Preventive and Therapeutic Effects of Quercetin on Experimental Radiation Induced Lung Injury in Mice

  • Objective: To investigate the protective effect of quercetin on radiation induced lung injury (RILI) and related mechanisms. Materials and Methods: Mice treated with radiation and/or quercetin were sacrificed at 1-8 weeks after irradiation under anesthesia. Lung tissues were collected for histological examination. Immunohistochemistry (IHC) and Western blotting were performed to detect the protein expression of nuclear factor-${\kappa}B$ ($NF-{\kappa}B$) and Mitogen-activated protein kinases (MAPK) pathway. Results: Hematoxylin and eosin (HE) staining showed that radiation controls displayed more severe lung damage than quercetin groups, either high or low dose. Results of IHC and Western blotting demonstrated the expression level of $NF-{\kappa}B$ to be decreased and that of an inhibitor of $NF-{\kappa}B$ ($I{\kappa}b-{\alpha}$) to be increased by the quercetin intervention compared with the radiation control group. Numbers of JNK/SAPK, p38 and p44/p42 positive inflammatory cells were decreased in the radiation+quercetin injection group (P<0.05). Conclusions: Quercetin may play a radio-protective role in mice lung via suppression of $NF-{\kappa}B$ and MAPK pathways.

An innovative BRB with viscoelastic layers: performance evaluation and numerical simulation

  • Energy induced by minor earthquake and micro vibration cannot be dissipated by traditional buckling-restrained braces (BRBs). To solve this problem, a new type of hybrid passive control device, named as VE-BRB, which is configured by a BRB with high-damping viscoelastic (VE) layers, is developed and studied. Theoretical analysis, performance tests, numerical simulation and case analysis are conducted to study the seismic behavior of VE-BRBs. The results indicate that the combination of hysteretic and damping devices lead to a multi-phased nature and good performance. VE-BRB's working state can be divided into three phases: before yielding of the steel core, VE layers provide sufficient damping ratio to mitigate minor vibrations; after yielding of the steel core, the steel's hysteretic deformations provide supplemental dissipative capacity for structures; after rupture of the steel core, VE layers are still able to work normally and provide multiple security assurance for structures. The simulation results agreed well with the experimental results, validating the finite element analysis method, constitutive models and the identified parameters. The comparison of the time history analysis on a 6-story frame with VE-BRBs and BRBs verified the advantages of VE-BRB for seismic protection of structures compared with traditional BRB. In general, VE-BRB had the potential to provide better control effect on structural displacement and shear in all stages than BRB as expected.

Molecular Cloning of Hemoglobin Alpha-chain Gene from Pantholops hodgsonii, a Hypoxic Tolerance Species

  • To investigate the possible mechanisms of high-altitude native animals in adapting to high altitude, we cloned hemoglobin alpha-chain (alpha-chain Hb) gene from Pantholops hodgsonii, an animal species that indigenously lives at elevations of 3700-5500 m on the Qinghai-Tibetan plateau. Using reverse transcription polymerase chain reaction (RT-PCR) technique, the alpha-chain Hb gene was amplified from total RNA in the liver of the Pantholops hodgsonii. TA cloning technique was used and the PCR product was cloned into pGEM-T vector. The DNA sequence of the gene was highly homologous with sheep (99.1%), goat (98.6%), cattle (95.6%) and human (86.5%). The alpha-chain Hb gene encoded a 142-amino acid protein that could be identified with the homology of alpha-chain Hb protein in sheep (98%), goat (96%), cattle (91%) and human (87%). However, 18 alternations were detected when compared with the alpha-chain Hb gene in human, and 2 in sheep. Moreover, the alterations of a117 GluAsp and $\alpha$132 AsnSer in important regions were noted in human and sheep, respectively. Phylogenetic analysis suggested that the structure of alpha-chain Hb was highly similar to that in sheep. This study provided essential information for elucidating the possible roles of hemoglobin in adapting to extremely high altitude in Pantholops hodgsonii.

An Alternative for the Enrichment of Information Literacy Instruction as a General Education (교양교육으로서 정보활용능력 교육 위상 강화 방안)

  • The major aim of this study is to discuss the possibility whether the merging of Information Literacy and rhetoric will be able to have essential elements for being a general education subject in universities. The characteristic of rhetorical information literacy (RIL) is organized as follows. Situated literacy using heuristic method, process oriented literacy attaching importance to research, and literacy relevant to ubiquitous environment. So, it is argued that rhetorical information literate citizenry is essential for a modern democratic society. As the information literacy instruction program can be various, for example, technology oriented instruction is needed to be compared. For the practical execution of the RILI program, 'asking situated questions to discover problem', 'selecting and writing review of meta study books', and 'writing visiting report of library', and 'instruction of information ethics for democratic citizens' have been proposed.
