독일 라이프치히그라시민속박물관 소장 유물을 중심으로 -)

  • Najang was the central Seori, affiliated to the Ministry of War during the Joseon dynasty. The objective of this study is to research the existing authentic Najang costumes from the late-Joseon dynasty by examining factors, such as the composition of the costume, size, and method of creation, and attempt to replicate it. The Leipzig Grassi Museum in Germany possesses an official uniform of Najang from the late-Joseon dynasty, and we visited the museum in May of 2013 to examine it for the study. Written records, or Uigwe, and other pictorial data from the Joseon Dynastry describe the Najang wearing black or navy clothing with white decoration and pointy hats. The most notable characteristic of the Najang uniform is that it has the cotton cords pattern. The hat has a brass ball attached, which was worn with the ball facing the front in the early Joseon Dynasty, and was worn facing the back in the later years. They usually wore black head cloths (Heuk-geon), but would attach feathers on the black hats (Heuk-rip) for special occasions. The Najang uniform preserved in the Leipsiz Grassi Muesim does not exist in Korea. It is made of cotton. The cotton cord pattern of the uniform of Najang was made using single-ply cords and double-ply cords. The hat worn by Najang is in a form of a cone that becomes narrower towards the top or is in a form with wide and open end. It was made of oiled paper covered with hemp, and two circular metal disks were attatched at the rear.

Antiviral Activity of a Type 1 Ribosome-inactivating Protein from Chenopodium album L.

  • The antiviral activity of CAP30 from Chenopodium album, a type1 ribosome-inactivating protein (RIP), was examined against 5 different plant viral pathogens, and its activity against Tobacco mosaic virus was compared to those of well known antiviral proteins such as Pokeweed Antiviral protein from leaves and seeds. When the inoculating concentration of Tobacco mosaic virus was varied from 0.4 to $400{\mu}g/ml$, it was observed that CAP30 at the concentration of $1{\mu}g/ml$ suppressed the viral infection of C. amaranthicolor and C. quinoa almost completely up to $40{\mu}g/ml$ Tobacco mosaic virus. Results from the assays for the inhibitions of in vitro translation of rabbit reticulocyte lysate and the suppression of Tobacco mosaic virus infection ($10{\mu}g/ml$) to C. quinoa indicated that CAP30 is a strong inhibitor of protein synthesis and virus infection. The infection of several viruses other than Tobacco mosaic virus to host plants were also inhibited by $5{\mu}g/ml$ CAP30, suggesting that a gene encoding CAP30 can be used to develop transgenic virus-resistant plants.

Progress and Prospect of Rice Biotechnology in Korea

  • This is a progress report of rice biotechnology including development of gene transformation system, gene cloning and molecular mapping in rice. The scope of the research was focused on the connection between conventional breeding and biotech-researches. Plant transformation via Agrobacterium or particle bombardment was developed to introduce one or several genes to recommended rice cultivars. Two chimeric genes containing a maize ribosome inactivating protein gene (RIP) and a gerbicide resistant gene (bar) were introduced to Nipponbare, a Japonica cultivar, and transmitted to Korean cultivars. The homozygous progenies of herbicide resistant transgenic plant showed good fertility and agronomic characters. To explore the genetic resourses in rice, over 8,000 cDNA clones from immature rice seed have been isolated and sequenced. About 13% of clones were identified as enzymes related to metabolic pathway. Among them, twenty clones have high homology with genes encoding enzymes in the photorespiratory carbon cycle reaction. Up to now about 100 clones were fully sequenced and registered at EMBL and GenBank. For the mapping of quantitative tarits loci (QTL) and eternal recombinant inbred population with 164 F13 lines (MGRI) was developed from a cross between Milyang 23 and Gihobyeo, Korean rice cultivars. After construction of fully saturated RFLP and AFLP map, quantitative traits using MGRI population were analyzed and integrated into the molecular map. Eighty seven loci were determined with 27 QTL characters including yield and yield components on rice chromosomes. Map based cloning was also tried to isolate semi-dwarf (sd-1) gene in rice. A DNA probe, RG 109, the most tightly linked to sd-1 gene was used to screen from bacterial artifical chromosome (BAC) libraries and five over lapping clones presumably containing sd-1 gene were isolated. Rice genetic database including results of biotech reasearch and classical genetics is provided at Korea Rice Genome Server which is accessible with world wide web (www) browser. The server provides rice cDNA sequences and map informations linked with phenotypic images.

Effect of 42 amino acid long amyloid-β peptides on Arabidopsis plants

  • Although the evolution of Arabidopsis thaliana and humans diverged approximately 1.6 billion years ago, recent studies have demonstrated that protein function and cellular processes involved in disease response remain remarkably conserved. Particularly, γ-secretase, a multisubunit protein complex that participates in intramembrane proteolysis (RIP) regulation, is also known to mediate the cleavage of more than 80 substrates including the amyloid precursor protein (APP) and the Notch receptor. Although the genes (PS1/2, APH-1, PEN-2, and NCT) coding for the γ-secretase complex components are present in plant genomes, their function remains largely uncharacterized. Given that the deposition of 42 amino acid long amyloid-β peptides (hAβ42) is thought to be one of the main causes of Alzheimer's disease, we aimed to examine the physiological effects of hAβ42 peptides on plants. Interestingly, we found that Arabidopsis protoplast death increased after 24 h of exposure to 3 or 5 µM hAβ42 peptides. Furthermore, transgenic Arabidopsis plants overexpressing the hAβ42 gene exhibited changes in primary root length and silique phyllotaxy. Taken together, our results demonstrate that hAβ42 peptides, a metazoan protein, significantly affect Arabidopsis protoplast viability and plant morphology.

Identification and Characterization of an Antifungal Protein, AfAFPR9, Produced by Marine-Derived Aspergillus fumigatus R9

  • A fungal strain, R9, was isolated from the South Atlantic sediment sample and identified as Aspergillus fumigatus. An antifungal protein, AfAFPR9, was purified from the culture supernatant of Aspergillus fumigatus R9. AfAFPR9 was identified to be restrictocin, which is a member of the ribosome-inactivating proteins (RIPs), by MALDI-TOF-TOF-MS. AfAFPR9 displayed antifungal activity against plant pathogenic Fusarium oxysporum, Alternaria longipes, Colletotrichum gloeosporioides, Paecilomyces variotii, and Trichoderma viride at minimum inhibitory concentrations of 0.6, 0.6, 1.2, 1.2, and 2.4 μg/disc, respectively. Moreover, AfAFPR9 exhibited a certain extent of thermostability, and metal ion and denaturant tolerance. The iodoacetamide assay showed that the disulfide bridge in AfAFPR9 was indispensable for its antifungal action. The cDNA encoding for AfAFPR9 was cloned from A. fumigatus R9 by RT-PCR and heterologously expressed in E. coli. The recombinant AfAFPR9 protein exhibited obvious antifungal activity against C. gloeosporioides, T. viride, and A. longipes. These results reveal the antifungal properties of a RIP member (AfAFPR9) from marine-derived Aspergillus fumigatus and indicated its potential application in controlling plant pathogenic fungi.

Antibacterial Effect of the Surface-Modified Biomedical Polyurethane against Staphylococcus aureus and Staphylococcus epidermidis

  • Staphylococal infection still remains to be one of the most serious infections, having various complications in the clinical use of indwelling polymeric medical devices. However, there are a few promising systems showing a high antibacterial effect without causing any demage of polymer backbone under biological environments such as blood or body fluid. In order to resolve this problem, we have designed a new antibiotic releasing system via a hydrolysis mechanism. The surface of biomedical polyurethane (PU) was modified by using 1,6-diisocyanatohexane (HMDI) to immobilize the rifampicon. Also, the immobilized rifampicin was designed to be released by a selective cleavage of the unstable carbamate linkage that exists on the rifampicin-immobilized polyurethane (PHR). The immobilization of rifampicin on the surface of polyurethane was confirmed by the disappearance of the characteristics IR absorbance peak of the isocyanate (-NCO) group at $2,267\;cm^{-1}$. The PHR showed a continuous rifampicin release profile under an aqueous environment of 10 mM of PBS (phosphate-buffered saline) for ove 6 days. The rifampicin molecules, which are released from PHR under an optimal bacterial infection environment, had a higher antibacterial activity against both S. aureus and S. epidermidis than rifampicin-incorporated polyurethane (RIP). In addition, the PHR maintained a stable antibacterial effect under a blood-mimic aqueous environment such as bovine calf serum.

Anti-cancer Effects of Dendropanax Morbifera Extract in MCF-7 and MDA-MB-231 Cells (황칠나무 줄기 추출물의 MCF-7과 MDA-MB-231 유방암 세포주에 대한 세포증식억제 효과)

  • Objectives : Dendropanax morbifera is known as a tree that has been used in traditional medicine for various diseases. However, its biological activities in cancer have not yet been clearly elucidated. In this study, we investigated the anti-cancer effects of water extract of Dendropanax morbifera (DP) on 2 human breast cancer cell lines (estrogen dependent MCF-7 and estrogen independent MDA-MB-231). Methods : The MTT assay and flow cytometry were used to assess cell proliferation, along with cell cycle analysis. Nitric oxide production was detected by Griess assay. The expression of apoptosis related gene was assessed by quantitative real-time PCR. Results : Our data revealed that DP inhibits the cell growth in a dose dependent manner (0, 50, 100, 250, and 500 μg/ml) of both estrogen independent MDA-MB-231 and estrogen dependent MCF-7 breast cancer cells. Also, LPS induced nitric oxide production was significantly reduced by DP. Cell cycle analysis showed an increased G1 phase in the MCF-7 cell and G2/M phase in the MDA-MB-231 cell. DP decreased mRNA expression of apoptotic suppressor gene Bcl-xL, and increased mRNA expression of pro-apoptotic genes. DP increased mRNA expression of p21, and Rip1 in both cell. And DP decreased mRNA expression of survivin in the MCF-7 cell. Conclusions : Taken together, these results indicate that DP extract are source of anti-cancer potential and could be developed botanical drug.

The application effective investigation of Organic materials from organic farming village (유기농업 시범마을에서 유기농자재의 활용 효과)

  • 지속 가능한 농업과 안전한 농산물의 생산을 유지시키면서 환경부담 물질 투입량을 절감시킬 수 있는 농법을 개발하기 위해서 유기농자재 사용실태 및 효과를 조사했다. 전북 익산시 웅포면 유기농업 시범마을 현지 농가에서 사용 중인 유기 농자재의 실태를 조사하고 또한 유기농자재의 성분을 분석하였다. 유기농 자재 시용효과를 검증하기 위해서 시범마을 현지 농가포장에서 삼광벼 품종으로 포트 육묘 후 기계이앙을 실시하고 유기농자재를 시용하여 조사하였다. 유기농업 종합기술 시범마을에서 활용중인 유기농자재는 27.5%가 유기농 목록에 공시된 자재를 사용하고 있었다. 시범마을에서 활용중인 유기농자재의 성분 중 수분은 평균 50.1%, 유기물함량은 36.0%, 총 질소함량은 1.71% 이며 염분은 0.56% 이였다. 경종방법과 유기농자재를 이용하여 새로운 유기농 벼 재배 기술을 개발할 목적으로 시험포를 조성한 지역에서 토양의 화학성분을 조사한 결과 pH, 마그네슘, 규산은 망성 단지에서 낮은 경향을 보였고, 유기물, 인산, 규산, 칼륨은 삼기단지에서 높았다. 포트육묘 후 이앙을 실시한 결과 단위면적당 이앙주수별 생육에서 재식밀도가 높을 수록 초장과 경수가 적었다. 산파육묘와 포트육묘 이앙 모두 도복 발생은 없었으며, 포트 육묘 3절의 두께가 양호하였다. 쌀겨 시용에서 잡초발 생량이 적었으며, 포트육묘 이앙에서 병해충이 적었다. 수량은 산파육묘에 비해 포트육묘 방식에서 5~10% 높았는데, 포트 육묘에서 주당 수수, 수당 립수가 많았고, 천립중이 무거웠다.
